We describe the design and tests of a set-up mounted in a conventional double beam spectrophotometer, which allows the determination of optical density of samples confined in a long liquid core waveguide (LCW) capillary. Very long optical path length can be achieved with capillary cell, allowing measurements of samples with very low optical densities. The device uses a custom optical concentrator optically coupled to LCW (TEFLON® AF). Optical density measurements, carried out using a LCW of ~ 45 cm, were in accordance with the Beer-Lambert Law. Thus, it was possible to analyze quantitatively samples at concentrations 45 fold lower than that regularly used in spectrophotometric measurements.

Capillary electrophoresis has become a well-established and routine-based separation technique. It is based on the differences between charged analyte mobility in aqueous or organic electrolytes. Its major limitation is the sensitivity due to small sample injection volumes and the narrow diameter of the capillaries, especially when UV detection is used. There are a number of ways to increase the concentration sensitivity. This report shows some on-line preconcentration strategies to perform it in free solution capillary electrophoresis that are based on manipulation of the analyte electrophoretic velocity during the sample introduction (stacking, field amplification and transient isotachophoresis).

The carrot leaf dehydration conditions in air circulation oven were optimized through response surface methodology (RSM) for minimizing the degradation of polyunsaturated fatty acids, particularly alpha-linolenic (LNA, 18:3n-3). The optimized leaf drying time and temperature were 43 h and 70 ºC, respectively. The fatty acids (FA) were investigated using gas chromatography equipped with a flame ionization detector and fused silica capillary column; FA were identified with standards and based on equivalent-chain-length. LNA and other FA were quantified against C21:0 internal standard. After dehydration, the amount of LNA, quantified in mg/100 g dry matter of dehydrated carrot leaves, were 984 mg.

This work describes the development of a home-made capillary electrophoresis (CE) system based on the capacitively coupled contactless conductivity detection (C4D) for the separation of the metallic species Zn2+, Cr3+, Pb2+, Cd2+, Co2+, Cu2+, Ni2+ e Tl+. A background electrolyte composed of MES/Histidine 0,02 mol L-1 (pH 5.0) was optimized for the separation of the metallic species by using organic solvents and complexing agents as additives. The system allowed the determination of the metallic species using MES/Histidine 0,02 mol L-1 and methanol 5% (pH 5.0) as a background electrolyte, 15 kV separation voltage and hydrodynamic injection by gravity.

A gas chromatographic method has been developed for the assay of fluvastatin sodium (FLU). FLU was silylated with N,O-bis(trimethylsilyl)trifluoroacetamide-1% trimethylchlorosilane at 90 ºC for 30 min and analysed in a DB-1 column by capillary gas chromatograph with a flame ionization detector. The method was validated. The assay was linear over the concentration range at 10.0 to 50.0 µg mL-1. The limit of detection and the limit of quantitation were 1.0 and 3.0 µg mL-1, respectively. The recoveries of FLU derivatives were in the range of 99.25-99.80%. In inter-day and intra-day analysis, the values of relative standard deviation (%) and the relative mean error (%) were found between 0.20-0.80% and -0.20-0.75%, respectively. The developed method was succesfully applied to analyze the FLU content in tablet formulation. The results were statistically compared with those obtained by the official method, and no significant difference was found between the two methods. Therefore, it can be recommended for the quality control assay of FLU in pharmaceutical industry.

A capillary electrophoresis (CE) method was developed and validated for determination of cetirizine dihydrochloride in tablets and compounded capsules. The electrophoretic separation was performed in an uncoated fused-silica capillary (40 cm x 50 μm i.d.) using 20 mmol L-1 sodium tetraborate buffer (pH 9.3) as background electrolyte, a hydrodinamic sample injection at 50 mBar for 5 s, 20 KV applied voltage at 25 °C, and detection at 232 nm. The proposed method was compared with the high performance liquid chromatographic (HPLC) method previously validated for this drug, and statistical analysis showed no significant difference between the techniques.

This work describes CE preconcentration strategies based on the effect of manipulation of the disperse/secondary velocity. Introduced by Terabe et al. in 1984, micellar electrokinetic chromatography is a powerful separation approach that increases the usage of electrokinetic phenomena for the separation of nonionic compounds. The main disadvantage of MEKC is the low concentration sensitivity associated with the limited optical path length for on-capillary photometric detection and the limited volume of sample solution that can be injected. This paper compiles on-line concentration strategies for neutral analytes by sample stacking and sweeping in micellar electrokinetic chromatography.

