The humoral immune response plays an important role in the clearance of Giardia lamblia. However, our knowledge about the specific antigens of G. lamblia that induce a protective immune response is limited. The purpose of this study was to identify and characterise the immunogenic proteins of G. lamblia in a mouse model. We generated monoclonal antibodies (moAbs) specific to G. lamblia (1B10, 2C9.D11, 3C10.E5, 3D10, 5G8.B5, 5F4, 4C7, 3C5 and 3C6) by fusing splenocytes derived from infected mice. Most of these moAbs recognised a band of ± 71 kDa (5G8 protein) and this protein was also recognised by serum from the infected mice. We found that the moAbs recognised conformational epitopes of the 5G8 protein and that this antigen is expressed on the cell surface and inside trophozoites. Additionally, antibodies specific to the 5G8 protein induced strong agglutination (> 70-90%) of trophozoites. We have thus identified a highly immunogenic antigen of G. lamblia that is recognised by the immune system of infected mice. In summary, this study describes the identification and partial characterisation of an immunogenic protein of G. lamblia. Additionally, we generated a panel of moAbs specific for this protein that will be useful for the biochemical and immunological characterisation of this immunologically interesting Giardia molecule.

This study aimed to analyse changes in the spatial distribution of Lutzomyia longipalpis in Posadas, an urban area located in northeastern Argentina. Data were obtained during the summer of 2007 and 2009 through two entomological surveys of peridomiciles distributed around the city. The abundance distribution pattern for 2009 was computed and compared with the previous pattern obtained in 2007, when the first human visceral leishmaniasis cases were reported in the city. Vector abundance was also examined in relation to micro and macrohabitat characteristics. In 2007 and 2009, Lu. longipalpis was distributed among 41.5% and 31% of the households in the study area, respectively. In both years, the abundance rates at most of the trapping sites were below 30 Lu. longipalpis per trap per night; however, for areas exhibiting 30-60 Lu. longipalpis and more than 60 Lu. longipalpis, the areas increased in both size and number from 2007-2009. Lu. longipalpis was more abundant in areas with a higher tree and bush cover (a macrohabitat characteristic) and in peridomiciles with accumulated unused material (a microhabitat characteristic). These results will help to prioritise and focus control efforts by defining which peridomiciles display a potentially high abundance of Lu. longipalpis.

Chagas disease, caused by Trypanosoma cruzi, represents an endemic among Latin America countries. The participation of free radicals, especially nitric oxide (NO), has been demonstrated in the pathophysiology of seropositive individuals with T. cruzi. In Chagas disease, increased NO contributes to the development of cardiomyopathy and megacolon. Metallothioneins (MTs) are efficient free radicals scavengers of NO in vitro and in vivo. Here, we developed a murine model of the chronic phase of Chagas disease using endemic T. cruzi RyCH1 in BALB/c mice, which were divided into four groups: infected non-treated (Inf), infected N-monomethyl-L-arginine treated (Inf L-NAME), non-infected L-NAME treated and non-infected vehicle-treated. We determined blood parasitaemia and NO levels, the extent of parasite nests in tissues and liver MT-I expression levels. It was observed that NO levels were increasing in Inf mice in a time-dependent manner. Inf L-NAME mice had fewer T. cruzi nests in cardiac and skeletal muscle with decreased blood NO levels at day 135 post infection. This affect was negatively correlated with an increase of MT-I expression (r = -0.8462, p < 0.0001). In conclusion, we determined that in Chagas disease, an unknown inhibitory mechanism reduces MT-I expression, allowing augmented NO levels.

Lutzomyia longipalpis was recorded for the first time in Argentina in 2004, in the province of Formosa. In the following years, the vector spread to the south and west in the country and was recorded in the province of Chaco in 2010. From November 2010-May 2012, captures of Phlebotominae were made in the city of Resistencia and its surroundings, to monitor the spread and possible colonisation of Lu. longipalpis in the province of Chaco. In this monitoring, Lu. longipalpis was absent in urban sampling sites and its presence was restricted to Barrio de los Pescadores. This suggests that the incipient colonisation observed in 2010 was not followed by continuous installation of vector populations and expansion of their spatial distribution as in other urban centres of Argentina.

