Eight bufadienolides were successfully isolated and purified from ChanSu by high-speed counter-current chromatography (HSCCC) combined with preparative HPLC (prep-HPLC). First, a stepwise elution mode of HSCCC with the solvent system composed of petroleum ether - ethyl acetate - methanol - water (4:6:4:6, 4:6:5:5, v/v) was employed and four bufadienolides, two partially purified fractions were obtained from 200 mg of crude extract. The partially purified fractions III and VI were then further separated by prep-HPLC, respectively, and another four bufadienolides were recovered. Their structures were confirmed by ESI-MS and ¹H-NMR spectra.

An efficient method for the rapid separation and purification of polyphenols from artichoke by polyamide column chromatography in combination with high-speed counter-current chromatography (HSCCC) was successfully built. The crude ethanol extracts from dry artichoke were first pre-separated by polyamide column chromatography and divided in two parts as sample 1 and sample 2. Then, the samples were further separated by HSCCC and yielded 7.8 mg of chlorogenic acid (compound I), 24.5 mg of luteolin-7-O-β-D-rutinoside (compound II), 18.4 mg of luteolin-7-O-β-D-glucoside (compound III), and 33.4 mg of cynarin (compound IV) with purity levels of 92.0%, 98.2%, 98.5%, and 98.0%, respectively, as determined by high-performance liquid chromatography (HPLC) method. The chemical structures of these compounds were identified by electrospray ionization-mass spectrometry (ESI-MS) and nuclear magnetic resonance (NMR).

An improved method based on reverse flow injection is proposed for determining sulfate concentration in the wet-process of phosphoric acid (WPA). The effect of reagent composition, flow rate, temperature, acid concentration, length of the reaction coil, and linear response range on the flow system is discussed in detail. Optimal conditions are established for determining sulfate in the WPA samples. Baseline drift is avoided by a periodic washing step with EDTA in an alkaline medium. A linear response is observed within a range of 20 - 360 mg L-1, given by the equation A = 0.0020C (mg L-1) + 0.0300, R² = 0.9991. The detection limit of the proposed method for sulfate analysis is 3 mg L-1, and the relative standard deviation (n = 12) of sulfate absorbance peak is less than 1.60%. This method has a rate of up to 29 samples per hour, and the results compare well with those obtained with gravimetric method.

OBJETIVO: Descrever os critérios para formação de grupos no internato e a avaliação que os alunos fazem do relacionamento grupal, analisando as variáveis associadas. MÉTODO: Trata-se de um estudo transversal, que utilizou questionário autopreenchido, com perguntas sobre dados sociodemográficos, autoavaliação do desempenho escolar, critérios para formação e satisfação com grupo de internato. Sofrimento psíquico foi avaliado a partir do Self Report Questionnaire. Utilizou-se o teste do qui-quadrado e regressão logística para análise multivariada. RESULTADOS:A taxa de resposta no internato foi de mais de 90%. A maioria dos alunos utilizou critérios ligados à rede social (82,6%) e predominaram sujeitos satisfeitos com seu grupo (81,2%). Após análise multivariada, apenas autoavaliação do desempenho escolar "boa ou ótima" e critérios de escolha relacionados à rede de apoio se mantiveram associados à satisfação com o grupo. CONCLUSÕES: Apesar de ser um estágio da formação profissional, os alunos se escolheram por questões ligadas à rede social. Sendo uma profissão na qual o trabalho em equipe é inerente, deveriam ser criadas estratégias durante o curso médico para elaborar as dificuldades de relacionamento grupal entre alunos de Medicina.

