OBJETIVO: Comparar os resultados maternos e perinatais de pacientes portadoras de placenta prévia, após adoção do internamento materno prolongado, com os de uma série histórica ocorrida em 1991. MÉTODOS: Estudo retrospectivo comparando 108 casos da doença - em pacientes hospitalizadas em uma instituição de ensino do estado do Ceará, nordeste do Brasil, no período de primeiro de janeiro de 2006 a 31 de dezembro de 2010 - com 101 casos ocorridos em 1991, na mesma instituição. Os seguintes dados maternos e perinatais foram coletados: idade materna, paridade, idade gestacional no momento do parto, via de parto, tempo de internamento materno, escores de Apgar ao primeiro e quinto minutos, peso ao nascimento, adequação do peso ao nascer, tempo de hospitalização neonatal, morbidade materna e neonatal e mortalidades (materna, fetal, neonatal e perinatal). As variáveis categóricas foram analisadas utilizando-se os testes do χ² de associação e exato de Fischer. Os resultados foram considerados significativos quando p<0,05. RESULTADOS: Em 1991, 1,1% dos casos (101/8.900) apresentou placenta prévia. No presente estudo, a prevalência foi de 0,4% (108/24.726). Nenhuma morte materna foi observada nas duas séries. Em relação às pacientes de 1991, as da série atual foram significativamente mais jovens, com menor paridade e ficaram mais tempo internadas. Para os resultados perinatais observaram-se melhores índices de Apgar ao primeiro e quinto minutos, maior tempo de internamento neonatal e redução das mortalidades fetal, neonatal e perinatal. CONCLUSÃO: Os resultados perinatais, em pacientes com placenta prévia, foram significativamente melhorados entre o ano de 1991 e os anos de 2006 e 2010. Não podemos afirmar, entretanto, ter sido esta melhora necessariamente decorrente do maior tempo de internamento materno.

Este trabalho teve por objetivo estudar os aspectos clínicos e as características físico-químicas do leite em ovelhas com mastite induzida experimentalmente com Staphylococcus aureus. Foram utilizados dez animais da raça Santa Inês, com peso médio de 30 kg, fêmeas, primíparas recém-paridas, mantidos em apriscos e clinicamente sadios. Após se estabelecer os padrões de normalidade para as variáveis estudadas, os animais foram inoculados experimentalmente numa mama com uma cepa de S. aureus, empregando-se o inóculo de 1,0x10(4)ufc/ml, enquanto a outra serviu como controle. As observações clínicas e laboratoriais foram realizadas nos intervalos de 12, 24, 36, 48, 60, 72, 84, 96, 108, 120, 132, 168, 180, 288 e 336 horas após a inoculação do agente etiológico (PI). Todos os animais apresentaram manifestações clínicas sistêmicas e nas glândulas inoculadas, observadas com mais intensidade a partir de 24 horas após a inoculação. Alterações significativas (P<0,05) na produção e nos componentes físico-químicos do leite em relação aos seus controles ocorreram, reduzindo o volume e gordura. O pH, o teor de cloretos e a CCS atingiram índices muito elevados (P<0,05), acompanhados da reação na prova do CMT. O exame bacteriológico revelou a presença do S. aureus durante a fase de infecção. Após a instituição do tratamento nas ovelhas às 36 h PI, um animal morreu 48h PI; nas demais ocorreu a recuperação clínica, no entanto não houve o restabelecimento fisiológico das mamas inoculadas, que perderam a sua funcionalidade.

