OBJETIVO: avaliar o padrão de diferenciação histológica em lesões de peritônio superficial e lesões de endometriose pélvica profunda (EPP), localizadas em ligamentos úteros-sacros, intestino (reto e sigmóide) e septo retovaginal. MÉTODOS: estudo prospectivo não randomizado, que incluiu 139 pacientes. Foram obtidas 234 biópsias (179 com EPP - Grupo Profundas - e 55 com endometriose superficial - Grupo Superficiais). Das 179 lesões de EPP (Grupo Profundas), 15 foram obtidas do septo retovaginal, 72 de nódulos do reto e sigmóide e 92 de ligamentos útero-sacrais. As biópsias foram classificadas em glandulares bem diferenciadas, glandulares indiferenciadas, glandulares mistas e estromal, com base em uma classificação morfológica específica. RESULTADOS: no Grupo Profundas, 33,5% das biópsias apresentaram padrão glandular indiferenciado e 46,9%, padrão glandular misto. No Grupo Superficiais, houve predominância do padrão glandular diferenciado (41,8%). Comparando especificamente as diferentes localizações das biópsias de lesão de EPP (Grupo Profundas), notou-se que o padrão glandular misto foi predominante nos nódulos intestinais (61,1%). CONCLUSÕES: foi possível concluir que há predomínio de endometriose glandular bem diferenciada na endometriose superficial, predomínio da doença mista e indiferenciada na EPP e, especificamente estudando a endometriose de reto e sigmóide, houve predomínio da endometriose glandular mista.

Para determinar as principais micoses e oomicoses que acometeram animais domésticos na área de abrangência do Laboratório de Patologia Veterinária (LPV) da Universidade Federal de Santa Maria (UFSM), foi realizado um estudo retrospectivo em 9.487 protocolos de necropsias e 20.199 exames histopatológicos (totalizando 29.686 casos), realizados no LPV-UFSM, entre janeiro de 1990 e dezembro de 2012. Do total de protocolos analisados, 230 apresentaram micoses ou pitiose (oomicose), sendo 179 casos (78%) de micoses e 51 casos (22%) de pitiose. Os protocolos foram revisados para determinar os principais achados referentes à epidemiologia, sinais clínicos e às alterações macroscópicas e microscópicas. Em dois casos (0,8%) não foi possível determinar o gênero ou o grupo do fungo observado. As principais doenças diagnosticadas, em ordem decrescente de prevalência, foram: pitiose, candidíase, aspergilose, zigomicose, dermatofitose, malasseziose, criptococose, megabacteriose e esporotricose. Outras doenças diagnosticadas numa única ocorrência cada foram histoplasmose e pneumocistose. Os equinos foram os mais acometidos pela pitiose e os animais de companhia (cães e gatos) foram os mais acometidos pelas micoses.

Abstract: Magellanic penguins (Spheniscus magellanicus) usually arrive in poor body conditions at Brazilian beaches during the winter. Hematology provides valuable information about clinical and immunity status of the animals. The aims of this study were to determine the hematologic, total plasma protein (TPP) and fibrinogen profiles of young and adult magellanic penguins in PROAMAR and CETAS-SC, relating these results with the state of health and survival possibility of the animals. In Paraná 14 animals were evaluated in pre and eight in post-rehabilitation and 29 animals were evaluated in Santa Catarina after rehabilitation. Before rehabilitation, all animals showed weakness. In hematological exams of these animals, we found that anemia was present in 83% of the penguins that died and 50% of those which survived. The heterophils/lymphocytes (H/L) ratio was 3.87±0.57 in animals that died, significantly higher than the average of 2.20±0.30 for animals that survived. These two parameters are useful to assess the survival possibility of animals to rehabilitation. The body condition score was positively correlated with hematocrit and TPP, and negatively correlated with H/L ratio. After rehabilitation, the values were similar to other animals of the family Spheniscidae, with averages ranging from 1.64 to 1.90x106 erythrocytes/μL; 43.38 to 48.80% of hematocrit; 12.45 to 13.52g/dL of hemoglobin; 8,684 to 14,011 leukocytes/μL; 4,767 to 8,041 heterophils/μL; 3,215 to 4,951 lymphocytes/μL; 95 to 655 eosinophils/μl; 179.8 to 277.9 monocytes/μL; 141 to 184.9 basophils/μL; and 1.26 to 1.74 of H/L ratio. These parameters can therefore be used as reference values and release parameters for young and adult Magellanic penguins in captivity on the rehabilitation centers.

