El objetivo de este trabajo fue estudiar las alteraciones producidas por el deterioro en semillas de soja (Glycine max L. Merrill) utilizando diferentes pruebas de vigor. Se utilizaron semillas de soja de diferentes cultivares en 1997 y 1998, las cuales fueron sometidas a la prueba de germinación y diferentes pruebas de vigor: envejecimiento acelerado; conductividad eléctrica; deterioro controlado y tetrazolio. El deterioro controlado en semillas con 22% de humedad mostró ser un mejor indicador de la disminución del vigor de la semilla de soja que el envejecimiento acelerado. La conductividad eléctrica en semillas individuales podría ser considerada una buena prueba de vigor para detectar alteraciones producidas en las membranas citoplasmáticas en un estadio temprano del deterioro de las semillas. La clasificación de la viabilidad y el vigor a través de la prueba de tetrazolio está dada por el lugar donde se produce el daño y no por el tipo de daño que sufre la semilla.

The objective of this work was to evaluate the width and length incidence in a single seed fraction of oat [Avena sativa (L.)] cv. Cristal. The seeds were selected by a mechanical divider and by hand, and their correspondence to radiographic images in seeds with glumes and their caryopses. The width and length of the seeds with glumes and their caryopses were measured with electronic calliper, and their weight, with precision balance. Radiographic images of seeds with glumes were taken with an X-ray experimental equipment. The analyst selected seeds with glumes by the width and by the length previously determined and so with more weight, than that obtained by hand selection was slightly narrower, larger and lighter. The presence of the glumes masked the caryopses real dimensions (width and length), and conduced the analyst to select seeds that differed more by the width than by the length. The radiographic images showed the presence, or not, of caryopses inside the seed and its real dimensions. The mechanical partition method for seeds showed to be more efficient because the analyst subjectivity was not considered when the selection upon its dimensions was done. The X-ray analysis was a useful tool that complements the pure seed fraction selection as another factor of seed quality.

The objective of this work was to evaluate the effects of selection for high protein on seed physiological quality and grain yield of soybean. Four populations of BC1F4 and four of F4, each from a cross between a commercial variety and a line bearing high protein seeds, were used. The high protein content selection has a tendency to affect negatively the seed physiological quality. Estimates of correlation coefficients between protein content and grain yield were mostly negative but varied among populations. It is possible to obtain lines with high protein content, keeping the grain yield and the seed physiological quality of their respective recurrent progenitors.

The aphid Aphis gossypii Glover (Hemiptera: Aphididae), a harmful pest on cotton, causes direct damage, reducing plant vigor, and indirect damage by honeydew secretion and transmission of several viruses. Due to the problem of pesticide resistance, alternative techniques for chemical control, such as the use of natural insecticides, have been object of research. The effects of aqueous extracts of neem (Azadirachta indica A. Juss) seed powder on the development, survival and fecundity of A. gossypii were evaluated. Treatments consisted of neem seed powder in the concentrations of 23.8, 122.0, 410.0 and 1,410.0 mg/100 mL of distilled water. Mortality rate during the nymphal development for aphids maintained on cotton leaf discs treated with the two highest concentrations were, respectively, 60.0% and 100.0%. With the exception of the highest concentration (1,410.0 mg/100 mL), neem concentrations did not extend the aphids' development period. The net reproductive rate (R0) was of 35.0 nymphs/female for control aphids and of 0.0 nymph/female when the group of females was exposed to neem seed powder at 1,410.0 mg/100 mL since birth. The aqueous extract of neem seeds is efficient against the aphid A. gossypii, causing nymph mortality and reducing their survival period and fecundity.

O objetivo deste trabalho foi avaliar o efeito do condicionamento e dos estresses salino e térmico sobre o vigor e a viabilidade de sementes de paineira (Chorisia speciosa St.-Hil) armazenadas em geladeira, em embalagens impermeáveis, durante três anos. Sementes com e sem punção do tegumento foram condicionadas em água destilada e em soluções de KNO3 a 0,1 M e 0,2 M, durante 24 horas, a 27ºC. O estresse salino foi simulado com soluções de NaCl em potenciais osmóticos variando de 0,0 a -1,2 MPa, a 27ºC. O estresse térmico de diferentes intensidades foi aplicado em sementes secas, que germinaram a 27ºC. Houve diminuição da viabilidade e do vigor das sementes com o aumento da intensidade dos estresses salino e térmico. Nas sementes condicionadas e intactas, o limite máximo de tolerância à salinidade não foi reduzido, porém houve diminuição da tolerância ao estresse térmico, em sementes condicionadas com ou sem punção. O teste de condutividade elétrica indicou as sementes condicionadas em água e KNO3 0,1 M como as de maior vigor. Sementes condicionadas em água apresentaram maior velocidade de germinação sob estresses salino e térmico. O condicionamento substituiu a punção do tegumento, que não é recomendada para sementes armazenadas.

