Para controlar a proliferação de Egeria densa e reduzir os prejuízos que vêm causando na geração de energia em Paulo Afonso, é necessário conhecer sua capacidade de regeneração. Para isso, foram marcados quadrados de 4 m², em diferentes épocas do ano e em três profundidades (0-2, 2-6 e 6-9 m), e retiradas as biomassas de E. densa iniciais e três meses depois. Na profundidade de 0-2 m, as retiradas de plantas foram repetidas por cinco vezes, em quatro dos mesmos quadrados. Nesta profundidade, as massas regeneradas (518 g m-2) foram semelhantes às iniciais (482 g m-2), independentemente da época do ano (estação seca, geral: 506 g m-2; chuvosa: 513 g m-2) e da repetição da retirada. Na de 2-6 m, as massas regeneradas foram apenas 66% das iniciais (150 e 229 g m-2) e na de 6-9 m, apenas 21% (11,6 e 54,9 g m-2). A rápida recolonização e o grande crescimento de E. densa tornam muito difícil erradicá-la dos reservatórios do São Francisco. Sugere-se que seja explorado esse grande potencial de produção de biomassa.

Autonomic neuropathy is a frequent complication of diabetes associated with higher morbidity and mortality in symptomatic patients, possibly because it affects autonomic regulation of the sinus node, reducing heart rate (HR) variability which predisposes to fatal arrhythmias. We evaluated the time course of arterial pressure and HR and indirectly of autonomic function (by evaluation of mean arterial pressure (MAP) variability) in rats (164.5 ± 1.7 g) 7, 14, 30 and 120 days after streptozotocin (STZ) injection, treated with insulin, using measurements of arterial pressure, HR and MAP variability. HR variability was evaluated by the standard deviation of RR intervals (SDNN) and root mean square of successive difference of RR intervals (RMSSD). MAP variability was evaluated by the standard deviation of the mean of MAP and by 4 indices (P1, P2, P3 and MN) derived from the three-dimensional return map constructed by plotting MAPn x [(MAPn+1) - (MAPn)] x density. The indices represent the maximum concentration of points (P1), the longitudinal axis (P2), and the transversal axis (P3) and MN represents P1 x P2 x P3 x 10-3. STZ induced increased urinary glucose in diabetic (D) rats compared to controls (C). Seven days after STZ, diabetes reduced resting HR from 380.6 ± 12.9 to 319.2 ± 19.8 bpm, increased HR variability, as demonstrated by increased SDNN, from 11.77 ± 1.67 to 19.87 ± 2.60 ms, did not change MAP, and reduced P1 from 61.0 ± 5.3 to 51.5 ± 1.8 arbitrary units (AU), P2 from 41.3 ± 0.3 to 29.0 ± 1.8 AU, and MN from 171.1 ± 30.2 to 77.2 ± 9.6 AU of MAP. These indices, as well as HR and MAP, were similar for D and C animals 14, 30 and 120 days after STZ. Seven-day rats showed a negative correlation of urinary glucose with resting HR (r = -0.76, P = 0.03) as well as with the MN index (r = -0.83, P = 0.01). We conclude that rats with short-term diabetes mellitus induced by STZ presented modified autonomic control of HR and MAP which was reversible. The metabolic control may influence these results, suggesting that insulin treatment and a better metabolic control in this model may modify arterial pressure, HR and MAP variability

The dorsal periaqueductal gray (DPAG) has been implicated in the behavioral and autonomic expression of defensive reactions. Several results suggest that, along with GABA, glutamate and serotonin, nitric oxide (NO) may play a role in defense reactions mediated by this region. To further investigate this possibility we microinjected methylene blue (MB; 10, 30 or 100 nmol/0.5 µl) into the DPAG of rats submitted to the elevated plus-maze test, an animal model of anxiety. MB has been used as an inhibitor of soluble guanylate cyclase (sGC) to demonstrate cGMP-mediated processes, and there is evidence that NO may exert its biological effects by binding to the heme part of guanylate cyclase, causing an increase in cGMP levels. The results showed that MB (30 nmol) significantly increased the percent of time spent in the open arms (saline = 11.57 ± 1.54, MB = 18.5 ± 2.45, P<0.05) and tended to do the same with the percentage of open arm entries (saline = 25.8 ± 1.97, MB = 33.77 ± 3.07, P<0.10), but did not change the number of enclosed arm entries. The dose-response curve, however, had an inverted U shape. These results indicate that MB, within a limited dose range, has anxiolytic properties when microinjected into the DPAG.