A soja é uma das principais culturas agrícolas do Brasil, sendo a sua produtividade muito influenciada pela competição exercida pelas plantas daninhas. Foram realizados dois experimentos em casa de vegetação, em Jaboticabal, SP, objetivando determinar o acúmulo de massa seca, assim como a distribuição e o acúmulo de macronutrientes em plantas de soja, no período de outubro de 2000 a fevereiro de 2001, e de Solanum americanum, no período de janeiro a maio de 1995. As plantas cresceram em vasos com capacidade de 7 litros, preenchidos com areia de rio lavada e peneirada; elas foram irrigadas diariamente com solução nutritiva. Os tratamentos foram representados pelas épocas de amostragem, realizada a intervalos de 14 dias, iniciando-se 21 dias após a emergência (DAE), até 161 DAE para S. americanum e 119 DAE para soja cv. BR-16 (precoce). O ponto de máximo acúmulo teórico de massa seca deu-se aos 104 DAE para a soja (35,0 g por planta) e 143 DAE para S. americanum (179,62 g por planta). Da emergência até 49 e 63 DAE, as folhas apresentaram maior participação no acúmulo de massa seca para soja e S. americanum, respectivamente. Após esses períodos, verificou-se, em ambas as espécies, inversão na representatividade das folhas por caules para a espécie daninha e por caules e, posteriormente, por estruturas reprodutivas, para a cultura. A taxa de absorção diária dos macronutrientes atingiu maiores valores entre 69 e 87 DAE para a soja e entre 105 e 119 DAE para a planta daninha. Considerando a média dos valores de pontos de inflexão observados para a cultura da soja, tem-se que aos 75 DAE uma planta de soja acumula teoricamente 23,9 g de massa seca, 564,4 mg de N, 7,1 mg de P, 490,8 mg de K, 487,0 mg de Ca, 156,6 mg de Mg e 36,0 mg de S. Para o mesmo período, uma planta de S. americanum acumula teoricamente 33,75 g de massa seca, 875,96 mg de N, 88,46 mg de P, 983,54 mg de K, 647,60 mg de Ca, 100,93 mg de Mg e 42,15 mg de S.

Realizou-se o levantamento florístico de uma área remanescente de cerrado sensu stricto no Município de Botucatu, região centro-oeste do Estado de São Paulo, Sudeste do Brasil. Foram registrados 179 táxons de angiospermas (177 espécies) distribuídas em 122 gêneros e 49 famílias e cinco espécies de pteridófitas pertencentes a quatro gêneros e duas famílias. O componente lenhoso foi predominante representando mais de 60% das espécies registradas. A proporção das espécies arbóreas para as não arbóreas foi de 1:3. As famílias com maior riqueza foram: Asteraceae (27 espécies), Fabaceae (22), Myrtaceae (11). A comparação entre seis áreas de cerrado revelou grande heterogeneidade florística uma vez que os índices de similaridade produziram valores baixos, mesmo entre áreas próximas.

The submucous plexus of the normal small and large intestine of Calomys callosus was studied by NADH and AChE histochemical techniques and by transmission and scanning electron microscopy. The plexus contains (mean ± SD) 7,488 ± 293 neurons/cm2 in the duodenum, 5,611 ± 836 in the jejunum, 2,741 ± 360 in the ileum, 3,067 ± 179 in the cecum, and 3,817 ± 256 in the proximal colon. No ganglia or nerve cell bodies were seen in the esophagus, stomach, distal colon or rectum. The neurons are pear-shaped with a round or oval nucleus and the neuronal cell profile areas were larger in the large intestine than in the small intestine. Most of the neurons display intense AChE activity in the cytoplasm. AChE-positive nerve fibers are present in a primary meshwork of large nerve bundles and in a secondary meshwork of finer nerve bundles. At the ultrastructural level, the ganglia are irregular in shape and covered with fibroblast-like cells. The nucleoplasm of the neurons is finely granular with a few condensations of chromatin attached to the nuclear envelope. In the neuropil numerous varicosities filled with vesicles of different size and electron densities are seen. The pre- and post-synaptic membrane thickenings are asymmetric. Characteristic glial cells with oval nuclei and few organelles are numerous. These data provide a detailed description of this submucosal meshwork.

