We determined whether over-expression of one of the three genes involved in reverse cholesterol transport, apolipoprotein (apo) AI, lecithin-cholesterol acyl transferase (LCAT) and cholesteryl ester transfer protein (CETP), or of their combinations influenced the development of diet-induced atherosclerosis. Eight genotypic groups of mice were studied (AI, LCAT, CETP, LCAT/AI, CETP/AI, LCAT/CETP, LCAT/AI/CETP, and non-transgenic) after four months on an atherogenic diet. The extent of atherosclerosis was assessed by morphometric analysis of lipid-stained areas in the aortic roots. The relative influence (R²) of genotype, sex, total cholesterol, and its main sub-fraction levels on atherosclerotic lesion size was determined by multiple linear regression analysis. Whereas apo AI (R² = 0.22, P < 0.001) and CETP (R² = 0.13, P < 0.01) expression reduced lesion size, the LCAT (R² = 0.16, P < 0.005) and LCAT/AI (R² = 0.13, P < 0.003) genotypes had the opposite effect. Logistic regression analysis revealed that the risk of developing atherosclerotic lesions greater than the 50th percentile was 4.3-fold lower for the apo AI transgenic mice than for non-transgenic mice, and was 3.0-fold lower for male than for female mice. These results show that apo AI overexpression decreased the risk of developing large atherosclerotic lesions but was not sufficient to reduce the atherogenic effect of LCAT when both transgenes were co-expressed. On the other hand, CETP expression was sufficient to eliminate the deleterious effect of LCAT and LCAT/AI overexpression. Therefore, increasing each step of the reverse cholesterol transport per se does not necessarily imply protection against atherosclerosis while CETP expression can change specific athero genic scenarios.

To efficiently examine the association of glutamic acid decarboxylase antibody (GADA) positivity with the onset and progression of diabetes in middle-aged adults, we performed a case-cohort study representing the ~9-year experience of 10,275 Atherosclerosis Risk in Communities Study participants, initially aged 45-64 years. Antibodies to glutamic acid decarboxylase (GAD65) were measured by radioimmunoassay in 580 incident diabetes cases and 544 non-cases. The overall weighted prevalence of GADA positivity (³1 U/mL) was 7.3%. Baseline risk factors, with the exception of smoking and interleukin-6 (P £ 0.02), were generally similar between GADA-positive and -negative individuals. GADA positivity did not predict incident diabetes in multiply adjusted (HR = 1.04; 95%CI = 0.55, 1.96) proportional hazard analyses. However, a small non-significant adjusted risk (HR = 1.29; 95%CI = 0.58, 2.88) was seen for those in the highest tertile (³2.38 U/mL) of positivity. GADA-positive and GADA-negative non-diabetic individuals had similar risk profiles for diabetes, with central obesity and elevated inflammation markers, aside from glucose, being the main predictors. Among diabetes cases at study's end, progression to insulin treatment increased monotonically as a function of baseline GADA level. Overall, being GADA positive increased risk of progression to insulin use almost 10 times (HR = 9.9; 95%CI = 3.4, 28.5). In conclusion, in initially non-diabetic middle-aged adults, GADA positivity did not increase diabetes risk, and the overall baseline profile of risk factors was similar for positive and negative individuals. Among middle-aged adults, with the possible exception of those with the highest GADA levels, autoimmune pathophysiology reflected by GADA may become clinically relevant only after diabetes onset.

We previously reported that a DNA vaccine constructed with the heat shock protein (HSP65) gene from Mycobacterium leprae (DNA-HSP65) was protective and also therapeutic in experimental tuberculosis. By the intramuscular route, this vaccine elicited a predominant Th1 response that was consistent with its protective efficacy against tuberculosis. It has been suggested that the immune response to Hsp60/65 may be the link between exposure to microorganisms and increased cardiovascular risk. Additionally, the high cholesterol levels found in atherosclerosis could modulate host immunity. In this context, we evaluated if an atherogenic diet could modulate the immune response induced by the DNA-HSP65 vaccine. C57BL/6 mice (4-6 animals per group) were initially submitted to a protocol of atherosclerosis induction and then immunized by the intramuscular or intradermal route with 4 doses of 100 µg DNA-HSP65. On day 150 (15 days after the last immunization), the animals were sacrificed and antibodies and cytokines were determined. Vaccination by the intramuscular route induced high levels of anti-Hsp65 IgG2a antibodies, but not anti-Hsp65 IgG1 antibodies and a significant production of IL-6, IFN-g and IL-10, but not IL-5, indicating a Th1 profile. Immunization by the intradermal route triggered a mixed pattern (Th1/Th2) characterized by synthesis of anti-Hsp65 IgG2a and IgG1 antibodies and production of high levels of IL-5, IL-6, IL-10, and IFN-g. These results indicate that experimentally induced atherosclerosis did not affect the ability of DNA-HSP65 to induce a predominant Th1 response that is potentially protective against tuberculosis.

