Resistance of the dengue vector to temephos stimulated its substitution for Bacillus thuringiensis var. israelensis (Bti) since 2001 in Brazil. The persistence of the two Bti formulations employed at that time by the Health Ministry, Vectobac G and Aquabac G, was assayed under laboratory and outdoor conditions. Both formulations were tested at 0.2 g/10 liters of water, the same concentration applied in the field for vector control. The tests were done against Ae. aegypti third instar larvae (Rockefeller strain). In the laboratory, Vectobac G and Aquabac G caused at least 95% mortality until 101 and 45 days after treatment, respectively. In the outdoor assays, test containers of different materials were treated with either formulation and placed in a shaded area. Larvae were introduced each 3-6 days and mortality was recorded 24 and 48 hours later. In the first set of assays, performed in June 2001, mortality levels of 70% or more were attained for 2-5 weeks for both formulations in all containers. The exception was for the iron one that rusted, resulting in low mortality after seven days. In the second set of assays (August 2001), 70% mortality was attained for just 1-2 weeks for all the containers and both formulations.

Antigenic preparations from Sporothrix schenckii usually involve materials from mixed cultures of yeast and mycelia presenting cross-reactions with other deep mycoses. We have standardized pure yeast phase with high viability of the cells suitable to obtain specific excretion-secretion products without somatic contaminations. These excretion-secretion products were highly immunogenic and did not produce noticeable cross-reactions in either double immunodiffusion or Western blot. The antigenic preparation consists mainly of proteins with molecular weights between 40 and 70 kDa, some of them with proteolytic activity in mild acidic conditions. We also observed cathepsin-like activity at two days of culture and chymotrypsin-like activity at four days of culture consistent with the change in concentration of different secreted proteins. The proteases were able to cleave different subclasses of human IgG suggesting a sequential production of antigens and molecules that could interact and interfere with the immune response of the host.

Foodborne diseases represent operational risks in industrial restaurants. We described an outbreak of nine clustered cases of acute illness resembling acute toxoplasmosis in an industrial plant with 2300 employees. These patients and another 36 similar asymptomatic employees were diagnosed with anti-T. gondii IgG titer and avidity by ELISA. We excluded 14 patients based on high IgG avidity and chronic toxoplasmosis: 13 from controls and one from acute disease other than T. gondii infection. We also identified another three asymptomatic employees with T.gondii acute infection and also anti-T. gondii IgM positive as remaining acute cases. Case control study was conducted by interview in 11 acute infections and 20 negative controls. The ingestion of green vegetables, but not meat or water, was observed to be associated with the incidence of acute disease. These data reinforce the importance of sanitation control in industrial restaurants and also demonstrate the need for improvement in quality control regarding vegetables at risk for T. gondii oocyst contamination. We emphasized the accurate diagnosis of indexed cases and the detection of asymptomatic infections to determine the extent of the toxoplasmosis outbreak.

INTRODUCTION: Listeria monocytogenes is a ubiquitous microorganism in nature and is responsible for listeriosis, an infectious disease caused by consumption of contaminated food. METHODS: Molecular characterization was performed on 19 strains of Listeria monocytogenes (serovars 1/2a, 1/2b, 4b and 4c), isolated from dairy products in Rio Grande do Sul, Brazil. The molecular techniques applied were random amplification of polymorphic DNA and restriction enzyme analysis. In addition to the molecular analysis, the antimicrobial resistance profile was determined. RESULTS: The strains studied showed a low degree of diversity. In relation to the antimicrobial resistance profile of those microorganisms from the samples analyzed, all of them were susceptible to the antimicrobials tested. CONCLUSIONS: The molecular techniques that were used presented good discriminatory power for the strains studied. Furthermore, all of the samples that were analyzed were susceptible to the antimicrobials tested.

