Twenty-eight Chagas disease patients (CD), 22 with the indeterminate clinical form (IND) and six with the cardiac or digestive form (CARD/DIG), were treated with benznidazole and underwent clinical and laboratorial analysis before (IND and CARD/DIG) and nine years after [patients after treatment (CDt), patients with the indeterminate clinical form at treatment onset (INDt) and with the cardiac or digestive form at treatment onset (CARD/DIGt)] treatment. The data demonstrate that 82.1% of CDt patients (23/28) remained clinically stable and 95.4% of the INDt (21/22) and 33.3% of the CARD/DIGt (2/6) patients showed unaltered physical and laboratorial examinations. The clinical evolution rate was 2%/year and was especially low in INDt patients (0.5%/year) relative to CARD/DIGt patients (7.4%/year). Positive haemoculture in treated patients was observed in 7.1% of the cases. None of the INDt (0/21) and 33.3% of the CARD/DIGt (2/6) patients displayed positive cultures. The PCR presented a positive rate significantly higher (85.2%, 23/27) than haemoculture and two samples from the same patient revealed the same result 57.7% of the patients. Conventional serology-ELISA on 16 paired samples remained positive in all individuals. Semi-quantitative ELISA highlighted significant decreases in reactivity, particularly in INDt relative to IND. Non-conventional serology-FC-ALTA-IgG, after treatment, showed positive results in all sera and 22 paired samples examined at seven and nine years after treatment, demonstrated significantly lower reactivity, particularly in INDt patients. This study was retrospective in nature, had a low number of samples and lacked an intrinsic control group, but the data corroborate other results found in the literature. The data also demonstrate that, even though a cure has not been detected in the none-treated patients, the benefits for clinical evolution were selectively observed in the group of INDt patients and did not occur for CARD/DIGt patients.

Several protozoan parasites exist in the Trypanosomatidae family, including various agents of human diseases. Multiple lines of evidence suggest that important differences are present between the translational and mRNA processing (trans splicing) systems of trypanosomatids and other eukaryotes. In this context, certain small complexes of RNA and protein, which are named small nuclear ribonucleoproteins (U snRNPs), have an essential role in pre-mRNA processing, mainly during splicing. Even though they are well defined in mammals, snRNPs are still not well characterized in trypanosomatids. This study shows that a U5-15K protein is highly conserved among various trypanosomatid species. Tandem affinity pull-down assays revealed that this protein interacts with a novel U5-102K protein, which suggests the presence of a sub-complex that is potentially involved in the assembly of U4/U6-U5 tri-snRNPs. Functional analyses showed that U5-15K is essential for cell viability and is somehow involved with the trans and cis splicing machinery. Similar tandem affinity experiments with a trypanonosomatid U5-Cwc21 protein led to the purification of four U5 snRNP specific proteins and a Sm core, suggesting U5-Cwc-21 participation in the 35S U5 snRNP particle. Of these proteins, U5-200K was molecularly characterized. U5-200K has conserved domains, such as the DEAD/DEAH box helicase and Sec63 domains and displays a strong interaction with U5 snRNA.

An online scheme to assign Stenotrophomonas isolates to genomic groups was developed using the multilocus sequence analysis (MLSA), which is based on the DNA sequencing of selected fragments of the housekeeping genes ATP synthase alpha subunit (atpA), the recombination repair protein (recA), the RNA polymerase alpha subunit (rpoA) and the excision repair beta subunit (uvrB). This MLSA-based scheme was validated using eight of the 10 Stenotrophomonas species that have been previously described. The environmental and nosocomial Stenotrophomonas strains were characterised using MLSA, 16S rRNA sequencing and DNA-DNA hybridisation (DDH) analyses. Strains of the same species were found to have greater than 95% concatenated sequence similarity and specific strains formed cohesive readily recognisable phylogenetic groups. Therefore, MLSA appeared to be an effective alternative methodology to amplified fragment length polymorphism fingerprint and DDH techniques. Strains of Stenotrophomonas can be readily assigned through the open database resource that was developed in the current study (www.steno.lncc.br/).