This report describes a leiomyoma of the inferior third section of the esophagus removed during laparoscopic cholecystectomy. The patient is a woman 55-years-age, carrying esophageal myoma of 40 mm in diameter wide, situated in the posterior wall of the lower esophagus. Indications for surgery were based mainly on the growth of the mass (6 mm when discovered 7 years previously, increased to 40 mm). Recently the patient returned suffering from pain, which could be attributed to his litiasic cholecystopaty. A small degree of low disphagia could also be observed. Radiologic imaging, direct endoscopic examination and endoscopic ultrasound showed that the mioma protruded on to the oesophagic lumen, discreetly diminishing there. A laparoscopic esophageal myomectomy was indicated at the same session of the laparoscopic cholecystectomy. Once the pneunoperitoneum was installed, five ports were placed as if for a hiatus hernia surgery. The cholecystectomy was uneventful. Next, an esophagoscopy was performed so as to determine the precise area covering the base of the tumour; at the right-lateral site. Longitudinal and circular fibres of the esophagus was severed over the lesion and the enucleation of the tumour was performed alternating the monopolar dissection, bipolar and hidrodisection. Control-endoscopy was carried out to verify mucosa integrity. Four suture points with poliglactine 3-0 string so as to close the musculature followed this. One suture was placed in for diminution of the size of the esophagean hiatus. Total time of intervention: two hours (30m for the cholecystectomy and one hour and thirty minutes for the myomectomy). Postoperative period: uneventful. Disappearance of the disphagia was observed. Radiologic transit control with water-soluble contrast at 4th post-operative day: good passage. Diagnosis from laboratory of pathology: conjunctive tumour formed by muscle non-striated cells: leiomyoma. The patient was re-examined on the two-month postoperative follow-up. General conditions were good and there were no complain of dysphagia. Neither there were any symptoms of gastro-esophageal reflux.

OBJETIVO: aplicar técnicas de geoprocessamento para a análise espacial do perfil de nascimentos por município. MÉTODOS: foi um estudo ecológico e exploratório com dados do Sistema de Informações de Saúde sobre nascidos vivos de 2004 e utilizando técnicas de geoprocessamento. Foram estimadas as autocorrelações espaciais das variáveis: parto cesáreo, escolaridade materna, baixo peso ao nascer, escore de Apgar de cinco minutos, prematuridade, número de consultas e mães adolescentes, além do mapa com o índice de desenvolvimento humano. Foi utilizada a estatística I de Moran para detecção de agregados de eventos no espaço, pelo programa TerraView 3.1.3 (desenvolvido pelo Instituto Nacional de Pesquisas Espaciais - INPE e de acesso público). Foram construídos mapas espaciais com estas variáveis e estimados os coeficientes de correlação de Pearson. RESULTADOS: os resultados mostraram que as proporções de nascidos vivos de mães com escolaridade acima de primeiro grau e de cesáreas, apresentaram padrão espacial visualmente identificável e autocorrelação espacial significativa. O baixo peso ao nascer, a prematuridade, o índice de Apgar, o número de consultas de pré-natal e o de mães adolescentes apresentaram padrão espacial aleatório, demonstrando que, nesta escala de análise, estes indicadores não discriminaram grupos de risco, apesar do seu inquestionável valor preditivo para morbimortalidade infantil em nível individual. Houve correlação positiva entre parto cesáreo e escolaridade, e entre parto cesáreo e índice de desenvolvimento humano e correlação negativa entre mães adolescentes e índice de desenvolvimento humano, com significância estatística (p<0,05). CONCLUSÕES: esta metodologia permitiu identificar aglomerados espaciais para as variáveis parto cesáreo e escolaridade materna, além de aprofundar o conhecimento sobre o perfil de nascimentos nos municípios, apresentando potencial no direcionamento de ações voltadas para áreas específicas.

OBJETIVO: elaborar e validar um questionário para medir os diferentes domínios da função sexual nas mulheres brasileiras pós-menopausa. MÉTODOS: participaram deste estudo 251 mulheres entre 2 e 15 anos após menopausa. Foi examinada a reprodutibilidade/confiabilidade do instrumento pelos coeficientes de correlação linear, intraclasse e concordância de Lin. A consistência interna foi examinada pelo coeficiente α de Cronbach. A verificação da face, conteúdo e construto seguiram diretrizes teóricas clássicas para validação de testes. RESULTADOS: foi gerado um instrumento inicial com 57 itens estruturados em nove domínios. Foram eliminados 14 (24,5%) questões por baixa correlação com a escala total ou pequeno poder de discriminação. A forma final com 43 itens demonstrou excelente reprodutibilidade (r=0,719; IC95%=0,690-0,750; pc=0,887; IC95%=0,850-0,930; p<0,001) e confiabilidade. A consistência interna da escala foi excelente (α de Cronbach 0,951) e cerca de 60% dos avaliadores confirmaram as validades de face e conteúdo. O exame da validade de construto mostrou adequação do instrumento (α=0,951). CONCLUSÕES: concluiu-se que o questionário proposto é adequado para examinar a função sexual feminina em mulheres pós-menopausa.

