The consumption of bottled mineral water has significantly increased in Brazil so that it is in the interest of public health to determine the parasitological and microbiological status of some brands of Brazilian mineral water available in the town of Campinas, São Paulo, Brazil. For this purpose, detection of protozoa by direct immunofluorescence technique and microbiological parameters were determined for each specimen after membrane filtration. Giardia cysts were not present while cryptosporidial oocysts were detected in two samples. The counts of protozoa varied from 0.2 to 0.5 oocysts/l. The detected level of Pseudomonas aeruginosa and heterotrophic bacteria reflected the level of organic enrichment of the water.

The purpose of this study was to investigate the genotypes of Giardia lamblia from human and animal feces and their epidemiological and clinical characteristics in Argentina, South America. Seventy isolates, 60 from humans (adults and children), eight from dogs and two from cows were processed by polymerase chain reaction-restriction fragment length polymorphism. Data corresponding to demographic, socio-cultural and environmental variables and presence/absence of signs/symptoms were collected. The triosephosphate isomerase gene was amplified from 43 (71.66%) of the 60 human fecal samples. Among these, 3/43 (6.98%) were genotype AII and 40/43 (93.02%) were genotype B. Assemblage AII was detected in three children who lived together in a shantytown and they were oligosymptomatic and none had diarrhea. This genotype was not found in animals. Genotype B showed a high prevalence in both adults and children. It was also found in polysymptomatic people, many of whom presented diarrhea. It was also found only in one dog. The present study represents the first contribution to the knowledge of G. lamblia genotypes in Argentina.

The reproductive mechanism of Giardia intestinalis, considered one of the earliest divergent eukaryotes, has not been fully defined yet. Some evidence supports the hypothesis that Giardia is an exclusively asexual organism with a clonal population structure. However, the high genetic variability, the variation in ploidy during its life cycle, the low heterozygosity and the existence of genes involved in the meiotic-like recombination pathway in the parasite's genome cast doubt on exclusively asexual nature of Giardia. In this work, semiquantitative RT-PCR analysis was used to assess the transcription pattern of three meiosis-like-specific genes involved in homologues recombination: dmc1, hop1 and spo11. The mRNAs were amplified during the parasite's differentiation processes, encystation and excystation, and expression was found at each stage of its life cycle. A semiquantitative assessment also suggests that expression of some of the genes is regulated during encystation process.

Stool is chemically complex and the extraction of DNA from stool samples is extremely difficult. Haemoglobin breakdown products, such as bilirubin, bile acids and mineral ions, that are present in the stool samples, can inhibit DNA amplification and cause molecular assays to produce false-negative results. Therefore, stool storage conditions are highly important for the diagnosis of intestinal parasites and other microorganisms through molecular approaches. In the current study, stool samples that were positive for Giardia intestinalis were collected from five different patients. Each sample was stored using one out of six different storage conditions [room temperature (RT), +4ºC, -20ºC, 70% alcohol, 10% formaldehyde or 2.5% potassium dichromate] for DNA extraction procedures at one, two, three and four weeks. A modified QIAamp Stool Mini Kit procedure was used to isolate the DNA from stored samples. After DNA isolation, polymerase chain reaction (PCR) amplification was performed using primers that target the β-giardin gene. A G. intestinalis-specific 384 bp band was obtained from all of the cyst-containing stool samples that were stored at RT, +4ºC and -20ºC and in 70% alcohol and 2.5% potassium dichromate; however, this band was not produced by samples that had been stored in 10% formaldehyde. Moreover, for the stool samples containing trophozoites, the same G. intestinalis-specific band was only obtained from the samples that were stored in 2.5% potassium dichromate for up to one month. As a result, it appears evident that the most suitable storage condition for stool samples to permit the isolation of G. intestinalis DNA is in 2.5% potassium dichromate; under these conditions, stool samples may be stored for one month.

