Durante o ano agrícola de 1978/79, foi realizado um experimento a campo, na re gião da Depressão Central do Rio Grande do Sul, com o objetivo de conhecer o comportamento das cultivares de soja BR1, Bragg, Davis, IAS4, IAS5, Paraná e Planalto em relação ao metribuzin aplicado em pré-emergência nas doses de 0, 490 e 980 g/ha. Os efeitos dos tratamentos foram estimados através de avaliação visual de fitotoxicidade, contagem da população de plantas, determinações do peso seco da parte aérea e do número de grãos e obtenção do rendimento de grãos. Para a maioria das variáveis em estudo, constatou-se que ocorreram reduções proporcionais aos acréscimos das doses do herbicida. Os resultados da avaliação visual de fitotoxicidade mostraram que houve diferenças significativas entre os tratamentos de doses, tendo as cultivares Bragg e Davis demonstrado o menor efeito fito-tóxico, enquanto BR1 foi a que apresentou maior grau de injúria. Quanto ao rendimento de grãos alcançado pelas cultivares, foi constatado que Bragg comportou-se como altamente tolerante; BR1 e Davis como moderadamente tolerantes; IAS5 como intermediária e Paraná, Planalto e IAS4 como moderadamente suscetíveis.

Este projeto teve por objetivo avaliar a eficácia de herbicidas para controle de plantas aquáticas submersas das espécies Egeria densa e Egeria najas. O estudo foi realizado em três etapas consecutivas: em laboratório, em caixas d'água e numa represa de pequeno porte, sem fluxo de água. Na primeira etapa, avaliou-se a eficiência de 23 herbicidas utilizados em áreas agrícolas e disponíveis no mercado brasileiro. As elevadas concentrações em que os herbicidas foram efetivos inviabilizam programas de controle fundamentados em uma única aplicação. No experimento em caixas d'água, foram avaliados os efeitos de doses crescentes de fluridone, nas formulações líquida e granulada, no controle de Egeria densa e Egeria najas. Foram testadas as concentrações de 10, 20, 40, 80 e 150 ppb de fluridone na formulação líquida e 20, 40, 80 e 150 ppb na granulada. Após uma única aplicação, os resultados indicaram que o fluridone, nas concentrações de 80 e 150 ppb, em ambas as formulações, foi eficaz no controle dessas duas espécies. No experimento em represa de pequeno porte sem fluxo de água, avaliou-se o efeito da manutenção das concentrações de fluridone no controle de Egeria najas. Uma represa de 1.980 m² foi tratada com sete aplicações de fluridone, na formulação líquida, visando a manutenção das concentrações do ingrediente ativo na água entre 10 e 20 ppb. Os resultados indicaram que o controle proporcionado pelo herbicida foi superior a 99%. Não houve modificação significativa nas características relacionadas à qualidade da água.

A suscetibilidade diferencial de espécies de plantas daninhas a herbicidas é uma importante variável que deve ser considerada na escolha do herbicida e dose a serem aplicados. Assim, dois experimentos foram desenvolvidos com o objetivo de avaliar a suscetibilidade de três espécies de plantas daninhas da família Convolvulaceae (Ipomoea quamoclit, I. triloba e Merremia cissoides) aos herbicidas sulfentrazone e amicarbazone, por meio de curvas de dose-resposta. Os experimentos foram instalados em área com solo argiloso, onde foram aplicadas oito doses de cada herbicida, em pré-emergência. As doses utilizadas foram 8D, 4D, 2D, D, 1/2D, 1/4D, 1/8D e testemunha sem aplicação, sendo D a dose recomendada de sulfentrazone (600 g ha-1) ou de amicarbazone (980 g ha-1). As doses recomendadas de ambos os herbicidas foram eficazes para controlar as três plantas daninhas nas avaliações realizadas até os 60 dias após aplicação. Observou-se suscetibilidade diferencial entre as espécies estudadas, em que I. quamoclit foi a mais sensível. As ordens de suscetibilidade das espécies aos herbicidas foram: I. quamoclit > M. cissoides > I. triloba para o sulfentrazone; e I. quamoclit > I. triloba > M. cissoides para o amicarbazone. Os níveis de controle indicam a viabilidade de aplicação de sulfentrazone e amicarbazone em pré-emergência para controle dessas espécies de plantas daninhas da família Convolvulaceae.

