The ID-Chagas test is a particle gel immunoassay (PaGIA). Red coloured particles are sensitised with three different synthetic peptides representing antigen sequences of Trypanosoma cruzi: Ag2, TcD and TcE. When these particles are mixed with serum containing specific antibodies, they agglutinate. The reaction mixture is centrifuged through a gel filtration matrix allowing free agglutinated particles to remain trapped on the top or distributed within the gel. The result can be read visually. In order to investigate the ability of the ID-PaGIA to discriminate negative and positive sera, 111 negative and 119 positive, collected in four different Brazilian institutions, were tested by each of the participants. All sera were previously classified as positive or negative according to results obtained with three conventional tests (indirect immunofluorescence, indirect hemaglutination, and enzime linked immunosorbent assay). Sensitivity rates of ID-PaGIA varied from 95.7% to 97.4% with mean sensitivity of 96.8% and specificity rates varied from 93.8 to 98.8% with mean specificity of 94.6%. The overall Kappa test was 0.94. The assay presents as advantages the simplicity of operation and the reaction time of 20 min. In this study, ID-PaGIA showed to be highly sensitive and specific.

The ultrastructure is described of the meronts, microgamonts and young oocyst stages of Isospora hemidactyli of the gecko Hemidactylus mabouia from Belém, PA, north Brazil. The endogenous stages all develop in the nucleus of the gut epithelial cells. The nucleus remains intact up to the latest stages of the parasite's development, but degenerates by the time the oocyst appears. Merogonic division appears to be asynchronous, and some of the differentiated merozoites contained more than one nucleus. Microgamonts conform in structure with those of other eimeriids. Some of the type 2 wall-forming bodies disintegrate into smaller globules and ground substance of lower density.

More than 85% of all cases of schistosomiasis in Cameroon occur in the northern sahelian half of the country representing 20% of the population. Several workers have advocated the integrated approach to schistosomiasis control, including snail control, but the death and decay of aquatic organisms, and fish kill that often follows Bayluscide application at the dose of 1g/m³ decrease its acceptability. The present study was designed to assess the effect of lower Bayluscide doses on snail host and non-target fish, frog, the tadpole kill. Bayluscide was applied to study ponds at concentrations of 0, 0.25, 0.5, and 1 g/m³ (ppm). Pre and post application assessment of snails hosts of schistosomes, fish, frog, and tadpole kill were carried out. All 0.25, 0.5, and 1 g/m³ Bayluscide concentrations reduced snail population significantly. Bayluscide concentration of 0.50 g/m³ applied in two rounds of 0.25 g/m³ resulted in high snail mortality and low lethality to fish, frogs, and tadpoles. Further studies are needed to assess the cost-effectiveness of Bayluscide in the control of schistosomiasis following the simplified approach.

A fish-infecting myxosporean, Henneguya hemiodopsis sp. n., found infecting the gills of Hemiodopsis microlepis and collected from the Poty River near the city of Teresina, Brazil, was described based on ultrastructural studies. The parasite occurred within large whitish polysporic plasmodia (up to 200 μm in diameter) containing asynchronous developmental sporogonic stages, mainly mature spores. The spores measured 19.7 ± 0.9 μm in total length (n = 30) and the ellipsoidal spore body was 10.8 ± 0.5 μm long, 3.3 ± 0.4 μm wide and 2.5 ± 0.5 μm thick. The spores were composed of two equal shell valves adhering together along the straight suture line, with each valve having equal-sized caudal tapering tails measuring 8.7 ± 0.6 μm in length. The spores were surrounded by a thin anastomosed network of microfibrils, more evident on the tails. There were two symmetric elongated bottle-like polar capsules 3.5 ± 0.3 μm long and 1.0 ± 0.2 μm wide, each with a polar filament with five to six coils. Given the morphological and ultrastructural differences from previously described parasites and the specificity of the host species, we propose a new species, named H. hemiodopsis sp. n.

A model of skin infection with Leishmania amazonensiswith low doses of parasites is compared to infection with high doses of L. amazonensis and low and high doses of Leishmania major. C57BL/6 mice were infected with 10³ or 10(6) parasites in the ear and the outcome of infection was assessed. The appearance of lesions in mice infected with 10³ parasites was delayed compared to mice infected with 10(6) Leishmania and parasites were detectable at the infection site before lesions became apparent. Mice infected with L. amazonensisdisplayed persistent lesions, whereas infection with L. major spontaneously healed in all groups, although lymphocytes persisted at the site of infection after healing. Macrophages persisted only in L. amazonensis-infected mice. High-dose L. amazonensis-infected mice produced lower levels of IFN-γ and TNF than mice infected with L. major. No correlation between the persistence of parasites and IL-10 levels and the production of nitric oxide or urea by macrophages was found. We conclude that infection with low doses of L. amazonensisin the dermis changes the course of infection by delaying the appearance of lesions. However, low-dose infection does not change the outcomes of susceptibility and cytokine production described for subcutaneous infection with high numbers of parasites.

