Eighteen young steers were inoculated with Toxoplasma gondii and randomly distributed into three groups of six animals each: GI, 2.5x10(5) "P" strain oocysts, GII, 5.0x10(6) "RH" strain tachyzoites, and GIII (Control). Clinical, serological and parasitemia exams were realized. Parasite investigation by bioassay and PCR was realized on semen and fragments of skeletal musculature, lymph nodes, brain, retina, spleen, liver, lung, testicle, epididymis and seminal vesicle. Blood and semen samples were collected on days -2, -1, 1, 3, 5, 7, 14 and weekly thereafter, up to postinfection day (PID) 84. The inoculated steers (GI and GII) presented hyperthermia from PID 3 to 16. Antibodies against T. gondii were detected through the indirect fluorescence antibody test (IFAT) on PID 5 (1:16) in both inoculated groups (oocysts and tachyzoites), reaching peaks of 1:4096 on PID 7. Parasitemia outbursts occurred in all infected bovines, principally from PID 7 to 28, independent of the strain and inoculate used. Bioassays revealed the presence of parasites in semen samples of animals infected with oocysts (GI) and tachyzoites (GII) on several experimental days between PID 7 and 84. Tissue parasitism by T. gondii was diagnosed by bioassay and the PCR technique in several organ and tissue fragments. These findings suggest the possibility of sexual transmission of T. gondii in the bovine species.

RESUMO: Este estudo descreveu as características seminais, da membrana plasmática e do acrossoma de espermatozoide congelado/descongelado de 19 ejaculados de garanhões da raça Nordestina. Os aspectos analisados incluíram os parâmetros físicos do sêmen fresco; a motilidade e a longevidade do sêmen diluído e descongelado; a morfologia espermática, integridade funcional e estrutural da membrana plasmática do espermatozoide e a habilidade de ligação do espermatozoide à membrana perivitelina da gema do ovo de galinha do sêmen descongelado. As variáveis foram avaliadas pela ANOVA com post hoc teste de Student Newman-Keuls (P<0,05). A MT e a MP foram maiores (P<0,05) no sêmen diluído do que no descongelado. A percentagem média de defeitos maiores, menores e totais foi muito inferior ao limite recomendado pelo CBRA. A porcentagem de reativos ao HOST foi de 14,21±1,12% e a porcentagem média de membranas íntegras detectadas pelo teste supravital de 62,22±9,06% e pela sonda SYBR-14 de 81,47±26,90. O número médio de espermatozoides ligados à MPV após a descongelação do sêmen foi de 230,39±57,09. A MT e MP no tempo 0 min do TTR foi superior (P<0,05) em relação a 150 min, não diferindo nos tempos 10 min e 30 min. Os resultados demonstram que a utilização dos testes laboratoriais adicionais ajudam no processo de avaliação das amostras, possibilitando a obtenção de informações mais confiáveis e precisas. Embora a criopreservação tenha provocado queda na motilidade seminal, o uso de diluidor contendo amidas minimizou os danos osmóticos nas células espermáticas e manteve a integridade morfológica, funcional e estrutural da membrana plasmática do espermatozoide. Estes resultados são um referencial em estudos futuros uma vez que, inexistem dados comparativos nesta raça.

The structure of the fruit and seed in development of Chorisia speciosa are described with the main purpose of clarifying the origin and nature of the hairs that cover the seeds and aiding future taxonomical and ecological studies of the group. The fruit is an ellipsoid loculicide capsule and presents the exocarp formed by 7-10 cells layers, with very thick walls and evident simple pits. A great number of mucilage secretory cavities and ramified vascular bundles, accompanied by fibers, occur in the parenchymatic mesocarp. The endocarp derives from the ventral epidermis of the ovary wall, whose cells undergo a gradual elongation, become lignified, and constitute the trichomes which cover the mature seeds. The fruit aperture occurs by means of a suture evident in the ovarian wall in the middle region of the carpel leaf. Anatropous and bitegmic ovules, provided by a hypostase, give rise to campilotropous and bitegmic seeds. The testa is uniseriate, the exotegmen is completely formed by macrosclereids, and mucilage secretory cavities occur in the mesotegmen. The endotegmen, which is differentiated in the endothelium, is crushed in the mature seed. The plicate embryo, which occupies practically the entire seminal cavity, is found between endosperm layers, both being rich in lipids.