Findings by our group have shown that the dorsolateral telencephalon of Gymnotus carapo sends efferents to the mesencephalic torus semicircularis dorsalis (TSd) and that presumably this connection is involved in the changes in electric organ discharge (EOD) and in skeletomotor responses observed following microinjections of GABA A antagonist bicuculline into this telencephalic region. Other studies have implicated the TSd or its mammalian homologue, the inferior colliculus, in defensive responses. In the present study, we explore the possible involvement of the TSd and of the GABA-ergic system in the modulation of the electric and skeletomotor displays. For this purpose, different doses of bicuculline (0.98, 0.49, 0.245, and 0.015 mM) and muscimol (15.35 mM) were microinjected (0.1 µL) in the TSd of the awake G. carapo. Microinjection of bicuculline induced dose-dependent interruptions of EOD and increased skeletomotor activity resembling defense displays. The effects of the two highest doses showed maximum values at 5 min (4.3 ± 2.7 and 3.8 ± 2.0 Hz, P < 0.05) and persisted until 10 min (11 ± 5.7 and 8.7 ± 5.2 Hz, P < 0.05). Microinjections of muscimol were ineffective. During the interruptions of EOD, the novelty response (increased frequency in response to sensory novelties) induced by an electric stimulus delivered by a pair of electrodes placed in the water of the experimental cuvette was reduced or abolished. These data suggest that the GABA-ergic mechanisms of the TSd inhibit the neural substrate of the defense reaction at this midbrain level.

Agaricus blazei Murill is a native Brazilian mushroom which functions primarily as an anticancer substance in transplanted mouse tumors. However, the mechanism underlying this function of A. blazei Murill remains obscure. The present study was carried out to investigate the effect of fraction FA-2-b-ß, an RNA-protein complex isolated from A. blazei Murill, on human leukemia HL-60 cells in vitro. Typical apoptotic characteristics were determined by morphological methods using DNA agarose gel electrophoresis and flow cytometry. The growth suppressive effect of fraction FA-2-b-ß on HL-60 cells in vitro occurred in a dose- (5-80 µg/mL) and time-dependent (24-96 h) manner. The proliferation of HL-60 cells (1 x 10(5) cells/mL) treated with 40 µg/mL of fraction FA-2-b-ß for 24-96 h and with 5-80 µg/mL for 96 h resulted in inhibitory rates ranging from 8 to 54.5%, and from 4.9 to 86.3%, respectively. Both telomerase activity determined by TRAP-ELISA and mRNA expression of the caspase-3 gene detected by RT-PCR were increased in HL-60 cells during fraction FA-2-b-ß treatment. The rate of apoptosis correlated negatively with the decrease of telomerase activity (r = 0.926, P < 0.05), but correlated positively with caspase-3 mRNA expression (r = 0.926, P < 0.05). These data show that fraction FA-2-b-ß can induce HL-60 cell apoptosis and that the combined effect of down-regulation of telomerase activity and up-regulation of mRNA expression of the caspase-3 gene could be the primary mechanism of induction of apoptosis. These findings provide strong evidence that fraction FA-2-b-ß could be of interest for the clinical treatment of acute leukemia.

Ectopic gastric mucosa (EGM) is considered to be a congenital condition. Rare cases of adenocarcinoma have been described. There are no data justifying regular biopsies or follow-up. Cyclooxygenase-2 (COX-2) is a protein involved in gastrointestinal tumor development by inhibiting apoptosis and regulating angiogenesis. The aim of this prospective study was to evaluate COX-2 expression in EGM and compare it with normal tissue and Barrett's esophagus. We evaluated 1327 patients. Biopsies were taken from the inlet patch for histological evaluation and from the gastric antrum to assess Helicobacter pylori infection. Biopsies taken from normal esophageal, gastric antrum and body mucosa and Barrett's esophagus were retrieved from a tissue bank. EGM biopsies were evaluated with respect to type of epithelium, presence of H. pylori, and inflammation. COX-2 was detected by immunohistochemistry using the avidin-biotin complex. EGM islets were found in 14 patients (1.1%). Histological examination revealed fundic type epithelium in 58.3% of cases, H. pylori was present in 50% and chronic inflammation in 66.7%. Expression of COX-2 was negative in normal distal esophagus, normal gastric antrum and normal gastric body specimens (10 each). In contrast, EGM presented over-expression of COX-2 in 41.7% of cases and Barrett's esophagus in 90% of cases (P = 0.04 and 0.03, respectively). COX-2 immunoexpression in EGM was not related to gender, age, epithelium type, presence of inflammation or intestinal metaplasia, H. pylori infection, or any endoscopic finding. Our results demonstrate up-regulation of COX-2 in EGM, suggesting a possible malignant potential of this so-called harmless mucosa.