The relationship between lipid serum levels and coronary atherosclerotic plaque fat content was studied in 51 necropsy patients. Serum lipids were measured by standard techniques, during life, in the absence of lipid-lowering drugs. Intima, intimal fat and media areas were measured using a computerized system in cryosections of the odd segments of the right, anterior descending and circumflex coronary arteries stained with Sudan-IV. Mean intimal and lipid areas were 5.74 ± 1.98 and 1.22 ± 0.55 mm² (22.12 ± 8.48%) in 26 cases with high cholesterol (³200 mg/dL) and 4.98 ± 1.94 and 1.16 ± 0.66 mm² (22.75 ± 9.06%) in 25 cases with normal cholesterol (<200 mg/dL; P > 0.05). Patients with high levels of low-density lipoprotein (³130 mg/dL, N = 15) had a higher intima/media area ratio than those with normal levels of low-density lipoprotein (<130 mg/dL, N = 13, P < 0.01). No significant difference in the morphometrical variables was found in groups with high or low serum levels of triglycerides (³200 mg/dL, N = 13 vs <200 mg/dL, N = 36) or high-density lipoprotein (³35 mg/dL, N = 11 vs <35 mg/dL, N = 17). The association between the morphological measurements and serum levels of cholesterol, its fractions, and triglycerides was also tested and the correlation coefficients were low. Although high cholesterol is a risk factor, we show here that in patients with severe atherosclerosis blood cholesterol and triglyceride levels seem to have little influence on coronary lipid content, indicating that other factors may contribute to arterial lipid deposition and plaque formation.

In order to determine the effect of antibodies against electronegative low-density lipoprotein LDL(-) on atherogenesis, five groups of LDL low receptor-deficient (LDLr-/-) mice (6 per group) were immunized with the following antibodies (100 µg each): mouse anti-LDL(-) monoclonal IgG2b, rabbit anti-LDL(-) polyclonal IgG or its Fab fragments and mouse irrelevant monoclonal IgG and non-immunized controls. Antibodies were administered intravenously one week before starting the hypercholesterolemic diet (1.25% cholesterol) and then every week for 21 days. The passive immunization with anti-LDL(-) monoclonal IgG2b, polyclonal antibody and its derived Fab significantly reduced the cross-sectional area of atherosclerotic lesions at the aortic root of LDLr-/- mice (28.8 ± 9.7, 67.3 ± 17.02, 56.9 ± 8.02 µm² (mean ± SD), respectively) compared to control (124.9 ± 13.2 µm²). Vascular cell adhesion molecule-1 protein expression, quantified by the KS300 image-analyzing software, on endothelium and the number of macrophages in the intima was also decreased in aortas of mice treated with anti-LDL(-) monoclonal antibody (3.5 ± 0.70 per field x 10) compared to controls (21.5 ± 3.5 per field x 10). Furthermore, immunization with the monoclonal antibody decreased the concentration of LDL(-) in blood plasma (immunized: 1.0 ± 1.4; control: 20.5 ± 3.5 RLU), the amount of cholesterol oxides in plasma (immunized: 4.7 ± 2.7; control: 15.0 ± 2.0 pg COx/mg cholesterol) and liver (immunized: 2.3 ± 1.5; control: 30.0 ± 26.0 pg COx/mg cholesterol), and the hepatic content of lipid hydroperoxides (immunized: 0.30 ± 0.020; control: 0.38 ± 0.15 ng/mg protein). In conclusion, antibodies against electronegative LDL administered intravenously may play a protective role in atherosclerosis.