Reveste-se de grande importância o conhecimento da marcha de absorção de nutrientes pelas culturas, principalmente visando uma aplicação racional dos fertilizantes. Utilizou-se no presente experimento a cultivar Roma VF, de porte determinado, que tem tido grande aceitação tanto pelos tomaticultores como pelas industrias processadoras. As amostras para analise foram coletadas no período junho a outubro de 1977, em uma área experimental instalada num solo representativo da região oeste do Estado de São Paulo (Latossol Vermelho Amarelo), no município de Narandiba (DIRA de Presidente Prudente)· A cultura recebeu uma adubação de 85--300-100 kg/ha de NPK, sendo 1/3 do N aplicado juntamente com todo o P e K no plantio e os 2/3 restantes aplicados em cobertura aos 30 e 35 dias após a germinação. A área foi de 1,25 x 0,30 m (26,666 covas/ha), com culturais foram os normalmente recomendados para a cultura na região. As amostras (4 repetições) foram coletadas aos 15, 30, 45, 60, 75, 90 e 105 dias após a germinação. As plantas foram separadas em folhas cotiledonares, folhas, caule e frutos. Determinou-se o peso de matéria seca e analisou-se para N, P, K, Ca, Mg, S, B, Cu, Fe, Mo e Zn. Os resultados analíticos obtidos revelaram um crescimento lento ate aos 30 dias, após o que houve um crescimento acelerado, com o peso da materia seca praticamente dobrando a cada quinzena no período dos 45 aos 75 dias, atingindo o máximo aos 105 dias (5.706,61 kg/ha). Quanto ao crescimento dos frutos, expresso em peso de materia seca, houve um aumento de cerca de 20 vezes no intervalo dos 45 aos 75 dias, praticamente duplicando o peso no período dos 75 aos 90 dias e estabilizando-se aos 105 dias (2.708,6 kg/ha). Na época de floração, as folhas apresentavam, em função da materia seca, 3,7% N; 0,50% P; 4,44% K; 3,24% Ca; 0,99% Mg; 0,46% S; 72 ppm B; 15 ppm Cu; 434 ppm Fe; 375 ppm Mn; 0,18 ppm Mo; 148 ppm Zn. A produção efetiva de 65 ton/ha obtida na área experimental, contem as seguintes quantidades de nutrientes nos frutos: 67,8 g N; 8,9 g P; 112,2g K; 7,7 g Ca; 6,0 g Mg; 3,1 g S; 93 g B; 45 g Cu; 547 g Fe; 163 g Mn; 485 mg Mo.

Neste trabalho foram propostas fórmulas para se calcular o rendimento esperado, em polpa concentrada, em industrias de processamento de tomate, comparando-o com o rendimento realmente obtido, e visando controlar o desempenho da indústria. Para testar estas fórmulas foi feito um estudo comparativo entre o rendimento esperado calculado e o rendimento real, tomando por base os assentamentos de uma indústria, de porte médio, de processamento de tomates, do Estado de São Paulo, nos anos agrícolas de 1981 e 1982. Concomitantemente foi feita uma análise das perdas, na forma de descartes, na esteira de seleção e de sementes e pele, nas despolpadeiras e também uma verificação do Brix médio ponderado, nas duas safras. Os resultados mostraram que as fórmulas são adequadas para o cálculo do rendimento esperado pois as diferenças, com o rendimento real, obtido foram de -1,49% e +1,74% em 1982. As perdas de tomate (descartes) foram consideradas elevadas em comparação com as de outros países o que significa tomate de baixa qualidade. O Brix médio ponderado mensal que alcançou até 5,62 foi significativamente maior que o de anos anteriores.

Several larval and pupal products of Aedes fluviatilis (Lutz) were tested for their influence on the oviposition behaviour of females of the same species. Significant (alfa = 0,05) atractiveness was shown by: larval water, previously containing 5 to 15 larvae/1,5 ml; larval water, preserved up to 38 days; evaporate and reconstructed larval water extracts up to 2 years after production and water filtered through fresh or dried ground larvae. hexanic larval water extracts and water filtered through fresh or dired ground pupae did not influence oviposition.