O objetivo deste trabalho foi avaliar o vigor de três clones de umezeiro (Prunus mume Sieb. et Zucc.) e do pessegueiro 'Okinawa' [Prunus persica (L.) Batsch], propagados por estacas herbáceas, em condições de campo. O experimento foi conduzido em blocos ao acaso, com quatro tratamentos (genótipos) e cinco repetições. As plantas foram espaçadas 0,5 m entre si. O pessegueiro 'Okinawa' apresentou maior diâmetro do tronco, em relação aos clones de umezeiro. Na análise conjunta das variáveis, o Clone 10 revela-se o menos vigoroso, indicando a possibilidade de sucesso como porta-enxerto ananizante para pessegueiro.

This work aimed to analyse Eucalyptus grandis W.Hill ex Maiden seed behaviour, under controlled deterioration, and to estimate viability equation constants for the species. Seeds were harvested in the growing season of 1999, and the moisture contents were adjusted from 11.3% to a range between 1.2 and 18.1% at 25ºC. The subsamples were sealed into laminate aluminium-foil packets, for storage in incubators at 40, 50 and 65±0.5ºC. The seeds presented orthodox performance, in which the constants for predicting seed longevity of E. grandis were K E = 9.661, C W = 6.467, C H = 0.03498 and C Q = 0.0002330. The usual and inverse relationship between water content and seed longevity was also observed. The lowest moisture content limit for application of the viability equation at 65ºC was 4.9%, estimated under hygroscopic equilibrium with 23% of relative humidity in the storage environment.

Mungbean cultivar MGS Esmeralda was developed by Asian Vegetable Research and Development Center (Shanhua, Taiwan), as a result of crossing between the lines VC 1973A and VC 2768A. In ten trials conducted in the State of Minas Gerais, Brazil, it produced 13.5% more grains than 'Ouro Verde MG-2' (control cultivar), and its highest yield was 2,550 kg ha-1. The cultivar MGS Esmeralda is more susceptible to lodging, and its pods mature more uniformly than Ouro Verde MG-2 pods. One hundred-seed mass of 'MGS Esmeralda' ranged between 5.5 and 6.8 g. Both cultivars are susceptible to powdery mildew and cercospora leaf spot.

The objective of this work was to determine the effects of seed priming and sulfur application on cell membrane characteristics, seedling emergence, chlorophyll content and grain yield of soybean (Glycine max) in saline soil. A complete-block design in 4x3 factorial arrangement with three replicates was used to test four types of seed priming (water, auxin, gibberellin and non-priming) and three levels of sulfate availability (0, 70 and 140 kg ha-1 K2SO4). The soil had a silty loam texture with an electrical conductivity of 3.61 ds m-1, a pH of 8.2 and a saturation percentage of about 46%. Seed priming had significant effects on mean emergence rate (MER), emergence percentage, relative water content (RWC) of leaves, relative chlorophyll content, time of maturity, shoot length and grain yield. The highest values for these variables were observed in the priming treatments, except for the time of maturity. Sulfur application had significant effects on MER, shoot length, RWC, membrane injury index and grain yield. Priming treatments provide greater emergence rates and grain yields and interact sinergicaly with sulfur rates.

The objective of this work was to quantify the accumulation of the major seed storage protein subunits, β-conglycinin and glycinin, and how they influence yield and protein and oil contents in high-protein soybean genotypes. The relative accumulation of subunits was calculated by scanning SDS-PAGE gels using densitometry. The protein content of the tested genotypes was higher than control cultivar in the same maturity group. Several genotypes with improved protein content and with unchanged yield or oil content were developed as a result of new breeding initiatives. This research confirmed that high-protein cultivars accumulate higher amounts of glycinin and β-conglycinin. Genotypes KO5427, KO5428, and KO5429, which accumulated lower quantities of all subunits of glycinin and β-conglycinin, were the only exceptions. Attention should be given to genotypes KO5314 and KO5317, which accumulated significantly higher amounts of both subunits of glycinin, and to genotypes KO5425, KO5319, KO539 and KO536, which accumulated significantly higher amounts of β-conglycinin subunits. These findings suggest that some of the tested genotypes could be beneficial in different breeding programs aimed at the production of agronomically viable plants, yielding high-protein seed with specific composition of storage proteins for specific food applications.