This review provides examples of the fact that different procedures for the measurement of atherosclerosis in mice may lead to interpretation of the extent of atherosclerosis having markedly different biological and clinical significance for humans: 1) aortic cholesterol measurement is highly sensitive for the detection of early and advanced atherosclerosis lesions, but misses the identification of the location and complexity of these lesions that are so critical for humans; 2) the histological analysis of the aortic root lesions in simvastatin-treated and control mice reveals similar lesion morphology in spite of the remarkable simvastatin-induced reduction of the aortic cholesteryl ester content; 3) in histological analyses, chemical fixation and inclusion may extract the tissue fat and also shrink and distort tissue structures. Thus, the method may be less sensitive for the detection of slight differences among the experimental groups, unless a more suitable procedure employing physical fixation with histological sample freezing using optimal cutting temperature and liquid nitrogen is employed. Thus, when measuring experimental atherosclerosis in mice, investigators should be aware of several previously unreported pitfalls regarding the extent, location and complexity of the arterial lesion that may not be suitable for extrapolation to human pathology.

The objective of the present study was to assess the effects of the immunosuppressant rapamycin (Rapamune®, Sirolimus) on both resistance vessel responsiveness and atherosclerosis in apolipoprotein E-deficient 8-week-old male mice fed a normal rodent diet. Norepinephrine (NE)-induced vasoconstriction, acetylcholine (ACh)- and sodium nitroprusside (SNP)-induced vasorelaxation of isolated mesenteric bed, and atherosclerotic lesions were evaluated. After 12 weeks of orally administered rapamycin (5 mg·kg-1·day-1, N = 9) and compared with untreated (control, N = 9) animals, rapamycin treatment did not modify either NE-induced vasoconstriction (maximal response: 114 ± 4 vs 124 ± 10 mmHg, respectively) or ACh- (maximal response: 51 ± 8 vs 53 ± 5%, respectively) and SNP-induced vasorelaxation (maximal response: 73 ± 6 vs 74 ± 6%, respectively) of the isolated vascular mesenteric bed. Despite increased total cholesterol in treated mice (982 ± 59 vs 722 ± 49 mg/dL, P < 0.01), lipid deposition on the aorta wall vessel was significantly less in rapamycin-treated animals (37 ± 12 vs 68 ± 8 µm² x 10³). These results indicate that orally administered rapamycin is effective in attenuating the progression of atherosclerotic plaque without affecting the responsiveness of resistance vessels, supporting the idea that this immunosuppressant agent might be of potential benefit against atherosclerosis in patients undergoing therapy.

Pequi is the fruit of Caryocar brasiliense and its oil has a high concentration of monounsaturated and saturated fatty acids, which are anti- and pro-atherogenic agents, respectively, and of carotenoids, which give it antioxidant properties. Our objective was to study the effect of the intake of a cholesterol-rich diet supplemented with pequi oil, compared to the same diet containing soybean oil, on atherosclerosis development, and oxidative stress in atherosclerosis-susceptible LDL receptor-deficient mice (LDLr-/-, C57BL/6-background). Female mice were fed a cholesterol-rich diet containing 7% soybean oil (Soybean group, N = 12) or 7% pequi oil (Pequi group, N = 12) for 6 weeks. The Pequi group presented a more atherogenic lipid profile and more advanced atherosclerotic lesions in the aortic root compared to the Soybean group. However, the Pequi group presented a less advanced lesion in the aorta than the Soybean group and showed lower lipid peroxidation (Soybean group: 50.2 ± 7.1; Pequi group: 30.0 ± 4.8 µmol MDA/mg protein) and anti-oxidized LDL autoantibodies (Soybean group: 35.7 ± 9.4; Pequi group: 15.6 ± 3.7 arbitrary units). Peritoneal macrophages from the Pequi group stimulated with zymosan showed a reduction in the release of reactive oxygen species compared to the Soybean group. Our data suggest that a pequi oil-rich diet slows atherogenesis in the initial stages, possibly due to its antioxidant activity. However, the increase of serum cholesterol induces a more prominent LDL migration toward the intimae of arteries, increasing the advanced atherosclerotic plaque. In conclusion, pequi oil associated with an atherogenic diet worsens the lipid profile and accelerates the formation of advanced atherosclerotic lesions despite its antioxidant action.

