Abstract: The objective of this work was to purify and standardize the rue (Ruta graveolens) extract and evaluate its effect on Magnaporthe oryzae as an alternative to the integrated management of rice blast. The drug was characterized, the liquid extract was obtained, and the methodology for quantifying the standard markers psoralen and bergapten was validated. Rue extract and the markers, solely or in combination, were assayed in vitro, as well as in greenhouse conditions, for their ability to suppress leaf blast, by the evaluation of mycelial growth, conidial germination, and appressorium formation. Rue extract inhibited M. oryzae mycelial growth (100%), conidial germination (LD50=0.237 mg), and the appressorium formation (LD50=0.121 mg); besides, the extract reduced leaf blast severity by 80.84%. Fluorescence microscopy showed that rue extract did not damage M. oryzae cell wall and plasma membrane, indicating another mode of action. Rue extract has a great potential for controlling rice leaf blast.

Mauritia vinifera (buriti) is a palm tree that grows wild in different areas of Brazil, particularly in the Amazonian region. The buriti oil is rich in carotenoids, especially in β-carotene. The growing interest in other natural sources of β-carotene has stimulated the industrial use of buriti as a raw material for pulp oil extraction. Most processes are based on the conventional technologies, involving drying and pressing the pulp for oil recovery and further separation of carotenoids in a liquid phase using organics solvents. In the present work, the ethanol-based process was evaluated for simultaneous carotenoids recovering and fractionating from buriti pulp. The raw material and ethanol, 1:4 ratio, were placed in an erlenmeyer flask and maintained at 30rpm for 1 hour in a temperature-controlled bath at 65ºC. The mixture was filtered under vacuum and cooling at 10ºC to allow for the separation of the solvent in two phases. Carotenoids composition, determined by HPLC, has indicated a β-carotene concentration about 12 times greater in the lower phase than in the upper phase. The profile of the carotenoids in the denser phase is quite similar to that of raw buriti oil, and the concentration of total carotenoids is 40% higher than that of the original raw oil, making the ethanol-based process particularly attractive for industrial applications.

The sublethal effect of extracts of Azadirachta indica on Ceratitis capitata was evaluated. Two pairs of flies were treated in plastic tubes with cotton placed in plastic cages. An artificial diet (hydrolyzed protein + sugar) was provided ad libitum. The extracts affected significantly the longevity of C. capitata. The pre-oviposition period were not significantly affected by the extracts. The A. indica branches extracted with dichloromethane (888 ppm) affected significantly the fecundity and fertility, reducing the number of eggs laid to approximately 80 % and the egg hatching by 30 % at the 8th day. Therefore, the neem branches extracted with dichloromethane affected the reproduction of C. capitata.

OBJETIVO: Apesar de os achados radiográficos de apendicite aguda serem bem documentados, o valor da radiografia simples ainda não foi completamente determinado. O objetivo do presente estudo foi estabelecer a freqüência da associação de apendicite aguda a um sinal radiográfico caracterizado por imagem de acúmulo fecal ocupando todo o ceco. MATERIAIS E MÉTODOS: Foram realizadas radiografias simples de abdome de 100 pacientes consecutivos com apendicite aguda, comprovada por operação e exame anatomopatológico. Pesquisou-se, nas radiografias, a presença de imagem de acúmulo fecal no ceco, caracterizada por hipotransparência ocupando todo o ceco e, eventualmente, também o cólon ascendente. RESULTADOS: A imagem de acúmulo fecal no ceco foi encontrada em 97% dos doentes, independentemente de idade, sexo, cor da pele ou estádio da apendicite. CONCLUSÃO: Este estudo sugere que a presença de imagem radiográfica de acúmulo fecal no ceco pode ser um sinal útil no diagnóstico de apendicite aguda.