Epidermal changes from 32 cutaneous and 3 mucosal American leishmaniasis (ACL) active lesions were studied for HLA-DR, -DP expression, Lanerhans cells and lymphocyte infiltration. In addition to a DR and DQ positivity at the surface of the cells of the inflammatory infiltrate, a strong reaction for DR antigens was detected on keratinocytes. Hyperplasia of Langerhans cells was present in al cutaneous lesions and epidermis was infiltrated by T lymphocytes. When healed lesions of 14 of these subjects were re-biopsied 1 to 12 months after the end of pentavalent antimonial therapy, MHC class antigens could no longer be seen on keratinocytes. Our data represrn evidence for hhe reversibility of the abnormal HLA-DR expression by keratinocytes in ACL after Glucantime therapy or spontaneous scar formation, demonstrating that this expresion is restricted to the period of active lesions. The present findings can be regarded as an indirect evidence that keratinocytes may be involved in the immunopathology of ACL.

In contrast to China where vegatation is predominantly herbaceous, vegetation in Brazil is commonly arboreous. This fact may explain why Chinese drugs are usually acetate derived, while actual and potential natural therapeutic agents from Brazil are mostly shikimate derived. Only relatively few compounds isolated from Brazilian plants have been submitted to adequate pharmacological testing

This paper recalls the outcoming of marine natural products research and reviews a selection of marirne bioactive metabolites in current use together with promising trends in marine pharmacology.

We have recenty studied several natural product constituents which have effects on the CNS. (1) Tetrahydropalmatine (THP) and its analogues were isolated from Corydalis ambigua and various species of Stephania. (+)-THP and (-)-THP posses not only analgesic activity, but also exert sedative-tranquillizing and hypnotic actions. Results of receptor binding assay and their pre-and post-synaptic effects on dopaminergic system indicate that (-)-THP and (-)-stepholidine are dopamine receptor antagonists while (+)-THP is a selective dopamine depletor. (2) 3-Acetylaconitine (AAC) is an alkaloid isolated from Aconitum flavum. The relative potency of analgesic action of AAC was 5.1-35.6 and 1250-3912 times that of morphine and aspirin, respectively. The analgesic effect of AAC was antagonized by naloxone, but was eliminated by reserpine. In monkeys, after AAC was injected for 92 days, no abstinence syndrome was seen after sudden AAC withdrawal or when challenged with nalorphine. (3) Huperzine A (Hup-A) is an alkaloid isolated from Huperzia serrata which was found to be a selective ChE inhibitor and could improve learning and retrieval process. Preliminary clinical studies showed that Hup-A improve short-and long-term memory in patients of cerebral arteriosclerosis with memory impairment. (4) Ranamargarin is a new tetradecapeptide isolated from the skin of the Chines frog Rana margaratae. This peptide may mainly act on NK-1 receptor.

In the present work we have described the in vivo antimalarial actrivity of six different plants. Two of them (Verninia brasiliana and Eupatorium squalidum) were tested in a randomic approach among 273 crude extracts from plants; four (Acanhospermum australe, Esenbeckia febrifuga, Lisianthus specious and Tachia guianensis) were selected after screening 22 crude extracts from different medicinal and some of them showed antimalarial activity in vitro. Some aspects of recent research with natural products aiming to produce drugs are discussed.

In order to evaluate the potential diuretic effect of two natural products, Elephantopus scaber and Alpinia speciosa, a trial administration was carried out in ten healthy volunteers and the effects compared to the those of a placebo. The substances were given on different days, with a seven day interval between doses. The amount of substance administered to the subjects was five times the usual dose i.e. 7.5 g/100 ml and 0.8 g/100 ml respectively. The following parameters parameteres were measured: urinary and plasma sodium, potassium, uric acid, calcium, phosphate, urea, creatinine. The subjects were also examined clinically and total urinary volume was assessed. The only significant finding (p < 0.05) was a slight diuresis with Alpinia speciosa, which also lowered the diastolic (p < 0.05) and systolic (p < 0.01) blood pressures. No effect on electrolytes or renal function parameters was observed, and this probably excludes any renal tubular or glomerular effect from these substances.