Rocio virus (ROCV) is an encephalitic flavivirus endemic to Brazil. Experimental flavivirus infections have previously demonstrated a persistent infection and, in this study, we investigated the persistence of ROCV infection in golden hamsters (Mesocricetus auratus). The hamsters were infected intraperitoneally with 9.8 LD50/0.02 mL of ROCV and later anaesthetised and sacrificed at various time points over a 120-day period to collect of blood, urine and organ samples. The viral titres were quantified by real-time-polymerase chain reaction (qRT-PCR). The specimens were used to infect Vero cells and ROCV antigens in the cells were detected by immunefluorescence assay. The levels of antibodies were determined by the haemagglutination inhibition technique. A histopathological examination was performed on the tissues by staining with haematoxylin-eosin and detecting viral antigens by immunohistochemistry (IHC). ROCV induced a strong immune response and was pathogenic in hamsters through neuroinvasion. ROCV was recovered from Vero cells exposed to samples from the viscera, brain, blood, serum and urine and was detected by qRT-PCR in the brain, liver and blood for three months after infection. ROCV induced histopathological changes and the expression of viral antigens, which were detected by IHC in the liver, kidney, lung and brain up to four months after infection. These findings show that ROCV is pathogenic to golden hamsters and has the capacity to cause persistent infection in animals after intraperitoneal infection.

This review investigates ancient infectious diseases in the Americas dated to the pre-colonial period and considers what these findings can tell us about the history of the indigenous peoples of the Americas. It gives an overview, but focuses on four microbial pathogens from this period: Helicobacter pylori, Mycobacterium tuberculosis, Trypanosoma cruzi and Coccidioides immitis, which cause stomach ulceration and gastric cancer, tuberculosis, Chagas disease and valley fever, respectively. These pathogens were selected as H. pylori can give insight into ancient human migrations into the Americas, M. tuberculosis is associated with population density and urban development, T. cruzi can elucidate human living conditions and C. immitis can indicate agricultural development. A range of methods are used to diagnose infectious disease in ancient human remains, with DNA analysis by polymerase chain reaction one of the most reliable, provided strict precautions are taken against cross contamination. The review concludes with a brief summary of the changes that took place after European exploration and colonisation.

Trypanosoma cruzi strains from distinct geographic areas show differences in drug resistance and association between parasites genetic and treatment response has been observed. Considering that benznidazole (BZ) can reduce the parasite burden and tissues damage, even in not cured animals and individuals, the goal is to assess the drug response to BZ of T. cruzi II strains isolated from children of the Jequitinhonha Valley, state of Minas Gerais, Brazil, before treatment. Mice infected and treated with BZ in both phases of infection were compared with the untreated and evaluated by fresh blood examination, haemoculture, polymerase chain reaction, conventional (ELISA) and non-conventional (FC-ALTA) serologies. In mice treated in the acute phase, a significant decrease in parasitaemia was observed for all strains. Positive parasitological and/or serological tests in animals treated during the acute and chronic (95.1-100%) phases showed that most of the strains were BZ resistant. However, beneficial effect was demonstrated because significant reduction (p < 0.05%) and/or suppression of parasitaemia was observed in mice infected with all strains (acute phase), associated to reduction/elimination of inflammation and fibrosis for two/eight strains. BZ offered some benefit, even in not cured animals, what suggest that BZ use may be recommended at least for recent chronic infection of the studied region.

Reports of triatomine infestation in urban areas have increased. We analysed the spatial distribution of infestation by triatomines in the urban area of Diamantina, in the state of Minas Gerais, Brazil. Triatomines were obtained by community-based entomological surveillance. Spatial patterns of infestation were analysed by Ripley’s K function and Kernel density estimator. Normalised difference vegetation index (NDVI) and land cover derived from satellite imagery were compared between infested and uninfested areas. A total of 140 adults of four species were captured (100 Triatoma vitticeps, 25Panstrongylus geniculatus, 8 Panstrongylus megistus, and 7 Triatoma arthurneivai specimens). In total, 87.9% were captured within domiciles. Infection by trypanosomes was observed in 19.6% of 107 examined insects. The spatial distributions ofT. vitticeps, P. geniculatus, T. arthurneivai, and trypanosome-positive triatomines were clustered, occurring mainly in peripheral areas. NDVI values were statistically higher in areas infested by T. vitticeps and P. geniculatus. Buildings infested by these species were located closer to open fields, whereas infestations of P. megistus andT. arthurneivai were closer to bare soil. Human occupation and modification of natural areas may be involved in triatomine invasion, exposing the population to these vectors.