The humoral immune response plays an important role in the clearance of Giardia lamblia. However, our knowledge about the specific antigens of G. lamblia that induce a protective immune response is limited. The purpose of this study was to identify and characterise the immunogenic proteins of G. lamblia in a mouse model. We generated monoclonal antibodies (moAbs) specific to G. lamblia (1B10, 2C9.D11, 3C10.E5, 3D10, 5G8.B5, 5F4, 4C7, 3C5 and 3C6) by fusing splenocytes derived from infected mice. Most of these moAbs recognised a band of ± 71 kDa (5G8 protein) and this protein was also recognised by serum from the infected mice. We found that the moAbs recognised conformational epitopes of the 5G8 protein and that this antigen is expressed on the cell surface and inside trophozoites. Additionally, antibodies specific to the 5G8 protein induced strong agglutination (> 70-90%) of trophozoites. We have thus identified a highly immunogenic antigen of G. lamblia that is recognised by the immune system of infected mice. In summary, this study describes the identification and partial characterisation of an immunogenic protein of G. lamblia. Additionally, we generated a panel of moAbs specific for this protein that will be useful for the biochemical and immunological characterisation of this immunologically interesting Giardia molecule.

Giardia lamblia trophozoites were incubated for 2 h with activated murine macrophages, nitric oxide (NO) donors or a superoxide anion generator (20 mU/ml xanthine oxidase plus 1 mM xanthine). Activated macrophages were cytotoxic to Giardia trophozoites (~60% dead trophozoites). This effect was inhibited (>90%) by an NO synthase inhibitor (200 µM) and unaffected by superoxide dismutase (SOD, 300 U/ml). Giardia trophozoites were killed by the NO donors, S-nitroso-acetyl-penicillamine (SNAP) and sodium nitroprusside (SNP) in a dose-dependent manner (LD50 300 and 50 µM, respectively). A dual NO-superoxide anion donor, 3-morpholino-sydnonimine hydrochloride (SIN-1), did not have a killing effect in concentrations up to 1 mM. However, when SOD (300 U/ml) was added simultaneously with SIN-1 to Giardia, a significant trophozoite-killing effect was observed (~35% dead trophozoites at 1 mM). The mixture of SNAP or SNP with superoxide anion, which yields peroxynitrite, abolished the trophozoite killing induced by NO donors. Authentic peroxynitrite only killed trophozoites at very high concentrations (3 mM). These results indicate that NO accounts for Giardia trophozoite killing and this effect is not mediated by peroxynitrite

Estudo da prevalência de parasitas intestinais realizado em 370 indivíduos residentes na CECAP, distrito-sede de Botucatu, Estado de São Paulo, permitiu verificar que 41,62% encontravam-se infestados por uma ou mais espécies de parasitas intestinais. Foram encontrados os seguintes parasitas: Entamoeba histolytica 0,54%, E. coli 6,21%, I. butschlii 0,27%, Giardia lamblia 9,72%, Ancylostomidae 5,94%, Strongyloides stercoralis 6,75%, Trichuris trichiura 17,29%, Ascaris lumbricoides 7,56%, Enterobius vermicularis 3,78%, Hymenolepis nana 5,40% e Taenia sp. 1,62%. Apresentam-se dados sobre a distribuição dos parasitas em relação à idade e ao sexo dos indivíduos. O atributo cor não permite maiores considerações por serem os não brancos significantemente pouco numerosos. Dos indivíduos examinados, 25,67% apresentavam apenas uma espécie de parasita e entre as associações parasitárias mais freqüentes encontramos as de Ascaris lumbricoides - Trichuris trichiura e Trichuris trichiura - Giardia lamblia.

