The effects of genetics and environmental factors on isoflavone content of soybean (Glycine max L.) cultivars grown in different locations in Brazil in 1993/94 were evaluated. Seeds of different cultivars were analised by high performance liquid chromatography (HPLC). In Rio Grande do Sul (RS), Paraná (PR), and Mato Grosso do Sul (MS) States, a significant difference in the isoflavone total content average of the cultivars IAS 5 and FT-Abyara (163.9, 116.4 and 79.5 mg/100 g, respectively) was observed. In general, IAS 5 contained higher isoflavone than FT-Abyara. Cultivars IAS 5 and FT-Abyara grown at Vacaria, RS (28°30' S latitude) with temperature average of 19°C, had the highest isoflavone concentrations (218.7 and 163.8 mg/100 g, respectively). In Palotina, PR (24°27' S latitude), where temperature average was 24°C, the isoflavone concentrations were 105.9 and 86.8 mg/100 g, respectively. The lowest isoflavone contents were observed for FT-Estrela and FT-Cristalina, (27.6 and 46.5 mg/100 g, repectively) at Rondonópolis, MT (16°20' S latitude), where the temperature was 27°C.

A correta quantificação da concentração de lignina em plantas forrageiras, pelo método espectrofotométrico, pressupõe a existência de um padrão de referência. Um padrão de referência deve ter composição fenólica semelhante à da lignina da parede celular. O objetivo deste trabalho foi verificar se ligninas extraídas com solução ácida de dioxano para serem utilizadas como padrão de referência, apresentariam variação na composição fenólica da mesma maneira que a lignina da parede celular. Amostras de parede celular de "bromegrass", milho e trevo-vermelho foram submetidas ao método para extração de ligninas com solução ácida de dioxano. A composição fenólica das ligninas foi analisada mediante oxidação alcalina pelo nitrobenzeno com posterior separação dos componentes monoméricos via HPLC. As ligninas extraídas confirmaram a existência de variação na composição fenólica da mesma maneira às ligninas intactas presentes na parede celular e de substancial presença de ácidos cinâmicos nas ligninas de gramíneas. No que diz respeito à composição fenólica, ligninas extraídas com solução ácida de dioxano podem ser consideradas potenciais padrões de referência paras as análises espectrofotométricas.

The objective of this work was to investigate glyphosate adsorption by soils and its relationship with unoccupied binding sites for phosphate adsorption. Soil samples of three Chilean soils series - Valdivia (Andisol), Clarillo (Inceptisol) and Chicureo (Vertisol) - were incubated with different herbicide concentrations. Glyphosate remaining in solution was determined by adjusting a HPLC method with a UV detector. Experimental maximum adsorption capacity were 15,000, 14,300 and 4,700 mg g¹ for Valdivia, Clarillo, and Chicureo soils, respectively. Linear, Freundlich, and Langmuir models were used to describe glyphosate adsorption. Isotherms describing glyphosate adsorption differed among soils. Maximum adjusted adsorption capacity with the Langmuir model was 231,884, 17,874 and 5,670 mg g-1 for Valdivia, Clarillo, and Chicureo soils, respectively. Glyphosate adsorption on the Valdivia soil showed a linear behavior at the range of concentrations used and none of the adjusted models became asymptotic. The high glyphosate adsorption capacity of the Valdivia soil was probably a result of its high exchangeable Al, extractable Fe, and alophan and imogolite clay type. Adsorption was very much related to phosphate dynamics in the Valdivia soil, which showed the larger unoccupied phosphate binding sites. However relationship between unoccupied phosphate binding sites and glyphosate adsorption in the other two soils (Clarillo and Chicureo) was not clear.

