Asthma is an inflammatory condition characterized by the involvement of several mediators, including reactive oxygen species. The aim of the present study was to investigate the superoxide release and cellular glutathione peroxidase (cGPx) activity in peripheral blood granulocytes and monocytes from children and adolescents with atopic asthma. Forty-four patients were selected and classified as having intermittent or persistent asthma (mild, moderate or severe). The spontaneous or phorbol myristate acetate (PMA, 30 nM)-induced superoxide release by granulocytes and monocytes was determined at 0, 5, 15, and 25 min. cGPx activity was assayed spectrophotometrically. The spontaneous superoxide release by granulocytes from patients with mild (N = 15), moderate (N = 12) or severe (N = 6) asthma was higher at 25 min compared to healthy individuals (N = 28, P < 0.05, Duncan test). The PMA-induced superoxide release by granulocytes from patients with moderate (N = 12) or severe (N = 6) asthma was higher at 15 and 25 min compared to healthy individuals (N = 28, P < 0.05 in both times of incubation, Duncan test). The spontaneous or PMA-induced superoxide release by monocytes from asthmatic patients was similar to healthy individuals (P > 0.05 in all times of incubation, Duncan test). cGPx activity of granulocytes and monocytes from patients with persistent asthma (N = 20) was also similar to healthy individuals (N = 10, P > 0.05, Kruskal-Wallis test). We conclude that, under specific circumstances, granulocytes from children with persistent asthma present a higher respiratory burst activity compared to healthy individuals. These findings indicate a risk of oxidative stress, phagocyte auto-oxidation, and the subsequent release of intracellular toxic oxidants and enzymes, leading to additional inflammation and lung damage in asthmatic children.

The causes of luteal phase progesterone deficiency in polycystic ovary syndrome (PCOS) are not known. To determine the possible involvement of hyperinsulinemia in luteal phase progesterone deficiency in women with PCOS, we examined the relationship between progesterone, luteinizing hormone (LH) and insulin during the luteal phase and studied the effect of metformin on luteal progesterone levels in PCOS. Patients with PCOS (19 women aged 18-35 years) were treated with metformin (500 mg three times daily) for 4 weeks prior to the test cycle and throughout the study period, and submitted to ovulation induction with clomiphene citrate. Blood samples were collected from control (N = 5, same age range as PCOS women) and PCOS women during the late follicular (one sample) and luteal (3 samples) phases and LH, insulin and progesterone concentrations were determined. Results were analyzed by one-way analysis of variance (ANOVA), Duncan's test and Karl Pearson's coefficient of correlation (r). The endocrine study showed low progesterone level (4.9 ng/ml) during luteal phase in the PCOS women as compared with control (21.6 ng/ml). A significant negative correlation was observed between insulin and progesterone (r = -0.60; P < 0.01) and between progesterone and LH (r = -0.56; P < 0.05) concentrations, and a positive correlation (r = 0.83; P < 0.001) was observed between LH and insulin. The study further demonstrated a significant enhancement in luteal progesterone concentration (16.97 ng/ml) in PCOS women treated with metformin. The results suggest that hyperinsulinemia/insulin resistance may be responsible for low progesterone levels during the luteal phase in PCOS. The luteal progesterone level may be enhanced in PCOS by decreasing insulin secretion with metformin.

To efficiently examine the association of glutamic acid decarboxylase antibody (GADA) positivity with the onset and progression of diabetes in middle-aged adults, we performed a case-cohort study representing the ~9-year experience of 10,275 Atherosclerosis Risk in Communities Study participants, initially aged 45-64 years. Antibodies to glutamic acid decarboxylase (GAD65) were measured by radioimmunoassay in 580 incident diabetes cases and 544 non-cases. The overall weighted prevalence of GADA positivity (³1 U/mL) was 7.3%. Baseline risk factors, with the exception of smoking and interleukin-6 (P £ 0.02), were generally similar between GADA-positive and -negative individuals. GADA positivity did not predict incident diabetes in multiply adjusted (HR = 1.04; 95%CI = 0.55, 1.96) proportional hazard analyses. However, a small non-significant adjusted risk (HR = 1.29; 95%CI = 0.58, 2.88) was seen for those in the highest tertile (³2.38 U/mL) of positivity. GADA-positive and GADA-negative non-diabetic individuals had similar risk profiles for diabetes, with central obesity and elevated inflammation markers, aside from glucose, being the main predictors. Among diabetes cases at study's end, progression to insulin treatment increased monotonically as a function of baseline GADA level. Overall, being GADA positive increased risk of progression to insulin use almost 10 times (HR = 9.9; 95%CI = 3.4, 28.5). In conclusion, in initially non-diabetic middle-aged adults, GADA positivity did not increase diabetes risk, and the overall baseline profile of risk factors was similar for positive and negative individuals. Among middle-aged adults, with the possible exception of those with the highest GADA levels, autoimmune pathophysiology reflected by GADA may become clinically relevant only after diabetes onset.

