The goal of this study was to investigate the pattern of inflammatory response induced by Lagochilascaris minor in murine experimental model. For this purpose 115 mice were given 1000-3000 L. minor infective eggs "per os" and 51 uninfected mice were considered as controls. Four hours post-inoculation (PI), 3rd stage larvae were seen passing through the mucosa of terminal ends of small intestine. Six hours PI larvae were observed as an embolus inside the portal vein and also migrating through the liver parenchyma. During the first 24 h larvae-containing eggs of L. minor were observed in the lumen of intestinal tract. Two days PI larvae were seen migrating through lung parenchyma associated with an initial neutrophilic perivasculitis. From the 13th day of this experimental study, L. minor larvae were found mainly in skeletal muscles, in the center of granulomas. Concentric fibrosis with mixed inflammatory infiltrate involved the larvae after the 47th day PI, persistently. This experimental murine study with L. minor indicated that the 3rd stage larvae penetrated via ileum-cecal mucosa reaching the liver and probably other tissues through the hematogenic via. Throughout its pathway the larvae induced a granulomatous reaction, with abundant polimorphonuclear cells.

The symptomatic phases of many inflammatory diseases are characterized by migration of large numbers of neutrophils (PMN) across a polarized epithelium and accumulation within a lumen. For example, acute PMN influx is common in diseases of the gastrointestinal system (ulcerative colitis, Crohn's disease, bacterial enterocolitis, gastritis), hepatobiliary system (cholangitis, acute cholecystitis), respiratory tract (bronchial pneumonia, bronchitis, cystic fibrosis, bronchiectasis), and urinary tract (pyelonephritis, cystitis). Despite these observations, the molecular basis of leukocyte interactions with epithelial cells is incompletely understood. In vitro models of PMN transepithelial migration typically use N-formylated bacterial peptides such as fMLP in isolation to drive human PMNs across epithelial monolayers. However, other microbial products such as lipopolysaccharide (LPS) are major constituents of the intestinal lumen and have potent effects on the immune system. In the absence of LPS, we have shown that transepithelial migration requires sequential adhesive interactions between the PMN beta2 integrin CD11b/CD18 and JAM protein family members. Other epithelial ligands appear to be abundantly represented as fucosylated proteoglycans. Further studies indicate that the rate of PMN migration across mucosal surfaces can be regulated by the ubiquitously expressed transmembrane protein CD47 and microbial-derived factors, although many of the details remain unclear. Current data suggests that Toll-like receptors (TLR), which recognize specific pathogen-associated molecular patterns (PAMPs), are differentially expressed on both leukocytes and mucosal epithelial cells while serving to modulate leukocyte-epithelial interactions. Exposure of epithelial TLRs to microbial ligands has been shown to result in transcriptional upregulation of inflammatory mediators whereas ligation of leukocyte TLRs modulate specific antimicrobial responses. A better understanding of these events will hopefully provide new insights into the mechanisms of epithelial responses to microorganisms and ideas for therapies aimed at inhibiting the deleterious consequences of mucosal inflammation.

Sepsis is a systemic inflammatory response commonly caused by bacterial infection. We demonstrated that the outcome of sepsis induced by cecal ligation and puncture (CLP) correlates with the severity of the neutrophil migration failure towards infectious focus. Failure appears to be due to a decrease in the rolling and adhesion of neutrophil to endothelium cells. It seems that neutrophil migration impairment is mediated by the circulating inflammatory cytokines, such as TNF-alpha and IL-8, which induce the nitric oxide (NO) production systemically. It is supported by the fact that intravenous administration of these cytokines reduces the neutrophil migration induced by different inflammatory stimuli, and in severe sepsis the circulating concentrations of the cytokines and chemokines are significantly increased. Moreover, the neutrophil migration failure and the reduction in the rolling/adhesion were not observed in iNOS-/- mice and, aminoguanidine prevented this event. We also demonstrated that the failure of neutrophil migration is a Toll-4 receptor (TLR4) dependent mechanism, since it was not observed in TLR4 deficient mice. Furthermore, it was also observed that circulating neutrophils obtained from septic patients present failure of neutrophil chemotaxis toward fMLP, IL-8, and LTB4 and an increased in sera concentrations of NO3 and cytokines. In conclusion, we demonstrated that, in sepsis, failure of neutrophil migration is critical for the outcome and that NO is involved in the process.