Objetivou-se, no presente estudo, pesquisar a prevalência de anticorpos anti-Neospora caninum em 812 amostras de soros sangüíneos de bovinos leiteiros procedentes de propriedades rurais de sete municípios das microrregiões de Itapecuru-Mirim, Médio Mearim e Presidente Dutra, estado do Maranhão, Brasil. Para o cálculo do tamanho da amostra, considerou-se um soroprevalência de 34,7% para N. caninum, com erro máximo de 9,5% e intervalo de confiança de 95%. Para a detecção da presença de anticorpos da classe IgG, utilizou-se a técnica de Imunofluorescência Indireta (IFI), com ponto de corte 1:200, usando como antígeno, taquizoítos da cepa NC-1, mantida em cultura celular no Laboratório de Diagnóstico das Parasitoses dos Animais da Escola de Medicina Veterinária da UFBA. Do total de amostras analisadas, encontrou-se uma prevalência de 50,74%. Os títulos variaram de 1:200 a 1:6400, assim distribuídos: 108 (26,21%) amostras de soro apresentaram título de 1:200; 132 (32,04%) 1:400; 94 (22,81%) 1:800; 46 (11,16%) 1:1600; 23 (5,58%) 1:3200 e nove (2,18%) com títulos de 1:6400. Dentre as microrregiões, a Itapecuru-Mirim apresentou o menor percentual de animais soropositivos (20,69%) e Presidente Dutra o maior (47,66%). Com relação à variável sexo, observou-se maior prevalência de sororreagentes nas fêmeas (46,80%) do que nos machos (52,46%). Não se verificou diferença significativa (P>0,05) para as variáveis microrregiões, sexo e idade. Conclui-se que os bovinos leiteiros das regiões estudadas estão expostos à infecção por N. caninum.

Este estudo teve como objetivo determinar a prevalência das dermatopatias tumorais de equinos diagnosticadas no Laboratório de Patologia Veterinária (LPV) da Universidade Federal de Santa Maria (UFSM), Rio Grande do Sul. Para isso, foram revisados os protocolos de biópsia de pele de equinos, arquivados no LPV-UFSM, entre janeiro de 1999 e dezembro de 2009. Em todos os casos, foram considerados os diagnósticos morfológicos que constavam nos protocolos. Durante esse período foram submetidas ao LPV-UFSM 139 amostras cutâneas de equinos. Dessas 139 amostras, 108 (77,6%) eram de tumores cutâneos, neoplásicos ou não; os outros 31 casos consistiam de dermatoses não tumorais. Dos 108 equinos, 53 (49,1%) eram fêmeas e 37 (34,2% %) eram machos, em 18 (16,7%) casos não estava descrito no protocolo o sexo. Quanto à raça, 66 (61,1%) eram de raça pura e 13 (12%) não tinham raça definida; em 29 (26,9%) protocolos não havia a descrição da raça. A raça mais prevalente foi a Crioula (44/108 [40,7%]), o restante dos 22 equinos de raça pura pertencia a outras sete raças diferentes. Quanto à idade, as categorias mais prevalentes em ordem decrescente foram a de 1-5 anos (47/108 [43,5%]) e a de 6-14 anos (21/108 [19,5%]), cavalos com 15 anos ou mais representaram 11,1% (12/108). Apenas um cavalo tinha menos de um ano de idade. Em 27 protocolos não constava a idade. Os tumores mais prevalentes incluíram: sarcoide (62/108 [57,4%]); carcinoma de células escamosas (11/108 [10,2%]); pitiose (9/108 [8,3%]); tecido de granulação (7/108 [6,5%]; e o granuloma eosinofílico (4/108 [3,8%]). Os outros tumores contaram com aproximadamente 14% dos casos.