This review describes the advantages and disadvantages of using capillary liquid chromatography (CLC), which is considered the newest member in the analytical separation science arsenal. Although CLC has tremendous potential for being the next major innovation in separatory analysis, it has not yet obtained great popularity compared to conventional high performance (and ultra-high performance) liquid chromatography. Comparisons are made between these techniques and some of the reasons that CLC has not yet reached its potential will be advanced.

In this work, a rapid and simple method using capillary electrophoresis (CE) was developed for the determination of the benzoate, sorbate, methyl and propylparaben in foodstuffs. A running buffer consisting of 20 mmol L-1 (pH = 9.3) tetraborate enabled separation of the analytes in less than 5 min. The detector wavelength was set at 220 nm. The method was successfully applied to the analysis of sodas, sweeteners, sauces and juices. The range of preservatives found were from 478.5-466.6 mg kg-1 for methylparaben , 83.7-231.3 mg kg-1 for sorbate and 336.7-428.3 mg kg-1 for benzoate.

A stability-indicating method using MEKC was validated for the analysis of olmesartan medoxomil in tablets. Successful separation was achieved using a fused silica capillary (40 cm x 50 µm i.d.); background electrolyte consisted of a combination of 10 mmol L-1 borate buffer and 5 mmol L-1 anionic detergent sodium dodecyl sulfate (95:5; v/v) pH 6.5; hydrodynamic mode at 50 mBar for 5 s; 25 kV separation voltage at 25 ºC; and column temperature 25 ºC with detection at 257 nm. The proposed method, validated following ICH guidelines, was applied to the determination of this antihypertensive with good results compared with an LC method.

Density of dilute aqueous solutions of 1,2 pentanediol, 2,4 pentanediol, 1,4 pentanediol and 1,5 pentanediol at 283.15, 288.15, 293.15, 298.15, 303.15 and 308.15 K in the concentration range 0.0000 to 0.0060 in mole fraction were determined by using Wood-Brusie type capillary neck pycnometers. The solute partial molar volume as a function of solute concentration for each system was correlated with a linear equation for each temperature to estimate the slope limit and the partial molar volume at infinite dilution, and the predominant hydrophilic-hydrophobic effect was found in this region.

The filling of capillaries via the sol-gel process is growing. Therefore, this technical note focuses on disseminating knowledge acquired in the Group of Analytical Chemistry and Chemometrics over seven years working with monolithic stationary phase preparation in fused silica capillaries. We believe that the detailed information presented in this technical note concerning the construction of an alternative high pressurization device, used to fill capillary columns via the sol-gel process, which has promising potential for applications involving capillary electrochromatography and liquid chromatography in nano scale, may be enlightening and motivating for groups interested in developing research activities within this theme.

A fast gas chromatography with a flame ionisation detector (GC-FID) method for the simultaneous analysis of methyl palmitate (C16:0), stearate (C18:0), oleate (C18:1), linoleate (C18:2) and linolenate (C18:3) in biodiesel samples was proposed. The analysis was conducted in a customised ionic-liquid stationary-phase capillary, SLB-IL 111, with a length of 14 m, an internal diameter of 0.10 mm, a film thickness of 0.08 µm and operated isothermally at 160 °C using hydrogen as the carrier gas at a rate of 50 cm s-1 in run time about 3 min. Once methyl myristate (C14:0) is present lower than 0.5% m/m in real samples it was used as an internal standard. The method was successful applied to monitoring basic and acidic catalysis transesterification reactions of vegetable oils such as soybean, canola, corn, sunflower and those used in frying process.

ABSTRACT A portable flow board system was developed in the present study with the aim to facilitate lab-scale experiments of controlled atmosphere (CA) with fruits and vegetables. This sturdy flow board combines ease fabrication, low cost and gas economy. Its functionality is provided by manifolds and gas mixers. Each gaseous component is supplied by a gas cylinder through a differential valve of adjusted pressure control, generally at 6 kPa, and forced through 13 standardized restrictors coupled to each manifold output. Controlled atmospheres are then formed with one, two or three gases in 13 gas mixers affixed to the flow board base, which are further conducted through flexible tubes to storage mini-chambers that can also be used to study metabolic consumption and production of gaseous components. The restrictors used in the flow gaseous components were manufactured from microhematocrit test-type capillary glass tubes following the hot forming method under continuous air flow. The portable flow board showed to be low cost and simple post-harvest equipment that allows preparing controlled atmospheres in open systems with stable composition and flow, in a manner similar to traditional flow boards with control of gas escape by barostats.