Human infection with the protozoa Trypanosoma cruzi extends through North, Central, and South America, affecting 21 countries. Most human infections in the Western Hemisphere occur through contact with infected bloodsucking insects of the triatomine species. As T. cruzi can be detected in the blood of untreated infected individuals, decades after infection took place; the infection can be also transmitted through blood transfusion and organ transplant, which is considered the second most common mode of transmission for T. cruzi. The third mode of transmission is congenital infection. Economic hardship, political problems, or both, have spurred migration from Chagas endemic countries to developed countries. The main destination of this immigration is Australia, Canada, Spain, and the United States. In fact, human infection through blood or organ transplantation, as well as confirmed or potential cases of congenital infections has been described in Spain and in the United States. Estimates reported here indicates that in Australia in 2005-2006, 1067 of the 65,255 Latin American immigrants (16 per 1000) may be infected with T. cruzi, and in Canada, in 2001, 1218 of the 131,135 immigrants (9 per 1000) whose country of origin was identified may have been also infected. In Spain, a magnet for Latin American immigrants since the 2000, 5125 of 241,866 legal immigrants in 2003 (25 per 1000), could be infected. In the United States, 56,028 to 357,205 of the 7,20 million, legal immigrants (8 to 50 per 1000), depending on the scenario, from the period 1981-2005 may be infected with T. cruzi. On the other hand, 33,193 to 336,097 of the estimated 5,6 million undocumented immigrants in 2000 (6 to 59 per 1000) could be infected. Non endemic countries receiving immigrants from the endemic ones should develop policies to protect organ recipients from T. cruzi infection, prevent tainting the blood supply with T. cruzi, and implement secondary prevention of congenital Chagas disease.

Aedes albopictus was responsible for transmission in the first outbreak of chikungunya (CHIK) on La Réunion Island, Indian Ocean, in 2005-2006. The magnitude of the outbreak on this island, which had been free of arboviral diseases for over 30 years, as well as the efficiency of Ae. albopictus as the main vector, raises questions about the maintenance of the CHIK virus (CHIKV) through vertical transmission mechanisms. Few specimens collected from the field as larvae were found to be infected. In this study, Ae. albopictus originating from La Réunion were orally infected with a blood-meal containing 10(8) pfu/mL of the CHIKV epidemic strain (CHIKV 06.21). Eggs from the first and second gonotrophic cycles were collected and raised to the adult stage. The infectious status of the progeny was checked (i) by immunofluorescence on head squashes of individual mosquitoes to detect the presence of viral particles or (ii) by quantitative RT-PCR on mosquito pools to detect viral RNA. We analysed a total of 1,675 specimens from the first gonotrophic cycle and 1,709 from the second gonotrophic cycle without detecting any viral particles or viral RNA. These laboratory results are compared to field records.

The general objective of this article was to study the process of illegal migration of Brazilian women to Switzerland and, more specifically, to analyze the motivations for migration, the difficulties found upon arrival, the circumstances that led them to get married and lastly an evaluation of the whole experience. The methodology was based in a qualitative approach. The participants were eight Brazilian women that illegally migrated to Switzerland but now are married with Swiss or European men. They responded to an interview focused in the objectives of the study. The appreciation of the data was realized with Minayo´s Content Analysis. The motivations were related to a bettering of the financial situation. In the difficulties encountered, we can bring out illegality, language and prejudice. Regarding marriage, they married to stay legally in the country. Finally, the evaluation of the experience was negative for most participants.

O objetivo deste trabalho foi verificar a forma de espacialização vertical e horizontal dos indicadores de qualidade química, física e hídrica de um Latossolo Vermelho distrófico textura média, cultivado com citros. O pomar de laranjeira Valência sobre limoeiro Cravo foi plantado em 1995, em Alto Paraná, região noroeste do Paraná. Em agosto de 2002, coletaram-se amostras deformadas e indeformadas de solo para realizar determinações químicas, físicas e hídricas. A amostragem foi feita em 24 transeções perpendiculares aos troncos das laranjeiras, em duas profundidades (0-0,20 e 0,20-0,40 m) e em três posições (projeção da copa da laranjeira, rodado da entrelinha e centro da entrelinha). Os indicadores avaliados foram submetidos às análises multivariadas, por meio dos componentes principais, e agrupados pelo método de Tocher. Os resultados demonstraram que a qualidade do solo apresentou espacialização heterogênea entre os indicadores de qualidade física, química e hídrica do solo, resultante das ações de manejo do pomar de laranjeira. A integração dos indicadores de qualidade física, química e hídrica do solo caracterizou a espacialização vertical da qualidade do solo (0-0,40 m) na projeção da copa da laranjeira e a espacialização vertical e horizontal (0-0,20 e 0,20-0,40 m) no rodado da entrelinha e centro da entrelinha. No rodado da entrelinha (0-0,20 m) ocorreu a maior restrição física e hídrica associada à melhor condição química do solo para as laranjeiras.

