Insects of the Simuliidae family have been the object of control in Rio Grande do Sul since the 70s. Their constant attacks became a social-economical problem as well as a problem of Public Health, with serious consequences to men and to the economy of the areas in which the insects develop. At first, the control was done with a chemical larvicide Themephos ABATE 500 E, but an imperfect measuring of outflow to determine the quantity of the product made Simulium spp. resistant to it. From 1983 on, following a study of a new method for the outflow measuring, we started to use a biological larvicide Bacillus thuringiensis serovar israelensis based. The biological control uses the new method in 36.4% of the state area, assisting about 3,500,000 inhabitants.

A polymerase chain reaction (PCR)-based assay which amplifies repetitive DNA elements present within bacterial genomes was used to characterize and differentiate Leptospira sp. Thirty-five strains from a reference culture collection and 18 clinical isolates which had been previously analyzed by cross agglutinin absorption test (CAAT) were evaluated by this technique. PCR results from analysis of the reference culture collection showed no bands corresponding to serogroups Australis, Autumnalis, Bataviae, Celledoni, Cynopteri, Djasiman, Panama, Pomona, Pyrogenes, and Tarassovi. However, the PCR method was able to clearly discriminate the serogroups Andamana, Ballum, Canicola, Grippotyphosa, Hebdomadis, Icterohaemorrhagiae, Javanica, Sejroe, Semaranga, and Shermani. Clinical isolates previously characterized by CAAT as serovar Copenhageni, serovar Castellonis, and as serovar Canicola were in agreement with PCR results. The clinical isolate previously characterized as serovar Pomona was not differentiated by PCR. Forty additional clinical isolates from patients with leptospirosis obtained in São Paulo, Brazil were also evaluated by this PCR method. Thirty-nine of these were determined to belong to serogroup Icterohaemorrhagiae (97.5%) and one to serogroup Sejroe (2.5%). These results demonstrate that the PCR method described in this study has utility for rapid typing of Leptospira sp. at the serogroup level and can be used in epidemiological survey.

Leptospirosis is an emerging infectious disease that has been identified as both a human and animal health problem worldwide. Regular outbreaks associated with specific risk factors have been reported in Argentina. However, there are no available data concerning the genetic population level for this pathogen. Therefore, the aim of this work was to describe the genetic diversity of Leptospira interrogans through the application of two molecular typing strategies: variable number of tandem repeats (VNTR) and multilocus sequence typing (MLST). For this purpose, seven reference strains and 18 non-epidemiologically related isolates from diverse hosts and Argentinean regions were analysed. Among them, nine genotypes and seven sequence types (STs), including three unreported STs, were described using VNTR and MLST, respectively. eBURST analysis demonstrated that ST37 was the most frequent and founder genotype of a clonal complex (CCs) containing STN1 and STN3, suggesting the importance of studying the serovars belonging to this CC in Argentina. The data from maximum parsimony analysis, which combined both techniques, achieved intra-serovar discrimination, surmounted microscopic agglutination test discrepancies and increased the discriminatory power of each technique applied separately. This study is the first to combine both strategies for L. interrogans typing to generate a more comprehensive molecular genotyping of isolates from Argentina in a global context.

The objective of the study was to determine the association between clinical ophthalmic alterations and seroreactivity to leptospirosis by serovar Icterohaemorrhagiae in horses in Rio de Janeiro, Brazil. A total of 199 horses were studied. A microscopic agglutination test (MAT) was used to detect specific anti-Leptospira antibodies in blood serum. A total of 107 (53.8%) horses were seroreactive (titres > 200); 54 had high (> 800) titres, of which 44 were against serovar Icterohaemorrhagiae. Forty-two out of these 44, plus 40 seronegative horses (titers < 100) were given detailed ophthalmic examinations. Epiphora, ocular congestion, blepharospasm, photophobia, and peripapillary focal depigmentation were the most frequent alterations in seroreactive horses. Most ocular alterations were significantly more frequent in seroreactive horses. Horses seroreactive for leptospirosis (serovar Icterohaemorrhagiae) had a significantly higher prevalence of ophthalmic alterations than seronegative horses, providing additional evidence for an association between leptospirosis and equine uveitis.

