Cucurbatacins are known to produce cytotoxic and anticancer activities. Two novel norcucurbitacin glucosides (Wvl and Wv2) have recently been isolated from a purified fraction obtained from the rhizome of Wilbrandia verticillata. The present study evaluates the cytotoxic and anti-tumour activities of the norcucurbitacins. We have found a regular cytotoxicity in KB cells (Cy50 = 12µg/ml) as well as a significant inhibition in the Walker 256 carcinosarcoma growth (approximately 75%).

A review of the role of the environment as a determinant of infectious and parasitic diseases is presented. Historical considerations and the several environmental classifications of diseases are introduced. In a broader perspective the subject is analyzed in view of the emergence of the environmental health area, with its new paradigms. A review of epidemiological studies about environmental sanitation conditions and measures is presented, analyzing the conclusions derived from 256 studies. Finally, an epidemiological study carried out in Betim, Minas Gerais is briefly described, in order to illustrate the potentiality of this kind of study. Setting priorities of interventions regarding diarrhea control was the aim of this investigation. Conclusion about the role of this approach to optimize preventive measures for the control of infectious and parasitic diseases, of sound importance to the reality of the developing world, is stated.

The complete life cycle of Triatoma flavida, weekly fed on hens, was studied at 28±2°C and 80±10% RH. Aspects related to hatching, life span, mortality and feeding behavior for each stage of its life cycle were evaluated. The hatching rate observed for 100 eggs was 93% with an average incubation period of 27.2 days. Sixty-two nymphs completed the cycle and the mean egg to adult development time was 230.4 days. Mean duration of 1st, 2nd, 3rd, 4th and 5th instar nymphs was 22.1, 25.3, 36.7, 49.7 and 69.4 days, respectively. The number of blood meals on each nymphal stage varied from 1 to 7. The mortality rate was 6.5% for NI, 23% for NIII and 7.5% for NV nymphs. Mean number of laid eggs per female was 283.1. Adult survival rates were 344.8 ± 256.4 days for males and 285.3 ± 201.8 days for females.

A total of 256 sites in 11 habitats were surveyed for Biomphalaria in Melquiades rural area (State of Minas Gerais) in August and November 1999 and in March 2000. Of the 1,780 Biomphalaria collected, 1,721 (96.7%) were B. glabrata and 59 (3.3%) B. straminea. Snails were found in all habitats except in wells, with the largest mean numbers in tanks, seepage ponds and canals, and the smallest numbers in springs, rice fields and fishponds. People's knowledge of the occurrence of Biomphalaria at the collection sites and the presence of Biomphalaria ova were strongly correlated with the occurrence of snails, and distance between houses and collection sites, as well as water velocity were inversely correlated with Biomphalaria occurrence (p < 0.001). The strongest predictor o f Biomphalaria occurrence was the presence of tilapia fish in fishponds. Fourteen Biomphalaria (0.8% of all snails) found at 6 sites were infected with Schistosoma mansoni. Suggestions are made for the utilization of local people's knowledge in snail surveys and further studies are recommended on the possible use of tilapia for biological control of Biomphalaria in fishponds, as well as modeling of S. mansoni transmission and reinfection.

The presence of anti-Toxoplasma gondii IgM and IgG antibodies was studied in samples of blood serum taken from eighty dogs with nervous symptoms at the Serviço de Enfermidades Infecciosas dos Animais, Faculdade de Medicina Veterinária e Zootecnia, Unesp, Botucatu, São Paulo, Brazil. The frequency of IgG titers were 16 (13.7%), 64 (13.7%), and 256 (5%), and for IgM titers were 16 (7.5%), 64 (15%), and 256 (8.7%). Positive reactions were more frequent in the older animals, males, from a rural environment, in constant contact with small animals, principally birds and rodents. There was a higher frequency of a positive reaction in dogs fed with kitchen food, especially in those fed with raw ingredients. The most common neurological pictures were alterations in consciousness, in movement, and in the hand-cart test. The percentage of reagents with specific IgM antibodies was high, indicating active infections, but the possibility of co-infection with the distemper virus can not be discarded, and this may be a predisposing factor for toxoplasmosis infection, once the distemper virus has a potent immunosupressive action.

Significant advances were made in the diagnosis of filariasis in the 1990s with the emergence of three new alternative tools: ultrasound and tests to detect circulating antigen using two monoclonal antibodies, Og4C3 and AD12-ICT-card. This study aimed to identify which of these methods is the most sensitive for diagnosis of infection. A total of 256 individuals, all male and carrying microfilariae (1-15,679 MF/mL), diagnosed by nocturnal venous blood samples, were tested by all three techniques. The tests for circulating filarial antigen concurred 100% and correctly identified 246/256 (96.69%) of the positive individuals, while ultrasound detected only 186/256 (73.44%). Of the circulating antigen tests, ICT-card was the most convenient method for identification of Wuchereria bancrofti carriers. It was easy to perform, practical and quick.