Atualmente, os critérios utilizados no Sul do Brasil para definição da dose de nitrogênio (N) a ser aplicada no milho são o teor de matéria orgânica no solo, a expectativa de rendimento da cultura e as características da cultura antecessora. Embora apresente alta relação carbono:nitrogênio (C/N), a aveia preta é a espécie de cobertura de solo de inverno mais utilizada como antecessora às culturas comerciais de verão. Essa característica pode resultar em imobilização do N do solo, deficiência de N na planta de milho e redução no rendimento de grãos. Embora na determinação da dose a ser aplicada sejam consideradas as espécies antecessoras, os avanços quanto à melhor época para aplicar N em cobertura em milho foram pequenos. Com o objetivo de avaliar a época mais adequada para aplicação da primeira dose de N em cobertura no milho cultivado em sucessão a espécies de inverno com distintas relações C/N, um experimento foi realizado em vasos em casa de vegetação em Porto Alegre-RS. Utilizaram-se colunas de solo não deformado, classificado como Argissolo Vermelho distrófico típico. Os tratamentos constaram de quatro sistemas de coberturas de solo de inverno (aveia preta, ervilhaca comum, nabo forrageiro e pousio) e três formas de manejo de N em cobertura (com aplicação de N nos estádios V3 ou V5 e sem aplicação de N em cobertura). O delineamento experimental foi o completamente casualizado, em esquema fatorial 4 x 3, com três repetições. Procedeu-se à análise de variância pelo teste F e à comparação de médias pelo teste de Tukey (p < 0,05). A aveia, que tem alta relação C/N, apresentou baixa taxa de mineralização e de liberação de N de seus resíduos, enquanto as culturas de ervilhaca comum e nabo forrageiro mostraram relação C/N mais estreita, estimulando esses processos. Com o uso de aveia como cultura antecessora ao milho, verificou-se diminuição dos teores de N mineral no solo e de N total na planta de milho, independentemente da época de aplicação de N em cobertura, diminuindo o desenvolvimento inicial da planta. Em sucessão à ervilhaca comum e ao nabo forrageiro, o teor relativo de clorofila na folha e a produção de massa seca de milho, avaliados no estádio V7, foram maiores em relação aos obtidos em sucessão à aveia preta, independentemente da época de aplicação da primeira dose de N em cobertura. Os dados obtidos evidenciaram ser possível retardar a época de aplicação da primeira dose de N em cobertura em milho do estádio V3 para V5, quando o milho for cultivado em sucessão a espécies de inverno com baixa relação C/N.

Os solos brasileiros, principalmente os do cerrado, são bastante intemperizados e pobres em alguns micronutrientes catiônicos na solução do solo. A utilização de técnicas de manejo, como a adubação verde, pode favorecer o fluxo difusivo (FD) e a disponibilidade desses nutrientes para as plantas. O presente trabalho visou avaliar se a incorporação de adubos verdes ao solo, em diferentes doses e épocas, modifica o FD e a forma iônica de transporte dos micronutrientes Zn, Cu, Fe e Mn no solo. Para tanto, foram incorporados dois resíduos vegetais largamente cultivados como adubo verde: o feijão guandu (Cajanus cajan) ou o milheto (Pennisetum americanum) por diferentes períodos (0, 15, 25, 35, 45 e 55 dias) e doses (0, 9, 18 e 36 t ha-1) num Latossolo Vermelho, argiloso, em condições de laboratório. Para avaliar o FD, utilizaram-se resinas de troca aniônica (positivamente carregada) e de troca catiônica (negativamente carregada) na forma de lâmina, incubadas junto ao solo em câmaras de difusão durante 15 dias. Os resultados obtidos demonstraram que houve aumento do FD do Cu e do Fe com o aumento das doses de material vegetal, principalmente no início do período de incubação, e maior fluxo desses dois micronutrientes para a resina aniônica em relação à catiônica, possivelmente por ser o seu transporte no solo mais dependente da formação de complexos organometálicos com carga líquida negativa. Já para Zn e Mn, o fluxo difusivo foi maior para a resina catiônica. O aumento do tempo de incubação favoreceu o fluxo difusivo de Mn e Zn e reduziu o do Cu e Fe.

