Systemic fungal infections are responsible for high mortality rates. Several species of fungi may be involved, but Candida spp. is the most prevalent. Simvastatin is used to lower cholesterol and also exhibits antifungal action. The aim of this study was to evaluate the synergistic action of simvastatin with fluconazole against strains of Candida spp. Susceptibility testing was performed according to protocol M27-A3, by broth microdilution method and the synergistic effect of simvastatin and fluconazole was calculated based on FICI (Fractional Inhibitory Concentration Index). Eleven strains were evaluated, and simvastatin showed a synergistic effect with fluconazole against 10 (91%) of the Candida spp. strains tested. Simvastatin may be a valuable drug in the treatment of systemic infections caused by Candida spp.

SUMMARY A food-borne trematode infection fascioliasis is one among common public health problems worldwide. It caused a great economic loss for the human race. Control of snail population below a certain threshold level is one of the important methods in the campaign to reduce the incidence of fascioliasis. The life cycle of the parasite can be interrupted by killing the snail or Fasciola larva redia and cercaria inside of the snail Lymnaea acuminata. In vitro toxicity of different binary combinations (1:1 ratio) of plant-derived larvicidal active components such as citral, ferulic acid, umbelliferone, azadirachtin and allicin against Fasciola redia and cercaria were tested. The mortality of larvae was observed at 2h, 4h, 6h and 8h of treatment. In in vitro condition azadirachtin + allicin (1:1 ratio) was highly toxic against redia and cercaria (8h LC50 0.006 and 0.005 mg/L). Toxicity of citral + ferulic acid was lowest against redia and cercaria larvae.

Currently multiresistant Staphylococcus aureus is one common cause of infections with high rates of morbidity and mortality worldwide, which directs scientific endeavors in search for novel antimicrobials. In this study, nine extracts from Bidens pilosa (root, stem, flower and leaves) and Annona crassiflora (rind fruit, stem, leaves, seed and pulp) were obtained with ethanol: water (7:3, v/v) and their in vitro antibacterial activity evaluated through both the agar diffusion and broth microdilution methods against 60 Oxacillin Resistant S. aureus (ORSA) strains and against S. aureus ATCC6538. The extracts from B. pilosa and A. crassiflora inhibited the growth of the ORSA isolates in both methods. Leaves of B. pilosa presented mean of the inhibition zone diameters significantly higher than chlorexidine 0.12% against ORSA, and the extracts were more active against S. aureus ATCC (p < 0.05). Parallel, toxicity testing by using MTT method and phytochemical screening were assessed, and three extracts (B. pilosa, root and leaf, and A. crassiflora, seed) did not evidence toxicity. On the other hand, the cytotoxic concentrations (CC50 and CC90) for other extracts ranged from 2.06 to 10.77 mg/mL. The presence of variable alkaloids, flavonoids, tannins and saponins was observed, even though there was a total absence of anthraquinones. Thus, the extracts from the leaves of B. pilosa revealed good anti-ORSA activity and did not exhibit toxicity.

Introduction: The essential oil Mentha x villosa (MVEO) has a wide range of actions, including antibacterial, antifungal, antiprotozoal and schistosomicidal actions. The present study aimed to investigate the ultrastructural changes of MVEO on the tegument of adult Schistosoma mansoni. Materials and Methods: Different concentrations of MVEO were tested on S. mansoni adult worms in vitro. Ultrastructural changes on the tegument of these adult worms were evaluated using scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Results: The MVEO caused the death of all worms at 500 μg mL-1 after 24 h. After 24h of 500 μg mL-1 MVEO treatment, bubble lesions were observed over the entire body of worms and they presented loss of tubercles in some regions of the ventral portion. In the evaluation by TEM, S. mansoni adult worms treated with MVEO, 500 μg mL-1, presented changes in the tegument and vacuoles in the syncytial matrix region. Glycogen granules close to the muscle fibers were visible. Conclusion: The ability of MVEO to cause extensive ultrastructural damage to S. mansoni adult worms correlates with its schistosomicidal effects and confirms earlier findings with S. mansoni.

Os autores, utilizando a técnica da diluição seriada em meio líquido, determinaram as concentrações mínimas inibitórias da ampicüina e da hetacilina em 150 amostras de Salmonella typhi, isoladas em sua totalidade a partir de hemoculturas, no Estado da Guanabara. Obteve-se como média das concentrações bacteriostáticas, 0,545 mcg/ml para ampicilina e 0,720 mcg/ml, no caso da hetacilina.