Between 1999-2002, Médécins Sans Frontières-Spain implemented a project seeking to determine the efficacy and safety of benznidazole in the treatment of recent chronic Chagas disease in a cohort of seropositive children in the Yoro Department, Honduras. A total of 24,471 children were screened for Trypanosoma cruzi IgG antibodies through conventional enzyme-linked immunosorbent assays (ELISA) on filter paper. Recombinant ELISA (0.93% seroprevalence) showed 256 initially reactive cases, including 232 confirmed positive cases. Of these, 231 individuals were treated with benznidazole (7.5 mg/kg/day) for 60 days and were followed with a strict weekly medical control and follow-up protocol. At the end of the project, 229 patients were examined by the Honduras Secretariat of Health for post-treatment serological assessments; 88.2% seroconverted after 18 months and 93.9% seroconverted after three years. No differences were found in the seroconversion rates according to age or sex. Most of the side effects of the treatment were minor. These results support the argument that in areas where T. cruzi I is predominant and in areas affected by T. cruzi II, when vector transmission has been interrupted, Chagas disease diagnosis and treatment are feasible, necessary and ethically indisputable.

Hantavirus disease is caused by the hantavirus, which is an RNA virus belonging to the family Bunyaviridae. Hantavirus disease is an anthropozoonotic infection transmitted through the inhalation of aerosols from the excreta of hantavirus-infected rodents. In the county of Itacoatiara in the state of Amazonas (AM), Brazil, the first human cases of hantavirus pulmonary and cardiovascular syndrome were described in July 2004. These first cases were followed by two fatal cases, one in the municipality of Maués in 2005 and another in Itacoatiara in 2007. In this study, we investigated the antibody levels to hantavirus in a population of 1,731 individuals from four different counties of AM. Sera were tested by IgG/IgM- enzyme-linked immune-sorbent assay using a recombinant nucleocapsid protein of the Araraquara hantavirus as an antigen. Ten sera were IgG positive to hantavirus (0.6%). Among the positive sera, 0.8% (1/122), 0.4% (1/256), 0.2% (1/556) and 0.9% (7/797) were from Atalaia do Norte, Careiro Castanho, Itacoatiara and Lábrea, respectively. None of the sera in this survey were IgM-positive. Because these counties are distributed in different areas of AM, we can assume that infected individuals are found throughout the entire state, which suggests that hantavirus disease could be a local emerging health problem.

Leprosy in children is correlated with community-level factors, including the recent presence of disease and active foci of transmission in the community. We performed clinical and serological examinations of 1,592 randomly selected school children (SC) in a cross-sectional study of eight hyperendemic municipalities in the Brazilian Amazon Region. Sixty-three (4%) SC, with a mean age of 13.3 years (standard deviation = 2.6), were diagnosed with leprosy and 777 (48.8%) were seropositive for anti-phenolic glycolipid-I (PGL-I). Additionally, we evaluated 256 house-hold contacts (HHCs) of the students diagnosed with leprosy; 24 (9.4%) HHC were also diagnosed with leprosy and 107 (41.8%) were seropositive. The seroprevalence of anti-PGL-I was significantly higher amongst girls, students from urban areas and students from public schools (p < 0.0001). Forty-five (71.4%) new cases detected amongst SC were classified as paucibacillary and 59 (93.6%) patients did not demonstrate any degree of physical disability at diagnosis. The results of this study suggest that there is a high rate of undiagnosed leprosy and subclinical infection amongst children in the Amazon Region. The advantages of school surveys in hyperendemic areas include identifying leprosy patients at an early stage when they show no physical disabilities, preventing the spread of the infection in the community and breaking the chain of transmission.