The role of sympathetic nerve activity in the changes in arterial blood pressure and renal function caused by the chronic administration of NG-nitro-L-arginine methyl ester (L-NAME), an inhibitor of nitric oxide (NO) synthesis, was examined in sham and bilaterally renal denervated rats. Several studies have demonstrated that sympathetic nerve activity is elevated acutely after L-NAME administration. To evaluate the role of renal nerve activity in L-NAME-induced hypertension, we compared the blood pressure response in four groups (N = 10 each) of male Wistar-Hannover rats weighing 200 to 250 g: 1) sham-operated vehicle-treated, 2) sham-operated L-NAME-treated, 3) denervated vehicle-treated, and 4) denervated L-NAME-treated rats. After renal denervation or sham surgery, one control week was followed by three weeks of oral administration of L-NAME by gavage. Arterial pressure was measured weekly in conscious rats by a tail-cuff method and renal function tests were performed in individual metabolic cages 0, 7, 14 and 21 days after the beginning of L-NAME administration. L-NAME (60 mg kg-1 day-1) progressively increased arterial pressure from 108 ± 6.0 to 149 ± 12 mmHg (P<0.05) in the sham-operated group by the third week of treatment which was accompanied by a fall in creatinine clearance from 336 ± 18 to 222 ± 59 µl min-1 100 g body weight-1 (P<0.05) and a rise in fractional urinary sodium excretion from 0.2 ± 0.04 to 1.62 ± 0.35% (P<0.05) and in sodium post-proximal fractional excretion from 0.54 ± 0.09 to 4.7 ± 0.86% (P<0.05). The development of hypertension was significantly delayed and attenuated in denervated L-NAME-treated rats. This was accompanied by a striking additional increase in fractional renal sodium and potassium excretion from 0.2 ± 0.04 to 4.5 ± 1.6% and from 0.1 ± 0.015 to 1.21 ± 0.37%, respectively, and an enhanced post-proximal sodium excretion compared to the sham-operated group. These differences occurred despite an unchanged creatinine clearance and Na+ filtered load. These results suggest that bilateral renal denervation delayed and attenuated the L-NAME-induced hypertension by promoting an additional decrease in tubule sodium reabsorption in the post-proximal segments of nephrons. Much of the hypertension caused by chronic NO synthesis inhibition is thus dependent on renal nerve activity.

The purpose of the present study was to evaluate the effects of aerobic physical training (APT) on heart rate variability (HRV) and cardiorespiratory responses at peak condition and ventilatory anaerobic threshold. Ten young (Y: median = 21 years) and seven middle-aged (MA = 53 years) healthy sedentary men were studied. Dynamic exercise tests were performed on a cycloergometer using a continuous ramp protocol (12 to 20 W/min) until exhaustion. A dynamic 24-h electrocardiogram was analyzed by time (TD) (standard deviation of mean R-R intervals) and frequency domain (FD) methods. The power spectral components were expressed as absolute (a) and normalized units (nu) at low (LF) and high (HF) frequencies and as the LF/HF ratio. Control (C) condition: HRV in TD (Y: 108, MA: 96 ms; P<0.05) and FD - LFa, HFa - was significantly higher in young (1030; 2589 ms²/Hz) than in middle-aged men (357; 342 ms²/Hz) only during sleep (P<0.05); post-training effects: resting bradycardia (P<0.05) in the awake condition in both groups; VO2 increased for both groups at anaerobic threshold (P<0.05), and at peak condition only in young men; HRV in TD and FD (a and nu) was not significantly changed by training in either groups. The vagal predominance during sleep is reduced with aging. The resting bradycardia induced by short-term APT in both age groups suggests that this adaptation is much more related to intrinsic alterations in sinus node than in efferent vagal-sympathetic modulation. Furthermore, the greater alterations in VO2 than in HRV may be related to short-term APT.

The objective of the present study was to characterize the heart rate (HR) patterns of healthy males using the autoregressive integrated moving average (ARIMA) model over a power range assumed to correspond to the anaerobic threshold (AT) during discontinuous dynamic exercise tests (DDET). Nine young (22.3 ± 1.57 years) and 9 middle-aged (MA) volunteers (43.2 ± 3.53 years) performed three DDET on a cycle ergometer. Protocol I: DDET in steps with progressive power increases of 10 W; protocol II: DDET using the same power values as protocol 1, but applied randomly; protocol III: continuous dynamic exercise protocol with ventilatory and metabolic measurements (10 W/min ramp power), for the measurement of ventilatory AT. HR was recorded and stored beat-to-beat during DDET, and analyzed using the ARIMA (protocols I and II). The DDET experiments showed that the median physical exercise workloads at which AT occurred were similar for protocols I and II, i.e., AT occurred between 75 W (116 bpm) and 85 W (116 bpm) for the young group and between 60 W (96 bpm) and 75 W (107 bpm) for group MA in protocols I and II, respectively; in two MA volunteers the ventilatory AT occurred at 90 W (108 bpm) and 95 W (111 bpm). This corresponded to the same power values of the positive trend in HR responses. The change in HR response using ARIMA models at submaximal dynamic exercise powers proved to be a promising approach for detecting AT in normal volunteers.