Os sedimentos transportados pelas águas barrentas do rio Guamá e a heterogeneidade dessas substâncias são responsáveis pela formação do solo e pela diversidade de características químicas na área inundável. Este trabalho teve como objetivo determinar os atributos físicos e químicos de um solo de várzea baixa do rio Guamá, em diferentes épocas, cultivado com canarana de Paramaribo (Echinochloa polystachya H.B.K) e canarana erecta lisa (Echinochloa pyramidales Lam). O delineamento experimental foi inteiramente casualizado, em esquema fatorial 4 x 4 (quatro profundidades e quatro épocas de amostragem), com seis repetições. Amostras compostas de solo foram coletadas em seis pontos eqüidistantes, a partir de uma transecção, nas profundidades de 0-10, 10-20, 20-30 e 30-40 cm, nos meses de maio, agosto e novembro/2002 e fevereiro/2003. Foram determinados a granulometria, matéria orgânica, pH em H2O, pH em KCl, P extraível, K trocável, Al trocável, Ca e Mg trocáveis e os micronutrientes: Cu, Mn, Zn e Fe. Os solos da área estudada estão inseridos na unidade dos Gleissolos e caracterizam-se por serem pouco desenvolvidos, mal drenados, normalmente ácidos, ocorrendo no perfil horizonte franco-argilo-siltoso. Houve influência da sazonalidade na composição química do solo; na época mais chuvosa e de inundação mais intensa da várzea, correspondente ao mês de fevereiro, observou-se aumento do pH nos teores de Cu e de Fe e decréscimos nos de Mg e de Al trocável; no período de menor umidade do solo, a saturação por bases e a CTC foram mais elevadas. O Fe foi o nutriente que apresentou maior variação nos seus teores com a inundação do solo, cujo aumento foi superior a 1.000 % no período de maior inundação do solo (fevereiro).

A magnitude da mobilidade vertical dos nutrientes no perfil afeta o contato destes com as raízes e a lixiviação e, por isso, influencia a época e o método de aplicação dos fertilizantes ao solo. O presente trabalho objetivou avaliar a mobilidade de K em solos de acordo com o método de aplicação e a dose de cloreto de potássio. O experimento foi realizado em 1998, em colunas de lixiviação com 7,5 cm de diâmetro e 35 cm de altura. Os tratamentos consistiram de doses de K (0, 150 e 300 mg kg-1), aplicadas sobre a superfície ou incorporadas até 15 cm de profundidade em dois solos ácidos. A cada sete dias, durante oito semanas, foram adicionados 300 mL de água destilada sobre a superfície de cada coluna. A solução percolada foi coletada no dia seguinte, e nela foram determinados o volume e as concentrações de Ca, Mg e K. A aplicação de KCl sobre a superfície dos solos promoveu a descida de K para profundidades superiores a 10 cm. Apesar disso, a lixiviação de K foi pequena, porém aumentou com a dose e com a incorporação do fertilizante ao solo e foi mais intensa nas primeiras percolações. A adição de KCl aumentou expressivamente a percolação de Ca e Mg durante as cinco primeiras percolações, o que pode representar aumento temporário na disponibilidade desses cátions às plantas, pois coincide com o período de implantação das culturas, em que a exigência das plantas por nutrientes é alta. Mesmo nesses solos com alto tamponamento, a adição de K sobre a superfície promoveu bom aprofundamento do nutriente no perfil sem, contudo, proporcionar grande lixiviação.