During the second half of 1986 the impact of the improvement of water supply and excreta disposal facilities on diarrheal diseases and intestinal parasitosis was studied in 254 children up to six years of age from two favelas (shanty towns) of Belo Horizonte, Brazil. The estimated incidence of diarrhea was 6.2 episodes/child year and the estimated period prevalence reached 31.0 episode days/ child/ year. The point prevalence of parasitosis was 70.7% (Ascaris lumbricoides: 55.4%, Trichuris trichiura: 19.6%, Giardia lamblia: 17.9%). The estimated prevalence of diarrhea decreased with improvement of water supply and sanitation facilities to 45% and 44% respectively, but no statistically significant impact was observed in the case of parasitosis. School education and weaning practice were found to be other important determinants of diarrhea.

Foram analisadas hortaliças "in natura", comercializadas na Região Metropolitana de São Paulo, SP-Brasil, visando à pesquisa e à identificação de cistos de protozoários de interesse médico. As hortaliças, constituídas de 50 amostras de cada variedade, consistiram em: alface (Lactuca sativa), variedades lisa e crespa, escarola (Chichorium sp) e agrião (Nasturtium officinale). Os resultados evidenciaram elevados percentuais de contaminação em todas as variedades de hortaliças analisadas, porém as freqüências de protozoários foram maiores no agrião. As amostras de alfaces apresentaram os menores percentuais de contaminação, enquanto que a escarola apresentou valores geralmente situados entre o agrião e as alfaces. Observou-se uma grande variedade de protozoários, cujas freqüências de ocorrência na população residente na Região Metropolitana de São Paulo são igualmente elevadas. Os mais freqüentes foram Entamoeba sp (com 4 e 8 núcleos) e Giardia sp. Foram também isolados cistos de Iodamoeba sp, Endolimax sp e Chilomastix sp. Os elevados níveis de contaminação fecal encontrados nas amostras analisadas apontam para a importância dos alimentos na transmissão de protozoários intestinais.

OBJETIVO: Evaluar la presencia de parásitos en efluentes semisólidos y líquidos en distintas localidades de la Patagonia argentina considerando que ésta es una de las restricciones para su uso. MÉTODOS: Las muestras tomadas en 4 Plantas de Tratamiento de Efluentes Domiciliarios se analizaron siguiendo las normativas de Environmental Protection Agency, Organización Mundial de la Salud, Standard Methods for Examination of Water and Wastewater y de algunas clasificaciones. RESULTADOS: Solamente 2 de las 6 muestras de semisólidos analizadas tenían huevos de Ascaris lumbricoides no viables. De las 10 muestras líquidas, solamente 2 no tenían huevos mientras las restantes tenían patógenos de las categorías I (Giardia sp., Hymenolepis diminuta y Enterobius vermicularis) y III (Ascaris lumbricoides, Ancylostoma duodenale y Trichuris trichiura). CONCLUSIONES: Todas las muestras de semisólidos analizadas fueron aptas para su uso como fertilizantes porque no se registró en ninguna de ellas la presencia de huevos viables de Ascaris lumbricoides y solamente 6 de las muestras líquidas fueron aptas para riego por carecer de huevos o por ser su concentración igual o inferior a 1 huevo por litro.

O objetivo do estudo foi estimar as freqüências de tuberculose e parasitoses intestinais na em comunidades indígenas da localidade de Iauareté (AM), em 2001. Estudo transversal (n=333) visando à obtenção de dados demográficos e amostras biológicas para exames de escarro e fezes. Dentre os 43 sintomáticos respiratórios, seis foram positivos na pesquisa de bacilos álcool-ácido resistentes no escarro. As parasitoses intestinais apresentaram freqüência significativamente maior entre a população Hüpda do que entre os índios que habitam os demais bairros (37,5% vs. 19,3% para Ascaris lumbricoides, 32,4% vs. 16,3% para Trichuris trichiura, 75% vs. 19,3% para ancilostomídeos, 75% vs. 35,4% para Entamoeba histolyticaD dispar e 33,3% vs. 10,7% para Giardia lamblia). Conclui-se que a tuberculose e o parasitismo intestinal são freqüentes nessas comunidades, exigindo medidas de controle e melhorias na assistência à saúde.