Este trabalho objetivou estudar a seletividade da associação de óleo fúsel e herbicidas sobre cana-de-açúcar, cultivar IAC93-3046, e a eficácia de controle sobre as espécies Panicum maximum, Amaranthus deflexus, Ipomoea quamoclit, Brachiaria decumbens e Euphorbia heterophylla. O experimento foi instalado no Centro de Cana do Instituto Agronômico, localizado em Ribeirão Preto-SP, em delineamento de blocos casualizados, com 10 tratamentos e quatro repetições. As parcelas foram constituídas de seis linhas de cana-de-açúcar com 4 m de comprimento, espaçadas de 1,50 m (36 m²), sendo metade semeada com plantas daninhas, para avaliação de controle, e a outra metade mantida no limpo, para avaliação da seletividade. Os tratamentos foram constituídos por herbicidas e sua associação com o óleo fúsel, sendo diuron+hexazinone (1.170+330 g ha-1); diuron+hexazinone (1.170+330 g ha-1) + óleo fúsel (25,0 L ha-1); diuron+hexazinone (819+231 g ha-1) + óleo fúsel (25,0 L ha-1); metribuzin (1.920 g ha-1); metribuzin (1.920 g ha-1) + óleo fúsel (25,0 L ha-1); metribuzin (1.344 g ha-1) + óleo fúsel (25,0 L ha-1); amicarbazone (1.400 g ha-1); amicarbazone (1.400 g ha-1) + óleo fúsel (25,0 L ha-1); amicarbazone (980 g ha-1) + óleo fúsel (25,0 L ha-1) e ausência de herbicidas. A aplicação dos tratamentos foi realizada com equipamento costal pressurizado e volume de calda correspondente a 250 L ha-1, em pós-emergência da cultura (30 cm) e plantas daninhas (até 20 cm). Os herbicidas diuron+hexazinone, metribuzin e amicarbazone isolados foram eficazes no controle de todas as espécies, mas, em dose completa e 70% da dose associados com óleo fúsel, não apresentaram controle satisfatório apenas às espécies I. quamoclit e E. heterophylla. Esses mesmos tratamentos foram seletivos ao cultivar IACSP 93-3046, ao comparar com a testemunha as variáveis altura, estande, diâmetro e produção de colmos, assim como aos atributos qualitativos. Entretanto, diuron+hexazinone e amicarbazone em dose completa associados com óleo fúsel não foram seletivos à concentração de sólidos solúveis no caldo e na produção de colmos, respectivamente.

Objetivou-se neste estudo avaliar a tensão superficial estática, o pH e a produção da espuma de misturas em tanque de glyphosate + chlorimuron-ethyl, associadas ou não com adjuvantes. Dois trabalhos foram realizados, sendo a primeira etapa desenvolvida em delineamento inteiramente casualizado com seis tratamentos e 20 repetições, representados por soluções dos herbicidas glyphosate (540,0 g e.a. ha-1) e chlorimuron-ethyl (7,5 g ha-1), isolados e em mistura em tanque nas formulações: Polaris®; Roundup Ready®; Classic®; Polaris® + Classic®; Roundup Ready® + Classic® e testemunha (água). Na segunda etapa foram estudados em DIC 70 tratamentos e 20 repetições, em fatorial 2 x 5 x 7, constituído por duas misturas de glyphosate com chlorimuron-ethyl (Polaris® + Classic® e Roundup Ready ® + Classic®), cinco adjuvantes (Joint Oil®; Nimbus®; Assist®; Natur' Oil® e Agr' Óleo®) e sete doses dos adjuvantes (0,000; 0,031; 0,062; 0,125; 0,250; 0,500 e 1,000% de v/v). As soluções de Polaris® e Roundup Ready®; isoladas ou em misturas com Classic®; caracterizaram-se por tensões superficiais estáticas de 43,2 e 35,9 mN m-1 e pH médios em torno de 4,5 a 4,6, respectivamente. As misturas de Polaris®+Classic®; Roundup Ready®+Classic® e Classic® apresentaram a maior quantidade e persistência da espuma ao longo do tempo, quando comparadas às formulações de Polaris® e Roundup Ready®. Os adjuvantes promoveram redução da tensão superficial quando associados às misturas de Polaris® + Classic® e Roundup Ready® + Classic®; caracterizando-se em termos de eficiência pela ordem decrescente: Nimbus® (36,5% e 25,8%) < Joint Oil® (32,2% e 25,2%) < Agr' Oil® (21,2% e 13,4%) < Natur' Oil® (17,4% e 10,6%) < Assist® (8,5% e 10,6%). Os adjuvantes promoveram redução da quantidade e persistência da espuma das misturas de Polaris® + Classic® e Roundup Ready® + Classic®; com pequenas variações dos níveis médios de pH das soluções inferiores a 0,5.