First remarks on the nesting biology of Hypodynerus andeus (Packard) (Hymenoptera, Vespidae, Eumeninae) in the Azapa valley, northern Chile. Some aspects about the nesting biology of the potter wasp Hypodynerus andeus (Packard, 1869) are reported for the first time. Observations were carried out at the Azapa valley, coastal desert of northern Chile. A total of sixty nests were collected and examined, each composed by 1-14 cells, most of them found attached to concrete lamp posts. The only preys recorded in the cells were Geometridae (Lepidoptera) caterpillars and the presence of the parasitoid Anthrax sp. (Diptera, Bombyliidae) was also recorded. A number of arthropods belonging to different groups, mainly spiders, were found occupying empty nests.

A new genus and species of leaf miner (Lepidoptera, Gracillariidae) for Chile associated to the native tree Lithraea caustica. We propose the new genus and species of Gracillariidae (Lepidoptera) Hualpenia lithraeophaga Mundaca, Parra &Vargas gen. nov., sp. nov., leaf miner of Lithraea caustica (Mol.) H. et Arn (Anacardiaceae) occurring in southern central Chile. Aspects of the life cycle, adult and larval morphology, development and feeding habits of the new genus and species are also presented. We emphasise the uniqueness and importance of this new species for broadening the current knowledge on the Chilean fauna of Gracillariidae.

Vihualpenia nom. nov. is proposed as a replacement name for Hualpenia Mundaca, Parra & Vargas, 2013 (Lepidoptera, Gracillariidae), in order to remove homonymy with Hualpenia Franz, 1996 (Coleoptera, Staphylinidae).

Egg laying site selection by a host plant specialist leaf miner moth at two intra-plant levels in the northern Chilean Atacama Desert. The spatial distribution of the immature stages of the leaf miner Angelabella tecomae Vargas & Parra, 2005 was determined at two intra-plant levels (shoot and leaflet) on the shrub Tecoma fulva fulva (Cav.) D. Don (Bignoniaceae) in the Azapa valley, northern Chilean Atacama Desert. An aggregated spatial pattern was detected for all the immature stages along the shoot, with an age dependent relative position: eggs and first instar larvae were clumped at apex; second, third and fourth instar larvae were mostly found at intermediate positions; meanwhile the spinning larva and pupa were clumped at basis. This pattern suggests that the females select new, actively growing leaflets for egg laying. At the leaflet level, the immature stages were found more frequently at underside. Furthermore, survivorship was higher for larvae from underside mines. All these results highlight the importance of an accurate selection of egg laying site in the life history of this highly specialized leaf miner. By contrast, eventual wrong choices in the egg laying site selection may be associated with diminished larval survivorship. The importance of the continuous availability of new plant tissue in this highly human modified arid environment is discussed in relation with the observed patterns.

Observations on the biology and distribution of Uresiphita reversalis (Lepidoptera, Crambidae), a defoliator of the native tree Calia secundiflora in México. Uresiphita reversalis (Guenée, 1854) feeding on Calia secundiflora (Ortega) Yakovlev is recorded for the first time in the state of Hidalgo, Mexico. New aspects regarding the life cycle, feeding behaviour, geographical distribution and host plant damage by U. reversalis on C. secundiflora are here presented and discussed.