We studied the development of the insulin secretion mechanism in the pancreas of fetal (19- and 21-day-old), neonatal (3-day-old), and adult (90-day-old) rats in response to stimulation with 8.3 or 16.7 mM glucose, 30 mM K+, 5 mM theophylline (Theo) and 200 µM carbamylcholine (Cch). No effect of glucose or high K+ was observed on the pancreas from 19-day-old fetuses, whereas Theo and Cch significantly increased insulin secretion at this age (82 and 127% above basal levels, respectively). High K+ also failed to alter the insulin secretion in the pancreas from 21-day-old fetuses, whereas 8.3 mM and 16.7 mM glucose significantly stimulated insulin release by 41 and 54% above basal levels, respectively. Similar results were obtained with Theo and Cch. A more marked effect of glucose on insulin secretion was observed in the pancreas of 3-day-old rats, reaching 84 and 179% above basal levels with 8.3 mM and 16.7 mM glucose, respectively. At this age, both Theo and Cch increased insulin secretion to close to two-times basal levels. In islets from adult rats, 8.3 mM and 16.7 mM glucose, Theo, and Cch increased the insulin release by 104, 193, 318 and 396% above basal levels, respectively. These data indicate that pancreatic B-cells from 19-day-old fetuses were already sensitive to stimuli that use either cAMP or IP3 and DAG as second messengers, but insensitive to stimuli such as glucose and high K+ that induce membrane depolarization. The greater effect of glucose on insulin secretion during the neonatal period indicates that this period is crucial for the maturation of the glucose-sensing mechanism in B-cells.

Systemic metabolic acidosis is known to cause a decrease in salt and water reabsorption by the kidney. We have used renal lithium clearance to investigate the effect of chronic, NH4Cl-induced metabolic acidosis on the renal handling of Na+ in male Wistar-Hannover rats (200-250 g). Chronic acidosis (pH 7.16 ± 0.13) caused a sustained increase in renal fractional Na+ excretion (267.9 ± 36.4%), accompanied by an increase in fractional proximal (113.3 ± 3.6%) and post-proximal (179.7 ± 20.2%) Na+ and urinary K+ (163.4 ± 5.6%) excretion when compared to control and pair-fed rats. These differences occurred in spite of an unchanged creatinine clearance and Na+ filtered load. A lower final body weight was observed in the acidotic (232 ± 4.6 g) and pair-fed (225 ± 3.6 g) rats compared to the controls (258 ± 3.7 g). In contrast, there was a significant increase in the kidney weights of acidotic rats (1.73 ± 0.05 g) compared to the other experimental groups (control, 1.46 ± 0.05 g; pair-fed, 1.4 ± 0.05 g). We suggest that altered renal Na+ and K+ handling in acidotic rats may result from a reciprocal relationship between the level of metabolism in renal tubules and ion transport.

The molecular basis for RHD pseudogene or RHDpsi is a 37-bp insertion in exon 4 of RHD. This insertion, found in two-thirds of D-negative Africans, appears to introduce a stop codon at position 210. The hybrid RHD-CE-Ds, where the 3' end of exon 3 and exons 4 to 8 are derived from RHCE, is associated with the VS+V- phenotype, and leads to a D-negative phenotype in people of African origin. We determined whether Brazilian blood donors of heterogeneous ethnic origin had RHDpsi and RHD-CE-Ds. DNA from 206 blood donors were tested for RHDpsi by a multiplex PCR that detects RHD, RHDpsi and the C and c alleles of RHCE. The RHD genotype was determined by comparison of size of amplified products associated with the RHD gene in both intron 4 and exon 10/3'-UTR. VS was determined by amplification of exon 5 of RHCE, and sequencing of PCR products was used to analyze C733G (Leu245Val). Twenty-two (11%) of the 206 D-negative Brazilians studied had the RHDpsi, 5 (2%) had the RHD-CE-Ds hybrid gene associated with the VS+V- phenotype, and 179 (87%) entirely lacked RHD. As expected, RHD was deleted in all the 50 individuals of Caucasian descent. Among the 156 individuals of African descent, 22 (14%) had inactive RHD and 3% had the RHD-CE-Ds hybrid gene. These data confirm that the inclusion of two different multiplex PCR for RHD is essential to test the D-negative Brazilian population in order to avoid false-positive typing of polytransfused patients and fetuses.

