The characteristics of tuberculosis (TB) patients related to a chain of recent TB transmissions were investigated. Mycobacterium tuberculosis (MTB) isolates (120) were genotyped using the restriction fragment length polymorphism-IS6110 (R), spacer oligotyping (S) and mycobacterial interspersed repetitive units-variable number of tandem repeats (M) methods. The MTB isolates were clustered and the clusters were grouped according to the similarities of their genotypes. Spearman’s rank correlation coefficients between the groups of MTB isolates with similar genotypes and those patient characteristics indicating a risk for a pulmonary TB (PTB) chain transmission were ana- lysed. The isolates showing similar genotypes were distributed as follows: SMR (5%), SM (12.5%), SR (1.67%), MR (0%), S (46.67%), M (5%) and R (0%). The remaining 35 cases were orphans. SMR exhibited a significant correlation (p < 0.05) with visits to clinics, municipalities and comorbidities (primarily diabetes mellitus). S correlated with drug consumption and M with comorbidities. SMR is needed to identify a social network in metropolitan areas for PTB transmission and S and M are able to detect risk factors as secondary components of a transmission chain of TB.

Chagas disease or American trypanosomiasis is, together with geohelminths, the neglected disease that causes more loss of years of healthy life due to disability in Latin America. Chagas disease, as determined by the factors and determinants, shows that different contexts require different actions, preventing new cases or reducing the burden of disease. Control strategies must combine two general courses of action including prevention of transmission to prevent the occurrence of new cases (these measures are cost effective), as well as opportune diagnosis and treatment of infected individuals in order to prevent the clinical evolution of the disease and to allow them to recuperate their health. All actions should be implemented as fully as possible and with an integrated way, to maximise the impact. Chagas disease cannot be eradicated due because of the demonstrated existence of infected wild triatomines in permanent contact with domestic cycles and it contributes to the occurrence of at least few new cases. However, it is possible to interrupt the transmission ofTrypanosoma cruziin a large territory and to eliminate Chagas disease as a public health problem with a dramatic reduction of burden of the disease.

A pesquisa teve por objetivo registrar eventuais mudanças nas decisões instrucionais e nas representações sobre o ensino, resultantes de um trabalho sistemático de reflexão sobre a prática, em um curso de formação inicial de professores de Física. Na Faculdade, os participantes assistiam aos vídeos de seus desempenhos em classes do secundário, analisando, problematizando e propondo reformulações para suas práticas docentes. O estudo descreve as características que marcaram a interação professor-licenciandos, facilitando o processo de (re)construção e (re)formulação das representações e das práticas docentes.

Os tricomas glandulares presentes nas folhas e ramos das plantas do gênero Lycopersicon são responsáveis pela secreção de metabólitos de diferentes naturezas. A presença de alguns desses compostos tem sido associada à resistência do tomate a diferentes insetos. A traça-do-tomateiro, Tuta absoluta (Meyrick), é uma das pragas mais nocivas da América do Sul. O adulto oviposita sobre as folhas do tomate e suas larvas abrem galerias no mesófilo das folhas, ramos, flores e frutos. As espécies silvestres do tomate conservam a capacidade de biossintetizar compostos químicos que lhes conferem resistência a esta praga. No presente trabalho, foi avaliada a preferência para oviposição desse inseto sobre folhas com e sem tricomas glandulares de L. esculentum (Mill.) cv. Uco Plata, suscetível, e de L. hirsutum f. glabratum (Mull.) PI 134417, espécie silvestre afim ao tomate, e resistente à traça. Os resultados sugerem que as fêmeas ovipositam indistintamente sobre as folhas de ambas espécies, independentemente da presença, ou não, dos tricomas glandulares. E a presença destes e de seus exsudatos não têm efeito inibidor na oviposição do inseto.