Osteoprotegerin (OPG) regulates bone mass by inhibiting osteoclast differentiation and activation, and plays a role in vascular calcification. We evaluated the relationship between osteoprotegerin levels and inflammatory markers, atherosclerosis, and mortality in patients with stages 3-5 chronic kidney disease. A total of 145 subjects (median age 61 years, 61% men; 36 patients on hemodialysis, 55 patients on peritoneal dialysis, and 54 patients with stages 3-5 chronic kidney disease) were studied. Clinical characteristics, markers of mineral metabolism (including fibroblast growth factor-23 [FGF-23]) and inflammation (high-sensitivity C-reactive protein [hsCRP] and interleukin-6 [IL-6]), and the intima-media thickness (IMT) in the common carotid arteries were measured at baseline. Cardiac function was assessed by color tissue Doppler echocardiography. After 36 months follow-up, the survival rate by Kaplan-Meier analysis was significantly different according to OPG levels (χ2=14.33; P=0.002). Increased OPG levels were positively associated with IL-6 (r=0.38, P<0.001), FGF-23 (r=0.26, P<0.001) and hsCRP (r=0.0.24, P=0.003). In addition, OPG was positively associated with troponin I (r=0.54, P<0.001) and IMT (r=0.39, P<0.0001). Finally, in Cox analysis, only OPG (HR=1.07, 95%CI=1.02-1.13) and hsCRP (HR=1.02, 95%CI=1.01-1.04) were independently associated with increased risk of death. These results suggested that elevated levels of serum OPG might be associated with atherosclerosis and all-cause mortality in patients with chronic kidney disease.

Foi feito um estudo sobre o comportamento de uvas finas de mesa (Vitis vinifera L.) var. Itália frente à armazenagem refrigerada e à utilização de embalagem, para aumentar a vida-de-prateleira. Cachos da uva no estádio verde maduro foram embalados individualmente em três diferentes filmes plásticos (Cryovac® PD-900; Cryovac® PD-955 e Cryovac® PD-961EZ ) de alta permeabilidade ao oxigênio e ao gás carbônico e armazenados a 1ºC (85-95% UR) por 63 dias e a 25ºC (80-90% UR) por 21 dias; frutas sem embalagem serviram de controle. Ao longo da armazenagem foram realizadas análises sensoriais de aceitação quanto ao sabor e aparência, utilizando escala hedônica não estruturada de 9cm e 30 provadores por sessão. Foram realizadas também análises do teor de sólidos solúveis, acidez titulável e vitamina C ao longo da armazenagem. As frutas embaladas apresentaram taxas de perda de massa 28 vezes menores que as controle, mas não foi observada diferença significativa (p>0,05) entre as diferentes embalagens. A combinação de armazenagem a 1°C com o filme PD-955 prolongou a vida de prateleira das uvas por 63 dias contra 11 a 21 dias do controle. Nas uvas embaladas, o fim da vida-de-prateleira foi determinado por deterioração microbiológica e no controle pela perda de massa, que causou o enrugamento, perda de turgidez das bagas e ressecamento dos engaços e pedicelos.

O trabalho teve como objetivo realizar a classificação dos tomates de mesa (Licopersicon esculentum Mill.) convencional e orgânico comercializados na Região Metropolitana de Curitiba. Foram avaliadas seis amostras de tomates do sistema convencional e três de tomates do sistema orgânico. Os frutos foram classificados em: grupos, oblongo e redondo; subgrupos: verde maduro, pintado, rosado, vermelho e vermelho maduro e classes: gigante, grande, médio e pequeno. De acordo com a ocorrência de defeitos graves e leves nos frutos, os tomates foram classificados também em tipos: extra, especial ou selecionado e comercial. As amostras cultivadas no sistema convencional foram classificadas como do grupo redondo e plurilocular, enquanto que as do sistema orgânico do grupo oblongo e bilocular. Na classe houve um predomínio de frutos médios e nos subgrupos de frutos rosado e vermelho. Em relação ao tipo, todas as amostras foram classificadas como fora do padrão ou do tipo.