Verificaram os autores que, "in vitro", concentrações de "Hycanthone" variáveis de 0,39 a 400 mcg/ml não impediram o desenvolvimento de Salmonella anatum, S. derby, S. typhi e S. typhimurium. Essa concentração pode, sem dúvida, representar apoio à hipótese segundo a qual, na salmonelose de curso prolongado, há carreamento das bactérias pelos helmintos, uma vez que Macêdo & cols. curaram, recentemente, a um só tempo, a esquistossomíase mansônica e a infecção bacteriana de pacientes acometidos pelo citado e intrigante problema da patologia regional brasileira.

Ascaris suum metabolic antigens were obtained frorn second and early third stage larvae cultured in vitro in supplemented Eagle's minimum essential medium. Metabolic antigens harvested after 12 and 16 days from in vitro cultures were eluted through Bio-Gel Al.5. Three main elution peaks were identified, dialysed, lyophilized and injected into mice with 4% sodium alginate. Peak 11 from elution of two preparations of metabolic antigens protected mice against a chállenge infection of 10,000 A. suum embryonated eggs.

Foi estudada a atividade "in vitro" de um novo antibiótico aminoglicosidico, a Tobramicina, e feita a comparação com a Gentamicina. Os resultados de 1.080 testes de sensibilidade pelo método de discos obtidos com os principais agentes infecciosos e segundo a origem do material indicam que o novo antibiótico possue atividade comparável à da Gentamicina.

Os autores estudaram a ação "in vitro" da oxamniquine sobre dez espécies de salmonelas. Verificaram que diluições de 1,56 a 400 mcg/ml não exerceram qualquer efeito no crescimento bacteriano. Estas observações têm implicações no tratamento da salmonelose de curso prolongado, assim como nos estudos da sua patogenia.

Comparamos diferentes medios artificiales y procedimientos de cultivo in vitro de Leishmania braziliensis braziliensis (Lbb) aislada de lesiones de hamsteres experimentalmente infectados. Observamos que la aspiración de material de la lesión, usando jeringa de 5 mlyaguja 22g(25 x 8mm), mostró resultados satisfactorios siendo un procedimiento conveniente de colecta. El medio de Agar Sangre Difco (DAB) es un medio eficiente para el aislamiento de Lbb. El medio de Evans demostro similares resultados al DAB, mientras que el medio líquido de Schneider y el químicamente definido AR-103 fueron nitidamente inferiores. En la propagación in vitro de Lbb, el medio de Schneider permitió un buen crecimiento del flagelado, alcanzando densidades de 0,21 - 2,57 x 10(8) promastigotes por ml ya, en el 5.° dia de cultivo. La adición de 5-fluorocytosine a los tubos de cultivo (150/jg/ml)para evitar la contaminación por hongos, no inhibió el crecimiento de Lbb.

O presente estudo avalia a resposta de cepas de Plasmodium falciparum às drogas antimaláricas, através de testes in vitro, isoladas em 7 municípios do sul do Estado do Pará. Foram efetuados 69 testes para cloroquina e mefloquina, 62 para amodiaquina e 61 para quinino. Os resultados mostram elevada resistência para cloroquina (71%), relativamente baixa resistência para amodiaquina com (25,8%) e para o quinino apenas 8,2%. Mefloquina revela ampla sensibilidade (100%), mas, demonstrando perda da mesma quando comparada em dois períodos distintos. Evidenciou-se também cepas multirresistentes em dois dos municípios estudados.

Este estudo avalia a evolução da sensibilidade in vitro do Plasmodium falciparum em uma área de prospecção de ouro no Estado do Amapá no período de 1983 a 1990. Foram efetuados 75 testes para cloroquina e quinino, 74 para amodiaquina e 76 para mefloquina. Os resultados revelaram 81% de resistência à cloroquina e 27% para a amodiaquina, enquanto que para quinino e mefloquina não foram evidenciadas cepas resistentes. Contudo, para estas duas últimas drogas identificou-se uma crescente perda da sensibilidade ao longo do tempo. Aparentemente observa-se uma associação entre resistência à cloroquina e a diminuição da sensibilidade ao quinino.