We determined the effect of long-term aerobic swimming training regimens of different intensities on colonic carcinogenesis in rats. Male Wistar rats (11 weeks old) were given 4 subcutaneous injections (40 mg/kg body weight each) of 1,2-dimethyl-hydrazine (DMH, dissolved in 0.9% NaCl containing 1.5% EDTA, pH 6.5), at 3-day intervals and divided into three exercise groups that swam with 0% body weight (EG1, N = 11), 2% body weight (EG2, N = 11), and 4% body weight of load (EG3, N = 10), 20 min/day, 5 days/week for 35 weeks, and one sedentary control group (CG, N = 10). At sacrifice, the colon was removed and counted for tumors and aberrant crypt foci. Tumor size was measured and intra-abdominal fat was weighed. The mean number of aberrant crypt foci was reduced only for EG2 compared to CG (26.21 ± 2.99 vs 36.40 ± 1.53 crypts; P < 0.05). Tumor incidence was not significantly different among groups (CG: 90%; EG1: 72.7%; EG2: 90%; EG3: 80%). Swimming training did not affect either tumor multiplicity (CG: 2.30 ± 0.58; EG1: 2.09 ± 0.44; EG2: 1.27 ± 0.19; EG3: 1.50 ± 0.48 tumors) or size (CG: 1.78 ± 0.24; EG1: 1.81 ± 0.14; EG2: 1.55 ± 0.21; EG3: 2.17 ± 0.22 cm³). Intra-abdominal fat was not significantly different among groups (CG: 10.54 ± 2.73; EG1: 6.12 ± 1.15; EG2: 7.85 ± 1.24; EG3: 5.11 ± 0.74 g). Aerobic swimming training with 2% body weight of load protected against the DMH-induced preneoplastic colon lesions, but not against tumor development in the rat.

Amiodarone-induced thyroid dysfunction (AITD) is a common complication of amiodarone therapy and its prevalence varies according to iodine intake, subclinical thyroid disorders and the definition of AITD. There is no consensus about the frequency of screening for this condition. We evaluated 121 patients on chronic regular intake of amiodarone (mean intake = 248.5 ± 89 mg; duration of treatment = 5.3 ± 3.9 years, range = 0.57-17 years) and with stable baseline cardiac condition. Those with no AITD were followed up for a median period of 3.2 years (range: 0.6-6.7) and the incidence rate of AITD, defined by clinical and laboratorial findings as proposed by international guidelines, was obtained (62.8 per 1000 patients/year). We applied the Cox proportional hazard model to adjust for potential confounding factors and used sensitivity analysis to identify the best screening time for follow-up. We detected thyroid dysfunction in 59 (48.7%) of the 121 patients, amiodarone-induced hypothyroidism in 50 (41.3%) and hyperthyroidism in 9 (7.5%). Compared with patients without AITD, there was no difference regarding dosage or duration of therapy, heart rhythm disorder or baseline cardiac condition. During the follow-up of the 62 patients without AITD at baseline evaluation, 11 developed AITD (interquartile range, IR: 62.8 (95%CI: 31.3-112.3) cases per 1000 patients/year), 9 of them with hypothyroidism - IR: 11.4 (95%CI: 1.38-41.2), and 2 hyperthyroidism - IR: 51.3 (95%CI: 23.4-97.5). Age, gender, dose, and duration of treatment were not significant after adjustment. During the first 6 months of follow-up the incidence rate for AITD was 39.3 (9.2-61.9) cases per 1000 patients/year. These data show that AITD is quite common, and support the need for screening at 6-month intervals, unless clinical follow-up dictates otherwise or further information regarding the prognosis of untreated subclinical AITD is available.