Terrestrial arthropods from tree canopies in the Pantanal of Mato Grosso, Brazil. This study represents a contribution to the knowledge of the diversity of arthropods associated to the canopy of Vochysia divergens Pohl (Vochysiaceae). Three trees individuals were sampled during two seasonal periods in this region: a) by spraying one tree canopy during high water (February); b) by fogging two tree canopies during low water (September/October). The 15,744 arthropods (183.2±38.9 individuals/m²) obtained from all three trees (86 m²) represented 20 taxonomic orders, 87.1% were Insecta, and 12.9% Arachnida. The dominant groups were Hymenoptera (48.5%; 88.9 individuals/m²), mostly Formicidae (44.5%; 81.4 individuals/m²), followed by Coleoptera (14.0%; 25.5 individuals/m²) and Araneae (10.2%; 19.5 individuals/m²), together representing 62.5% of the total catch. Fourteen (70%) of all orders occurred on three trees. Dermaptera, Isoptera, Neuroptera, Odonata, Plecoptera and Trichoptera were collected from only one tree. Of the total, 2,197 adult Coleoptera collected (25.5±11.3 individuals/m²), 99% were assigned to 32 families and 256 morphospecies. Nitidulidae (17.9% of the total catch; 4.6 individuals/m²), Anobiidae (16.7%; 4.3 individuals/m²), Curculionidae (13.2%; 3.4 individuals/m²) and Meloidae (11.4%; 2.9 individuals/m²) dominated. The communitiy of adult Coleoptera on V. divergens indicated a dominance of herbivores (37.8% of the total catch, 127 spp.) and predators (35.2%, 82 spp.), followed by saprophages (16.2%, 32 spp.) and fungivores (10.8%, 15 spp.). The influence of the flood pulse on the community of arboreal arthropods in V. divergens is indicated by the seasonal variation in evaluated groups, causing changes in their structure and composition.

Protein content of leaf-cutting ant queens before the nuptial flight and during the post-claustral phase. This study evaluated the crude protein content of queens of Atta sexdens before the nuptial flight and after the claustral phase in laboratory and field colonies. The hypothesis was that protein is used for survival of the queen and for early colony growth during the claustral phase. Additionally, the nest morphology, live biomass and adult population of field colonies were evaluated. Crude protein was determined by digestion of the organic material with sulfuric acid at high temperatures. The mean crude protein content was 123.23 ± 11.20 mg for females before the nuptial flight and 70.44 ± 12.21 mg for laboratory-reared queens after the claustral phase. The post-claustral crude protein content of field-collected queen was 55.90 ± 9.18 mg. With respect to the loss of crude protein as a function of duration of the claustral phase, laboratory-reared queens lost 52.79 mg and field-collected queens lost 67.33 mg compared to females before the nuptial flight. A positive linear correlation was observed between the weight of field-collected queens (256.4 ± 36.3 mg) and colony biomass (13.02 ± 9.12 g), but there was no correlation between biomass and nest depth (13.11 ± 3.82 cm). As expected, the present results support the hypothesis that protein is used for survival of the queen and for early colony growth, as demonstrated by the reduction in crude protein content as a function of duration of the claustral phase. To our knowledge, this is the first study to provide data of the dynamics of protein reserves in leaf-cutting ant queens during the claustral phase.

Os efeitos adversos dos metais pesados para as diversas formas de vida dificultam a recuperação de solos contaminados por estes elementos. Neste trabalho, avaliaram-se os efeitos da inoculação com fungos micorrízicos arbusculares no crescimento e absorção de metais de mudas de cinco espécies arbóreas, transplantadas para misturas que continham diferentes proporções de um solo contaminado (PSC). Mudas de Senna multijuga (L.C. Rich.) Irwin et Barneby (cássia verrugosa), Luehea grandiflora Mart. et Zucc. (açoita-cavalo), Enterolobium contortisiliquum (Vell.) Morong (tamboril), Albizia lebbeck (L.) Benth. (albizia) e Senna macranthera (Collard.) Irwin et Barneby (fedegoso), inoculadas e sem inoculação, foram transplantadas para as misturas de solos e desenvolvidas por 180 dias, no período de abril a novembro de 1996, em vasos, em casa de vegetação do Departamento de Ciência do Solo da UFLA, Lavras (MG). Verificou-se que a elevação na PSC na mistura reduziu o desenvolvimento das mudas e a colonização micorrízica (CM), sendo isto causado pela elevada absorção de metais pelas plantas, especiamente, de Cd e Zn. A inoculação favoreceu o crescimento das mudas após transplantio, sendo esse efeito mais evidente nas misturas de solo com baixa PSC. A CM foi reduzida de 70 a 90% no solo não contaminado para valores próximos de zero na mistura com alta PSC. Os níveis críticos de toxidez (redução de 10% na matéria seca das plantas inoculadas) dos metais no solo foram, em mg dm-3, de 83, 57, 153, 256 e 16, para o Zn, e de 1,3; 0,9; 0,8; 4,0 e 1,6, para Cd, para açoita-cavalo, cássia verrugosa, fedegoso, tamboril e albizia, respectivamente. Observando esses níveis críticos, as plantas não inoculadas apresentaram produção de matéria seca relativa, média para todas as espécies, de apenas 39%, evidenciando os benefícios da inoculação para o crescimento pós-transplantio das mudas. Esses benefícios relacionaram-se com menores teores de metais na parte aérea. Mesmo desconhecendo os mecanismos envolvidos nestas respostas, os resultados deste trabalho evidenciaram a importância das micorrizas arbusculares para o crescimento de mudas de árvores e para a recuperação de áreas tropicais contaminadas com metais pesados.

