Fibrocytes are important for understanding the progression of many diseases because they are present in areas where pathogenic lesions are generated. However, the morphology of fibrocytes and their interactions with parasites are poorly understood. In this study, we examined the morphology of peripheral blood fibrocytes and their interactions with Leishmania (L.) amazonensis . Through ultrastructural analysis, we describe the details of fibrocyte morphology and how fibrocytes rapidly internaliseLeishmania promastigotes. The parasites differentiated into amastigotes after 2 h in phagolysosomes and the infection was completely resolved after 72 h. Early in the infection, we found increased nitric oxide production and large lysosomes with electron-dense material. These factors may regulate the proliferation and death of the parasites. Because fibrocytes are present at the infection site and are directly involved in developing cutaneous leishmaniasis, they are targets for effective, non-toxic cell-based therapies that control and treat leishmaniasis.

Several different models of Trypanosoma cruzi evolution have been proposed. These models suggest that scarce events of genetic exchange occurred during the evolutionary history of this parasite. In addition, the debate has focused on the existence of one or two hybridisation events during the evolution of T. cruzi lineages. Here, we reviewed the literature and analysed available sequence data to clarify the phylogenetic relationships among these different lineages. We observed that TcI, TcIII and TcIV form a monophyletic group and that TcIII and TcIV are not, as previously suggested, TcI-TcII hybrids. Particularly, TcI and TcIII are sister groups that diverged around the same time that a widely distributed TcIV split into two clades (TcIVS and TcIVN). In addition, we collected evidence that TcIII received TcIVSkDNA by introgression on several occasions. Different demographic hypotheses (surfing and asymmetrical introgression) may explain the origin and expansion of the TcIII group. Considering these hypotheses, genetic exchange should have been relatively frequent between TcIII and TcIVS in the geographic area in which their distributions overlapped. In addition, our results support the hypothesis that two independent hybridisation events gave rise to TcV and TcVI. Consequently, TcIVS kDNA was first transferred to TcIII and later to TcV and TcVI in TcII/TcIII hybridisation events.

T-cell based vaccines against human immunodeficiency virus (HIV) generate specific responses that may limit both transmission and disease progression by controlling viral load. Broad, polyfunctional, and cytotoxic CD4+T-cell responses have been associated with control of simian immunodeficiency virus/HIV-1 replication, supporting the inclusion of CD4+ T-cell epitopes in vaccine formulations. Plasmid-encoded granulocyte-macrophage colony-stimulating factor (pGM-CSF) co-administration has been shown to induce potent CD4+ T-cell responses and to promote accelerated priming and increased migration of antigen-specific CD4+ T-cells. However, no study has shown whether co-immunisation with pGM-CSF enhances the number of vaccine-induced polyfunctional CD4+ T-cells. Our group has previously developed a DNA vaccine encoding conserved, multiple human leukocyte antigen (HLA)-DR binding HIV-1 subtype B peptides, which elicited broad, polyfunctional and long-lived CD4+ T-cell responses. Here, we show that pGM-CSF co-immunisation improved both magnitude and quality of vaccine-induced T-cell responses, particularly by increasing proliferating CD4+ T-cells that produce simultaneously interferon-γ, tumour necrosis factor-α and interleukin-2. Thus, we believe that the use of pGM-CSF may be helpful for vaccine strategies focused on the activation of anti-HIV CD4+ T-cell immunity.

Insects are essential to tropical ecosystems functioning. In semi-arid regions, the increase in abundance and/or activity (e.g. reproduction and foraging behavior) of insects is usually associated with climatic variables. The present study investigates which climatic variables are best predictors of insect abundance in an area of Caatinga in northeastern Brazil. Individuals were sampled for 24 months using Malaise and pitfall traps, and beating trays. A total of 58925 individuals belonging to 20 insect orders were collected. The most abundant orders were Hymenoptera, Diptera, Collembola and Coleoptera. Most orders studied showed a clear maximum abundance in the rainy season. Rainfall and humidity were the best predictors of insect abundance in the Caatinga. However, no climatic variable could explain Psocoptera and Blattodea variance in abundance/activity. Our results suggest that climatic changes associated with rainfall patterns in the Caatinga may affect ecosystem processes and services that depend direct or indirectly on insect abundance/activity.

This study reports for the first time the occurrence of Hypatropis inermis on upland rice crops (variety Cambará), in Novo Progresso, state of Pará, Brazil (7°07'45.71"S 55°23'21.13"W). The inventory of insect pests was conducted between November 2010 and March 2011 with entomological sweep nets and visual search on stems of rice plants. This record indicates that rice crops may represent important feeding and mating sites for this species.