Endochondral calcification involves the participation of matrix vesicles (MVs), but it remains unclear whether calcification ectopically induced by implants of demineralized bone matrix also proceeds via MVs. Ectopic bone formation was induced by implanting rat demineralized diaphyseal bone matrix into the dorsal subcutaneous tissue of Wistar rats and was examined histologically and biochemically. Budding of MVs from chondrocytes was observed to serve as nucleation sites for mineralization during induced ectopic osteogenesis, presenting a diameter with Gaussian distribution with a median of 306 ± 103 nm. While the role of tissue-nonspecific alkaline phosphatase (TNAP) during mineralization involves hydrolysis of inorganic pyrophosphate (PPi), it is unclear how the microenvironment of MV may affect the ability of TNAP to hydrolyze the variety of substrates present at sites of mineralization. We show that the implants contain high levels of TNAP capable of hydrolyzing p-nitrophenylphosphate (pNPP), ATP and PPi. The catalytic properties of glycosyl phosphatidylinositol-anchored, polidocanol-solubilized and phosphatidylinositol-specific phospholipase C-released TNAP were compared using pNPP, ATP and PPi as substrates. While the enzymatic efficiency (k cat/Km) remained comparable between polidocanol-solubilized and membrane-bound TNAP for all three substrates, the k cat/Km for the phosphatidylinositol-specific phospholipase C-solubilized enzyme increased approximately 108-, 56-, and 556-fold for pNPP, ATP and PPi, respectively, compared to the membrane-bound enzyme. Our data are consistent with the involvement of MVs during ectopic calcification and also suggest that the location of TNAP on the membrane of MVs may play a role in determining substrate selectivity in this micro-compartment.

Apolipoprotein CIII (apo-CIII) participates in the regulation of triglyceride-rich lipoprotein metabolism. Several polymorphic sites have been detected within and around the apo-CIII gene. Here, we examined the relationship between apo-CIII SstI polymorphism (CC, CG, GG genotypes) and plasma triglyceride (TG) levels in a group of 159 Japanese individuals living in Southern Brazil. The sample was divided into a group of Japanese descendants (N = 51) with high TG (HTG; >200 mg/dL) and a group of Japanese descendants (N = 108) with normal TG (NTG; <200 mg/dL). TG and total cholesterol levels were analyzed by an enzymatic method using the Labtest-Diagnostic kit and high- and low-density lipoproteins by a direct method using the Labtest-Diagnostic kit and DiaSys Diagnostic System International kit, respectively. A 428-bp sequence of apo-CIII gene was amplified using oligonucleotide primers 5' GGT GAC CGA TGG CTT CAG TTC CCT GA 3' and 5' CAG AAG GTG GAT AGA GCG CTG GCC T 3'. The PCR products were digested with a restriction endonuclease SstI. Rare G allele was highly prevalent in our study population (0.416) compared to Caucasians (0.00-0.11). G allele was almost two times more prevalent in the HTG group compared to the NTG group (P < 0.001). The genotype distribution was consistent with the Hardy-Weinberg equilibrium. There was a significant association between rare G allele and HTG in Japanese individuals living in Southern Brazil as indicated by one-way ANOVA, P < 0.05.