This study aimed to describe the occurrence of Leptospira interrogans serovars Icterohaemorrhagiae and Canicola, in coastal zone and in southern grasslands of Rio Grande do Sul, Brazil. In each one of the four analyzed farms blood samples were collected from free-living wild animals, domestic animals and humans to perform serological testing for leptospirosis. The presence of antibodies was verified by microscopic agglutination test (MAT). The criterion adopted to consider a serum as agglutination reactant was at least 50% of leptospira for a microscopic field of 100x. From 17 blood samples collected at Chuí, five (29.41%) were positive, three (60.00%) for serovar Icterohaemorrhagiae and two (40.00%) for Canicola. From 21 samples collected in the County of Santana da Boa Vista, six (28.57%) were positive, four (66.67%) for serovar Canicola and two (33.33%) for serovar Icterohaemorrhagiae. From 32 samples collected at Alegrete, 10 (31.25%) were positive, seven (70.00%) for serovar Icterohaemorrhagiae and three (30.00%) foro serovar Canicola. From 17 blood samples collected in Cruz Alta, three (17.64%) were positive, two (66.67%) for serovar Icterohaemorrhagiae and one (33.33%) for Canicola. It is necessary to improve sanitary practices on farms in the state of Rio Grande do Sul, in order to achieve success in leptospirosis control programs.

The purpose of this study was to report the first recovery and characterization of Leptospira interrogans (serogroup Australis) from urine of swine in Brazil. The isolate was studied by serogrouping, MLVA, PGFE, and partial sequencing of rrs and secY. It was serogrouped as serogroup Australis, probably serovar Bratislava (titre 1,600), and sequenced as Leptospira interrogans. The MLVA and PGFE profiles also suggested the isolate as serovar Bratislava, since they were indistinguishable from reference strains Balico and Jez Bratislava. This is the first Leptospira interrogans serogroup Australis isolate, probably serovar Bratislava, obtained in Brazil.

Glycolipoprotein (GLP) from pathogenic serovars of Leptospira has been implicated in the pathogenesis of leptospirosis by its presence in tissues of experimental animals with leptospirosis, the inhibition of the Na,K-ATPase pump activity, and induced production of cytokines. The aims of the present study were to investigate the induction of IL-6 by GLP in peripheral blood mononuclear cells (PBMC) and to demonstrate monocyte stimulation at the cellular level in whole blood from healthy volunteers. PBMC were stimulated with increasing concentrations (5 to 2500 ng/ml) of GLP extracted from the pathogenic L. interrogans serovar Copenhageni, lipopolysaccharide (positive control) or medium (negative control), and supernatants were collected after 6, 20/24, and 48 h, and kept at -80ºC until use. Whole blood was diluted 1:1 in RPMI medium and cultivated for 6 h, with medium, GLP and lipopolysaccharide as described above. Monensin was added after the first hour of culture. Supernatant cytokine levels from PBMC were measured by ELISA and intracellular IL-6 was detected in monocytes in whole blood cultures by flow-cytometry. Monocytes were identified in whole blood on the basis of forward versus side scatter parameters and positive reactions with CD45 and CD14 antibodies. GLP ( > or = 50 ng/ml)-induced IL-6 levels in supernatants were detected after 6-h incubation, reaching a peak after 20/24 h. The percentage of monocytes staining for IL-6 increased with increasing GLP concentration. Thus, our findings show a GLP-induced cellular activation by demonstrating the ability of GLP to induce IL-6 and the occurrence of monocyte activation in whole blood at the cellular level.

O objetivo deste trabalho foi avaliar doses de lodo de curtume incorporadas ao solo sobre a nodulação e o controle de Meloidogyne spp. em soja. Foram empregadas doses crescentes (0; 5,0; 10,0; 20; 40,0; e 80 Mg ha-1) de lodo no solo. O solo de cada vaso foi infestado com 5.000 ovos de Meloidogyne spp após a semeadura da soja. As plantas foram conduzidas durante 70 dias em casa de vegetação e após esse período foram coletadas para avaliação da nodulação, massa de matéria seca da parte aérea e reprodução dos nematóides nas raízes. O aumento da dose de lodo de curtume aplicada ao solo reduziu de forma quadrática a reprodução do nematóide nas raízes de soja. Entretanto, o aumento da concentração de lodo no solo reduziu exponencialmente a nodulação da cultura e não proporcionou benefícios ao crescimento da planta.

