Growth of seedlings of fifteen tropical tree species representative, at the adult stage, of different successional positions, was studied under field conditions. Seedlings were grown in three treatments: full sun (FS), artificial shade imposed by neutral screens (AS) and natural shade imposed by a closed canopy in a Forest Reserve in Southeast Brazil (NS). Most of the studied species survived in both shade treatments, although their growth was severely affected. Decreases in height, internode numbers, dry weight, leaf area, root:shoot ratio (R:S) and increases in leaf mass ratio (LMR), leaf area ratio (LAR) and specific leaf area (SLA) were common responses to shade. Relative growth rates (RGRs) and net assimilation rates (NARs) were consistently lower in the shaded treatments than in full sun. RGR was significantly correlated with NAR in the FS and NS treatments, whereas it was correlated with LAR in the AS treatment. Natural shade had more severe effects than artificial shade on leaf area reduction and RGR. Between-species differences in R:S, LMR, SLA and LAR were not related to the successional status of species. However, there was a tendency for early-successional species to have higher RGRs than late successional ones, regardless of the light environment. Late-successional species also showed less pronounced responses to shade than early ones. The characteristics presented by the late-successional species may be associated with shade tolerance, enabling their persistence under dense canopies.

O objetivo deste artigo é mostrar como pessoas auto-identificadas como fãs de Gilda, colaboram, por meio de diversas práticas, no processo de consagração da cantora falecida. Respeitando a diferenciação nativa entre fã e devoto, veremos como Gilda é inscrita em uma textura diferencial do mundo-habitado que denominaremos de sagrado, a partir de performances que nao poderiam facilmente se circunscrever à esfera da religião. Ao invés de serm consideradas uma metáfora religiosa, as práticas dos fãs configuram modos diferentes de consagrar Gilda.

The objectives of the present study were to identify the cis-elements of the promoter absolutely required for the efficient rat NHE3 gene transcription and to locate positive and negative regulatory elements in the 5’-flanking sequence (5’FS), which might modulate the gene expression in proximal tubules, and to compare this result to those reported for intestinal cell lines. We analyzed the promoter activity of different 5’FS segments of the rat NHE3 gene, in the OKP renal proximal tubule cell line by measuring the activity of the reporter gene luciferase. Because the segment spanning the first 157 bp of 5’FS was the most active it was studied in more detail by sequential deletions, point mutations, and gel shift assays. The essential elements for gene transcription are in the region -85 to -33, where we can identify consensual binding sites for Sp1 and EGR-1, which are relevant to NHE3 gene basal transcription. Although a low level of transcription is still possible when the first 25 bp of the 5’FS are used as promoter, efficient transcription only occurs with 44 bp of 5’FS. There are negative regulatory elements in the segments spanning -1196 to -889 and -467 to -152, and positive enhancers between -889 and -479 bp of 5’FS. Transcription factors in the OKP cell nuclear extract efficiently bound to DNA elements of rat NHE3 promoter as demonstrated by gel shift assays, suggesting a high level of similarity between transcription factors of both species, including Sp1 and EGR-1.

Psychological factors can be correlated with temporomandibular disorders (TMDs), but the mechanisms are unknown. In the present study, we examined the microstructural changes and expression of proinflammatory cytokines in mandibular condylar cartilage of the temporomandibular joint (TMJ) in a psychological stress animal model. Male Sprague-Dawley rats (8 weeks old, 210 ± 10 g) were randomly divided into 3 groups: psychological stress (PS, N = 48), foot shock (FS, N = 24), and control (N = 48). After inducing psychological stress using a communication box with the FS rats for 1, 3, or 5 weeks, PS rats were sacrificed and compared to their matched control littermates, which received no stress and were killed at the same times as the PS rats. Body and adrenal gland weight were measured and corticosterone and adrenocorticotropic hormone levels were determined by radioimmunoassay. After hematoxylin-eosin staining for histological observation, the ultrastructure of the TMJ was examined by scanning electron microscopy. Transcription and protein levels of interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α) were evaluated by ELISA and semi-quantitative RT-PCR. The PS group showed a significantly higher adrenal gland weight after 3 weeks of stress and higher hormone levels at weeks 1, 3, and 5. Histopathological changes and thinning cartilage were apparent at weeks 3 and 5. In the PS group, TNF-α increased at 1, 3, and 5 weeks and IL-1β increased significantly after 1 and 3 weeks of stress, and then decreased to normal levels by 5 weeks. Psychological stress increased plasma hormone levels and RT-PCR indicated increased IL-1β and TNF-α expression in the TMJ in a time-dependent manner. These results suggest that cytokine up-regulation was accompanied by stress-induced cartilage degeneration in the mandibular condyle. The proinflammatory cytokines play a potential role in initiating the cartilage destruction that eventually leads to the TMDs.