A morfologia e anatomia de muitas espécies do cerrado são desconhecidas, sendo os frutos e sementes ainda menos estudados. Assim, esse trabalho objetivou estudar morfoanatomicamente os frutos e sementes de Byrsonima intermedia, uma das espécies mais freqüentes dos cerrados de São Paulo, descrevendo-os ontogeneticamente. Verificou-se que o ovário é súpero, tricarpelar e trilocular, com um óvulo por lóculo; a epiderme externa é unisseriada, com cutícula espessa no ápice e delgada na base do órgão; o mesofilo ovariano é multisseriado, parenquimático e vascularizado; a epiderme interna é unisseriada, com células que se dividem precocemente. Os óvulos são subcampilótropos, bitegumentados, com nucelo projetado através da micrópila; hipóstase e epístase são observadas. Durante o desenvolvimento do pericarpo, as divisões celulares restringem-se à fase inicial do desenvolvimento, ocorrendo previamente à diferenciação seminal. O fruto maduro é do tipo drupóide, carnoso, com pirênio lenhoso formando três lóculos. O exocarpo é unisseriado; o mesocarpo externo é parenquimático, ocorrendo esclereídes dispersas na região apical do fruto; no mesocarpo interno ocorrem várias camadas de esclereídes alongadas em diversos sentidos; o endocarpo é multisseriado, com as esclereídes alongadas longitudinalmente. A origem meso-endocárpica dos tecidos lignificados deste pericarpo contraria a definição clássica dos frutos drupóides, que destaca apenas o endocarpo lenhoso. A semente apresenta tegumentos e endosperma reduzidos. O embrião tem eixo hipocótilo-radicular contínuo, com dois cotilédones enrolados em espiral.

Estudou-se o comportamento germinativo de quatro espécies de Bromeliaceae [Alcantarea imperialis (Carrière) Harms, Pitcairnia flammea Lindl., Vriesea heterostachys (Baker) L. B. Sm. e Vriesea penduliflora L. B. Sm.], oriundas do Parque Estadual do Ibitipoca - MG, com preferências contrastantes de habitat e representando duas formas de vida diferentes. Os objetivos do presente estudo foram: a) caracterizar as sementes quanto ao tipo de germinação e descrever a morfologia do desenvolvimento pós-seminal; b) avaliar o efeito das temperaturas constantes (10 °C a 40 °C) e alternada (20-30 °C); c) da razão vermelho:vermelho-extremo (V:VE) e d) dos diferentes potenciais de água na porcentagem e na velocidade de germinação das sementes. Os resultados demonstraram que todas as espécies produzem sementes pequenas e muito leves, variando de 0,2 a 1,7 mg; a germinação é epígea com plântulas criptocotiledonares e apresentaram valores ótimos de temperatura entre 20 °C e 30 °C. Vriesea heterostachys e V. penduliflora foram as únicas que germinaram com sucesso no escuro, mas as sementes das quatro espécies requerem luz para a máxima germinação. Os valores de V:VE que resultaram em 50% da máxima germinação variaram entre as espécies. As condições de estresse hídrico reduziram a porcentagem e a velocidade de germinação de forma distinta entre as espécies. A. imperialis e P. flammea foram menos sensíveis aos baixos potenciais hídricos. Os requerimentos germinativos contrastantes entre as espécies estudadas estão relacionados à preferência de habitat de cada uma delas.