Este trabalho tem por objetivo caracterizar a desnutrição protéico-energética associada a parasitose intestinal em grupo de 149 crianças de ambos os sexos, na faixa etária de 3 a 72 meses, da cidade de Mirassol D'Oeste, na região do Projeto Polonoroeste em Mato Grosso. De cada criança foram coletados os seguintes dados: sexo, peso, idade e amostra de fezes para exame parasitológico. Os dados peso/idade obtidos foram analisados pelos critérios de GOMEZ. Utilizou-se como padrão de referência o National Center for Health Statistic (NCHS). Para diagnóstico dos parasitas intestinais executou-se o método de Hoffman, Pons e Janer. O grupo estudado constitui-se em sua maioria de crianças desnutridas, sendo a forma leve de desnutrição mais comum que as formas moderada e grave. As enteroparasitoses foram encontradas em 69% das amostras examinadas. A "Giardia lamblia" foi o protozoário mais comum e o "Ancilostomídeo" o helminto mais encontrado. O teste X² não mostrou relação de dependência entre o estado nutricional e a freqüência de enteroparasitoses.

Parasitological examinations were carried out during April to August, 1987, with 187 out-patients of the IMIP hospital, located in the center of Recife City, and 464 inhabitants of several villages around Cabo City, 50 Km southeast of Recife, Pernambuco, Brazil. Approximately 71% of the IMIP patients and 92% of the Cabo inhabitants were infected with at least one species of intestinal parasite. There was minimum difference in the prevalence rate of Trichuris trichiura between two areas, whereas the prevalence rates of Ascaris lumbricoides, hookworms, Strongyloides stercoralis, Schistosoma mansoni and Entamoeba histolytica were higher in the inhabitants of the Cabo City area. Only Giardia lamblia was more prevalent in the out-patients of IMIP hospital. Test tube cultivation revealed that the prevalence rate of Necator americanus in both areas was much higher than that of Ancylostoma duodenale , and also that the prevalence rate of S. stercoralis of the IMIP patients and Cabo inhabitants were 4.5% and 9.6%, respectively. Six hundred and fifteen sera were serologically examined for amoebiasis by the gel diffusion precipitation test (GDP) and enzyme linked immunosorbent assay (ELISA) using the antigen prepared from axenically cultured trophozoite of E. histolytica (strain HM-ITMSS). No positive reaction was observed in all of the sera as examined by GDP, while 32 out of 615 sera were positive on ELISA.

Durante os anos de 1982 e 1983, foram realizados exames parasitológicos de fezes em 3.076 crianças de 0-6 anos, matriculadas em 52 creches mantidas pela Prefeitura do Município de Sáo Paulo e em 50 manipuladores de alimentos (funcionários da cozinha), relativos a 18 creches. Estudou-se a prevalência de infecção por Giardia lamblia conforme sexo e faixa etária do hospedeiro e sazonalidade da infecção. A giardíase foi mais freqüente nas faixas etárias compreendidas entre 1-4 anos. A prevalência média foi 32,8% entre as crianças e 4,0% entre os manipuladores de alimentos. O estudo da sazonalidade de infecção revelou ser a giardíase, em 1982, mais prevalente no Outono e Primavera; em 1983, as maiores prevalências ocorreram no Outono e Verão. Estudos estatísticos revelaram não haver associação entre sexo e prevalência de infecção, não ocorrendo também nenhuma diferença significativa entre as quatro regiões metropolitanas estudadas (p > 0,05). O modo mais provável de aquisição de G. lamblia nas creches estudadas foi através do contato inter-humano, uma vez que tanto os fatores ambientais, como outros, incriminados na veiculação da giardíase através da água em outros países, diferem dos encontrados em nosso Município. Os manipuladores de alimentos tiveram papel pouco expressivo como fonte de infecção, pois a prevalência de infecção nesses indivíduos foi de apenas 4,0%.