Both N excess and deficiency may affect cotton yield and quality. It would therefore be useful to base the N management fertilization on the monitoring of the nutritional status. This study investigated the correlations among the following determination methods of the N nutritional status of cotton (Gossypium hirsutum L., var. Latifolia): chlorophyll readings (SPAD-502®, Minolta), specific-ion nitrate meter (Nitrate Meter C-141, Horiba-Cardy®), and laboratory analysis (conventional foliar diagnosis). Samples were taken weekly from two weeks before flowering to the fifth week after the first flower. The experiment was conducted on the Fazenda Santa Tereza, Itapeva, State of São Paulo, Brazil. The crop was fertilized with 40 kg ha-1 N at planting and 0, 30, 60, 90, and 120 kg ha-1 of side-dressed N. The range of leaf N contents reported as adequate for samples taken 80-90 days after plant emergence (traditional foliar diagnosis) may be used as reference from the beginning of flowering when the plant is not stressed. Specific-ion nitrate meter readings can be used as a nutritional indicator of cotton nutrition from one week after pinhead until the third week of flowering. In this case, plants are well-nourished when readings exceed 8,000 mg L-1 NO3-. The chlorophyll meter can also be used to estimate the nutritional status of cotton from the third week of flowering. In this case the readings should be above 48 in well-nourished plants.

Although silicon is not recognized as a nutrient, it may benefit rice plants and may alleviate the Mn toxicity in some plant species. The dry matter yield (root, leaf, sheaths and leaf blade) and plant architecture (angle of leaf insertion and leaf arc) were evaluated in rice plants grown in nutrient solutions with three Mn doses, with and without Si addition. The treatments were arranged in a 2 x 3 factorial [with and without (2 mmol L-1) Si; three Mn doses (0.5; 2.5 and 10 µmol L-1)], in a randomized block design with 4 replications. The experimental unit was a 4 L plastic vase with 4 rice (Metica-1 cultivar) plants. Thirty nine days after keeping the seedlings in the nutrient solution the plant dry matter yield was determined; the angle of leaf insertion in the sheath and the leaf arc were measured; and the Si and Mn concentrations in roots, sheaths and leaves were determined. The analysis of variance (F test at 5 and 1 % levels) and the regression analysis (for testing plant response to Mn with the Si treatments) were performed. The Si added to the nutrient solution increased the dry matter yield of roots, sheaths and leaf blades and also decreased the angle of leaf blade insertion into the sheath and the foliar arc in the rice plant. Additionally, it ameliorated the rice plant architecture which allowed an increase in the dry matter yield. Similarly, the addition of Mn to the solution improved the architecture of the rice plants with gain in dry matter yield. As Si was added to the nutrient solution, the concentration of Mn in leaves decreased and in roots increased thus alleviating the toxic effects of Mn on the plants.

Nitrogen is the main limiting factor in crop productivity and thereby soil management systems may change the mineralization and nitrification rates. In an experiment on soil management systems implemented in 1988 at the experimental station Fundação ABC, Ponta Grossa, in the central South region of the State of Paraná, inorganic N dynamics were examined to find a soil management strategy with a view to a sustainable environment. The objective of this study was to calculate the net mineralization and nitrification rates of soil N and the correlation with soil pH under management systems. Randomized complete block design was used, in split plots, in three replications. The following soil management systems (SMSs) were adopted in the plots: 1) conventional tillage (CT); 2) minimum tillage (MT); 3) no-tillage with chisel plow every three years (NT CH); and 4) continuous no-tillage (CNT). To evaluate the dynamics of inorganic N, samples were collected from sub-plots at different times (11 sampling times - T1 to T11). In the CNT and NT CH, the net mineralization rates were higher in the MT and CT systems in the 0-2.5 cm soil layer, while the nitrification rate was higher in the 2.5-5 cm layer. Soon after implementing the white oat management, the mineralization and nitrification rates in all soil layers were higher in the MT and CT systems. In the period of soybean development, in the 0-2.5 and 2.5-5 cm soil layers, the mineralization and nitrification rates were higher in the CNT and NT CH than in the MT and CT systems.