O objetivo deste trabalho foi determinar a composição de aminoácidos em grãos de cultivares de feijão e a presença de interação cultivares x locais, nos teores de aminoácidos, e identificar cultivares para uso direto na alimentação e derivações em programas de melhoramento. Os aminoácidos foram determinados por cromatografia líquida de alta performance (HPLC), em grãos de 19 cultivares de feijão cultivadas em dois locais. Os dados médios obtidos para cada cultivar, em duplicata, foram comparados entre si com a utilização do teste t, a 5% de probabilidade para cada local. Os grãos das cultivares analisadas apresentaram teores de aminoácidos essenciais e não-essenciais, adequados às necessidades diárias de um indivíduo adulto, o que indica alta qualidade da proteína do feijão. As cultivares de feijão apresentam, em ordem decrescente, os aminoácidos essenciais: leucina, lisina, fenilalanina, valina, isoleucina, treonina, histidina e metionina; e os aminoácidos não-essenciais: ácido glutâmico, ácido aspártico, arginina, serina, alanina, glicina, tirosina, prolina e cisteína. Os teores de leucina, isoleucina, histidina, valina, treonina, glicina e alanina foram afetados pela interação cultivares x locais. A cultivar Iraí apresenta composição de aminoácidos adequada e é indicada para uso em dietas e derivações em programas de melhoramento.

The objective of this work was to determine the contents of methylxanthines, caffeine and theobromine, and phenolic compounds, chlorogenic and caffeic acids, in 51 mate progenies (half-sib families) and estimate the heritability of genetic parameters. Mate progenies were from five Brazilian municipalities: Pinhão, Ivaí, Barão de Cotegipe, Quedas do Iguaçu, and Cascavel. The progenies were grown in the Ivaí locality. The contents of the compounds were obtained by high performance liquid chromatography (HPLC). The estimation of genetic parameters by the restricted maximum likelihood (REML) and the prediction of genotypic values via best linear unbiased prediction (BLUP) were obtained by the Selegen - REML/BLUP software. Caffeine (0.248-1.663%) and theobromine (0.106-0.807%) contents were significantly different (p<0.05) depending on the region of origin, with high individual heritability (ĥ²>0.5). The two different progeny groups determined for chlorogenic (1.365-2.281%) and caffeic (0.027-0.037%) acid contents were not significantly different (p<0.05) depending on the locality of origin. Individual heritability values were low to medium for chlorogenic (ĥ²<0.4) and caffeic acid (ĥ²<0.3). The content of the compounds and the values of genetic parameters could support breeding programs for mate.

The objective of this work was to evaluate the biochemical composition of six berry types belonging to Fragaria, Rubus, Vaccinium and Ribes genus. Fruit samples were collected in triplicate (50 fruit each) from 18 different species or cultivars of the mentioned genera, during three years (2008 to 2010). Content of individual sugars, organic acids, flavonols, and phenolic acids were determined by high performance liquid chromatography (HPLC) analysis, while total phenolics (TPC) and total antioxidant capacity (TAC), by using spectrophotometry. Principal component analysis (PCA) and hierarchical cluster analysis (CA) were performed to evaluate the differences in fruit biochemical profile. The highest contents of bioactive components were found in Ribes nigrum and in Fragaria vesca, Rubus plicatus, and Vaccinium myrtillus. PCA and CA were able to partially discriminate between berries on the basis of their biochemical composition. Individual and total sugars, myricetin, ellagic acid, TPC and TAC showed the highest impact on biochemical composition of the berry fruits. CA separated blackberry, raspberry, and blueberry as isolate groups, while classification of strawberry, black and red currant in a specific group has not occurred. There is a large variability both between and within the different types of berries. Metabolite fingerprinting of the evaluated berries showed unique biochemical profiles and specific combination of bioactive compound contents.

The objective of this work was to evaluate the influence of different combinations of grape cultivars and rootstocks on chemical characteristics of grape juices. Six treatments were evaluated, consisting of combinations between the Isabel Precoce and BRS Cora grape cultivars and the 'IAC 766', 'IAC 313', and 'IAC 572' rootstocks. Approximately 10 L of juice were obtained per treatment. Analyses of color, total soluble solids content, pH, anthocyanins, total phenolics, total sugars, and quantification and identification of biogenic amines by HPLC were performed. Biogenic amines, such as putrescine, cadaverine, spermidine, and spermine, were found in all evaluated cultivars. By principal component analysis (PCA), treatments can be divided into two groups, according to the cultivar. Juices obtained from 'Isabel Precoce' are characterized by higher levels of total sugar content and soluble solids; however, juices from 'BRS Cora' are positively correlated with phenolic content, anthocyanins, and color and acidity parameters. The differences found by PCA for juices from the Isabel Precoce and BRS Cora cultivars indicate that, regardless of the rootstock used, the most important factor in the chemical characterization of juices is the grape cultivar.