A associação das helicobacterias com doença gástrica em humanos e em alguns animais domésticos e selvagens sugere a participação dessas na patogênese da gastrite em cães. Neste artigo procurou-se verificar a presença de Helicobacter spp. na mucosa gástrica de cães e avaliar sua associação com os achados macro e microscópicos, considerando a idade. Coletaram-se amostras das regiões cárdica, fúndica, do corpo e pilórica dos estômagos de 60 cães para a realização de exame histopatológico, utilizando-se as colorações pela hematoxilina-eosina (HE) e carbolfucsina (CF). Tais exames revelaram Helicobacter spp. em 96,7% dos animais, sendo observados infiltrados inflamatórios, predominantemente mononucleares (100%), hiperplasia de nódulos linfoides (98,3%), erosões/ulcerações (6,7%), hemorragia (5%) e hiperemia (95%) em amostras coradas por HE. Não houve correlação da infecção por helicobacter spp. com a idade do animal e da idade com alterações inflamatórias na presença da bactéria. As amostras das regiões do corpo e pilórica apresentaram maior presença de bactérias a histopatologia (CF) (ambos 95%), em seguida vindo as regiões fúndica (91,7%) e cárdica (56,7%). Houve correlação do número de Helicobacter spp. com o de células inflamatórias e nódulos linfoides nessas regiões, sugerindo que as bactérias encontradas na mucosa gástrica dos cães poderiam ser responsáveis pelas alterações que caracterizam a gastrite.

As orquídeas no ambiente natural sofrem exploração devido a sua importância ornamental, levando algumas espécies à extinção. O cultivo in vitro é uma forma alternativa para a conservação ex-situ. Procurou-se determinar um meio de cultura eficiente para a germinação in vitro de sementes e o crescimento inicial de plântulas de Cattleya forbesii, bem como para o crescimento de plântulas in vitro de Cattleya harrisoniana. No primeiro caso, sementes foram inoculadas em meio de cultura básico de Murashige & Skoog (MS) = T1 e MS básico acrescido de 2,5 g L-1 de carvão ativado (CA) = T2. No segundo, plântulas com 1 ± 0,2 cm de altura foram submetidas aos tratamentos T1, T2, MS com a metade da concentração original de macro-micronutrientes (T3) e MS com a metade da concentração original de macro-micronutrientes suplementado com 1,25 g L-1 de CA (T4). Verificou-se aos 30 dias em C. forbesii uma porcentagem de germinação de 45% em T1 e 90% em T2. A adição de CA ao meio de cultura trouxe aumento na altura de plântulas de C. forbesii de acordo com análises realizadas aos 180 dias de cultivo. Em relação ao crescimento de C. harrisoniana, aos 240 dias observou-se que todos os parâmetros médios avaliados (altura da parte aérea, massa de matéria fresca total, número de raízes e folhas, comprimento da maior raiz e diâmetro do pseudocaule) foram significativamente maiores em T2. Dessa forma, sugere-se o uso do meio MS acrescido de 2,5 g L-1 de CA (T2), uma vez que é significativamente favorável tanto para a germinação de sementes quanto para o crescimento de ambas as espécies.