The influence of afterload on the rate of force generation by the myocardium was investigated using two types of preparations: the in situ dog heart (dP/dt) and isolated papillary muscle of rats (dT/dt). Thirteen anesthetized, mechanically ventilated and thoracotomized dogs were submitted to pharmacological autonomic blockade (3.0 mg/kg oxprenolol plus 0.5 mg/kg atropine). A reservoir connected to the left atrium permitted the control of left ventricular end-diastolic pressure (LVEDP). A mechanical constriction of the descending thoracic aorta allowed to increase the systolic pressure in two steps of 20 mmHg (conditions H1 and H2) above control values (condition C). After arterial pressure elevations (systolic pressure C: 119 ± 8.1; H1: 142 ± 7.9; H2 166 ± 7.7 mmHg; P<0.01), there were no significant differences in heart rate (C: 125 ± 13.9; H1: 125 ± 13.5; H2: 123 ± 14.1 bpm; P>0.05) or LVEDP (C: 6.2 ± 2.48; H1: 6.3 ± 2.43; H2: 6.1 ± 2.51 mmHg; P>0.05). The values of dP/dt did not change after each elevation of arterial pressure (C: 3,068 ± 1,057; H1: 3,112 ± 996; H2: 3,086 ± 980 mmHg/s; P>0.05). In isolated rat papillary muscle, an afterload corresponding to 50% and 75% of the maximal developed tension did not alter the values of the maximum rate of tension development (100%: 78 ± 13; 75%: 80 ± 13; 50%: 79 ± 11 g mm-2 s-1, P>0.05). The results show that the rise in afterload per se does not cause changes in dP/dt or dT/dt

To determine the influence of residual ß-cell function on retinopathy and microalbuminuria we measured basal C-peptide in 50 type 1 diabetic outpatients aged 24.96 ± 7.14 years, with a duration of diabetes of 9.1 ± 6.2 years. Forty-three patients (86%) with low C-peptide (<0.74 ng/ml) had longer duration of diabetes than 7 patients (14%) with high C-peptide (³0.74 ng/ml) (9 (2-34) vs 3 (1-10) years, P = 0.01) and a tendency to high glycated hemoglobin (HBA1) (8.8 (6-17.9) vs 7.7 (6.9-8.7)%, P = 0.08). Nine patients (18%) had microalbuminuria (two out of three overnight urine samples with an albumin excretion rate (AER) ³20 and <200 µg/min) and 13 (26%) had background retinopathy. No association was found between low C-peptide, microalbuminuria and retinopathy and no difference in basal C-peptide was observed between microalbuminuric and normoalbuminuric patients (0.4 ± 0.5 vs 0.19 ± 0.22 ng/ml, P = 0.61) and between patients with or without retinopathy (0.4 ± 0.6 vs 0.2 ± 0.3 ng/ml, P = 0.43). Multiple regression analysis showed that duration of diabetes (r = 0.30, r2 = 0.09, P = 0.031) followed by HBA1 (r = 0.41, r2 = 0.17, P = 0.01) influenced basal C-peptide, and this duration of diabetes was the only variable affecting AER (r = 0.40, r2 = 0.16, P = 0.004). In our sample of type 1 diabetic patients residual ß-cell function was not associated with microalbuminuria or retinopathy.