O objetivo deste trabalho foi identificar e quantificar os açúcares em frutos de pessegueiro da variedade 'Biuti', armazenados sob condições de ambiente (27,3ºC; 70% UR) e sob refrigeração (4ºC; 90%), comparar as diferenças entre os teores de açúcares nas duas condições de armazenamento. A identificação e a quantificação precisa dos açúcares (glicose, frutose e sacarose) foi realizada por cromatografia em fase líquida HPLC. Verificou-se que a sacarose foi o açúcar encontrado em maior quantidade, sendo verificado apenas traços de glicose e frutose em alguns frutos. Sob condições ambientais, os teores de sacarose do 3º até o 9º dia de armazenamento não diferiram significativamente entre si; entretanto, no 12º dia, os frutos obtiveram baixos teores de sacarose, pois os mesmos já estavam em processo de senescência. Sob refrigeração, o aumento nos teores de açúcares dos frutos ocorreu gradativamente durante todo o armazenamento, e ao final do mesmo, verificaram-se os maiores teores de sacarose, frutose e glicose, os quais eram mais elevados do que os encontrados nos frutos armazenados sob condições ambientais.

Mauritia vinifera (buriti) is a palm tree that grows wild in different areas of Brazil, particularly in the Amazonian region. The buriti oil is rich in carotenoids, especially in β-carotene. The growing interest in other natural sources of β-carotene has stimulated the industrial use of buriti as a raw material for pulp oil extraction. Most processes are based on the conventional technologies, involving drying and pressing the pulp for oil recovery and further separation of carotenoids in a liquid phase using organics solvents. In the present work, the ethanol-based process was evaluated for simultaneous carotenoids recovering and fractionating from buriti pulp. The raw material and ethanol, 1:4 ratio, were placed in an erlenmeyer flask and maintained at 30rpm for 1 hour in a temperature-controlled bath at 65ºC. The mixture was filtered under vacuum and cooling at 10ºC to allow for the separation of the solvent in two phases. Carotenoids composition, determined by HPLC, has indicated a β-carotene concentration about 12 times greater in the lower phase than in the upper phase. The profile of the carotenoids in the denser phase is quite similar to that of raw buriti oil, and the concentration of total carotenoids is 40% higher than that of the original raw oil, making the ethanol-based process particularly attractive for industrial applications.

O objetivo foi avaliar, por HPLC, a composição qualitativa e quantitativa de carotenoides em maracujás do cerrado. Frutos procedentes de acessos nativos de quatro espécies (Passiflora cincinnata, P. nitida, P. setacea e P. edulis) foram analisados, utilizando, como referência, o maracujá-amarelo comercial (P. edulis). As polpas de maracujá apresentaram neoxantina, violaxantina, cis-violaxantina, anteraxantina, luteína, zeaxantina, β-criptoxantina, poli-cis-caroteno, prolicopeno, cis-ζ-caroteno, trans-ζ-caroteno, trans-β-caroteno, 13-cis-β-caroteno e fitoflueno. Em geral, os teores de carotenoides entre as espécies e entre os acessos da mesma espécie foram significativamente diferentes. A espécie P. edulis apresentou o maior número de carotenoides, com diferença entre os acessos. Em um acesso de P. edulis comercial, foi encontrado o trans-β-caroteno como o carotenoide principal (7,8±0,8 µg g-1) e no outro o trans-ζ-caroteno (11,4±0,4 µg g-1). Dois acessos de P. edulis nativos do Cerrado apresentaram cis-ζ-caroteno como carotenoide majoritário (6,28±0,15 µg g-1 e 12,1±0,7 µg g-1, casca amarela e roxa, respectivamente). O perfil de carotenoides em frutos de espécies de maracujá apresentou diversidade de composição, com potencial de uso para melhoramento genético para agregar maior valor ao produto e estimular o seu consumo.