Este estudo teve por objetivo compreender o cuidado as pessoas com HIV/aids, na perspectiva de profissionais de saúde, em Portugal. Foi desenvolvido através do método da história oral, de Thompson, com a participação de 22 profissionais de saúde. Os dados foram obtidos através de entrevista semiestruturada e analisados com base na perspectiva de coletânea de narrativas, propostas pelo autor com o apoio do programa QSR NVivo. Os aspectos éticos foram obedecidos ao longo do estudo. O cuidado foi agrupado em três dimensões: cognitiva, afetiva-relacional e técnico-instrumental. A dimensão cognitiva destacou-se no período da revelação do diagnóstico de HIV/aids e ao longo da evolução da doença. A dimensão afetivo-relacional foi transversal e valorizada em todo o processo, desde o diagnóstico até à morte das pessoas com HIV/aids. A dimensão técnico-instrumental foi mais expressiva na fase mais avançada da doença, em situação de dependência e de terminalidade. Diante do exposto, podemos concluir que estas três dimensões são fundamentais para o cuidado à pessoa com HIV/aids.

Taxonomic study of Leschenaultia Robineau-Desvoidy (Diptera, Tachinidae). The genus Leschenaultia Robineau-Desvoidy, 1830 is redescribed. Two genera are considered as its junior synonyms: Echinomasicera Townsend, 1915 syn. nov. and Parachaetopsis Blanchard, 1959 syn. nov. Thirty two especies are treated, as follows: 18 described as new, Leschenaultia aldrichi, sp. nov. (Brazil, Santa Catarina), L. arnaudi sp. nov. (Haiti, La Salle), L. bergenstammi sp. nov. (Peru, San Martin), L. bessi sp. nov. (Brazil, Santa Catarina), L. bigoti sp. nov. (Peru, Huanuco), L. blanchardi sp. nov. (Equador, Cuenca), L. braueri sp. nov. (Brazil, Mato Grosso), L. brooksi sp. nov. (Brazil, Rio de Janeiro), L. coquilletti sp. nov. (Brazil, Santa Catarina); L. cortesi sp. nov. (Venezuela, Maracay), L. currani sp. nov. (Brazil, São Paulo), L. loewi sp. nov. (Mexico, Vera Cruz), L. macquarti sp. nov. (U. S. A., Arizona), L. reinhardi sp. nov. (Canada, Quebec), L. sabroskyi sp. nov. from (U. S. One species was not examined, Leschenaultia nigrisquamis (Townsend, 1892), and two were not recognized, L. trichopsis (Bigot, 1887) and L. hirta Robineau-Desvoidy, 1830. Keys for Nearctic and Neotropical species (only for males) are provided, as well as geographical distribution and illustrations for each species.

Com o objetivo de caracterizar a fauna local de insetos foram obtidas amostras semanais, de setembro/1999 a agosto/2000, utilizando-se armadilhas Malaise instaladas na borda da floresta e no seu interior. Uma análise temporal foi realizada com as espécies de Syrphidae coletadas há, aproximadamente, dezessete anos no mesmo local, dentro da floresta. A abundância e a riqueza de espécies também foram avaliadas. Tanto a riqueza quanto a abundância foram maiores na borda da floresta. Comparando-se os dados atuais com aqueles obtidos em 1986/1987, observa-se um decréscimo na abundância e também na riqueza de espécies de Syrphidae. A espécie mais abundante na borda foi Allograpta neotropica Curran, 1936 e no interior (1999/2000), Ocyptamus sativus (Curran, 1941). Os espécimens de Toxomerus Macquart, 1855 foram os mais abundantes na armadilha localizada na borda da floresta e os de Ocyptamus Macquart, 1834 no interior. Noventa e cinco espécies foram identificadas em 22 gêneros. Ocyptamus foi o gênero com maior riqueza de espécies (23). Na seqüência estão Copestylum Macquart, 1846 (15), Toxomerus (15) e Microdon Meigen, 1803 (10). Sete espécies foram comuns aos três levantamentos: Allograpta neotropica; Copestylum selectum (Curran, 1939); Leucopodella gracilis (Williston, 1891); Mixogaster polistes Hull, 1954; Ocyptamus funebris Macquart, 1834; Toxomerus procrastinatus Metz, 2001 e Toxomerus tibicen (Wiedemann, 1830). Três novas espécies de Microdon, uma de Toxomerus, uma de Aristosyrphus Curran, 1941 e uma de Myolepta Newman, 1838 foram identificadas.