One of the main negative anthropic effects on soil is the formation of crusts, resulting in soil degradation. This process of physical origin reduces soil water infiltration, causing increased runoff and consequently soil losses, water erosion and/or soil degradation. The study and monitoring of soil crusts is important for soil management and conservation, mainly in tropical regions where research is insufficient to explain how soil crusts are formed and how they evolve. The purpose of this study was to monitor these processes on soils with different particle size distributions. Soil crusts on a sandy/sandy loam Argissolo Vermelho-Amarelo (Typic Hapludult), sandy loam Latossolo Vermelho-Amarelo (Typic Hapludox) and a clayey Nitossolo Vermelho eutroférrico (Rhodic Kandiudalf) were monitored. The soil was sampled and data collected after 0, 3, 5 and 10 rain storms with intensities above 25 mm h-1, from December 2008 to May 2009. Soil chemical and particle size distribution analysis were performed. The changes caused by rainfall were monitored by determining the soil roughness, hydraulic conductivity and soil water retention curves and by micromorphological analysis. Reduced soil roughness and crust formation were observed for all soils during the monitored rainfall events. However, contrary to what was expected according to the literature, crust formation was not always accompanied by reductions in total porosity, hydraulic conductivity and soil water retention.

Because of the climate changes occurring across the planet, especially global warming, the different forms of agricultural soil use have attracted researchers´ attention. Changes in soil management may influence soil respiration and, consequently, C sequestration. The objectives of this study were to evaluate the long-term influence of liming on soil respiration and correlate it with soil chemical properties after two years of liming in a no-tillage system. A randomized complete block design was used with six replications. The experimental treatments consisted of four lime rates and a control treatment without lime. Two years after liming, soil CO2 emission was measured and the soil sampled (layers 0-5, 5-10, 10-20, and 20-30 cm). The P, Ca2+ e Mg2+ soil contents and pH and base saturation were determined. CO2 emission from soil limed at the recommended rate was 24.1 % higher, and at twice the recommended rate, 47.4 % higher than from unlimed soil. Liming improved the chemical properties, and the linear increase in soil respiration rate correlated positively with the P, Ca2+ and Mg2+ soil contents, pH and base saturation, and negatively with H + Al and Al3+ contents. The correlation coefficient between soil respiration rate and chemical properties was highest in the 10-20 cm layer.

Pig slurry applied to soil at different rates may affect soil properties and the mobility of chemical compounds within the soil. The purpose of this study was to evaluate the effects of rates of pig slurry application in agricultural areas on soil physical and chemical properties and on the mobility of glyphosate through the soil profile. The study was carried out in the 12th year of an experiment with pig slurry applied at rates of 0 (control), 50, 100 and 200 m³ ha-1 yr-1 on a Latossolo Vermelho distrófico (Hapludox) soil. In the control, the quantities of P and K removed by harvested grains were replaced in the next crop cycle. Soil physical properties (bulk density, porosity, texture, and saturated hydraulic conductivity) and chemical properties (organic matter, pH, extractable P, and exchangeable K) were measured. Soil solution samples were collected at depths of 20, 40 and 80 cm using suction lysimeters, and glyphosate concentrations were measured over a 60-day period after slurry application. Soil physical and chemical properties were little affected by the pig slurry applications, but soil pH was reduced and P levels increased in the surface layers. In turn, K levels were increased in sub-surface layers. Glyphosate concentrations tended to decrease over time but were not affected by pig slurry application. The concentrations of glyphosate found in different depths show that the pratice of this application in agricultural soils has the potential for contamination of groundwater, especially when the water table is the surface and heavy rains occur immediately after application.

Soil management, in terms of tillage and cropping systems, strongly influences the biological properties of soil involved in the suppression of plant diseases. Fungistasis mediated by soil microbiota is an important component of disease-suppressive soils. We evaluated the influence of different management systems on fungistasis against Fusarium graminearum, the relationship of fungistasis to the bacterial profile of the soil, and the possible mechanisms involved in this process. Samples were taken from a long-term experiment set up in a Paleudult soil under conventional tillage or no-tillage management and three cropping systems: black oat (Avena strigose L.) + vetch (Vicia sativa L.)/maize (Zea mays L.) + cowpea (Vigna sinensis L.), black oat/maize, and vetch/maize. Soil fungistasis was evaluated in terms of reduction of radial growth of F. graminearum, and bacterial diversity was assessed using ribosomal intergenic spacer analysis (RISA). A total of 120 bacterial isolates were obtained and evaluated for antibiosis, and production of volatile compounds and siderophores. No-tillage soil samples showed the highest level of F. graminearum fungistasis by sharply reducing the development of this pathogen. Of the cropping systems tested, the vetch + black oat/maize + cowpea system showed the highest fungistasis and the oat/maize system showed the lowest. The management system also affected the genetic profile of the bacteria isolated, with the systems from fungistatic soils showing greater similarity. Although there was no clear relationship between soil management and the characteristics of the bacterial isolates, we may conclude that antibiosis and the production of siderophores were the main mechanisms accounting for fungistasis.