O metabólito secundário 2-tridecanona, secretado pelos tricomas glandulares tipo VI das folhas de tomateiro silvestre, Lycopersicon hirsutum L., confere-lhe resistência a uma grande variedade de insetos, inclusive a traça-do-tomateiro, Tuta absoluta (Meyrick). O objetivo do trabalho foi avaliar a concentração de 2- tridecanona, a densidade de tricomas glandulares tipo VI e o grau de infestação da traça-do-tomateiro na cultivar suscetível 'Uco Plata' (Lycopersicon esculentum Mill.), na linhagem resistente PI 134417 (L. hirsutum f. glabratum) e em suas progênies F1 e F2. Foram avaliadas ainda as possíveis associações entre a concentração de 2-tridecanona, a densidade de tricomas glandulares tipo VI e o grau de infestação da traça-do-tomateiro. O grau médio de infestação da traça-do-tomateiro em 'Uco Plata' foi significativamente superior ao obtido em PI 134417. Ainda que a concentração de 2-tridecanona tenha sido significativamente superior no parental resistente, a presença deste metabólito somente explicaria parcialmente a resistência (R² = 8,17%). Não se detectaram diferenças significativas na densidade de tricomas tipo VI entre o parental suscetível e o resistente. Este comportamento ocorreu independentemente do conteúdo de 2-tridecanona e do grau de infestação da traça-do-tomateiro.

The objective of the present work was to determine the inheritance and stability of transgenes of a transgenic bean line expressing the genes rep-trap-ren from Bean golden mosaic virus and the bar gene. Crosses were done between the transgenic line and four commercial bean cultivars, followed by four backcrosses to the commercial cultivars. Progenies from each cross were evaluated for the presence of the transgenes by brushing the leaves with glufosinate ammonium and by polymerase chain reaction using specific oligonucleotides. Advanced generations were rub-inoculated with an isolate of Bean common mosaic necrosis virus (BCMNV). The transgenes were inherited consistently in a Mendelian pattern in the four crosses studied. The analyzed lines recovered close to 80% of the characteristics of the recurrent parent, as determined by the random amplified DNA markers used, besides maintaining important traits such as resistance to BCMNV. The presence of the transgene did not cause any detectable undesirable effect in the evaluated progenies.

This work investigates the adsorption of Alizarin, Eriochrome Blue Black R and Fluorescein using chitosan, goethite and magnetite as adsorbents. For Alizarin, the best adsorbent is chitosan with a Langmuir parameter of 15.8 mmol dye/g adsorbent. For Eriochrome Blue Black R only 1.94 mmol dye/g chitosan is adsorbed. Langmuir parameters for the Alizarin adsorption on both iron oxides display one or two orders of magnitude lower than for chitosan and two orders of magnitude lower in the case of Eriochrome Blue Black R. Fluorescein does not adsorb in appreciable amounts on chitosan and it presents the lower affinity on the iron oxides.

This work describes the creation of an very simple calculation algorithm, based in basic chemical and mathematic principles, for the calculation of weak diprotic acid dissociation constants as, for example, amino acids, from potentiometric titrations. For an easier understanding of the algorithm the logical reasoning of this calculus is schematized in a diagram of blocks. In the second part of the work the algorithm is applied to an Excel calculation sheet to determine the dissociation constants of Nicotinic Acid and Glycine, from the respective potentiometric titration curves. The values obtained using this algorithm are compared with those estimated by Hyperquad2008 (program generally used for this type of calculus) and also with the values of a stability constants database.

This work studies the effect of NTMP (nitrilotris(methylenephosphonic acid)) on the adsorption of Cu(II), Zn(II), and Cd(II) onto boehmite in the pH range 5-9.5. The data were analyzed using the 2-pK constant capacitance model (CCM) assuming ternary surface complex formation. Under stoichiometric conditions, NTMP is more effective for removing Cu(II) than Zn(II) from solution and the contribution of ternary surface complexes are important to model the adsorption of both metals. Under nonstoichiometric conditions and high surface loading with a Me(II)/NTMP ratio of 1:5, Cu(II) and Zn(II) adsorption is significantly suppressed. In the case of Cd(II) the free metal adsorption is the most dominant species.

This study aims to synthesize and characterize organoclays developed from an Argentinian montmorillonite (Bent) using hexadecyltrimethylammonium bromide (HDTMA-Br) as the intercalation agent. Subsequently, an adsorption mechanism is proposed. The obtained organoclays were more hydrophobic than the starting clay. Surfactant molecules were adsorbed initially through cation exchange in sites placed in the interlayer space of the clay. Adsorption in such sites continued until the interlayer space was saturated. Depending on the surfactant loading introduced during the intercalation process, different organizations of surfactant in the interlayer were obtained. Further adsorption of surfactant occurred in the mesopores generated by tactoids in the "house of cards" organization. This process kept surfactant molecules relatively free and out of the interlayer space.