The objectives of this work were to determine whether and at which seed physiological maturity stage the diquat desiccant affects the tall oat-grass (Arrhenatherum elatius) seed quality and yield, as well as the proper storage period between harvest and sowing. Diquat desiccation was evaluated in applications during milk or dough seed maturation stages. Seeds conventionally produced and stored under traditional storage conditions were used for the analyses. Seed samples were drawn every 30th day after harvest (DAH). After the 240th DAH, samples were drawn every 90th day up the to 690th DAH. The highest yield were obtained by desiccation applied at the beginning of the seed dough stage, with a satisfactory seed quality. Both final germination and seedling growth parameters achieved their maximum values between 180th and 240th DAH. Oat-grass seeds preserved satisfactory level of final germination (75%) up to 420th DAH. The application of diquat desiccant at the beginning of seed dough maturity stage can be a good solution for seed production of tall oat-grass. Early spring is the best sowing period for freshly harvested seeds of tall oat-grass regarding germination and seedling growth.

The objective of this study was to assess genotype by environment interaction for seed yield per plant in rapeseed cultivars grown in Northern Serbia by the AMMI (additive main effects and multiplicative interaction) model. The study comprised 19 rapeseed genotypes, analyzed in seven years through field trials arranged in a randomized complete block design, with three replicates. Seed yield per plant of the tested cultivars varied from 1.82 to 19.47 g throughout the seven seasons, with an average of 7.41 g. In the variance analysis, 72.49% of the total yield variation was explained by environment, 7.71% by differences between genotypes, and 19.09% by genotype by environment interaction. On the biplot, cultivars with high yield genetic potential had positive correlation with the seasons with optimal growing conditions, while the cultivars with lower yield potential were correlated to the years with unfavorable conditions. Seed yield per plant is highly influenced by environmental factors, which indicates the adaptability of specific genotypes to specific seasons.

The objective of this work was to evaluate isolates of Trichoderma harzianum regarding biocontrol of common bean seed-borne pathogens, plant growth promotion, and rhizosphere competence. Five isolates of T. harzianum were evaluated and compared with commercial isolate (Ecotrich), Carboxin+Thiram, and an absolute control. Bean seeds of the cultivar Jalo Precoce, contaminated with Aspergillus, Cladosporium, and Sclerotinia sclerotiorum, were microbiolized with antagonists, and seed health tests were carried out. Isolates were evaluated on autoclaved substrate and in field conditions. Ten days after sowing (DAS), plant length was measured. To test rhizosphere competence, isolates were applied in boxes containing autoclaved washed sand, and root colonization was evaluated at 10 DAS, using five plants per box. The most effective isolates in the seed health tests were: CEN287 and CEN289 to control Aspergillus; the commercial isolate to control Cladosporium; and CEN287 and CEN316 to control S. sclerotiorum. Isolates CEN289 and CEN290 promoted bean growth in greenhouse and field. Seed treatment with T. harzianum reduces the incidence of Aspergillus, Cladosporium, and S. sclerotiorum in 'Jalo Precoce' common bean seeds.

O objetivo deste trabalho foi avaliar metodologias para realização do teste de tetrazólio, na avaliação da viabilidade e do vigor de sementes de girassol, e estabelecer classes de qualidade para interpretação do teste. Foram avaliadas diferentes formas de extração do pericarpo e do tegumento das sementes, além do pré-condicionamento em água por 16 e 18 horas (25°C) e da coloração em solução de tetrazólio (30ºC), nas concentrações de 0,1 e 0,5%, por 2, 3 e 4 horas; e de 1,0%, por 1, 2 e 3 horas. Após a definição da metodologia mais adequada para realização do teste, classes de vigor foram estabelecidas de acordo com a emergência das plântulas no campo. Foram identificadas cinco classes, as quais apresentaram correlação perfeita com a emergência. Para avaliar a viabilidade das sementes de girassol, o pericarpo e o tegumento devem ser retirados após corte longitudinal com até 1/3 do comprimento da semente, o pré-condicionamento deve ser feito pela imersão em água por 16 horas, e a coloração deve ser realizada em solução de tetrazólio a 0,1%, durante 3 horas.

The objective of this work was to determine the inheritance mode of seed coat color in sesame. Two crosses and their reciprocals were performed: UCLA37 x UCV3 and UCLA90 x UCV3, of which UCLA37 and UCLA90 are white seed, and UCV3 is brown seed. Results of reciprocal crosses within each cross were identical: F1 seeds had the same phenotype as the maternal parent, and F2 resulted in the phenotype brown color. These results are consistent only with the model in which the maternal effect is the responsible for this trait. This model was validated by recording the seed coat color of 100 F2 plants (F3 seeds) from each cross with its reciprocal, in which the 3:1 expected ratio for plants producing brown and white seeds was tested with the chi-square test. Sesame seed color is determined by the maternal genotype. Proposed names for the alleles participating in sesame seed coat color are: Sc1, for brown color; and Sc2, for white color; Sc1 is dominant over Sc2.