Crude extract and fractions of Buddleja thyrsoides were investigated regarding antioxidant activities by DPPH, total phenolic contents by Folin-Ciocalteau and antimicrobial activity by the broth microdilution method. Total phenolics varied from 214.07 ± 3.6 to 438.4 ± 0.3 mg g-1. Crude extract, ethyl acetate, dichloromethane and butanolic fractions exhibited a weak scavenging activity (SC50=186.04 ± 10.8, 137.70 ± 8.5, 146.89 ± 9.0 and 165.71 ± 3.2 µg mL-1, respectively). A correlation between the antioxidant activities and total phenolic contents could be shown (r=0.857, p<0.01). The lowest value of MIC was observed with butanolic fraction against Saccharomyces cerevisiae (MIC and MFC at 62.5 µg mL-1). Dichloromethane and ethyl acetate fractions were effective against Staphylococcus aureus with MIC value at 250 and 500 µg mL-1 respectively.

A chromatographic technique for determination of rutin and narcissin in marigold extract and topical formulations was developed and validated. The method shows linearity over the concentration range of 0.2 - 6.0 μg/mL of rutin (r = 0.9986) and 0.8 - 12.0 μg/mL of narcissin (r = 0.9951). The values obtained for precision and accuracy are in agreement with ICH guidelines. Both the formulation excipients and the porcine ear skin samples did not interfere with the flavonoids determination. The recovery of rutin and narcissin in skin samples added with marigold extract was 81.41% and 83.35%, respectively, which demonstrate the applicability of this method to perform skin penetration studies.

The species Eremanthus mattogrossensis, known as "veludo do cerrado" (cerrado velvet), is native to the Brazilian Cerrado. Because the amount of metabolites present in plants may be influenced by biological and environmental factors, here we conducted an HPLC-DAD-MS/MS investigation of the metabolite concentrations found in the MeOH/H2O extract of the leaves of this species. The main compounds were identified and quantified, and the metabolites were grouped by chemical class (caffeoylquinic acids, flavonoids, and sesquiterpene lactone). Statistical analysis indicated a straight correlation between the quantity of metabolites and seasonality, suggesting that environmental properties elicit important metabolic responses.

A statistical mixture-design technique was used to study the effects of different solvents and their mixtures on the yield, total polyphenol content, and antioxidant capacity of the crude extracts from the bark of Schinus terebinthifolius Raddi (Anacardiaceae). The experimental results and their response-surface models showed that ternary mixtures with equal portions of all the three solvents (water, ethanol and acetone) were better than the binary mixtures in generating crude extracts with the highest yield (22.04 ± 0.48%), total polyphenol content (29.39 ± 0.39%), and antioxidant capacity (6.38 ± 0.21). An analytical method was developed and validated for the determination of total polyphenols in the extracts. Optimal conditions for the various parameters in this analytical method, namely, the time for the chromophoric reaction to stabilize, wavelength of the absorption maxima to be monitored, the reference standard and the concentration of sodium carbonate were determined to be 5 min, 780 nm, pyrogallol, and 14.06% w v-1, respectively. UV-Vis spectrophotometric monitoring of the reaction under these conditions proved the method to be linear, specific, precise, accurate, reproducible, robust, and easy to perform.

This article describes the isolation and identification of flavonoids in the hydroethanolic extract of the aerial parts from Tonina fluviatilis and evaluation of their antiradical activity. A method based on HPLC-DAD was developed and validated for detecting and quantifying flavonoids in hydroethanolic extracts. The flavonoids identified and quantified in the extract were 6,7-dimethoxyquercetin-3-O-β-D-glucopyranoside (1), 6-hydroxy-7-methoxyquercetin-3-O-β-D-glucopyranoside (2), and 6-methoxyquercetin-3-O-β-D-glucopyranoside (3). The developed method presented good validation parameters, showing that the results obtained are consistent and can be used in ensuring the quantification of these constituents in the extracts. Compounds 2 and 3 showed strong antiradical activity when compared with the positive controls (quercetin and gallic acid).