The current in vitro study was designed to investigate the anti-inflammatory, cytotoxic and antioxidant activities of boesenbergin A (BA), a chalcone derivative of known structure isolated from Boesenbergia rotunda. Human hepatocellular carcinoma (HepG2), colon adenocarcinoma (HT-29), non-small cell lung cancer (A549), prostate adenocarcinoma (PC3), and normal hepatic cells (WRL-68) were used to evaluate the cytotoxicity of BA using the MTT assay. The antioxidant activity of BA was assessed by the ORAC assay and compared to quercetin as a standard reference antioxidant. ORAC results are reported as the equivalent concentration of Trolox that produces the same level of antioxidant activity as the sample tested at 20 µg/mL. The toxic effect of BA on different cell types, reported as IC50, yielded 20.22 ± 3.15, 10.69 ± 2.64, 20.31 ± 1.34, 94.10 ± 1.19, and 9.324 ± 0.24 µg/mL for A549, PC3, HepG2, HT-29, and WRL-68, respectively. BA displayed considerable antioxidant activity, when the results of ORAC assay were reported as Trolox equivalents. BA (20 µg/mL) and quercetin (5 µg/mL) were equivalent to a Trolox concentration of 11.91 ± 0.23 and 160.32 ± 2.75 µM, respectively. Moreover, the anti-inflammatory activity of BA was significant at 12.5 to 50 µM and without any significant cytotoxicity for the murine macrophage cell line RAW 264.7 at 50 µM. The significant biological activities observed in this study indicated that BA may be one of the agents responsible for the reported biological activities of B. rotunda crude extract.

Os carotenóides são pigmentos naturais, responsáveis pela cor em muitos alimentos como frutas e vegetais, gema de ovo, pele e músculo de alguns peixes. Porém, sua importância também está relacionada a funções ou ações biológicas nos seres humanos, como atividade pró-vitamínica A, fortalecimento do sistema imunológico, diminuição do risco de doenças degenerativas como câncer, doenças cardiovasculares, degeneração macular e catarata. A determinação da composição de carotenóides em alimentos, portanto, é considerada prioritária mundialmente. O objetivo do presente estudo foi analisar quantitativamente, por cromatografia líquida de alta eficiência, os principais carotenóides presentes em três cultivares de mamão (Formosa, Golden e Sunrise) e três cultivares de pêssego (Xiripá, Coral e Diamante). Para cada cultivar, foram analisados cinco lotes durante o ano para mamão e durante a safra para pêssego. Os três cultivares de mamão apresentaram composição parecida com as médias dos conteúdos totais de licopeno, beta-criptoxantina e beta-caroteno variando de 18,5 a 23,9, 8,2 a 11,7 e 0,5 a 1,2 µg.g -1, respectivamente. Em relação ao pêssego, os cultivares Coral e Xiripá tiveram níveis muito baixos de carotenóides. O cultivar Diamante teve um teor total médio de 6,4 µg.g -1 de beta-criptoxantina, o carotenóide principal.

Este trabalho teve como objetivo avaliar a eficiência da extração de compostos nitrogenados, como proteínas solúveis (PS), nitrogênio total (NT), nitrogênio não proteico (NNP) e nitrogênio proteico (NP) da polpa de anchoíta (Engraulis anchoita), bem como obter informações a respeito de sua composição proximal e do frescor em relação ao seu local de captura. A polpa de anchoíta foi submetida a tratamento de extração de nitrogenados utilizando como soluções NaHCO3 (0,1; 0,2; 0,3 e 0,5%), NaCl 0,3% e água destilada. O ciclo de lavagem da polpa utilizando 0,1% de NaHCO3, dois ciclos de água destilada e um ciclo de NaCl 0,3% demonstrou maior eficiência na extração dos compostos nitrogenados, assim como das proteínas sarcoplasmáticas. Na determinação da composição proximal, a anchoíta in natura apresentou valores de umidade de 77,2%, proteína 16,8%, lipídios 3,4% e cinzas 2,4% e, para a polpa de anchoíta, foram encontrados valores de umidade de 78,1%, proteína 17,5%, lipídios 2,4% e cinzas 2,0%. A avaliação do frescor foi determinada através do pH, bases voláteis totais (N-BVT) e trimetilamina (N-TMA), encontrando-se valores de 6,3; 11,5 mg.100 g-1 e 2,8 mg.100 g-1 para a anchoíta in natura e 6,7; 20,2 mg.100 g-1; 3,1 mg.100 g-1 para a polpa, respectivamente.