O uso de leguminosas para cobertura do solo e suprimento de N é uma técnica importante na conservação do solo e redução dos custos em fertilizantes minerais. No sul do Brasil, o enfoque principal da pesquisa tem sido sobre espécies de ciclo hibernal. No presente estudo, avaliou-se o efeito do cultivo intercalar de quatro leguminosas estivais [feijão-de-porco (Canavalia ensiformis ), guandu anão (Cajanus cajan), mucuna cinza (Stizolobium niveum ) e soja preta (Glycine sp.)] no fornecimento de N e rendimento de grãos do milho, em três doses de N mineral (0, 60 e 120 kg ha-1), em comparação a um tratamento-testemunha, sem leguminosa. Dois experimentos foram realizados, em sistema de preparo mínimo-PM (1993 a 1997), com abertura de sulcos para semeadura do milho feita com arado de tração animal, e em sistema de preparo convencional-PC (1994 a 1997), com aração e duas gradagens na primavera-verão, num Latossolo Vermelho distroférrico, em Chapecó (SC). Na média das safras, a produção de matéria seca das leguminosas estivais variou de 1,26 a 5,48 Mg ha-1, e o N na fitomassa da parte aérea, de 31 a 132 kg ha-1. Em comparação ao tratamento-testemunha, sem N mineral, as leguminosas aumentaram o rendimento do milho em 17 a 93 % (423 a 2.256 kg ha-1), no PC, e em 12 a 43 % (281 a 1.030 kg ha-1), no PM, sendo estes incrementos equivalentes à aplicação de 12 a > 120 kg ha-1 de N mineral no PC e de 50 a 86 kg ha-1 de N mineral no PM. Portanto, o cultivo intercalar de leguminosas estivais, além de proteger o solo, representa uma técnica promissora no forneci-mento de N e aumento de rendimento do milho, recomendada principalmente para pequenas propriedades rurais, onde a colheita do milho é manual.

A capacidade de troca de cátions (CTC) é uma propriedade físico-química intrínseca aos constituintes minerais e orgânicos do solo. Apesar do uso de diferentes extratores e procedimentos, a CTC é normalmente expressa considerando apenas o controle ou não do pH na solução extratora. O objetivo deste trabalho foi discutir o significado da contribuição da matéria orgânica do solo prepresentada pelo carbono orgânico total (COT) e da argila à capacidade de troca de cátions de um Argissolo quando diferentes métodos estão envolvidos na determinação desse parâmetro. Para isso, utilizaram-se 75 amostras de um Argissolo Vermelho-Amarelo distrófico abrúptico da área do Departamento de Solos da Universidade Federal de Santa Maria, representando, em triplicata, cinco profundidades e cinco sistemas de uso e manejo de solo. A CTC efetiva (CTC E) foi estimada pelo cloreto de hexamina cobalto (CTC E Cohex) e pela soma de cátions Al3+, Ca2+, Mg2+ e K+ (CTC E SB + AlKCl), os três primeiros extraídos por KCl e o último por Mehlich-1; a CTC em pH 7,0 (CTC7) foi estimada por acetato de amônio (CTC7 Metson) e pela soma de cátions Ca2+, Mg2+ e K+ e H + Al estimado pelo índice SMP (CTC7 SB + H + AlSMP). Os valores de CTC obtidos pelos diferentes métodos se relacionam entre si, com coeficientes de correlação linear simples acima de 0,93. Os valores de CTC7 Metson são subestimados quando comparados com o método CTC7 (SB + H + AlSMP). Nesse sentido, as contribuições da argila e do COT à CTC7 foram, respectivamente, menores para a CTC7 Metson, 19 e 256 cmol c kg-1, que para a CTC7 (SB + H + AlSMP), 23 e 399 cmol c kg-1. A contribuição dos constituintes de solo depende, então, do cátion extrator e da capacidade de extração dos métodos empregados.

Random amplified polymorphic DNA markers (RAPD) were used to estimate the variability of 14 genotypes of Brazilian wheat (Triticum aestivum L.), using a set of 50 random 10mer primers. A total of 256 reproducibly scorable DNA amplification products were obtained from 48 of the primers, 83% of which were polymorphic. Genetic distances among genotypes were calculated and a dendrogram and a principal coordinates analysis showing the genetic relationships among them were obtained. Despite the low variability found (average genetic distance of 27%), two groups of genotypes could be identified, which probably reflect how they were formed. Studies such as this one may be important in the planning and development of future improvement programs for this plant species.