PURPOSE: To examine the expression of AKT and PTEN in a series of HER2-positive primary invasive breast tumors using immunohistochemistry, and to associate these expression profiles with classic pathologic features such as tumor grade, hormone receptor expression, lymphatic vascular invasion, and proliferation.METHODS: A total of 104 HER2-positive breast carcinoma specimens were prepared in tissue microarrays blocks for immunohistochemical detection of PTEN and phosphorylated AKT (pAKT). Original histologic sections were reviewed to assess pathological features, including HER2 status and Ki-67 index values. The associations between categorical and numeric variables were identified using Pearson's chi-square test and the Mann-Whitney, respectively.RESULTS: Co-expression of pAKT and PTEN was presented in 59 (56.7%) cases. Reduced levels of PTEN expression were detected in 20 (19.2%) cases, and these 20 tumors had a lower Ki-67 index value. In contrast, tumors positive for pAKT expression [71 (68.3%)] were associated with a higher Ki-67 index value.CONCLUSION: A role for AKT in the proliferation of HER2-positive breast cancers was confirmed. However, immunohistochemical detection of PTEN expression did not correlate with an inhibition of cellular proliferation or control of AKT phosphorylation, suggesting other pathways in these mechanisms of control.

O presente estudo investigou o uso da cito-(73,0%), Arcanobacterium pyogenes (6,0%), Streptococlogia aspirativa com agulha fina no diagnóstico da lin-cus spp. β hemolítico (5,0%) e Escherichia coli (4,0%) fadenite em ovinos e a ocorrência de microrganismos foram os microrganismos mais frequentes nos animais nos linfonodos com lesões, com ênfase no isolamento de com linfadenite. Streptococcus spp. (21,0%) e Staphylo-Corynebacterium pseudotuberculosis. Foram utilizados coccus spp. (7,0%) foram as bactérias isoladas com 100 linfonodos de ovinos com aumento de volume su-maior frequência nos linfonodos sem lesões colhidos em gestivos de linfadenite e 100 linfonodos de ovinos sem abatedouro. A punção aspirativa com agulha fina perlesões, colhidos em abatedouro. C. pseudotuberculosis mitiu identificar microrganismos "corineformes" em 79 (79,0%) animais com linfadenite e, destes, 73 (73,0%) foram identificados como C. pseudotuberculosis. Nenhuma linhagem de C. pseudotuberculosis foi isolada dos linfonodos dos animais sem lesões. Concluiu-se que C. pseudotuberculosis foi o microrganismo mais freqüente nos ovinos com linfadenite, e que a citologia aspirativa pode ser utilizada como método de triagem no diagnóstico da linfadenite caseosa ovina

O controle de plantas daninhas e a tolerância de gladíolos, cultivares Spick and Span e White Friendship, a tratamentos herbicidas foi estudado em um experimento de campo em solo areno-argiloso. Foram aplicados os herbicidas trifluralin e EPTC em pré-plantio com incorporação e chloroxuron, DCPA, diphenamid, pendimethalin e diuron em pré -emergência. Houve maior incidência de gramíneas que foram controladas por todos os herbicidas, exceto chloroxuron, sendo o controle pelo EPTC de 83% e pelos demais, acima de 90%. Para as dicotiledóneas, o controle foi na seguinte ordem decrescente: pendimethalin, chloroxuron, diuron, trifluralin, EPTC, DCPA e diphenamid. O EPTC afetou a germinação de bulbos, causando, também, retenção de crescimento e atraso no ciclo. No florescimento, as hastes florais foram colhidas, medidas e pesadas, verificando-se redução significativa de peso de matéria fresca com o tratamento EPTC para o cultivar Spick and Span, e com trifluralin, EPTC, chloroxuron e pendimetha lin para o White Friendship.