In the present experimental study we assessed induced osteoarthritis data in rabbits, compared three diagnostic methods, i.e., radiography (XR), computed tomography (CT) and magnetic resonance imaging (MRI), and correlated the imaging findings with those obtained by macroscopic evaluation. Ten young female rabbits of the Norfolk breed were used. Seven rabbits had the right knee immobilized in extension for a period of 12 weeks (immobilized group), and three others did not have a limb immobilized and were maintained under the same conditions (control group). Alterations observed by XR, CT and MRI after the period of immobilization were osteophytes, osteochondral lesions, increase and decrease of joint space, all of them present both in the immobilized and non-immobilized contralateral limbs. However, a significantly higher score was obtained for the immobilized limbs (XT: P = 0.016, CT: P = 0.031, MRI: P = 0.0156). All imaging methods were able to detect osteoarthritis changes after the 12 weeks of immobilization. Macroscopic evaluation identified increased thickening of joint capsule, proliferative and connective tissue in the femoropatellar joint, and irregularities of articular cartilage, especially in immobilized knees. The differences among XR, CT and MRI were not statistically significant for the immobilized knees. However, MRI using a 0.5 Tesla scanner was statistically different from CT and XR for the non-immobilized contralateral knees. We conclude that the three methods detected osteoarthritis lesions in rabbit knees, but MRI was less sensitive than XR and CT in detecting lesions compatible with initial osteoarthritis. Since none of the techniques revealed all the lesions, it is important to use all methods to establish an accurate diagnosis.

The objective of the present study was to assess the incidence, risk factors and outcome of patients who develop acute renal failure (ARF) in intensive care units. In this prospective observational study, 221 patients with a 48-h minimum stay, 18-year-old minimum age and absence of overt acute or chronic renal failure were included. Exclusion criteria were organ donors and renal transplantation patients. ARF was defined as a creatinine level above 1.5 mg/dL. Statistics were performed using Pearsons' chi2 test, Student t-test, and Wilcoxon test. Multivariate analysis was run using all variables with P < 0.1 in the univariate analysis. ARF developed in 19.0% of the patients, with 76.19% resulting in death. Main risk factors (univariate analysis) were: higher intra-operative hydration and bleeding, higher death risk by APACHE II score, logist organ dysfunction system on the first day, mechanical ventilation, shock due to systemic inflammatory response syndrome (SIRS)/sepsis, noradrenaline use, and plasma creatinine and urea levels on admission. Heart rate on admission (OR = 1.023 (1.002-1.044)), male gender (OR = 4.275 (1.340-13642)), shock due to SIRS/sepsis (OR = 8.590 (2.710-27.229)), higher intra-operative hydration (OR = 1.002 (1.000-1004)), and plasma urea on admission (OR = 1.012 (0.980-1044)) remained significant (multivariate analysis). The mortality risk factors (univariate analysis) were shock due to SIRS/sepsis, mechanical ventilation, blood stream infection, potassium and bicarbonate levels. Only potassium levels remained significant (P = 0.037). In conclusion, ARF has a high incidence, morbidity and mortality when it occurs in intensive care unit. There is a very close association with hemodynamic status and multiple organ dysfunction.

Stretching has been widely used to increase the range of motion. We assessed the effects of a stretching program on muscle-tendon length, flexibility, torque, and activities of daily living of institutionalized older women. Inclusion/exclusion criteria were according to Mini-Mental State Examination (MMSE) (>13), Barthel Index (>13) and Lysholm Scoring Scale (>84). Seventeen 67 ± 9 year-old elderly women from a nursing home were divided into 2 groups at random: the control group (CG, N = 9) participated in enjoyable cultural activities; the stretching group (SG, N = 8) performed active stretching of hamstrings, 4 bouts of 1 min each. Both groups were supervised three times per week over a period of 8 weeks. Peak torque was assessed by an isokinetic method. Both groups were evaluated by a photogrammetric method to assess muscle-tendon length of uni- and biarticular hip flexors and hamstring flexibility. All measurements were analyzed before and after 8 weeks by two-way ANOVA with the level of significance set at 5%. Hamstring flexibility increased by 30% in the SG group compared to pre-training (76.5 ± 13.0° vs 59.5 ± 9.0°, P = 0.0002) and by 9.2% compared to the CG group (76.5 ± 13.0° vs 64.0 ± 12.0°, P = 0.0018). Muscle-tendon lengths of hip biarticular flexor muscles (124 ± 6.8° vs 118.3 ± 7.6°, 5.0 ± 7.0%, P = 0.031) and eccentric knee extensor peak torque were decreased in the CG group compared to pre-test values (-49.4 ± 16.8 vs -60.5 ± 18.9 Nm, -15.7 ± 20%, P = 0.048). The stretching program was sufficient to increase hamstring flexibility and a lack of stretching can cause reduction of muscle performance.