Eucalyptus grandis and other clonal plantations cover about 3.5 million ha in Brazil. The impacts of intensively-managed short-rotation forestry on soil aggregate structure and Carbon (C) dynamics are largely undocumented in tropical ecosystems. Long-term sustainability of these systems is probably in part linked to maintenance of soil organic matter and good soil structure and aggregation, especially in areas with low-fertility soils. This study investigated soil aggregate dynamics on a clay soil and a sandy soil, each with a Eucalyptus plantation and an adjacent primary forest. Silvicultural management did not reduce total C stocks, and did not change soil bulk density. Aggregates of the managed soils did not decrease in mass as hypothesized, which indicates that soil cultivation in 6 year cycles did not cause large decreases in soil aggregation in either soil texture. Silt, clay, and C of the sandy plantation soil shifted to greater aggregate protection, which may represent a decrease in C availability. The organic matter in the clay plantation soil increased in the fractions considered less protected while this shift from C to structural forms considered more protected was not observed.

O nitrogênio é um dos elementos de grande impacto na produtividade da batata, e seu efeito nas plantas pode ser avaliado por meio de técnicas de diagnóstico do estado nutricional. Este trabalho teve o objetivo de determinar a dose ótima econômica de N para a produção de tubérculos e estimar o nível crítico de índices do estado de N na folha de duas cultivares de batata. O experimento foi conduzido de maio a agosto de 2008, em delineamento em blocos ao acaso em esquema fatorial 4 x 2, sendo quatro doses de N (0, 100, 200 e 400 kg ha-1 de N, aplicadas em pré-plantio como ureia) e duas cultivares de batata (Ágata e Asterix), com quatro repetições. Aos 21 dias após a emergência foram determinadas a leitura SPAD e os teores de N e de clorofila na quarta folha a partir do ápice (QF). A produtividade de tubérculos comerciais aumentou até 45.065 e 46.500 kg ha-1 com o aumento da dose de N até 297 e 250 kg ha-1 para Ágata e Asterix, respectivamente. Para essas cultivares, a dose para obtenção da máxima eficiência econômica foi de 290 e 245 kg ha-1 de N, respectivamente. Nas duas cultivares, houve efeito positivo de doses de N sobre o índice SPAD e teores de N e de clorofila na QF. Os valores críticos foram de 40,5 e 43,7 para o índice SPAD, de 66,7 e 75,2 g kg-1 para o teor foliar de N e de 6,13 e 6,96 mg g-1 para o teor de clorofila total na matéria fresca das folhas, respectivamente, para Ágata e Asterix. Os valores da leitura do índice SPAD correlacionaram-se com os valores de clorofila total extraível na quarta folha e com a produção de tubérculos de batata, indicando a possibilidade de medir o valor SPAD aos 21 DAE para prognosticar a produtividade de tubérculos de batata.

O rendimento e a conservação da cebola são influenciados pela disponibilidade de nitrogênio (N) no solo, o qual é requerido em grande quantidade. O presente trabalho objetivou avaliar o efeito de doses e do número de aplicações de N no estado nutricional das plantas, no rendimento e na conservação pós-colheita de bulbos de cebola. Foram realizados três experimentos, em campo, em Cambissolos catarinenses. Os tratamentos consistiram de uma combinação fatorial (4 x 3) envolvendo quatro doses de N (0, 50, 100 e 200 kg ha-1) e três modos de aplicação (aos 45, aos 45 e 75, e aos 30, 60 e 90 dias após o transplante). De cada dose, 25 % foram adicionados no plantio e o restante aplicado em uma ou dividido em duas ou três aplicações iguais. O transplante das mudas foi sempre realizado na segunda quinzena de julho, e a colheita foi efetuada aproximadamente 115 dias depois. O rendimento de bulbos aumentou de forma quadrática com o aumento da quantidade de N aplicada. A dose de N estimada que proporcionou a máxima produtividade econômica variou de 249 kg ha-1 em 2006/07, onde o solo era arenoso, a 116 e 142 kg ha-1 em 2008/09 e 2009/10, respectivamente, em solo mais argiloso. Os rendimentos máximos obtidos foram de 38, 46 e 30 t ha-1 em 2006/07, 2008/09 e 2009/10, respectivamente, e as doses de N correspondentes à máxima produtividade econômica promoveram incrementos de 42, 10 e 17 %, respectivamente, no tratamento sem N. O aumento do número de aplicações de N em cobertura, de uma para duas ou três, não alterou o rendimento e a conservação pós-colheita em nenhum ano. Em solos arenosos com baixo teor de matéria orgânica, é necessário aplicar maior quantidade de N para obter alto rendimento de cebola do que em solos argilosos com médios teores de matéria orgânica. A adição de N ao solo pode alterar negativamente a conservação dos bulbos em anos chuvosos.