The objective of the present study was to identify metabolic, cardiovascular and autonomic changes induced by fructose overload administered in the drinking water of rats for 8 weeks. Female Wistar rats (200-220 g) were divided into 2 groups: control (N = 8) and fructose-fed rats (N = 5; 100 mg/L fructose in drinking water for 8 weeks). The autonomic control of heart rate was evaluated by pharmacological blockade using atropine (3 mg/kg) and propranolol (4 mg/kg). The animals were submitted to an intravenous insulin tolerance test (ITT) and to blood glucose measurement. The fructose overload induced a significant increase in body weight (~10%) and in fasting glycemia (~28%). The rate constant of glucose disappearance (KITT) during ITT was lower in fructose-fed rats (3.25 ± 0.7%/min) compared with controls (4.95 ± 0.3%/min, P < 0.05) indicating insulin resistance. The fructose-fed group presented increased arterial pressure compared to controls (122 ± 3 vs 108 ± 1 mmHg, P < 0.05) and a reduction in vagal tonus (31 ± 9 vs 55 ± 5 bpm in controls, P < 0.05). No changes in sympathetic tonus were observed. A positive correlation, tested by the Pearson correlation, was demonstrable between cardiac vagal tonus and KITT (r = 0.8, P = 0.02). These data provided new information regarding the role of parasympathetic dysfunction associated with insulin resistance in the development of early metabolic and cardiovascular alterations induced by a high fructose diet.

Carbon nanotubes are highly versatile materials; new applications using them are continuously being developed. Special attention is being dedicated to the possible use of multiwalled carbon nanotubes in biomaterials contacting with bone. However, carbon nanotubes are also controversial in regards to effects exerted on living organisms. Carbon nanotubes can be used to improve the tribological properties of polymer/composite materials. Ultrahigh molecular weight polyethylene (UHMWPE) is a polymer widely used in orthopedic applications that imply wear and particle generation. We describe here the response of human osteoblast-like MG63 cells after 6 days of culture in contact with artificially generated particles from both UHMWPE polymer and multiwalled carbon nanotubes (MWCNT)/UHMWPE nanocomposites. This novel composite has superior wear behavior, having thus the potential to reduce the number of revision hip arthroplasty surgeries required by wear failure of acetabular cups and diminish particle-induced osteolysis. The results of an in vitro study of viability and proliferation and interleukin-6 (IL-6) production suggest good cytocompatibility, similar to that of conventional UHMWPE (WST-1 assay results are reported as percentage of control ± SD: UHMWPE = 96.19 ± 7.92, MWCNT/UHMWPE = 97.92 ± 8.29%; total protein: control = 139.73 ± 10.78, UHMWPE = 137.07 ± 6.17, MWCNT/UHMWPE = 163.29 ± 11.81 µg/mL; IL-6: control = 90.93 ± 10.30, UHMWPE = 92.52 ± 11.02, MWCNT/UHMWPE = 108.99 ± 9.90 pg/mL). Standard cell culture conditions were considered as control. These results, especially the absence of significant elevation in the osteolysis inductor IL-6 values, reinforce the potential of this superior wear-resistant composite for future orthopedic applications, when compared to traditional UHMWPE.

The objective of this study was to identify intravascular ultrasound (IVUS), angiographic and metabolic parameters related to restenosis in patients with dysglycemia. Seventy consecutive patients (77 lesions) selected according to inclusion and exclusion criteria were evaluated by the oral glucose tolerance test and the determination of insulinemia after a successful percutaneous coronary intervention (PCI) with a bare-metal stent. The degree of insulin resistance was calculated by the homeostasis model assessment of insulin resistance (HOMA-IR). Six-month IVUS and angiogram follow-up were performed. Thirty-nine patients (55.7%) had dysglycemia. The restenosis rate in the dysglycemic group was 37.2 vs 23.5% in the euglycemic group (P = 0.299). The predictors of restenosis using bivariate analysis were reference vessel diameter (RVD): £2.93 mm (RR = 0.54; 95%CI = 0.05-0.78; P = 0.048), stent area (SA): <8.91 mm² (RR = 0.66; 95%CI = 0.24-0.85; P = 0.006), stent volume (SV): <119.75 mm³ (RR = 0.74; 95%CI = 0.38-0.89; P = 0.0005), HOMA-IR: >2.063 (RR = 0.44; 95%CI = 0.14-0.64; P = 0.027), and fasting plasma glucose (FPG): ≤108.8 mg/dL (RR = 0.53; 95%CI = 0.13-0.75; P = 0.046). SV was an independent predictor of restenosis by multivariable analysis. Dysglycemia is a common clinical condition in patients submitted to PCI. The degree of insulin resistance, FPG, RVD, SA, and SV were correlated with restenosis. SV was inversely correlated with an independent predictor of restenosis in patients treated with a bare-metal stent.