We investigated the contribution of the duration of overdistention (DOD) to rat bladder function and morphology and explored its possible molecular mechanisms. Bladder overdistention was induced in male Sprague-Dawley rats (200-250 g) by an infusion of saline. Forty rats were divided into 5 groups submitted to different DOD, i.e., 1, 2, 4, and 8 h, and control. Bladder function was evaluated by cystometry. Morphological changes were observed by light and transmission electron microscopy. Compared to control (44.567 ± 3.472 cmH2O), the maximum detrusor pressure of groups with 2-, 4- and 8-h DOD decreased significantly (means ± SEM): 32.774 ± 3.726, 31.321 ± 2.847, and 29.238 ± 3.724 cmH2O. With the increase of DOD, inflammatory infiltration and impairment of ultrastructure were more obvious in bladder tissue. Compared to control (1.90 ± 0.77), the apoptotic indexes of groups with 1-, 2-, 4-, and 8-h DOD increased significantly (6.47 ± 2.10, 10.66 ± 1.97, 13.91 ± 2.69, and 18.33 ± 3.28%). Compared to control (0.147 ± 0.031/0.234 ± 0.038 caspase 3/β-actin and Bax/Bcl-2 ratios), both caspase 3/β-actin and Bax/Bcl-2 ratios of 1-, 2-, 4-, and 8-h DOD increased significantly (0.292 ± 0.037/0.508 ± 0.174, 0.723 ± 0.173/1.745 ± 0.471, 1.104 ± 0.245/4.000 ± 1.048, and 1.345 ± 0.409/8.398 ± 3.332). DOD plays an important role in impairment of vesical function and structure. With DOD, pro-apoptotic factors increase and anti-apoptotic factors decrease, possibly contributing to the functional deterioration and morphological changes of the bladder.

Multidrug resistance (MDR) poses a serious impediment to the success of chemotherapy for laryngeal cancer. To identify microRNAs and mRNAs associated with MDR of human laryngeal cancer Hep-2 cells, we developed a multidrug-resistant human laryngeal cancer subline, designated Hep-2/v, by exposing Hep-2 cells to stepwise increasing concentrations of vincristine (0.02-0.96'µM). Microarray assays were performed to compare the microRNA and mRNA expression profiles of Hep-2 and Hep-2/v cells. Compared to Hep-2 cells, Hep-2/v cells were more resistant to chemotherapy drugs (∼45-fold more resistant to vincristine, 5.1-fold more resistant to cisplatin, and 5.6-fold more resistant to 5-fluorouracil) and had a longer doubling time (42.33±1.76 vs 28.75±1.12'h, P<0.05), higher percentage of cells in G0/G1 phase (80.98±0.52 vs69.14±0.89, P<0.05), increased efflux of rhodamine 123 (95.97±0.56 vs 12.40±0.44%, P<0.01), and up-regulated MDR1 expression. A total of 7 microRNAs and 605 mRNAs were differentially expressed between the two cell types. Of the differentially expressed mRNAs identified, regulator of G-protein signaling 10, high-temperature requirement protein A1, and nuclear protein 1 were found to be the putative targets of the differentially expressed microRNAs identified. These findings may open a new avenue for clarifying the mechanisms responsible for MDR in laryngeal cancer.
Altered mean platelet volume in patients with polymyositis and its association with disease severity