Subjects with chronic obstructive pulmonary disease (COPD) present breathing pattern and thoracoabdominal motion abnormalities that may contribute to exercise limitation. Twenty-two men with stable COPD (FEV1 = 42.6 ± 13.5% predicted; age 68 ± 8 years; mean ± SD) on usual medication and with at least 5 years of diagnosis were evaluated at rest and during an incremental cycle exercise test (10 watts/2 min). Changes in respiratory frequency, tidal volume, rib cage and abdominal motion contribution to tidal volume and the phase angle that measures the asynchrony were analyzed by inductive respiratory plethysmography at rest and during three levels of exercise (30-50, 70-80, and 100% maximal work load). Repeated measures ANOVA followed by pre-planned contrasts and Bonferroni corrections were used for analyses. As expected, the greater the exercise intensity the higher the tidal volume and respiratory frequency. Abdominal motion contributed to the tidal volume increase (rest: 49.82 ± 11.19% vs exercise: 64.15 ± 9.7%, 63.41 ± 10%, and 65.56 ± 10.2%, respectively, P < 0.001) as well as the asynchrony [phase angle: 11.95 ± 7.24° at rest vs 22.2 ± 15° (P = 0.002), 22.6 ± 9° (P < 0.001), and 22.7 ± 8° (P < 0.001), respectively, at the three levels of exercise]. In conclusion, the increase in ventilation during exercise in COPD patients was associated with the major motion of the abdominal compartment and with an increase in the asynchrony independent of exercise intensity. It suggests that cycling exercise is an effective way of enhancing ventilation in COPD patients.

The objective of the present study was to compare the effect of acute exercise performed at different intensities in relation to the anaerobic threshold (AT) on abilities requiring control of executive functions or alertness in physically active elderly females. Forty-eight physically active elderly females (63.8 ± 4.6 years old) were assigned to one of four groups by drawing lots: control group without exercise or trial groups with exercise performed at 60, 90, or 110% of AT (watts) and submitted to 5 cognitive tests before and after exercise. Following cognitive pretesting, an incremental cycle ergometer test was conducted to determine AT using a fixed blood lactate concentration of 3.5 mmol/L as cutoff. Acute exercise executed at 90% of AT resulted in significant (P < 0.05, ANOVA) improvement in the performance of executive functions when compared to control in 3 of 5 tests (verbal fluency, Tower of Hanoi test (number of movements), and Trail Making test B). Exercising at 60% of AT did not improve results of any tests for executive functions, whereas exercise executed at 110% of AT only improved the performance in one of these tests (verbal fluency) compared to control. Women from all trial groups exhibited a remarkable reduction in the Simple Response Time (alertness) test (P = 0.001). Thus, physical exercise performed close to AT is more effective to improve cognitive processing of older women even if conducted acutely, and using a customized exercise prescription based on the anaerobic threshold should optimize the beneficial effects.