INTRODUÇÃO: A doença renal crônica (DRC) é um crescente problema de saúde pública. Ainda assim, há poucos dados sobre DRC no Brasil, principalmente nos seus estágios não dialíticos. OBJETIVO: Conhecer aspectos demográficos, clínicos e laboratoriais de pacientes com DRC não dialítica e avaliar o impacto dessas variáveis na progressão da doença. MÉTODOS: Estudo de coorte retrospectiva, composta de 65 pacientes adultos com DRC nos estágios 2-4, acompanhados e tratados ambulatorialmente por média de 28,24 ± 13,3 meses. RESULTADOS: A idade média foi de 64,6 ± 12,6 anos. As principais etiologias de DRC foram doença renal diabética (DRD) (47,7%) e nefroesclerose hipertensiva (34,2%). A maioria dos pacientes encontrava-se no estágio 3 da DRC (44,6%) e a minoria alcançou os alvos terapêuticos no controle de suas comorbidades, 40% para pressão arterial e 38,7% para o controle glicêmico. A perda média anual da taxa de filtração glomerular (TFG) foi 3,1 ± 7,3 mL/min/1,73 m² (mediana 1,4 mL/min/1,73 m²) e 21,5% dos pacientes evoluíram com DRC Progressiva. Pressão arterial diastólica (PAD) > 90 mmHg aumentou 2,7 vezes o risco de evoluir com DRC progressiva (IC 95%; 1,14-6,57; p = 0,0341), assim como pressão arterial sistólica (PAS) > 160 mmHg (RR = 3,64; IC 95%; 1,53-8,65; p = 0,0053) e proteinúria (RR = 4,05; IC 95%; 1,5510,56; p = 0,0031). Foi observada também média de PAS maior (p = 0,0359) e mediana de HDL-c menor (p = 0,0047) nos pacientes com DRC Progressiva. CONCLUSÃO: Neste estudo, hipertensão e proteinúria foram fatores de risco para evolução com DRC progressiva. Apesar do difícil controle clínico, a minoria dos pacientes evoluiu com a forma progressiva da DRC.

Introdu231;227;o e Objetivos: Comparar caracter237;sticas cl237;nicas e evolu231;227;o dos pacientes com e sem inj250;ria renal aguda (IRA), avaliar a incid234;ncia, mortalidade da IRA e fatores de risco de IRA e de 243;bito em pacientes em uma Unidade de Terapia Intensiva (UTI). M233;todos: Estudo retrospectivo que analisou 152 pacientes em uma 250;nica UTI. Avaliamos a idade, o sexo, o motivo do internamento, fatores de risco para IRA, dados laboratoriais, a necessidade de terapia renal substitutiva e a mortalidade. Acute Physiology and Chronic Health Evaluation (APACHE II), Sequential Organ Failure Assessment (SOFA) e RIFLE foram registrados no dia de admiss227;o na UTI. Determinamos a incid234;ncia da IRA, mortalidade e os preditores independentes de IRA e de 243;bito utilizando o modelo de regress227;o log237;stica. Resultados: A idade m233;dia foi de 57,1 ± 20 anos e 60,1% eram masculinos. IRA n227;o dial237;tica ocorreu em 81 pacientes (53,2%) e a IRA dial237;tica ocorreu em 19 pacientes (12,4%). A mortalidade global foi de 35,9%, enquanto que a taxa de mortalidade nos pacientes com IRA n227;o dial237;tica foi de 43,2% e a dos com IRA dial237;tica de 84,2%. Na an225;lise multivariada, a ventila231;227;o mec226;nica invasiva, a creatinina e a ureia elevadas na admiss227;o foram fatores de risco independentes para IRA, enquanto que diagn243;stico cl237;nico, uso de ventila231;227;o mec226;nica invasiva, ureia e lactato aumentados e hipernatremia foram fatores de risco independentes para mortalidade na UTI. Conclus227;o: A incid234;ncia e a mortalidade de IRA na UTI foram elevadas neste estudo, apesar dos avan231;os que v234;m surgindo no seu manejo.