A revegetação com espécies arbóreas é uma estratégia adequada, quando se pretende a reabilitação de terras degradadas. Entretanto, o estabelecimento e o crescimento destas em solos pobres em nutrientes dependem de diversos fatores como a capacidade de formar e beneficiar-se das micorrizas. No presente estudo, realizado em casa de vegetação na Universidade Federal de Lavras, Minas Gerais- Brasil, avaliaram-se aspectos da relação fungos micorrízicos arbusculares e espécies arbóreas do sudeste brasileiro. Os fungos estudados foram: Scutellospora pellucida, Acaulospora scrobiculata, Entrophospora colombiana, Gigaspora gigantea, Gigaspora margarita, Glomus etunicatum, Scutellospora gregaria, Glomus clarum e isolados oriundos de agrossistemas e de mata, os quais foram inoculados em dezesseis espécies vegetais: Luehea grandiflora, Cecropia pachystachya, Schinus terebinthifolius, Machaerium nyctitans, Senna macranthera, Senna spectabilis, Solanum granuloso-leprosum, Caesalpinea ferrea, Tabebuia serratifolia, Maclura tinctoria, Guazuma ulmifolia, Acacia polyphylla, Mimosa caesalpiniaefolia, Enterolobium contortisiliquum, Trema micrantha e Cedrela fissilis. Estas espécies apresentaram comportamento diferenciado em relação à susceptibilidade aos fungos e às respostas em crescimento decorrentes da inoculação. A amplitude de eficiência simbiótica dos fungos variou muito, sendo o Gl. clarum, E. colombiana, S. pellucida e Gl. etunicatum os fungos de maior amplitude, beneficiando mais de 80 % das espécies vegetais estudadas, e a A. scrobiculata a espécie de comportamento mais restrito em relação aos hospedeiros. A análise da compatibilidade fungo-hospedeiro e o comportamento geral das espécies em associação indicam a ocorrência de seletividade diferenciada na relação fungo-hospedeiro, existindo combinações de eficiência simbiótica muito variável para as plantas hospedeiras. A importância destes resultados para a revegetação de áreas degradadas é discutida.

A compactação do solo restringe o rendimento de culturas de grãos. Dentre outros fatores, a calagem pode favorecer a dispersão de microagregados e a formação de camadas compactadas. O objetivo deste trabalho foi avaliar os efeitos do calcário e da esterilização em atributos físicos e químicos do solo relacionados com a compactação do solo. O experimento foi efetuado em colunas de PVC, em casa de vegetação, durante 18 meses. Amostras esterilizadas e não esterilizadas de um Latossolo Vermelho distrófico foram incubadas com doses equivalentes a 0, 1,9, 3,8, 5,7, 7,6 e 15,2 Mg ha-1. Após 18 meses, foram avaliados a macro e microporosidade, a densidade do solo, teor de argila dispersa, a condutividade hidráulica saturada, o pH e os teores de Al, Ca e Mg trocáveis e de matéria orgânica do solo. A calagem aumentou a densidade do solo, o teor de argila dispersa em água, o pH e os teores de Ca2+ e Mg2+ trocáveis, e decresceu a micro e macroporosidade, a condutividade hidráulica saturada, os teores de matéria orgânica e de Al trocável. O aumento de pH e a redução da condutividade hidráulica saturada foram maiores no solo esterilizado. Os efeitos da calagem foram mais pronunciados na quantidade correspondente a 3,8 Mg ha-1. A esterilização aumentou o valor do pH em água e reduziu os teores de matéria orgânica e de Ca2+, além da condutividade hidráulica saturada. A dispersão de microagregados pela calagem pode, em parte, contribuir para a compactação do solo.

A utilização de resíduos orgânicos, como fertilizantes e condicionadores de solo, requer sua maturação e monitoramento da qualidade do material resultante. A vermicompostagem é uma técnica que, ao longo do processo, elimina os potenciais efeitos adversos dos resíduos à saúde humana e ao solo. A avaliação das substâncias húmicas (SH) nos produtos finais quanto à quantidade e à qualidade permite inferir sobre o grau de estabilidade e maturidade dos vermicompostos. Este trabalho objetivou avaliar a qualidade de seis vermicompostos das seguintes matérias-primas: esterco bovino (EB), esterco ovino (EO), esterco suíno (ES), esterco de codorna (EC), borra de café (BC) e de erva-mate (BE) com respeito ao teor de ácidos húmicos (AH) e de ácidos fúlvicos (AF) e às suas características químicas, após 70 dias de compostagem. O fracionamento químico da matéria orgânica foi realizado com base na solubilidade em meio básico e ácido e a distribuição de cada fração calculada como percentual do C total. Foram calculados os índices de humificação: percentual de AH e razão AH/AF. Nos AH e AF purificados, determinaram-se a composição elementar (CHNO), a composição química por espectroscopia de infravermelho com transformada de Fourier (FTIR) e o índice de aromaticidade I1630/I2920. O teor de substâncias húmicas (AH+AF) decresceu na seqüência: BC > EO≈ BE≈ EB > ES > EC, tendo sua composição química também diferido entre os vermicompostos. O grau de maturidade foi superior nos vermicompostos de resíduos de origem vegetal (borra de café e de erva-mate). Os AH desses dois vermicompostos apresentaram menor proporção de grupos oxigenados e menor caráter aromático.