Este artigo examina a visão do Banco Mundial sobre a primeira infância como exemplo da globalização da infância. Argumenta que a globalização é caracterizada por um ponto de vista econômico neoliberal que exacerba a desigualdade entre nações ricas e pobres e entre ricos e pobres, dentro de cada país, bem como por um otimismo tecnológico-científico que enfatiza os benefícios das intervenções tecnológicas. O artigo mostra como esses dois aspectos da globalização influenciaram as políticas para crianças pequenas que o Banco Mundial vem promovendo ativamente. Recorre aos indícios apresentados pelo Banco Mundial para exemplificar as contradições inerentes a essa abordagem.

Este artigo tem a finalidade de apresentar e descrever o funcionamento do Banco de Dados do jornal Zero Hora. Usando tecnologia avançada, seus sistemas de processamento, armazenamento, controle, recuperação e disseminação da informação permitem gerenciar as bases de dados e material informacional em texto e imagem

Este trabalho teve como objetivo determinar as curvas de dessorção das sementes de milho-doce (Zea mays L.), cultivares Superdoce e Doce Cristal, e ajustar diferentes modelos matemáticos aos dados obtidos. As sementes das duas cultivares foram submetidas à dessorção em diversos níveis de temperatura (30, 40, 50 e 60°C), combinados com diferentes umidades relativas do ar (30, 40, 50 e 60%), até atingirem a umidade de equilíbrio. Os seguintes modelos matemáticos foram ajustados por análise de regressão: Henderson-Thompson, Chung-Pfost, Copace, Sigma-Copace, Sabbah e Smith. As sementes das duas cultivares apresentaram umidades de equilíbrio higroscópico semelhantes. Os valores da variância explicada e do desvio-padrão, bem como a distribuição dos resíduos, das duas cultivares, indicam que as equações de Chung-Pfost, Sabbah e Smith foram as que melhor se ajustaram aos dados experimentais, com pequena superioridade da primeira.

Resumo: O objetivo deste trabalho foi quantificar o efeito do tipo de diesel e da concentração de biodiesel na mistura combustível sobre as emissões de gases por motor agrícola submetido a diferentes cargas. Foram testados dois tipos de diesel: S10, com baixo teor de enxofre; e S500, com alto teor de enxofre; bem como suas misturas com 5, 10, 15, 20, 50 e 100% de biodiesel. O motor, de quatro cilindros, turboalimentado e com 105 cv de potência máxima, foi submetido a oito modos de operação, sob diferentes rotações e torques. O ensaio seguiu a metodologia prevista na norma NBR ISSO 8174-4:2012, tendo-se analisado as emissões de O2, CO2, NO2, NO, CO e de hidrocarbonetos (HC). Somente as emissões de HC foram influenciadas pelo tipo de diesel utilizado. As emissões de NO2 e O2 aumentaram com o aumento da concentração de biodiesel na mistura, enquanto as emissões de CO2, NO, CO e HC diminuíram. A carga aplicada ao motor esteve diretamente relacionada às emissões, exceto para O2. O combustível S10, com 20% de biodiesel, reduz as emissões, em comparação ao diesel comercial (S500), sem perda no desempenho do motor.

Com o objetivo de introduzir, avaliar e selecionar variedades de uva sem sementes, adaptadas às condições tropicais semi-áridas e oferecer novas alternativas aos viticultores do Vale do São Francisco, implantou-se, em 1994, uma coleção com dezenove variedades de uvas sem sementes no Campo Experimental de Bebedouro, da Embrapa Semi-Árido, em Petrolina - PE. Foram avaliadas treze variedades ao longo dos anos de 1997 e 1998, correspondendo a cinco ciclos de produção. As variedades utilizadas foram Vênus, Arizul, Beauty Seedless, Thompson Seedless, Marroo Seedless, Canner, CG 39915, Pasiga, Saturn, Emperatriz, A1581, Paulistinha e Loose Perlette, enxertadas sobre o porta-enxerto IAC 572 ('Campinas'). Foram avaliados aspectos relacionados ao desenvolvimento vegetativo e produtivo das plantas e características e composição química dos frutos. Todas as variedades apresentaram cachos com tamanho pequeno. As variedades Vênus e Marroo Seedless destacaram-se em relação ao diâmetro de bagas, apresentando, respectivamente, 17,83 e 18,26 mm, sem a necessidade de aplicação de reguladores de crescimento. O teor de sólidos solúveis totais foi elevado na maioria das variedades, enquanto a acidez total titulável foi reduzida, resultando em relações SST/ATT satisfatórias. As variedades Vênus e Marroo Seedless foram as mais produtivas, com produtividades anuais de 24 t/ha e 20 t/ha, respectivamente.