A intensa mobilização do solo no sistema cultivo convencional causou degradação da estrutura, compactação e adensamento do solo abaixo da camada arável, reduzindo a macroporosidade e a taxa de infiltração de água no solo, com conseqüente aumento de escoamento superficial, de erosão e de assoreamento de rios e de reservatórios. A semeadura direta, que protege a superfície do solo, praticamente controlou a perda por erosão hídrica, os terraços foram eliminados, pelos agricultores, e a conseqüência foi maior escoamento superficial do que no sistema cultivo convencional. Com o objetivo de avaliar o comportamento hidrológico do vertical mulching em semeadura direta, em relação ao escoamento superficial, realizou-se esta pesquisa nos anos agrícolas 2002/2003 e 2003/2004, em Latossolo Vermelho distrófico típico, na região fisiográfica do planalto médio do Rio Grande do Sul, isto é, um experimento em escala de campo, com parcelas sem vertical mulching, com vertical mulching a cada 10 m e com vertical mulching a cada 5 m, no delineamento de blocos ao acaso com três repetições. Sulcos em nível, perpendiculares ao declive, com dimensões de 0,08 m de largura por x 0,38 m de profundidade, foram abertos e preenchidos com palha compactada o suficiente para prevenir desmoronamento das paredes do sulco. No ciclo da cultura da soja e do trigo foram simuladas chuvas com duas intensidades, 70 e 106 mm h-1. Foram determinados o escoamento superficial, a taxa de infiltração básica de água no solo e a concentração de nutrientes e de CO no escoamento superficial. Os resultados indicam que o vertical mulching na semeadura direta reduz, significativamente, o volume do escoamento superficial aumenta a taxa de infiltração básica de água no solo e reduz também as perdas totais de nutrientes e de CO devido ao menor volume de água no escoamento superficial.
"Vertical mulching" como prática conservacionista para manejo de enxurrada em sistema plantio direto

Nas regiões brasileiras de clima temperado e subtropical úmido, situadas ao sul do paralelo 24° de latitude sul, as características fundamentais da chuva apresentam potencial para superar a taxa de infiltração de água no solo e gerar enxurrada, em qualquer época do ano, independentemente do uso e do manejo de solo. O sistema plantio direto não tem sido implementado com a plenitude das práticas conservacionistas requeridas pelas condições de solo dessas regiões do País para conter o potencial erosivo de chuvas intensas. A enxurrada resulta na produção de sedimento enriquecido por agroquímicos, que, além de promover prejuízos econômicos ao sistema agrícola produtivo, pode afetar a ambiência. Este trabalho teve por objetivo avaliar a eficiência da prática conservacionista vertical mulching no manejo de enxurrada em lavouras sob sistema plantio direto, por meio da quantificação do índice de enriquecimento do sedimento. O estudo contemplou amostragens pareadas de solo em lavouras sem e com a prática conservacionista vertical mulching. As amostras pareadas foram constituídas por solo representativo de cada lavoura e pelo respectivo sedimento gerado por erosão hídrica. As amostras foram submetidas à determinação de pH em água, índice SMP, P disponível, K, Ca, Mg e Al trocáveis e matéria orgânica, e calculadas a soma de bases, a saturação por bases e a capacidade de troca de cátions efetiva. Os resultados indicam que a prática conservacionista vertical mulching reduz o grau de enriquecimento do sedimento produzido e previne o carreamento deste para fora da lavoura, minimizando perdas econômicas e riscos à ambiência. Em adição, infere-se que o sistema plantio direto, sem práticas complementares para manejo de enxurrada, não representa manejo conservacionista de solo capaz de prevenir degradação ambiental.

The deficiency or excess of micronutrients has been determined by analyses of soil and plant tissue. In Brazil, the lack of studies that would define and standardize extraction and determination methods, as well as lack of correlation and calibration studies, makes it difficult to establish limits of concentration classes for analysis interpretation and fertilizer recommendations for crops. A specific extractor for soil analysis is sometimes chosen due to the ease of use in the laboratory and not in view of its efficiency in determining a bioavailable nutrient. The objectives of this study were to: (a) evaluate B concentrations in the soil as related to the fertilizer rate, soil depth and extractor; (b) verify the nutrient movement in the soil profile; (c) evaluate efficiency of Hot Water, Mehlich-1 and Mehlich-3 as available B extractors, using sunflower as test plant. The experimental design consisted of complete randomized blocks with four replications and treatments of five B rates (0, 2, 4, 6, and 8 kg ha-1) applied to the soil surface and evaluated at six depths (0-0.05, 0.05-0.10, 0.10-0.15, 0.15-0.20, 0.20-0.30, and 0.30-0.40 m). Boron concentrations in the soil extracted by Hot Water, Mehlich-1 and Mehlich-3 extractors increased linearly in relation to B rates at all depths evaluated, indicating B mobility in the profile. The extractors had different B extraction capacities, but were all efficient to evaluate bioavailability of the nutrient to sunflower. Mehlich-1 and Mehlich-3 can therefore be used to analyze B as well as Hot Water.