Six brachytic maize varieties were crossed in a diallel mating scheme. Both varieties and crosses were grown hydroponically in a greenhouse, in randomized complete blocks with three replications in two seasons. Four brachytic double cross hybrids were used as checks. Twenty-eight days after planting, data for eight traits were taken for weights of the total plant (TPW), top plant (TOW), total roots (TRW), seminal roots (SRW), and nodal roots (NRW) and number of total roots (TRN), seminal roots (SRN), and nodal roots (NRN). Ten plants were measured in each plot and all the analyses were accomplished with plot means. In the diallel cross the top plant contributed 57.6% of the total plant weight, for seminal roots 15.4%, and for nodal roots 27.0%. Root number distribution was 36.7% seminal roots and 63.3% nodal roots. Approximately the same ratios were observed in the checks. The average heterosis effects were nonsignificant for all traits; the other components of heterosis (variety and specific heterosis) also were not important sources of variation in young plants. The overall results suggest that nonadditive gene action is not an important source of variation for the plant and root system of young plants. The positive correlation coefficients for combinations of traits indicated that they are under the control of a polygenic system

The objective of this study was to identify restriction fragment length polymorphism (RFLP) markers linked to QTLs that control aluminum (Al) tolerance in maize. The strategy used was bulked segregant analysis (BSA) and the genetic material utilized was an F2 population derived from a cross between the Al-susceptible inbred line L53 and Al-tolerant inbred line L1327. Both lines were developed at the National Maize and Sorghum Research Center - CNPMS/EMBRAPA. The F2 population of 1554 individuals was evaluated in a nutrient solution containing a toxic concentration of Al and relative seminal root length (RSRL) was used as a phenotypic measure of tolerance. The RSRL frequency distribution was continuous, but skewed towards Al-susceptible individuals. Seedlings of the F2 population which scored the highest and the lowest RSRL values were transplanted to the field and subsequently selfed to obtain F3 families. Thirty F3 families (15 Al-susceptible and 15 Al-tolerant) were evaluated in nutrient solution, using an incomplete block design, to identify those with the smallest variances for aluminum tolerance and susceptibility. Six Al-susceptible and five Al-tolerant F3 families were chosen to construct one pool of Al-susceptible individuals, and another of Al-tolerant, herein referred as "bulks", based on average values of RSRL and genetic variance. One hundred and thirteen probes were selected, with an average interval of 30 cM, covering the 10 maize chromosomes. These were tested for their ability to discriminate the parental lines. Fifty-four of these probes were polymorphic, with 46 showing codominance. These probes were hybridized with DNA from the two contrasting bulks. Three RFLPs on chromosome 8 distinguished the bulks on the basis of band intensity. DNA of individuals from the bulks was hybridized with these probes and showed the presence of heterozygous individuals in each bulk. These results suggest that in maize there is a region related to aluminum tolerance on chromosome 8

We investigated whether chronic stress applied from prepuberty to full sexual maturity interferes with spermatogenic and androgenic testicular functions. Male Wistar rats (40 days old) were immobilized 6 h a day for 60 days. Following immobilization, plasma concentrations of corticosterone and prolactin increased 135% and 48%, respectively, while plasma luteinizing hormone and testosterone presented a significant decrease of 29% and 37%, respectively. Plasma concentration of follicle-stimulating hormone was not altered in stressed rats. Chronic stress reduced the amount of mature spermatids in the testis by 16% and the spermatozoon concentration in the cauda epididymidis by 32%. A 17% reduction in weight and a 42% decrease in DNA content were observed in the seminal vesicle of immobilized rats but not in its fructose content. The growth and secretory activity of the ventral prostate were not altered by chronic stress.

The surgical specimens from 51 men submitted to radical prostatectomy for localized prostate cancer were examined by immunohistochemistry using proliferation cell nuclear antigen (PCNA) monoclonal antibody to evaluate the proliferative index (PI). The relationship between PI, biological variables and p53 protein expression was evaluated by immunohistochemistry. PI was low in invasive localized prostate carcinoma (mean, 12.4%) and the incidence of PCNA-positive cells was significantly higher in tumors with p53 expression (P = 0.0226). There was no statistical difference in PCNA values when biological parameters such as Gleason score, tumor volume, extraprostatic involvement, seminal vesicle infiltration or lymph node metastasis were considered. We conclude that proliferative activity is usually low in prostate carcinoma but is correlated with p53 immune staining, indicating that p53 is important in cell cycle control in this neoplasm.