The dorsal raphe nucleus (DRN) is the origin of ascending serotonergic projections and is considered to be an important component of the brain circuit that mediates anxiety- and depression-related behaviors. A large fraction of DRN serotonin-positive neurons contain nitric oxide (NO). Disruption of NO-mediated neurotransmission in the DRN by NO synthase inhibitors produces anxiolytic- and antidepressant-like effects in rats and also induces nonspecific interference with locomotor activity. We investigated the involvement of the 5-HT1A autoreceptor in the locomotor effects induced by NO in the DRN of male Wistar rats (280-310 g, N = 9-10 per group). The NO donor 3-morpholinosylnomine hydrochloride (SIN-1, 150, and 300 nmol) and the NO scavenger S-3-carboxy-4-hydroxyphenylglycine (carboxy-PTIO, 0.1-3.0 nmol) were injected into the DRN of rats immediately before they were exposed to the open field for 10 min. To evaluate the involvement of the 5-HT1A receptor and the N-methyl-D-aspartate (NMDA) glutamate receptor in the locomotor effects of NO, animals were pretreated with the 5-HT1A receptor agonist 8-hydroxy-2-(di-n-propylamino)tetralin (8-OH-DPAT, 8 nmol), the 5-HT1A receptor antagonist N-(2-[4-(2-methoxyphenyl)-1-piperazinyl]ethyl)-N-2-pyridinyl-cyclohexanecarboxamide maleate (WAY-100635, 0.37 nmol), and the NMDA receptor antagonist DL-2-amino-7-phosphonoheptanoic acid (AP7, 1 nmol), followed by microinjection of SIN-1 into the DRN. SIN-1 increased the distance traveled (mean ± SEM) in the open-field test (4431 ± 306.1 cm; F7,63 = 2.44, P = 0.028) and this effect was blocked by previous 8-OH-DPAT (2885 ± 490.4 cm) or AP7 (3335 ± 283.5 cm) administration (P < 0.05, Duncan test). These results indicate that 5-HT1A receptor activation and/or facilitation of glutamate neurotransmission can modulate the locomotor effects induced by NO in the DRN.

The present study investigated the effect of thioperamide (THIO), an H3 histaminergic receptor antagonist, microinjected into the cerebellar vermis on emotional memory consolidation in male Swiss albino mice re-exposed to the elevated plus-maze (EPM). We implanted a guide cannula into the cerebellar vermis using stereotactic surgery. On the third day after surgery, we performed behavioral tests for two consecutive days. On the first day (exposure), the mice (n=10/group) were exposed to the EPM and received THIO (0.06, 0.3, or 1.5 ng/0.1 µL) immediately after the end of the session. Twenty-four hours later, the mice were re-exposed to the EPM under the same experimental conditions, but without drug injection. A reduction in the exploration of the open arms upon re-exposure to the EPM (percentage of number of entries and time spent in open arms) compared with the initial exposure was used as an indicator of learning and memory. One-way analysis of variance (ANOVA) followed by the Duncan post hoc test was used to analyze the data. Upon re-exposure, exploratory activity in the open arms was reduced in the control group, and with the two highest THIO doses: 0.3 and 1.5 ng/0.1 µL. No reduction was seen with the lowest THIO dose (0.06 ng/0.1 µL), indicating inhibition of the consolidation of emotional memory. None of the doses interfered with the animals' locomotor activity. We conclude that THIO at the lowest dose (0.06 ng/0.1 µL) microinjected into the cerebellum impaired emotional memory consolidation in mice.

This study investigated the effects of histamine H1 or H2 receptor antagonists on emotional memory consolidation in mice submitted to the elevated plus maze (EPM). The cerebellar vermis of male mice (Swiss albino) was implanted using a cannula guide. Three days after recovery, behavioral tests were performed in the EPM on 2 consecutive days (T1 and T2). Immediately after exposure to the EPM (T1), animals received a microinjection of saline (SAL) or the H1 antagonist chlorpheniramine (CPA; 0.016, 0.052, or 0.16 nmol/0.1 µL) in Experiment 1, and SAL or the H2 antagonist ranitidine (RA; 0.57, 2.85, or 5.7 nmol/0.1 µL) in Experiment 2. Twenty-four hours later, mice were reexposed to the EPM (T2) under the same experimental conditions but they did not receive any injection. Data were analyzed using one-way ANOVA and the Duncan test. In Experiment 1, mice microinjected with SAL and with CPA entered the open arms less often (%OAE) and spent less time in the open arms (%OAT) in T2, and there was no difference among groups. The results of Experiment 2 demonstrated that the values of %OAE and %OAT in T2 were lower compared to T1 for the groups that were microinjected with SAL and 2.85 nmol/0.1 µL RA. However, when animals were microinjected with 5.7 nmol/0.1 µL RA, they did not show a reduction in %OAE and %OAT. These results demonstrate that CPA did not affect behavior at the doses used in this study, while 5.7 nmol/0.1 µL RA induced impairment of memory consolidation in the EPM.