Abnormal riboflavin status in the absence of a dietary deficiency was detected in 31 consecutive outpatients with Parkinson's disease (PD), while the classical determinants of homocysteine levels (B6, folic acid, and B12) were usually within normal limits. In contrast, only 3 of 10 consecutive outpatients with dementia without previous stroke had abnormal riboflavin status. The data for 12 patients who did not complete 6 months of therapy or did not comply with the proposed treatment paradigm were excluded from analysis. Nineteen PD patients (8 males and 11 females, mean age ± SD = 66.2 ± 8.6 years; 3, 3, 2, 5, and 6 patients in Hoehn and Yahr stages I to V) received riboflavin orally (30 mg every 8 h) plus their usual symptomatic medications and all red meat was eliminated from their diet. After 1 month the riboflavin status of the patients was normalized from 106.4 ± 34.9 to 179.2 ± 23 ng/ml (N = 9). Motor capacity was measured by a modification of the scoring system of Hoehn and Yahr, which reports motor capacity as percent. All 19 patients who completed 6 months of treatment showed improved motor capacity during the first three months and most reached a plateau while 5/19 continued to improve in the 3- to 6-month interval. Their average motor capacity increased from 44 to 71% after 6 months, increasing significantly every month compared with their own pretreatment status (P < 0.001, Wilcoxon signed rank test). Discontinuation of riboflavin for several days did not impair motor capacity and yellowish urine was the only side effect observed. The data show that the proposed treatment improves the clinical condition of PD patients. Riboflavin-sensitive mechanisms involved in PD may include glutathione depletion, cumulative mitochondrial DNA mutations, disturbed mitochondrial protein complexes, and abnormal iron metabolism. More studies are required to identify the mechanisms involved.

Aging is accompanied by a decrease in several physiological functions that make older individuals less responsive to environmental challenges. In the present study, we analyzed the immune response of female BALB/c mice (N = 6) of different ages (from 2 to 96 weeks) and identified significant age-related alterations. Immunization with hapten-protein (trinitrophenyl-bovine serum albumin) conjugates resulted in lower antibody levels in the primary and secondary responses of old mice (72 weeks old). Moreover, young mice (2, 16, and 32 weeks old) maintained specific antibodies in their sera for longer periods after primary immunization than did old mice. However, a secondary challenge efficiently induced memory in old mice, as shown by the increased antibody levels in their sera. The number of CD4+ and CD8+ T cells in the spleen increased until 8 weeks of age but there was no change in the CD4+/CD8+ ratio with aging. Splenic T cells from old mice that had or had not been immunized were less responsive to concanavalin-A and showed reduced cytokine production compared to young mice (IL-2: 57-127 vs 367-1104 pg/mL, IFN-g: 2344-12,836 vs 752-23,106 pg/mL and IL-10: 393-2172 vs 105-2869 pg/mL in old and young mice, respectively). These data suggest that there are significant changes in the organization of the immune system throughout life. However, the relevance of these alterations for the functioning of the immune system is unknown.

The cardiovascular electrophysiologic basis for the action of pyridostigmine, an acetylcholinesterase inhibitor, has not been investigated. The objective of the present study was to determine the cardiac electrophysiologic effects of a single dose of pyridostigmine bromide in an open-label, quasi-experimental protocol. Fifteen patients who had been indicated for diagnostic cardiac electrophysiologic study underwent two studies just before and 90-120 min after the oral administration of pyridostigmine (45 mg). Pyridostigmine was well tolerated by all patients. Wenckebach nodal anterograde atrioventricular point and basic cycle were not altered by pyridostigmine. Sinus recovery time (ms) was shorter during a 500-ms cycle stimulation (pre: 326 ± 45 vs post: 235 ± 47; P = 0.003) but not during 400-ms (pre: 275 ± 28 vs post: 248 ± 32; P = 0.490) or 600-ms (pre: 252 ± 42 vs post: 179 ± 26; P = 0.080) cycle stimulation. Pyridostigmine increased the ventricular refractory period (ms) during the 400-ms cycle stimulation (pre: 238 ± 7 vs post: 245 ± 9; P = 0.028) but not during the 500-ms (pre: 248 ± 7 vs post: 253 ± 9; P = 0.150) or 600-ms (pre: 254 ± 8 vs post: 259 ± 8; P = 0.255) cycle stimulation. We conclude that pyridostigmine did not produce conduction disturbances and, indeed, increased the ventricular refractory period at higher heart rates. While the effect explains previous results showing the anti-arrhythmic action of pyridostigmine, the clinical impact on long-term outcomes requires further investigation.