Pereskia aculeata Mill. (Ora-pro-nóbis) is a native cactaceae from tropical America, whose leaves have high protein content. In Brazil it is found in all territorial extension between the states of Bahia and Rio Grande do Sul. Most studies have focused on chemical characterization of the leaves of this specie. The objective was to assess the carotenoids profile and the total polyphenols present in the fruits of P. aculeate. Carotenoids were determined by HPLC-PAD (high performance liquid chromatography - photodiode array detector), total polyphenols were determined by Folin-Ciocalteu and vanillin methods. Trans-β-carotene was the main carotenoid, followed by α-carotene, lutein and other minor carotenoids. It was found 64.9 ± 1.1 mg.100g-1 of gallic acid equivalent, 14.8 ± 0.2 mg.100g-1 of catechin equivalent. Carotenoid identification of P. aculeate fruits are presented here by the first time and indicate that these fruits can be researched as source of bioactive substances, especially antioxidant and provitamin A carotenoids.

O desenvolvimento de tecnologias que permitam o aproveitamento integral dos frutos é de grande importância para as indústrias. Uma possibilidade de valorização da cultura do abacaxi seria o estímulo ao processamento do fruto para a obtenção do suco associado ao aproveitamento das cascas para a elaboração de farinha, produto de mercado crescente. Um dos fatores de grande importância para a aceitação do suco é o sabor, que se encontra diretamente relacionado ao perfil de ácidos e açúcares presentes. Nesta linha, este trabalho visou a processar frutos de abacaxi cv Smooth Cayenne colhidos em dois estádios de maturação para a obtenção e a caracterização do suco e da farinha de cascas. Os sucos obtidos foram analisados em HPLC para o perfil de açúcares e ácidos orgânicos. As farinhas de casca foram analisadas para rendimento, umidade, cinzas, fibras solúvel e insolúvel, lipídeos, proteína e carboidratos totais. Os resultados obtidos mostraram que frutos em estádio mais avançado de maturação produzem sucos com maior teor de açúcares e menor teor de ácidos, sendo o principal ácido o cítrico, e os açúcares frutose, glicose e sacarose. As farinhas de abacaxi apresentam-se como fontes de fibras insolúveis.

Several species of Annona (Annonaceae) are used in traditional Mexican medicine by their anti-anxiety, anticonvulsant and tranquilizing properties. It has been reported that the alkaloids isolated from some species of the Annona have affinity to serotonergic 5-HT1A receptors and modulate dopaminergic transmission, which is involved in depressive disorders. In this review it is showed the results of the antidepressant-like effect of an alkaloid extract from the aerial parts of Annona cherimola (TA) in mice. The antidepressant-like effect was evaluated in the forced swimming test. To elucidate a possible mechanism of action, experiments of synergism with antidepressant drugs, such as imipramine (IMI), clomipramine (CLIMI), and fluoxetine (FLX), were carried out. The neurotransmitter content (DA: dopamine, 5HT: serotonin and its metabolites, HVA: homovanillic acid and 5HIAA:5-hydroxyindoleacetic) in the whole brain of mice were also determined by HPLC method. The results showed that repeated treatment with TA produced antidepressant-like effects in mice. This effect was not related to an increase in locomotor activity. Administration of TA facilitated the antidepressant effect of IMI and CLIMI as well as increased the turnover of DA and 5-HT. The alkaloids: 1,2-dimethoxy-5, 6.6 to 7-tetrahydro-4H-dibenzoquinoline-3,8,9,10-tetraol, anonaine, liriodenine, and nornuciferine were the main constituents of TA.

The content of digoxin and lanatoside C (jointly quantified), lanatoside A, lanatoside B, glucoevatromonoside, odorobioside G, glucogitoroside, glucoverodoxine, glucodigifucoside and digitalinum verum was determined by HPLC in Digitalis lanata harvested in Brazil, as well as in clones industrially employed for the production of cardenolides. The Brazilian plants presented greater variation in the contents of cardenolides than the analyzed clones. Lanatoside C and digoxin concentrations were higher in the clones (6120±640 nmol/g dry leaf) than in the Brazilian plants (1820±900 nmol/g dry leaf). The concentrations of these glycosides were found to be within the range described for native species and also were similar to an European cultivar, what makes possible its industrial use for the cardenolides production.