Com o objetivo de fornecer subsídios para o controle de microrganismos em abatedouro de aves, avaliou-se a ação dos agentes químicos sanificantes comprovadamente mais eficientes (hipoclorito de sódio, ácido peracético, e dicloroisocianurato de sódio) em suspensões de esporos de bactérias aeróbias mesófilas e termófilas, isoladas de equipamentos de abatedouro de aves. Pelo teste de Duncan, constatou-se que dentre os esporos isolados o mais resistente obteve 1,5 e 1,2 RD quando em contato com o hipoclorito de sódio e ao ácido peracético com 30 minutos de tempo de contato, que foram os sanificantes mais eficientes (P > 0,05). O dicloroisocianurato de sódio proporcionou 0,1 RD em 30 minutos de tempo de contato com a mesma suspensão de esporos. Os esporos isolados apresentaram diferentes resistências aos sanificantes avaliados, indicando a necessidade de uma seleção criteriosa de agentes químicos para o procedimento de sanificação, sendo importante um rodízio entre os sanificantes mais eficientes testados, que foram o hipoclorito de sódio e o ácido peracético.

Avaliou-se a produção de aflatoxina B1 pelo Aspergillus flavus IMI 190443, inoculado em sementes de genótipos de amendoim: Tatu Vermelho, VRR-245, 2117 e 2155, in natura e autoclavado a 1210 C por 20 minutos, previamente cultivados no Centro Experimental do Instituto Agronômico de Campinas, em 1995/96. A aflatoxina B1 foi quantificada por cromatografia em camada delgada, utilizando comparação visual com padrões. Os níveis de aflatoxina B1 das amostras autoclavadas e in natura dos genótipos Tatu Vermelho, VRR-245 e 2155 foram significativamente diferentes (p <0,05, Teste de Duncan) quando comparados aos níveis do genótipo 2117. O genótipo 2117, originário da Índia, apresentou independente do processo com e sem aquecimento os menores níveis de aflatoxina B1 em relação aos dos outros genótipos. Os resultados obtidos mostram perspectivas de exploração da resistência varietal no controle da aflatoxina.

Maçãs, cv. Fuji, fisiologicamente maduras e super maduras, foram fatiadas na forma de palitos, concentradas osmoticamente e desidratadas, objetivando-se avaliar suas características físicas, químicas e sensoriais. A concentração osmótica foi com soluções a 50% de sacarose, maltodextrina de milho (Mor-rex 1920) e glicose de milho (Excell 1040), sendo os dois últimos produzidos pela Refinações de Milho Brasil. Os palitos de maçãs foram imersos nas soluções concentradoras, por seis horas, drenados e desidratados a 70º C até 10% de umidade pelo controle de peso, seguindo-se armazenamento ambiental em vidros. As avaliações da matéria-prima e produto final foram de umidade, sólidos solúveis totais (º Brix), açúcares, pH, acidez titulável, aceitação pelo consumidor e rendimento. A avaliação estatística dos dados seguiu o delineamento inteiramente ao acaso, com médias submetidas a comparações múltiplas através do teste de Duncan ao nível de 5% de probabilidade. A composição da maçã "in natura" foi similar a encontrada na literatura. Os agentes de concentração osmótica passaram a fazer parte da composição dos produtos desidratados, alterando as características físicas e químicas em ambos estádios de maturação. O rendimento melhorou nos três tratamentos, sendo o melhor com sacarose que, também, resultou na preferência dos consumidores que optaram por "gosto muito", independentemente dos estádios de maturação das maçãs. O estádio de maturação não interferiu nas características dos produtos processados.

Os consumidores estão preocupados em adquirir produtos que atendam as questões ambientais, sociais e conseqüentemente sejam saudáveis, contribuindo para o crescimento da comercialização de produtos orgânicos em todo mundo. O café, produto tradicionalmente cultivado no Brasil e de grande aceitação (70% da população brasileira consome café diariamente), é um dos produtos cultivados sob manejo orgânico. Este trabalho teve por objetivo avaliar as características temporais do gosto amargo na bebida de café orgânico. Foram avaliadas quatro marcas de café orgânico (ORG-1, ORG-2, ORG-3 e ORG-4) e uma marca de café convencional (CON) por meio da Análise Tempo-Intensidade. Sete provadores selecionados e treinados avaliaram as amostras de café utilizando o programa "Sistema de Coleta de Dados Tempo-Intensidade-SCDTI" para Windows. Os resultados obtidos foram estatisticamente analisados por Análise de Variância (ANOVA), teste de Duncan e Análise de Componente Principal (ACP). A marca ORG-3 apresentou maior intensidade máxima (Imax), apresentando diferença estatística significativa (p<0,05) em relação as demais amostras para a percepção desse atributo. Entre as amostras ORG-1, ORG-2, ORG-4 e CON não houve diferença estatística significativa (p>0,05) em relação aos seis parâmetros avaliados.