Tropical high altitude grasslands present several species with both microphyllous and highly sclerophyllous leaves, and co-occur in specific soil patches, thus exposed to identical environments. In this article we describe herbivory among co-occurring microphyllous species in a tropical high altitude grassland ecosystem of Serra do Cipó, Minas Gerais state, and we tested the effect of variable anatomic traits on leaf herbivory patterns. Leaf anatomical traits were investigated for Baccharis imbricata Heering , Lavoisiera imbricata DC. and L. subulata Triana (focal species). Herbivory was measured from branches and leaves of individual plants and compared among co-occurring species within one multispecific shrub patch and among L. subulata individuals from this patch and an adjacent monospecific patch. For all present plant species and individuals we estimated the proportion of leaves with different levels of area lost. For the focal species, six leaves were sorted and taken for histological sectioning, in order to allow precise measures of defensive structures. Relative mean leaf area lost differed significantly among the six species found in the multispecific patch. Lavoisiera subulata individuals were more attacked in the multispecific than in the monospecific patch. Leaf margin protection traits in both B. imbricata and L. imbricata showed significant effect against herbivory. Data suggest that some anatomic traits have direct effect against herbivory but their effect are not clearly perceptible among branches within individual plants or among plants within the same species.

Air pollution has been associated with health effects on different age groups. The present study was designed to assess the impact of daily changes in air pollutants (NO2, SO2, CO, O3, and particle matter (PM10)) on total number of daily neonatal deaths (those that occur between the first and the 28th days of life) in São Paulo, from January 1998 to December 2000, since adverse outcomes such as neonatal deaths associated with air pollution in Brazil have not been evaluated before. Generalized additive Poisson regression models were used and nonparametric smooth functions (loess) were adopted to control long-term trend, temperature, humidity, and short-term trends. A linear term was used for holidays. The association between air pollutants and neonatal deaths showed a short time lag. Interquartile range increases in PM10 (23.3 µg/m³) and SO2 (9.2 µg/m³) were associated with increases of 4% (95% CI, 2-6) and 6% (95% CI, 4-8), respectively. Instead of adopting a two-pollutant model we created an index to represent PM10 and SO2 effects. For an interquartile range increase in the index an increase of 6.3% (95% CI, 6.1-6.5) in neonatal deaths was observed. These results agree with previous studies performed by our group showing the deleterious effects of air pollutants during the perinatal period. The method reported here represents an alternative approach to analyze the relationship between highly correlated pollutants and public health problems, reinforcing the idea of the synergic effects of air pollutants in public health.

In order to assess the effect of air pollution on pediatric respiratory morbidity, we carried out a time series study using daily levels of PM10, SO2, NO2, ozone, and CO and daily numbers of pediatric respiratory emergency room visits and hospital admissions at the Children's Institute of the University of São Paulo Medical School, from August 1996 to August 1997. In this period there were 43,635 hospital emergency room visits, 4534 of which were due to lower respiratory tract disease. The total number of hospital admissions was 6785, 1021 of which were due to lower respiratory tract infectious and/or obstructive diseases. The three health end-points under investigation were the daily number of emergency room visits due to lower respiratory tract diseases, hospital admissions due to pneumonia, and hospital admissions due to asthma or bronchiolitis. Generalized additive Poisson regression models were fitted, controlling for smooth functions of time, temperature and humidity, and an indicator of weekdays. NO2 was positively associated with all outcomes. Interquartile range increases (65.04 µg/m³) in NO2 moving averages were associated with an 18.4% increase (95% confidence interval, 95% CI = 12.5-24.3) in emergency room visits due to lower respiratory tract diseases (4-day moving average), a 17.6% increase (95% CI = 3.3-32.7) in hospital admissions due to pneumonia or bronchopneumonia (3-day moving average), and a 31.4% increase (95% CI = 7.2-55.7) in hospital admissions due to asthma or bronchiolitis (2-day moving average). The study showed that air pollution considerably affects children's respiratory morbidity, deserving attention from the health authorities.