One of the expected benefits of no-tillage systems is a higher rate of soil C sequestration. However, higher C retention in soil is not always apparent when no-tillage is applied, due e.g., to substantial differences in soil type and initial C content. The main purpose of this study was to evaluate the potential of no-tillage management to increase the stock of total organic C in soils of the Pampas region in Argentina. Forty crop fields under no-tillage and conventional tillage systems and seven undisturbed soils were sampled. Total organic C, total N, their fractions and stratification ratios and the C storage capacity of the soils under different managements were assessed in samples to a depth of 30 cm, in three layers (0-5, 5-15 and 15-30 cm). The differences between the C pools of the undisturbed and cultivated soils were significant (p < 0.05) and most pronounced in the top (0-5 cm) soil layer, with more active C near the soil surface (undisturbed > no-tillage > conventional tillage). Based on the stratification ratio of the labile C pool (0-5/5-15 cm), the untilled were separated from conventionally tilled areas. Much of the variation in potentially mineralizable C was explained by this active C fraction (R² = 0.61) and by total organic C (R² = 0.67). No-till soils did not accumulate more organic C than conventionally tilled soils in the 0-30 cm layer, but there was substantial stratification of total and active C pools at no till sites. If the C stratification ratio is really an indicator of soil quality, then the C storage potential of no-tillage would be greater than in conventional tillage, at least in the surface layers. Particulate organic C and potentially mineralizable C may be useful to evaluate variations in topsoil organic matter.

The objective of this work was to evaluate the density, dynamics and vertical distribution of a Hrabeiella periglandulata population in a forest soil at Brno, Czech Republic. From December 2003 to November 2004, two plots covered by mixed stands and two covered by coniferous stands were sampled monthly. Six soil cores per plot were taken down to 15 cm and subdivided into layers, which were subjected to wet funnel extraction. Missing in one of the coniferous stands H. periglandulata was abundant in the mixed stand with the highest soil pH. In this stand, monthly sampling continued until November 2005, with three additional samplings up to January 2007. Mean annual density was 2,672±1,534 individuals m-2. Population dynamics differed from those reported from Germany. Highest densities were reached in early summer, lowest between August and December. Due to aggregated horizontal distribution, differences between monthly values were often nonsignificant. No significant correlation with climatic data was found. Nevertheless, the observed dynamics corresponded to the climatic conditions, showing particularly the negative effect of drought. The population was evenly distributed in the sampled soil profile, only avoiding the organic layer. Except for a locality in Poland, this is the easternmost record of the species.

The objective of this work was to determine the contribution of dissolved organic carbon (DOC) from a biochar mineral complex (BMC), so as to better understand the interactions between DOC, biochar, clay, and minerals during thermal treatment, and the effects of BMC on amended soils. The BMC was prepared by heating a mixture of a H3PO4-treated saligna biochar from Acacia saligna, clays, other minerals, and chicken manure. The BMC was applied to a sandy loam soil in Western Australia, where wheat was grown. Liquid chromatography-organic carbon detection (LC-OCD) tests were carried out on water extracts from the untreated biochar, the BMC, the BMC-amended soil, and on a control soil to measure the DOC concentration. LC-OCD tests provide a fingerprint of the DOC, which allows the fractions of DOC to be determined. Thermal processing enhanced the reaction of the A. saligna biochar with manure, clays and minerals, and affected the distribution of the DOC fractions. Notably, the process leads to immobilization of hydrophobic DOC and to an increase in the concentration of low-molecular-weight neutrals in the BMC. The application of the BMC to soil increases the DOC in the amended soil, especially the biopolymer fraction.