O trabalho consiste na implementação de um controle convencional PID/SISO-feedback para obter um ajuste fino na temperatura de entrada da água de aquecimento em um processo de pasteurização. Para isto utilizou-se uma resistência de 2500 Watts instalada na linha do fluido secundário da seção de aquecimento do pasteurizador e um Pt100 para a medição de sua temperatura. Como o comportamento desta temperatura em função de uma mesma perturbação degrau de potência na resistência é dependente da vazão de trabalho, objetivou-se encontrar um controle único para que a mesma fosse mantida no set-point desejado na faixa de operação de vazão da água do processo (300 a 700L/h). Três sintonias para o controlador adaptativo PID foram testadas: a primeira consistiu na implementação de uma função adaptativa dos parâmetros PID, ajustada através dos valores individuais obtidos para cada vazão de trabalho conforme metodologia da curva de reação do processo; a segunda consistiu em configurar os parâmetros do PID com os valores médios destes calculados individualmente para cada vazão, e a terceira consistiu na sintonia através de uma função adaptativa ajustada pelos parâmetros de sintonia obtidos pela metodologia de Aström & Hägglund. A avaliação do desempenho das sintonias dos controladores adaptativos foi realizada por comparação dos valores dos índices de erro, obtidos por perturbações do sistema em malha fechada na vazão de água. Os resultados obtidos mostraram que dentre as sintonias testadas, a terceira sintonia, popularmente conhecida como "Bang-Bang", apresentou menores oscilações e os menores valores dos índices de erros.

As concentrações de compostos fenólicos totais e antocianinas em sucos de uva reconstituídos e simples, de diferentes marcas, disponíveis no comércio varejista da região metropolitana de Belo Horizonte (Minas Gerais) foram quantificadas. Também se determinou a contribuição das antocianinas poliméricas à cor e o índice de degradação das antocianinas. Os teores de compostos fenólicos totais variaram entre 0,27 e 2,41 g/L e as concentrações de antocianinas de 1,17 a 66,80 mg/L. Foram observadas diferenças significativas (Teste de Duncan, p<0,05) nas concentrações de fenólicos totais e de antocianinas entre as diferentes marcas, tanto para o suco de uva reconstituído quanto para o simples. O percentual de contribuição das antocianinas poliméricas à cor também variou significativamente entre as marcas, sendo que os sucos de uva simples tiveram percentuais superiores (81,6%) aos dos sucos de uva reconstituídos (77,5%). Os sucos de uva reconstituídos apresentaram valor médio total de degradação das antocianinas (3,92) maior que os sucos de uva simples (3,44).

Vários fatores influenciam a percepção do produto pelo consumidor e, conseqüentemente, sua intenção de compra. Dentre tais fatores pode estar a expectativa criada pelas características da embalagem e do rótulo, pois representam o primeiro contato entre o indivíduo e o produto. O objetivo desse trabalho foi avaliar o efeito das características da embalagem na intenção de compra de couve minimamente processada, considerando as diferenças individuais dos consumidores para processar as informações. Cinco características da embalagem de couve minimamente processada (informação, tipo de produção, cor, preço e visibilidade do produto) foram manipuladas e 12 embalagens criadas seguindo delineamento fatorial incompleto. A intenção de compra para o produto foi avaliada por 144 consumidores baseada apenas na observação das referidas embalagens. Os dados foram analisados utilizando Conjoint e Cluster Analyses. Os resultados apontaram para um segmento único de consumidores, composto por indivíduos com percepção do produto bastante similar quanto à intenção de compra. Dentre as características da embalagem, a informação foi a que obteve maior importância relativa (77%), enfatizando seu papel na intenção de compra para esse grupo de consumidores. Em seguida, o tipo de produção, cor e preço também contribuíram significativamente (p<0,0001) na intenção de compra de couve minimamente processada, havendo maior intenção de compra para a embalagem com a característica "sem produtos químicos" e preço baixo.