As concentrações de inosina-5'-monofosfato e seus derivados (adenosina monofosfato, hipoxantina e inosina) nos músculos Psoas major, Latissimus dorsi, Tensor fascia e Extensor carpi radialis de bovinos, suínos e ovinos foram determinadas através de Cromatografia Líquida de Alta Eficiência (CLAE). A Análise de Variância (ANOVA) dos dados mostraram que as concentrações de 5'-IMP e seus derivados foram significativamente diferentes entre as três espécies (p < 0,001). A maior concentração de 5'-IMP foi detectada nos músculos de ovinos (média de 108,2 mg/100g) seguida pelos de suínos (média de 97 mg/100g) e pelos de bovinos (média de 69,3 mg/100g). Houve variações significativas nas concentrações no músculo, entre os animais nas espécies bovina (p < 0,01) e ovina (p < 0,05), porém não entre os da espécie suína (p > 0,05). Não houve diferenças significativas entre os tipos de músculo em bovinos e ovinos (p > 0,05), mas em suínos (p < 0,01) observou-se variação significativa.

Foram analisadas 20 amostras de queijo de leite de cabra" tipo coalho", elaborados de forma artesanal e armazenados durante 28 dias sob refrigeração a 10 ± 2 ºC, em intervalos de 0, 7, 14, 21 e 28 dias, tendo sido avaliados os seguintes parâmetros: contagem de bactérias aeróbias mesófilas (UFC/g) número mais provável de coliformes totais e fecais (NMP/g), contagem de bolores e leveduras (UFC/g) e contagem de Staphylococcus aureus (UFC/g). As contagens de bactérias aeróbias mesófilas variaram de 2,35 x 103 a 1,5 x 109 UFC/g; enquanto que o número de coliformes totais oscilaram entre 2,4 x 103 a 2,4 x 106 e 2,4 x 109 NMP/g. Por outro lado, a contagem de coliformes fecais a apresentou variações de 2,4 x 103 a 2,4 x 106 NMP/g e bolores e leveduras < 30 a 5,5 x 108 UFC/g. Durante o armazenamento não foi evidenciada a presença de Staphylococcus aureus, constatando-se a necessidade de reavaliar as técnicas utilizadas na manipulação e processamento artesanal do queijo elaborado.

INTRODUÇÃO: No contexto atual da elevada escassez de órgãos para o transplante renal e do reconhecimento cada vez maior da rejeição crônica mediada por anticorpos anti-HLA como uma importante causa de perda do enxerto, uma contínua demonstração da boa evolução a longo prazo de transplantes renais com doadores vivos não aparentados (DVNA) é de suma importância. OBJETIVOS: Analisar a sobrevida do enxerto e dos pacientes transplantados com DVNA, e compará-la com doadores vivos aparentados (DVA). MÉTODOS: Foram analisados 389 primeiros transplantes renais com doador vivo realizados em um único centro, entre janeiro de 1998 e dezembro de 2007, 281 com DVA e 108 com DVNA. RESULTADOS: Não houve diferença significativa na sobrevida dos pacientes (89,1% vs. 84,7%, p = 0,40) e do enxerto (81,1% vs. 68,9%, p = 0,77), em 10 anos de seguimento, entre DVA e DVNA, respectivamente. Na análise multivariada do modelo de regressão proporcional de Cox, a reatividade contra painel (PRA) > 10% e a ocorrência de rejeição aguda no 1º ano após o transplante foram os únicos preditores independentes de perda do enxerto (OR 2,54, IC 95% 1,35 - 4,78; p < 0,05 e OR 4,1, IC 95% 2,04 -4,78; p < 0,05, respectivamente). CONCLUSÃO: Transplantes renais com DVNA representam uma importante fonte de órgãos para suprir uma crescente demanda, com resultados semelhantes aos transplantes com DVA, independente da compatibilidade HLA.