Crop-livestock integration represents an interesting alternative of soil management, especially in regions where the maintenance of cover crops in no-tillage systems is difficult. The objective of this study was to evaluate soil physical and chemical properties, based on the hypothesis that a well-managed crop-livestock integration system improves the soil quality and stabilizes the system. The experiment was set up in a completely randomized design, with five replications. The treatments were arranged in a 6 x 4 factorial design, to assess five crop rotation systems in crop-livestock integration, and native forest as reference of soil undisturbed by agriculture, in four layers (0.0-0.05; 0.05-0.10; 0.10-0.15 and 0.15-0.20 m). The crop rotation systems in crop-livestock integration promoted changes in soil physical and chemical properties and the effects of the different systems were mainly detected in the surface layer. The crops in integrated crop-livestock systems allowed the maintenance of soil carbon at levels equal to those of the native forest, proving the efficiency of these systems in terms of soil conservation. The systems influenced the environmental stability positively; the soil quality indicator mineral-associated organic matter was best related to aggregate stability.

Is it possible to build predictive models (PMs) of soil particle-size distribution (psd) in a region with complex geology and a young and unstable land-surface? The main objective of this study was to answer this question. A set of 339 soil samples from a small slope catchment in Southern Brazil was used to build PMs of psd in the surface soil layer. Multiple linear regression models were constructed using terrain attributes (elevation, slope, catchment area, convergence index, and topographic wetness index). The PMs explained more than half of the data variance. This performance is similar to (or even better than) that of the conventional soil mapping approach. For some size fractions, the PM performance can reach 70 %. Largest uncertainties were observed in geologically more complex areas. Therefore, significant improvements in the predictions can only be achieved if accurate geological data is made available. Meanwhile, PMs built on terrain attributes are efficient in predicting the particle-size distribution (psd) of soils in regions of complex geology.

The use of pig slurry (PS) as fertilizer can affect the soil quality and increase total stocks of soil organic carbon (TOC). However, the effects of PS on TOC amount and forms in the soil are not fully understood, particularly in areas under no-tillage (NT). The purpose of this study was to determine TOC contents and stocks in the particulate (POC) and mineral-associated C fractions (MAC) of an Oxisol after nine years of maize-oat rotation under NT, with annual applications of PS, soluble fertilizer and combined fertilization (pig slurry + soluble fertilizer). The experiment was initiated in 2001 in Campos Novos, Santa Catarina, with the following treatments: PS at rates of 0 (without fertilization - PS0); 25 (PS25); 50 (PS50); 100 (PS100); and 200 m3 ha-1yr-1 (PS200); fertilization with soluble fertilizer (SF); and mixed fertilization (PS + SF). The TOC content was determined in samples of six soil layers to a depth of 40 cm, and the POC and MAC contents in four layers to a depth of 20 cm. From the rate of 50 m3 ha-1yr-1 and upwards, the soil TOC content and stock increased according to the PS rates in the layers to a depth of 10 cm. The POC and MAC contents and stocks were higher in the surface layers, with a clear predominance of the second fraction, but a greater relative amplitude in the contents of the first fraction.

A plant regeneration method with cell suspension cultures of banana, and the effect of biobalistic on regeneration potential are described in this report. Somatic embryos of banana were obtained from indirect embryogenesis of male inflorescence of banana cultivar Maçã (AAB group). Part of the calluses formed (40%) showed embryogenic characteristics (nonfriable, compact and yellow color). The cell suspension, originated from embryogenic calluses, contained clusters of small tightly packed cells with dense cytoplasms, relatively large nuclei and very dense nucleoli. After four months of culture, somatic embryos started to regenerate. The maximum number of regenerated plants was observed between 45 and 60 days after embryo formation.In the first experiment, 401 plants were regenerated from approximately 10 mL of packed cells. In the second experiment, 399 plants were regenerated from a cell suspension six months older than that of the first experiment. Cell transformation using particle bombardment with three different plasmid constructions, containing the uid-A gene, resulted in a strong GUS expression five days after bombardment; however, plant regeneration from bombarded cells was much lower than nonbombarded ones.

The objective of this work was to develop a genetic transformation system for tropical maize genotypes via particle bombardment of immature zygotic embryos. Particle bombardment was carried out using a genetic construct with bar and uidA genes under control of CaMV35S promoter. The best conditions to transform maize tropical inbred lines L3 and L1345 were obtained when immature embryos were cultivated, prior to the bombardment, in higher osmolarity during 4 hours and bombarded at an acceleration helium gas pressure of 1,100 psi, two shots per plate, and a microcarrier flying distance of 6.6 cm. Transformation frequencies obtained using these conditions ranged from 0.9 to 2.31%. Integration of foreign genes into the genome of maize plants was confirmed by Southern blot analysis as well as bar and uidA gene expressions. The maize genetic transformation protocol developed in this work will possibly improve the efficiency to produce new transgenic tropical maize lines expressing desirable agronomic characteristics.