This study aimed to evaluate the effect of Moringa oleifera Lam extract on the removal of total solids (TS), total suspended solids (TSS) and chemical oxygen demand (COD), in different filter media for treating wastewater of dairy cattle breeding (DCW). The moringa seed extract was obtained by grinding 50 g of seeds in one liter of distilled water and, after passing the solution through a quantitative paper filter of 25 microns, 60 mL of the extract were added to wastewater from cattle breeding before the filtration process in organic filters made of thin coal, bamboo leaves, eucalyptus leaves, gliricidia branches and sawdust. This was followed by the completely randomized experimental design, adopting a factorial of 5 x 2. Aliquots of filtered effluent were collected and the total solids (TS) concentrations, total suspended solids (TSS) and chemical oxygen demand (COD) were determined. It was found that the increase in the efficiency of removal of COD and total solids can be attributed to the coagulating power of the moringa seed extract, wherein the filter medium with bamboo leaves presented the best performance, showing potential for use as alternative filter material in the primary treatment of DCW.

A colite de derivação fecal (CD) é um processo inflamatório que ocorre no segmento colorretal desfuncionalizado, após uma cirurgia de desvio do trânsito intestinal. As principais características dessa entidade clínica são: apresenta-se na desfuncionalização do cólon ou reto; não há doença inflamatória intestinal preexistente; nunca acomete o sítio proximal à colostomia e ocorre resolução do processo após a restauração do trânsito intestinal. Diversas são as hipóteses postuladas para explicar o seu aparecimento; todavia, a deficiência nutricional do epitélio colônico, pela ausência dos ácidos graxos de cadeia curta (AGCC), no segmento desfuncionalizado, é a mais aceita na atualidade. Os autores fazem uma revisão da literatura enfocando os aspectos clínicos, histopatológicos e terapêuticos desta doença.

OBJETIVO: A peritonite é responsável por aproximadamente 50% das mortes por sepse e, apesar de avanços nos métodos usados para o seu diagnóstico e tratamento, cerca de um terço dos pacientes ainda morre de peritonite secundária grave. O objetivo do presente trabalho foi comparar a eficácia de diversos tipos de tratamento para a peritonite fecal grave e estabelecida. MÉTODOS: Foram usadas 40 ratas adultas, submetidas à peritonite fecal com injeção intraperitoneal de uma suspensão de fezes de ratos. Os animais foram divididos em oito grupos (n = 5): Grupo 1, controle; Grupo 2, limpeza mecânica intraperitoneal com gaze; Grupo 3, lavagem com solução salina a 0,9%, à temperatura ambiente; Grupo 4, lavagem com solução salina a 0,9%, a 37,8ºC; Grupo 5, lavagem com povidona-iodo a 0,5%; Grupo 6, lavagem com clorexidina a 0,05%; Grupo 7, injeção intramuscular de gentamicina e clindamicina; Grupo 8, introdução intraperitoneal de açúcar. RESULTADOS: Os grupos 5 e 8 foram os que apresentaram a mortalidade mais rápida (menos de 24 horas). Após 72 horas, permaneceu viva uma rata em cada um dos grupos 2, 3, 4 e 6. Nos grupos 1, 5, 7 e 8 não houve sobrevida. Apesar de todos os animais do Grupo 7 morrerem, o óbito ocorreu em um período mais longo (72 horas) do que o dos demais grupos. CONCLUSÃO: Somente ocorreu sobrevida nos grupos submetidos à limpeza peritoneal menos agressiva. Além disso, um procedimento terapêutico único de limpeza ou antibiótico sistêmico por um dia não é suficiente para prevenir a morte em ratos com peritonite fecal grave e estabelecida.