Map units directly related to properties of soil-landscape are generated by local soil classes. Therefore to take into consideration the knowledge of farmers is essential to automate the procedure. The aim of this study was to map local soil classes by computer-assisted cartography (CAC), using several combinations of topographic properties produced by GIS (digital elevation model, aspect, slope, and profile curvature). A decision tree was used to find the number of topographic properties required for digital cartography of the local soil classes. The maps produced were evaluated based on the attributes of map quality defined as precision and accuracy of the CAC-based maps. The evaluation was carried out in Central Mexico using three maps of local soil classes with contrasting landscape and climatic conditions (desert, temperate, and tropical). In the three areas the precision (56 %) of the CAC maps based on elevation as topographical feature was higher than when based on slope, aspect and profile curvature. The accuracy of the maps (boundary locations) was however low (33 %), in other words, further research is required to improve this indicator.

The relationships between nutrient contents and indices of the Diagnosis and Recommendation Integrated System (DRIS) are a useful basis to determine appropriate ranges for the interpretation of leaf nutrient contents. The purpose of this study was to establish Beaufils ranges from statistical models of the relationship between foliar concentrations and DRIS indices, generated by two systems of DRIS norms - the F value and natural logarithm transformation - and assess the nutritional status of cotton plants, based on these Beaufils ranges. Yield data from plots (average acreage 100 ha) and foliar concentrations of macro and micronutrients of cotton (Gossypium hirsutum r. latifolium) plants, in the growing season 2004/2005, were stored in a database. The criterion to define the reference population consisted of plots with above-average yields + 0.5 standard deviation (over 4,575 kg ha-1 seed cotton yield). The best-fitting statistical model of the relationship between foliar nutrient concentrations and DRIS indices was linear, with R² > 0.8090, p < 0.01, except for N, with R² = 0.5987, p < 0.01. The two criteria were effective to diagnose the plant nutritional status. The diagnoses were not random, but based on the effectiveness of the chi-square-tested method. The agreement between the methods to assess the nutritional status was 92.59-100 %, except for S, with 74.07 % agreement.

In order to select soil management practices that increase the nitrogen-use efficiency (NUE) in agro-ecosystems, the different indices of agronomic fertilizer efficiency must be evaluated under varied weather conditions. This study assessed the NUE indices in no-till corn in southern Paraguay. Nitrogen fertilizer rates from 0 to 180 kg ha-1 were applied in a single application at corn sowing and the crop response investigated in two growing seasons (2010 and 2011). The experimental design was a randomized block with three replications. Based on the data of grain yield, dry matter, and N uptake, the following fertilizer indices were assessed: agronomic N-use efficiency (ANE), apparent N recovery efficiency (NRE), N physiological efficiency (NPE), partial factor productivity (PFP), and partial nutrient balance (PNB). The weather conditions varied largely during the experimental period; the rainfall distribution was favorable for crop growth in the first season and unfavorable in the second. The PFP and ANE indices, as expected, decreased with increasing N fertilizer rates. A general analysis of the N fertilizer indices in the first season showed that the maximum rate (180 kg ha-1) obtained the highest corn yield and also optimized the efficiency of NPE, NRE and ANE. In the second season, under water stress, the most efficient N fertilizer rate (60 kg ha-1) was three times lower than in the first season, indicating a strong influence of weather conditions on NUE. Considering that weather instability is typical for southern Paraguay, anticipated full N fertilization at corn sowing is not recommended due the temporal variability of the optimum N fertilizer rate needed to achieve high ANE.