Low-level lasers are used at low power densities and doses according to clinical protocols supplied with laser devices or based on professional practice. Although use of these lasers is increasing in many countries, the molecular mechanisms involved in effects of low-level lasers, mainly on DNA, are controversial. In this study, we evaluated the effects of low-level red lasers on survival, filamentation, and morphology of Escherichia colicells that were exposed to ultraviolet C (UVC) radiation. Exponential and stationary wild-type and uvrA-deficientE. coli cells were exposed to a low-level red laser and in sequence to UVC radiation. Bacterial survival was evaluated to determine the laser protection factor (ratio between the number of viable cells after exposure to the red laser and UVC and the number of viable cells after exposure to UVC). Bacterial filaments were counted to obtain the percentage of filamentation. Area-perimeter ratios were calculated for evaluation of cellular morphology. Experiments were carried out in duplicate and the results are reported as the means of three independent assays. Pre-exposure to a red laser protected wild-type and uvrA-deficient E. coli cells against the lethal effect of UVC radiation, and increased the percentage of filamentation and the area-perimeter ratio, depending on UVC fluence and physiological conditions in the cells. Therapeutic, low-level red laser radiation can induce DNA lesions at a sub-lethal level. Consequences to cells and tissues should be considered when clinical protocols based on this laser are carried out.

Semiconductor laser devices are readily available and practical radiation sources providing wavelength tenability and high monochromaticity. Low-intensity red and near-infrared lasers are considered safe for use in clinical applications. However, adverse effects can occur via free radical generation, and the biological effects of these lasers from unusually high fluences or high doses have not yet been evaluated. Here, we evaluated the survival, filamentation induction and morphology of Escherichia coli cells deficient in repair of oxidative DNA lesions when exposed to low-intensity red and infrared lasers at unusually high fluences. Cultures of wild-type (AB1157), endonuclease III-deficient (JW1625-1), and endonuclease IV-deficient (JW2146-1) E. coli, in exponential and stationary growth phases, were exposed to red and infrared lasers (0, 250, 500, and 1000 J/cm2) to evaluate their survival rates, filamentation phenotype induction and cell morphologies. The results showed that low-intensity red and infrared lasers at high fluences are lethal, induce a filamentation phenotype, and alter the morphology of the E. coli cells. Low-intensity red and infrared lasers have potential to induce adverse effects on cells, whether used at unusually high fluences, or at high doses. Hence, there is a need to reinforce the importance of accurate dosimetry in therapeutic protocols.

Isoflavonas, associadas ao consumo de proteína de soja, constituem um grupo de substâncias envolvidas em atividades anti-carcinogênicas, na redução da perda de massa óssea e na diminuição do colesterol do sangue. A concentração de isoflavonas foi determinada em produtos comerciais à base de soja produzidos no Brasil. Foram analisados cinco farinhas de soja (FS), quatro proteínas texturizadas (PTS), dois extratos hidrossolúveis (EH) e quatro formulados infantis (FI). A extração foi com etanol e ácido acético e a quantificação e identificação das isoflavonas foram realizadas por CLAE. Em farinha de soja e em proteína texturizada predominaram os compostos malonil-conjugados. Farinha de soja (96mg de agliconas equivalentes/100g) e PTS (70mg de agliconas equivalentes/100g), obtidas a partir da mesma cultivar apresentaram diferenças na concentração e distribuição dos isômeros devido ao processamento. Extratos hidrossolúveis e formulados infantis apresentaram agliconas, variando de 8% a 28% do total de isoflavonas, mas os principais isômeros foram os beta-glicosídios. O consumo diário de 25g de proteína de farinha de soja integral ou de PTS contribuem com mais de 50mg de isoflavonas totais na dieta. Em formulados infantis a concentração de isoflavonas totais foi menor que nos demais produtos devido à adição de ingredientes não derivados de soja.