We investigated whether stress interferes with fertility during adulthood. Male Wistar rats (weighing 220 g in the beginning of the experiment) were forced to swim for 3 min in water at 32ºC daily for 15 days. Stress was assessed by the hot-plate test after the last stressing session. To assess fertility, control and stressed males (N = 15 per group) were mated with sexually mature normal females. Males were sacrificed after copulation. Stress caused by forced swimming was demonstrated by a significant increase in the latency of the pain response in the hot-plate test (14.6 ± 1.25 s for control males vs 26.0 ± 1.53 s for stressed males, P = 0.0004). No changes were observed in body weight, testicular weight, seminal vesicle weight, ventral prostate weight or gross histological features of the testes of stressed males. Similarly, no changes were observed in fertility rate, measured by counting live fetuses in the uterus of normal females mated with control and stressed males; no dead or incompletely developed fetuses were observed in the uterus of either group. In contrast, there was a statistically significant decrease in spermatid production demonstrated by histometric evaluation (154.96 ± 5.41 vs 127.02 ± 3.95 spermatids per tubular section for control and stressed rats, respectively, P = 0.001). These data demonstrate that 15 days of forced swimming stress applied to adult male rats did not impair fertility, but significantly decreased spermatid production. This suggests that the effect of stress on fertility should not be assessed before at least the time required for one cycle of spermatogenesis.

Coronary artery disease (CAD) is a worldwide leading cause of death. The standard method for evaluating critical partial occlusions is coronary arteriography, a catheterization technique which is invasive, time consuming, and costly. There are noninvasive approaches for the early detection of CAD. The basis for the noninvasive diagnosis of CAD has been laid in a sequential analysis of the risk factors, and the results of the treadmill test and myocardial perfusion scintigraphy (MPS). Many investigators have demonstrated that the diagnostic applications of MPS are appropriate for patients who have an intermediate likelihood of disease. Although this information is useful, it is only partially utilized in clinical practice due to the difficulty to properly classify the patients. Since the seminal work of Lotfi Zadeh, fuzzy logic has been applied in numerous areas. In the present study, we proposed and tested a model to select patients for MPS based on fuzzy sets theory. A group of 1053 patients was used to develop the model and another group of 1045 patients was used to test it. Receiver operating characteristic curves were used to compare the performance of the fuzzy model against expert physician opinions, and showed that the performance of the fuzzy model was equal or superior to that of the physicians. Therefore, we conclude that the fuzzy model could be a useful tool to assist the general practitioner in the selection of patients for MPS.

It has been reported that mental stress causes abnormality of spermiogram parameters. We investigated the effect of psychological stress on the L-arginine-nitric oxide (NO) pathway. Semen samples were collected from 29 healthy fourth semester medical students just before (stress) and 3 months after (non-stress) the final examinations. Psychological stress was measured by the State Anxiety Inventory questionnaire. After standard semen analysis, arginase activity and NO concentration were measured spectrophotometrically in the seminal plasma. Measurements were made in duplicate. During the stress period, sperm concentration (41.28 ± 3.70 vs 77.62 ± 7.13 x 10(6)/mL), rapid progressive motility of spermatozoa (8.79 ± 1.66 vs 20.86 ± 1.63%) and seminal plasma arginase activity (0.12 ± 0.01 vs 0.22 ± 0.01 U/mL) were significantly lower than in the non-stress situation, whereas seminal plasma NO (17.28 ± 0.56 vs 10.02 ± 0.49 µmol/L) was higher compared to the non-stress period (P < 0.001 for all). During stress there was a negative correlation between NO concentration and sperm concentration, the percentage of rapid progressive motility and arginase activity (r = -0.622, P < 0.01; r = -0.425, P < 0.05 and r = -0.445, P < 0.05, respectively). These results indicate that psychological stress causes an increase of NO level and a decrease of arginase activity in the L-arginine-NO pathway. Furthermore, poor sperm quality may be due to excessive production of NO under psychological stress. In the light of these results, we suggest that the arginine-NO pathway, together with arginase and NO synthase, are involved in semen quality under stress conditions.