Nasopharyngeal carcinoma (NPC) is notorious for the metastases, which are in close association with Epstein-Barr virus-encoded latent membrane protein 1 (LMP1). Arsenic trioxide (As2O3) has been shown to induce apoptosis and differentiation in NPC xenografts. Then, can it repress the cancer cells' metastasis potential? To elucidate this issue, the present study was performed. LMP1-negative cell line HNE1 and LMP1-positive cell line HNE1-LMP1 were used as in vitro model. Cells (1 x 10(5)/mL) were cultured with or without 3 µM As2O3 for 48 h. Then the survival cells were collected to investigate their potential of colony formation, attachment, invasion, and migration. Both confocal immunofluorescence staining and Western blot were used to detect the changes of LMP1 expression. The changes of MMP-9 were examined by RT-PCR assay and Western blot. The results were as follow: i) the colony formation inhibition rate (75.41 ± 3.9% in HNE1-LMP1 cells vs 37.89 ± 4.9% in HNE1 cells), the rate of attachment (HNE1-LMP1 vs HNE1: 56.40 ± 3.5 vs 65.87 ± 5.9%), the invasion inhibitory rate (HNE1-LMP1 vs HNE1: 56.50 ± 3.7 and 27.91 ± 2.1%), and the migration inhibitory rate (HNE1-LMP1 vs HNE1: 48.70 ± 3.9 vs 29.19 ± 6.27%) were all significantly different between the two cell lines (P < 0.01). ii) LMP1 was down-regulated in As2O3-treated HNE1-LMP1 cells. iii) The reduction of MMP-9 was found in As2O3-treated groups, more evident in HNE1-LMP1 cells. Thus, we conclude that As2O3 can reduce metastasis potential of NPC cells, involving inhibition of MMP-9 expression. LMP1 were also reduced in this process and seemed to enhance anti-metastasis activity of As2O3.

Our objective was to clone, express and characterize adult Dermatophagoides farinae group 1 (Der f 1) allergens to further produce recombinant allergens for future clinical applications in order to eliminate side reactions from crude extracts of mites. Based on GenBank data, we designed primers and amplified the cDNA fragment coding for Der f 1 by nested-PCR. After purification and recovery, the cDNA fragment was cloned into the pMD19-T vector. The fragment was then sequenced, subcloned into the plasmid pET28a(+), expressed in Escherichia coli BL21 and identified by Western blotting. The cDNA coding for Der f 1 was cloned, sequenced and expressed successfully. Sequence analysis showed the presence of an open reading frame containing 966 bp that encodes a protein of 321 amino acids. Interestingly, homology analysis showed that the Der p 1 shared more than 87% identity in amino acid sequence with Eur m 1 but only 80% with Der f 1. Furthermore, phylogenetic analyses suggested that D. pteronyssinus was evolutionarily closer to Euroglyphus maynei than to D. farinae, even though D. pteronyssinus and D. farinae belong to the same Dermatophagoides genus. A total of three cysteine peptidase active sites were found in the predicted amino acid sequence, including 127-138 (QGGCGSCWAFSG), 267-277 (NYHAVNIVGYG) and 284-303 (YWIVRNSWDTTWGDSGYGYF). Moreover, secondary structure analysis revealed that Der f 1 contained an a helix (33.96%), an extended strand (17.13%), a ß turn (5.61%), and a random coil (43.30%). A simple three-dimensional model of this protein was constructed using a Swiss-model server. The cDNA coding for Der f 1 was cloned, sequenced and expressed successfully. Alignment and phylogenetic analysis suggests that D. pteronyssinus is evolutionarily more similar to E. maynei than to D. farinae.