Apesar de oferecer uma série de vantagens, quanto à qualidade do produto final, a técnica de cultivo in vitro de plantas ainda é considerada dispendiosa, por causa, dentre outros fatores, da utilização de reagentes com alto grau de pureza no preparo de meios nutritivos. Entre as alternativas que podem ser adotadas para a redução dos custos relacionados com a produção de mudas, apresenta potencial a substituição de produtos PA por aqueles de menor custo. Baseado nestas informações, o objetivo deste estudo foi avaliar o efeito do melado de cana-de-açúcar sobre o desenvolvimento in vitro de bananeira, cv. Maçã. Para tal propósito, plantas de bananeira foram inoculadas em meios nutritivos, formulados à base de melado de cana-de-açúcar, com quatro concentrações distintas (brix de 1,5; 3,0; 4,5 e 6,0), e os dados obtidos foram comparados com aqueles das plantas cultivadas em meio MS (controle), perfazendo um total de cinco tratamentos. Apesar de terem-se desenvolvido em todos os tratamentos testados, observou-se que, para as variáveis número médio de folhas e biomassa da matéria fresca, as plantas cultivadas no meio MS (controle) apresentaram valores superiores. Entretanto, para o número médio de raízes, não houve diferença estatística entre os tratamentos utilizados. O melado de cana-de-açúcar não favorece o desenvolvimento de plântulas de bananeira, mas pode ser utilizado para o enraizamento dessas plantas in vitro.

Objetivando avaliar a densidade populacional de Pratylenchus brachyurus e Pratylenchus zeae, associados à Brachiaria brizantha, B. decumbens e B. humidicola, e sua influência na disponibilidade e na qualidade da forragem, realizou-se este trabalho. O experimento foi conduzido na Agropecuária Hisaeda, em Terenos, MS. Coletaram-se amostras de solo, raízes e parte aérea, em dez repetições ao acaso, de 1 m² cada, dos seguintes tratamentos: Bom, Intermediário e Ruim, caracterizados visualmente, considerando-se a percentagem de material verde das plantas. As densidades de P. brachyurus e P. zeae foram avaliadas nas amostras de solo e raízes. Na parte aérea, avaliou-se a disponibilidade de matéria seca dos materiais verde, morto e da rebrota, estado nutricional das plantas e qualidade da forragem. A fertilidade do solo foi determinada em todas as unidades amostrais. Ambos os nematoides foram identificados em todas as amostras, com maior número nas raízes (entre 87-311 P. brachyurus e 1-61 P. zeae.10 g-1) que no solo (de 0-8 P. brachyurus a 1-39 P. zeae.200 cm-3), no entanto, não houve diferenças significativas, na quantidade de espécimes, entre os tratamentos. Considerando-se que essas forrageiras são perenes e hospedam Pratylenchus spp., há tendência ao aumento da população desses patógenos, ao longo do tempo, podendo se tornar um sério problema fitossanitário.

Morpho-physiological characteristics and chemical composition are directly related to superior competitive ability of crops. This study intended to make a comparative analysis of dry matter production, leaf area and amount of epicuticular wax of three species of Sida spp: S. urens L., S. rhombifolia L. and S. spinosa L. Plants were collected at three growth stages: V1: stage described as up to 10 fully expanded leaves; V2: between 11 leaves and flowering; and R: after flowering. At stages V2 and R, the highest number of leaves was recorded for S. rhombifolia, followed by S. spinosa at V2 and S. urens at R. These results were relatively proportional to leaf area for all species. S. spinosa at the vegetative stage produced the highest values of specific leaf area (SLA), with no significant differences between species at the stage R. The amount of wax per unit of leaf area between species at the same developmental stage was significantly different only at the reproductive stage, where S. spinosa produced 23.18 and 6.23 fold more wax than S. urens and S. rhombifolia respectively. Between the growth stages of each species, there was decrease in the amount of wax with plant age and increase in leaf area (AFE), number of leaves and dry matter. The leaves of the Sida species exhibit different characteristics and this information can be used to optimize the use of herbicides in the control of these weeds.

Studies have been un dertaken into on the diversity and relative abundance of larvae of Anopheles (Nyssorhynchus) spp. in 22 permanent or temporary pools in an area of 70 km² in the eastern piedmont of the Venezuela Andes, between the mountains and the plains, an area in which malaria is refractory and A. nuñeztovari is present. Twelve species were identified, the most frequent, abundant and sympatric being A. triannulatus, A. albitarsis, A. nuñeztovari, A. oswaldoi and A. strodei. The samples from the permanent pools showed greater diversity of species and greater numbers of larvae than the samples from the temporary pools. The existence of the same larval associations in pools of other localities in the eastern piedmont of the Venezuelan Andes suggests the possibility of the making an ecological map of the breeding sites of A. nuñeztovari and for these anophelines in a region extending for 430 km.