To determine if Butea superba Roxb., a traditional Thai male potency herb, has androgenic activity in 60-day-old male Wistar rats, we measured its effects on the pituitary-testicular axis and sex organs. Intact and orchidectomized adult male rats were subdivided into five groups (10 rats/group): distilled water, Butea superba (BS)-10, BS-50, BS-250, and testosterone propionate (TP). They received 0, 10, 50, and 250 mg·kg body weight-1·day-1 BS in distilled water by gavage and 6 mg·kg body weight-1·day-1 TP sc, respectively, during the 30-day treatment period. Blood was collected every 15 days and luteinizing hormone (LH), follicle-stimulating hormone (FSH) and testosterone were measured. Changes of weight and histological appearance of sex organs were determined at the end of the 30-day treatment and 15-day post-treatment periods. TP treatment reduced serum FSH and LH levels and significantly increased the weight of the seminal vesicles and epididymis, in accordance with histopathological changes, in both intact and orchidectomized rats. No changes in serum testosterone, LH, and FSH levels were observed in any of the intact rats treated with BS, but a significant increase in seminal vesicle weight was observed only in the BS-250 group. Although a significant reduction in serum LH was detected in the BS-50 and BS-250 groups of orchidectomized rats, no significant change in weight or histology of sex organs was observed. Thus, we conclude that B. superba needs endogenous testosterone to work synergistically to stimulate the accessory sex organ of intact animals and can potentially exhibit an LH reduction effect in orchidectomized animals.

O trabalho foi desenvolvido em casa de vegetação no Campus do Centro de Ciências Agrárias da Universidade Federal do Espírito Santo, Alegre-ES, com o objetivo avaliar a influência do substrato e da lâmina de água na germinação de sementes e no desenvolvimento pós-seminal de plantas de pimenta-malagueta. As sementes foram distribuídas em vasos contendo os substratos Latossolo Vermelho puro (LP), Latossolo Vermelho + cama de galinheiro (A) e Latossolo Vermelho + esterco bovino (B), aos quais foram aplicadas lâminas de água equivalentes a 25, 50, 75, 100 e 125% da evapotranspiração da cultura. O delineamento experimental foi em blocos casualizados, com quatro repetições, em esquema fatorial 3x5. Foram avaliadas as seguintes características: germinação, índice de velocidade de emergência (IVE), massa fresca e seca, altura de planta, diâmetro de coleto e volume de raiz. Os substratos A e B apresentaram melhores resultados em relação ao LP para todas as características avaliadas, sendo que para todas as características houve aumento nos valores em resposta ao aumento no fornecimento de lâminas de água.

O objetivo deste trabalho foi avaliar a presença da dormência tegumentar, identificando tratamentos pré-germinativos que possam acelerar e uniformizar a germinação, bem como conhecer a morfologia do desenvolvimento pós seminal em sementes de Operculina macrocarpa. As sementes foram submetidas aos seguintes tratamentos pré-germinativos: imersão em água por 1 hora; imersão em solução de hipoclorito por 1 hora e testemunha, representada por sementes sem tratamento prévio. Os testes de germinação e de emergência foram conduzidos com quatro repetições de 25 e de 20 sementes, em laboratório e em casa-de-vegetação, respectivamente. As avaliações foram realizadas diariamente, durante 30 dias, onde foi calculada a porcentagem de plântulas normais aos 15 e 30 dias e o índice de velocidade de germinação/emergência. A germinação de Operculina macrocarpa é epígea-fanerocotiledonar e a semente apesar de apresentar um tegumento duro não se mostra impermeável à entrada de água, dispensando a adoção de tratamentos pré-germinativos.