Dentre os conceitos modernos de controle de pragas, o uso de inseticidas no tratamento de sementes constitui-se num dos métodos mais eficientes. Deve-se, entretanto, conhecer a influência desses produtos com relação à qualidade fisiológica das sementes tratadas. Objetivou-se através desta pesquisa avaliar a qualidade fisiológica de sementes de soja tratadas com inseticidas, sob quatro períodos de armazenamento (0, 15, 30 e 45 dias após o tratamento). O delineamento experimental foi o inteiramente casualizado, em esquema fatorial 4 x 7, com quatro repetições, onde as sementes do cultivar M-SOY 6101 foram tratadas com os inseticidas thiamethoxam (Cruiser 700 WS), na dose de 250 mL de produto comercial (p.c.)/100 kg de sementes; fipronil (Standak), na dose de 150 mL de p.c./100 kg de sementes; imidacloprid (Gaucho FS), na dose de 150 ml de p.c./100 kg de sementes; [imidacloprid + thiodicarb] (CropStar FS), na dose de 0,3 L. ha-1; carbofuran (Furadan 350 TS), na dose de 1,5 L/100 kg de sementes; acefato (Orthene 750 BR), na dose de 1 kg/ 100 kg de sementes e uma testemunha, sem tratamento. As variáveis analisadas foram: germinação, velocidade de emergência, comprimento de raiz e de plântula e porcentagem de plântulas normais no teste de envelhecimento acelerado. A aplicação dos inseticidas carbofuran e acefato é prejudicial à qualidade fisiológica das sementes de soja cultivar M-SOY 6101, por um período de armazenamento de até 45 dias. A redução da qualidade fisiológica das sementes, condicionada pelos inseticidas avaliados, intensifica-se com o prolongamento do armazenamento das sementes tratadas, recomendando-se, portanto, que o tratamento inseticida das sementes seja realizado próximo ao momento da semeadura.

Novos produtos para incorporação às sementes vêm sendo disponibilizados anualmente. Bioativadores, como o inseticida tiametoxam, são utilizados para aumentar o potencial produtivo das plantas, através de modificações no metabolismo vegetal. Objetivou-se neste trabalho avaliar os efeitos do tiametoxam na qualidade fisiológica de sementes de algodoeiro, particularmente no crescimento radicular de plântulas de algodoeiro e estabelecer concentrações do produto para pré-tratamento de sementes. Foram utilizados três lotes de sementes de algodoeiro, cultivar CD 408, tratadas com Cruiser® 350 FS contendo 35 gramas de ingrediente ativo (i.a.) de tiametoxam por litro, sendo, submetidas a seis doses: 0, 2, 4, 6, 8 e 10 mL de Cruiser® 350 FS/kg de semente. As avaliações utilizadas foram: germinação, primeira contagem de germinação, envelhecimento acelerado, comprimento e massa de matéria seca do sistema radicular, índice de velocidade de emergência e emergência final de plântulas. O tratamento de sementes de algodoeiro com tiametoxam favorece positivamente a qualidade fisiológica das sementes, e as doses de 5,0 a 7,0 mL de Cruiser® 350 FS/kg de semente mostram-se mais eficientes em melhorar o desempenho fisiológico das sementes de algodoeiro.

As sementes de amendoim são consideradas de difícil produção, pois apresentam frequentemente índices insatisfatórios de germinação e de vigor devido a muitos fatores, dentre eles, problemas associados à ocorrência de fungos e à alta sensibilidade às injúrias mecânicas. Assim, o tratamento das sementes com fungicida, juntamente com inseticidas, visando o controle de pragas iniciais na cultura, é recomendável. Na avaliação do potencial fisiológico de sementes de amendoim, além dos métodos rotineiramente utilizados, a análise computadorizada de plântulas tem grande potencial, pois é método rápido, preciso e sem erros de interpretação humana. O presente trabalho teve como objetivo avaliar a qualidade fisiológica de sementes de amendoim tratadas com fungicida ou inseticida, bem como a associação de ambos, utilizando-se a técnica de análise computadorizada de plântulas. Sementes do cultivar IAC 886 não tratadas e tratadas com o fungicida Maxim XL (Fludioxonil + Metalaxyl), com o inseticida Cruiser 350 FS (Tiametoxam) e com a mistura de ambos os produtos foram submetidas aos períodos de armazenamento de 0, 15, 30, 45, 60, 90 e 180 dias (20 °C, 50% UR do ar). Após cada período de armazenamento, as sementes foram submetidas à avaliação do grau de umidade, germinação, envelhecimento acelerado, emergência de plântulas em campo, condutividade elétrica e sanidade, além da análise computadorizada de plântulas. Concluiu-se que a análise computadorizada de plântulas apresenta eficiência equivalente aos testes de envelhecimento acelerado e de emergência de plântulas em campo na identificação de diferenças entre tratamentos com ausência ou presença de fungicida e de fungicida associado a inseticida nas sementes de amendoim.