OBJETIVO: Há poucos estudos sobre os efeitos de uma nova infecção após peritonite séptica. Com o objetivo de compreender melhor esta situação, realizou-se o presente experimento tendo como parâmetro o papel do tempo neste fenômeno. MÉTODO: Foram utilizadas 36 ratas Wistar adultas, submetidas a peritonite fecal com injeção intraperitoneal de uma solução de fezes de ratos. Os animais foram divididos em quatro grupos (n = 9): Grupo 1A - controle: injeção intraperitoneal de solução de fezes com uma quantidade sabidamente letal (10 ml/kg); Grupo 1B - reinfecção: injeção intraperitoneal de solução de fezes com uma quantidade sabidamente não letal (2 ml/kg) e, após 30 dias, injeção de solução de fezes (10 ml/kg); Grupo 2A - controle da reinfecção tardia: injeção intraperitoneal de fezes a 10 ml/kg; Grupo 2B - reinfecção tardia: injeção intraperitoneal de fezes a 2ml/kg e, após quatro meses, injeção de 10ml/kg. RESULTADOS: Todos os nove animais do Grupo 1A morreram no período de sete dias após a injeção da solução de fezes. Já no Grupo 1B, pré-infectado, apenas um animal morreu, 24 horas após a injeção da solução de fezes a 10 ml/kg (p < 0,001). Em relação ao Grupo 2, oito dos nove animais de cada subgrupo morreram no período de sete dias. CONCLUSÕES: Uma sepse peritoneal menor por fezes eleva a resistência orgânica a nova contaminação fecal mais intensa que ocorra após um período curto. Contudo, essa defesa não persiste por tempo mais prolongado.

OBJETIVO: Apesar das características radiográficas da apendicite aguda estarem bem documentadas, o valor da radiografia simples de abdome ainda não foi completamente estudado. Nesse sentido, o objetivo do presente trabalho foi verificar um novo sinal radiográfico caracterizado pela imagem de acúmulo fecal no ceco. MÉTODO: Foram estudados prospectivamente 170 pacientes consecutivos, de ambos os sexos, internados com abdome agudo e dor localizada no flanco direito, distribuídos em dois grupos: Grupo 1 (n = 100) - portadores de apendicite aguda, submetidos a um estudo radiográfico do abdome antes do tratamento cirúrgico, Grupo 2 (n = 70) - submetidos a dois estudos radiográficos do abdome:antes da operação e outro no dia seguinte à cirurgia. Todas as radiografias foram simples, em incidência ântero-posterior. RESULTADOS: A presença do sinal de acúmulo fecal no ceco esteve presente em 97 (97%) pacientes do Grupo 1 e em 68 (97,14%) pacientes do Grupo 2. No pós-operatório, dos 68 pacientes que apresentaram o sinal radiográfico, esse desapareceu em 66 casos. A sensibilidade do sinal radiográfico para apendicite aguda foi de 97,05 %. CONCLUSÕES: A imagem radiográfica de acúmulo fecal no ceco associa-se a apendicite aguda. Essa imagem geralmente desaparece após o apêndice cecal ser removido.

Objective: to evaluate the effect of the Euphorbia tirucalli hydroalcoholic extract (ETHE) on the development of Ehrlich Tumor, in its ascitic form. Methods: we intraperitoneally inoculated 15 Swiss mice with 10.44 x 107 cells of Ehrlich Tumor and divided them in two groups one day after: ETHE Group (eight mice), treated with a dosage of 125 mg/kg/day of EHTE for five days; and Control Group (seven mice), treated only with 0.9% isotonic saline solution over the same period. The treatment was done by gavage. Ten days after inoculation, four mice from each group were sacrificed for quantification of tumor cell number, ascitic fluid volume and bone marrow cell number. The remaining animals were maintained to evaluate survival. Results: The ascitic fluid volume and the tumor cell number were decreased in the ETHE group when compared with the control group, but with no statistical significance. On the other hand, survival was higher in the ETHE group, as well as the number of bone marrow cells. Conclusion: Treatment with ETHE after inoculation of Ehrlich Tumor decreases its development and increases survival and the bone marrow cellularity, thus reducing the myelosuppression present in the Ehrlich Tumor bearing mice.