Brain-derived neurotrophic factor (BDNF) is associated with neuroplasticity and synaptic strength, and is decreased in conditions associated with chronic stress. Nevertheless, BDNF has not yet been investigated in psoriasis, a chronic inflammatory systemic disease that is exacerbated by stress. Therefore, our aim was to determine BDNF plasma levels in psoriasis patients and healthy controls. Adult patients (n=94) presenting with psoriasis for at least 1 year were enrolled, and age- and gender-matched with healthy controls (n=307) from the Brazilian Longitudinal Study of Adult Health (ELSA-Brasil). Participants had neither a previous history of coronary artery disease nor current episode of major depression. BDNF plasma levels were determined using the Promega ELISA kit. A general linear model was used to compare BDNF levels in psoriasis patients and controls, with age, gender, systolic blood pressure, serum fasting glucose, blood lipid levels, triglycerides, smoking status, and body mass index examined. After adjusting for clinical and demographic variables, significantly decreased BNDF plasma levels were observed in psoriasis patients (P=0.01) (estimated marginal means of 3922 pg/mL; 95%CI=2660-5135) compared with controls (5788 pg/mL; 95%CI=5185-6442). Similar BDNF levels were found in both mild and severe cases of psoriasis. Our finding, that BDNF is decreased in psoriasis, supports the concept of a brain-skin connection in psoriasis. Further studies should determine if BDNF is increased after specific psoriasis treatments, and associated with different disease stages.

O objetivo deste trabalho foi o desenvolvimento de imunorreagentes policlonais convenientes para uso em um teste imunoenzimático para detecção rápida de Salmonella em alimentos. Foram produzidos seis anti-soros policlonais anti-Salmonella contendo as aglutininas e, h; 1,6; i; 1,2; f,g,s e m,t. Como antígenos, foram empregadas culturas formolizadas de quatro sorotipos de Salmonella (S. Typhimurium fases 1 e 2, S. Anatum fases 1 e 2, S. Agona monofásica e S. Oranienburg monofásica) cultivadas em caldo tripticase de soja. O teste imunoenzimático utilizado foi o indireto, empregando-se anti-IgG-peroxidase como conjugado e OPD-H2O2 como sistema cromógeno. Os testes imunoenzimáticos foram conduzidos com os anti-soros policlonais individuais absorvidos e não-absorvidos, bem como empregando-se um anti-soro polivalente, correspondente a um "pool" de anti-soros absorvidos e não-absorvidos com culturas puras de Salmonella e outras enterobactérias. A absorção dos soros eliminou as reações cruzadas com todas as enterobactérias testadas nesse estudo, apresentadas tanto por alguns anti-soros individuais quanto pelo polivalente. Entre os seis anti-soros estudados, os que apresentaram melhor desempenho foram o f,g,s não-absorvido e o polivalente absorvido.

A metodologia convencional utilizada para detecção de Salmonella em alimentos é trabalhosa, apresenta custo elevado e os resultados definitivos somente estão disponíveis após 96 horas. Vários métodos rápidos têm sido propostos, sendo os testes imunoenzimáticos os mais empregados. Este estudo relata o desenvolvimento de um teste imunoenzimático para detecção de Salmonella em alimentos, empregando-se um anti-soro policlonal monovalente contendo aglutininas f,g,s, não absorvido, e um anti-soro polivalente absorvido contendo as aglutininas e,h; 1,6; i; 1,2; f,g,s e m,t. A eficiência foi comparada com a da metodologia de cultivo tradicional. O teste imunoenzimático foi empregado para a detecção de Salmonella em amostras de alimentos infantis experimentalmente inoculadas com este patógeno e com outras enterobactérias, em diferentes proporções. O teste imunoenzimático revelou-se significativamente mais sensível que o método de cultivo. Esse mesmo teste, utilizando-se o anti-soro f,g,s não absorvido com antígenos heterólogos revelou concordância de 89,6% com o método de cultivo e sensibilidade de 100,0%. Por outro lado, empregando-se o anti-soro polivalente absorvido, a concordância com o método de cultivo foi de 81,3% embora a sensibilidade tenha se mantido no mesmo nível. O desempenho do teste imunoenzimático empregando-se um desses dois anti-soros indica um grande potencial de aplicação como método de triagem na detecção de Salmonella em alimentos.