Polymyositis (PM) is an autoimmune disease characterized by chronic inflammation in skeletal muscle. Mean platelet volume (MPV), a marker in the assessment of systemic inflammation, is easily measured by automatic blood count equipment. However, to our knowledge, there are no data in the literature with respect to MPV levels in PM patients. Therefore, in this study we aimed to investigate MPV levels in patients with PM. This study included 92 newly diagnosed PM patients and 100 healthy individuals. MPV levels were found to be significantly lower compared with healthy controls (10.3±1.23 vs 11.5±0.74 fL, P<0.001). Interestingly, MPV was found to be positively correlated with manual muscle test (MMT) score and negatively correlated with erythrocyte sedimentation rate (ESR) in patients with PM (r=0.239, P=0.022; r=−0.268, P=0.010, respectively). In addition, MPV was significantly lower in active PM patients compared with inactive PM patients (9.9±1.39 vs 10.6±0.92 fL, P=0.010). MPV was independently associated with PM in multivariate regression analyses, when controlling for hemoglobin and ESR (OR=0.312, P=0.031, 95%CI=0.108 to 0.899). The ROC curve analysis for MPV in estimating PM patients resulted in an area under the curve of 0.800, with sensitivity of 75.0% and specificity of 67.4%. Our results suggest that MPV is inversely correlated with disease activity in patients with PM. MPV might be a useful tool for rapid assessment of disease severity in PM patients.

Propolis is a resinous substance collected by honeybees to seal honeycomb, which has been used in folk medicine due to its antimicrobial and antioxidant properties. In the present study, water and methanol were used to extract phenols and flavonoids from propolis collected in thirteen different areas in the Algarve region during the winter and spring. The ABTS•+, DPPH•, and O2•- scavenging capacity, and metal chelating activity were also evaluated in the propolis samples. Methanol was more effective than water in extracting total phenols (2.93-8.76 mg/mL) (0.93-2.81 mg/mL). Flavones and flavonols were also better extracted with methanol (1.28-2.76 mg/mL) than with water (0.031-0.019 mg/mL). The free radical scavenging activity, ABTS (IC50=0.006-0.036 mg/mL), DPPH (IC50=0.007-0.069 mg/mL) and superoxide (IC50=0.001-0.053 mg/mL), of the samples was also higher in methanolic extracts. The capacity for chelating metal ions was higher in aqueous extracts (41.11-82.35%) than in the methanolic ones (4.33-29.68%). Propolis from three locations of Algarve region were richer in phenols and had better capacity for scavenging free ABTS and DPPH radicals than the remaining samples. These places are part of a specific zone of Algarve known as Barrocal.

INTRODUÇÃO: A hepatite B pode evoluir para cirrose e hepatocarcinoma. Sua prevalência estimada é de 3,2% em pacientes em hemodiálise (HD). A vacina para hepatite B (HB), quando aplicada por via intramuscular (IM) em pacientes com insuficiência renal crônica fase V, frequentemente não induz produção adequada de anticorpos. A injeção intradérmica (ID) foi sugerida como sendo o método de inoculação mais eficiente. OBJETIVO: Comparar a resposta imune à injeção IM ou ID da vacina em indivíduos em HD. PACIENTES E MÉTODOS: Trinta e um pacientes incidentes em HD foram randomizados alternativamente para vacinação contra HB via IM ou ID. Dezesseis foram designados aleatoriamente para receber vacina IM (40 mg/dose) e 15 ID (4mg /dose). Os níveis de anticorpos de superfície do vírus da hepatite B, parâmetros hematimétricos, ureia sérica, e Kt/V foram avaliados mensalmente. Proteína-C reativa, paratormônio, ferritina, aminotransferases e albumina foram avaliados antes da inoculação inicial e seis meses após a mesma. RESULTADOS: Os níveis de uréia foram maiores no grupo ID (P(1) = 0,031); os níveis de ferritina foram mais elevados no IM (P(2) = 0,037). Houve tendência a aumento nos níveis de proteína C reativa no grupo ID. A avaliação do Comitê de Monitoramento de Segurança dos indivíduos expostos recomendou a suspensão do estudo já que a inoculação por via IM converteu 62,5% e a ID converteu apenas 13,3% dos pacientes expostos. CONCLUSÃO: Com a metodologia utilizada, os resultados da vacina contra HB aplicada por via ID foi inferior à inoculação IM. Tais resultados podem ser decorrentes das doses inoculadas ou de outros fatores, como inflamação.