Obesity is a multifactorial disorder often associated with many important diseases such as diabetes, hypertension and other metabolic syndrome conditions. Argyrophil cells represent almost the total population of endocrine cells of the human gastric mucosa and some reports have described changes of specific types of these cells in patients with obesity and metabolic syndrome. The present study was designed to evaluate the global population of argyrophil cells of the gastric mucosa of morbidly obese and dyspeptic non-obese patients. Gastric biopsies of antropyloric and oxyntic mucosa were obtained from 50 morbidly obese patients (BMI >40) and 50 non-obese patients (17 dyspeptic overweight and 33 lean individuals) and processed for histology and Grimelius staining for argyrophil cell demonstration. Argyrophil cell density in the oxyntic mucosa of morbidly obese patients was higher in female (238.68 ± 83.71 cells/mm2) than in male patients (179.31 ± 85.96 cells/mm2) and also higher in female (214.20 ± 50.38 cells/mm2) than in male (141.90 ± 61.22 cells/mm2) morbidly obese patients with metabolic syndrome (P = 0.01 and P = 0.02, respectively). In antropyloric mucosa, the main difference in argyrophil cell density was observed between female morbidly obese patients with (167.00 ± 69.30 cells/mm2) and without (234.00 ± 69.54 cells/mm2) metabolic syndrome (P = 0.001). In conclusion, the present results show that the number of gastric argyrophil cells could be under gender influence in patients with morbid obesity. In addition, gastric argyrophil cells seem to behave differently among female morbidly obese patients with and without metabolic syndrome.

Durante um ano, trabalhou-se com o leite de conjunto produzido por três grupos distintos de cabras "Saanen", perfazendo um total variável entre 44 e 56 animais em lactação e um total de 179 amostras analisadas, nas quais determinou-se a composição química, o perfil nitrogenado e as principais características físico-químicas. Para a composição geral, os valores determinados foram: 3,27% para proteína total, 3,74% para gordura, 4,35% para lactose, 0,74% para cinzas e 88,49% para água. O perfil nitrogenado distribuiu-se em: 3,27% para proteína bruta (PB), 2,97% para proteína verdadeira (TP), 2,43% para a fração caseínas (C), e 0,84% para as proteínas do soro (PS), incluindo 0,30% para a fração nitrogenada não protéica (NNP). As características determinadas foram: 1,0324 para a densidade a 15ºC, 6,65 para o pH a 25ºC, 16,11ºD para a acidez, 0,172 g% para o teor de cloretos, -0,574ºH para o ponto de congelamento, 11,51% para sólidos totais (ST), 7,77% para sólidos desengordurados (SD), 12,45% para o extrato seco total (EST) calculado e 8,90% para o extrato seco desengordurado (ESD), também calculado.

O trabalho objetivou estudar o efeito do período de maturação de grãos nas propriedades físicas e reológicas da farinha de trigo, avaliado através das determinações de peso do hectolitro, peso de mil grãos, grau de extração, proteínas, cinzas, umidade, cor, número de queda, alveografia, farinografia e amido danificado. Amostras dos genótipos BRS 179, Rubi, Fundacep 30 e Fundacep 51, safra 2006/2007 foram armazenadas em sacos de papel Kraft com 10 kg de trigo, em ambiente com temperatura de 22 °C e umidade relativa do ar de 65-75%. Cada tratamento foi composto por um genótipo e as avaliações realizadas nos tempos 0, 15, 30, 60 e 90 dias de maturação. O experimento foi conduzido em delineamento inteiramente casualizado em arranjo fatorial 4 x 5. Os resultados foram analisados pelo emprego da análise de variância (Anova) e nos modelos significativos pelo teste F, e a comparação de médias pelo teste de Tukey a 5% de probabilidade. Com o aumento do período de maturação do trigo a farinha apresentou melhores propriedades físicas e reológicas avaliadas pelas alterações de número de queda, cor, alveografia e farinografia. Os resultados de atividade enzimática, luminosidade, intensidade de amarelo e força geral do glúten se mostraram efetivos na avaliação de propriedades físicas e reológicas, enquanto as avaliações de amido danificado e estabilidade não foram indicadores adequados. O trigo não deve ser comercializado após a colheita, pois apresenta melhoria na classificação comercial, sendo indicado período de maturação de 60 dias. Os genótipos de trigo BRS 179 e Fundacep 51 passaram das classes brando para pão, enquanto o Rubi de pão para melhorador, indicando maior valor tecnológico e comercial com o aumento do período de maturação dos grãos de trigo.