Nos últimos vinte anos, o consumo de alimentos diet e light tem aumentado sistematicamente, o que tem propiciado o constante desenvolvimento de produtos desse gênero. Grande ênfase tem sido dada àqueles produtos que substituem sacarose por edulcorantes de baixos conteúdos calóricos ou não calóricos. Seguindo esta tendência, adoçantes de mesa têm sido desenvolvidos variando-se amplamente o veículo e o tipo de edulcorante empregado. Neste trabalho, a análise de componentes principais associada à espectroscopia na região do infravermelho médio foi utilizada com sucesso para diferenciar os veículos empregados na produção destes adoçantes, sendo que esta metodologia quimiométrica reduziu o espaço dimensional para dois fatores, explicando cerca de 82-% da variância total dos dados. As variáveis responsáveis por esta discriminação estão localizadas na região da impressão digital do espectro de infravermelho (752,2 a 1284,5 cm-1). A análise exploratória mostrou-se útil para a visualização destes dados, gerando informações semiquantitativas para os adoçantes constituídos por lactose/aspartame, observações que seriam dificilmente visualizadas sem o recurso quimiométrico aplicado.

A presença dos produtos orgânicos nas gôndolas das grandes redes de supermercados indica que existe um potencial de mercado para esses produtos, no entanto poucas são as informações técnico-científicas sobre eles. Em razão disso, o presente trabalho teve como objetivo determinar a qualidade do tomate de mesa cultivado nos sistemas convencional (SC) e orgânico (SO) comercializado na Região Metropolitana de Curitiba. As amostras foram avaliadas pela massa, peso específico, cinzas, sólidos totais, sólidos solúveis totais, acidez titulável total, relação sólidos solúveis totais/acidez titulável total, pH, vitamina C, nitratos, nitritos, multirresíduos, benzimidazóis e ditiocarbamatos. Os resultados da análise físico-química mostraram que somente na umidade os tomates não apresentaram diferença significativa ao nível de 5% quando comparados pelo Teste de Tukey. Em relação à análise toxicológica, não foram detectados multirresíduos e benzimidazóis até os limites de 0,04 mg.kg-1 e 0,1 mg.kg-1 de carbendazim, respectivamente. Nos resíduos de pesticidas do grupo químico ditiocarbamatos, foi identificado 0,01 mg.kg-1 (CS2) nas amostras de SC3 e SC4 do tomate de mesa cultivado no sistema convencional, abaixo do limite máximo recomendado (LMR) de 2,0 mg.kg-1 (CS2) de mancozebe.

O presente trabalho teve como objetivo avaliar a qualidade pós-colheita de frutos de tomate de mesa de diferentes sistemas de cultivos. As amostras de tomate convencional, cv. Raísa (LV), e orgânico, cv. Santa Clara, foram mantidas a uma temperatura de 23,5 ± 2 ºC, com UR de 74% ± 5 e submetidas a análise da massa, peso específico, cinzas, sólidos totais, sólidos solúveis totais, acidez titulável total, relação sólidos solúveis totais/acidez titulável total, pH, vitamina C, Salmonella spp., coliformes totais, coliformes fecais, bolores e leveduras e análise sensorial, que foi realizada pela Análise Descritiva Quantitativa - ADQ. O tempo de armazenagem foi de 13 e 14 dias para o tomate de mesa cultivado nos sistemas convencional e orgânico, respectivamente. A perda de massa foi significativamente inferior (3,74%) no tomate convencional. Ambas as amostras apresentaram similar comportamento na análise física, química sensorial e microbiológica nos estádios de maturação

Sementes de feijão 'IAC-Carioca SH', após terem sido submetidas aos processos de classificação por peneiras e de separação por densidade em mesa gravitacional, foram avaliadas quanto à qualidade fisiológica e sanitária das sementes, contidas em três frações de descarga da mesa gravitacional. O teste envelhecimento acelerado não foi influenciado pelo tamanho da semente e o seu desempenho está relacionado ao grau de umidade das sementes. As sementes de maior densidade podem ser armazenadas por doze meses para posterior plantio, com pequena perda do poder germinativo e que não chegou a comprometer o estande. Sementes de densidade intermediária retidas nas peneiras 10, 12 e 13 e armazenadas até o décimo mês podem ser utilizadas como semente. Sementes de densidade menor, não são recomendadas para plantio e apresentam maior índice de contaminação por fungos de campo. A incidência dos fungos de armazenamento Aspergillus spp. e Penicillium spp., foi relacionada à umidade relativa no local de armazenamento da semente.
