O efeito de diferentes concentrações de ANA e BAP na micropropagação do abacaxizeiro, bem como no cultivo em hidroponia das plântulas obtidas in vitro, foram estudados em brotos de abacaxizeiro da variedade Pérola, inoculados em meio de cultura básico MS, suplementado com os fitorreguladores BAP e ANA em diferentes concentrações. Caracteres morfológicos quantitativos relacionados ao crescimento dos brotos e das plântulas de abacaxizeiro foram avaliados respectivamente durante os cultivos in vitro e em hidroponia e mostraram que, o tratamento T1 (BAP = 1,0 mg L-1e ANA = 0,5 mg L-1) proporcionou a maior taxa média de regeneração de brotos e conseqüentemente uma maior produção de matéria fresca. Entretanto, a altura dos brotos e a formação de suas raízes foram maiores nos tratamentos T2 (BAP = 0,5 mg L-1 e ANA = 0,25 mg L-1 ) e T3 (BAP = 0,25 mg L-1 e ANA = 0,12 mg L-1 ). Após sessenta dias de cultivo em hidroponia, todas as plântulas oriundas do tratamento T1 apresentaram um bom desenvolvimento, expresso pela maioria dos caracteres morfológicos avaliados. O sistema de micropropagação utilizado neste trabalho possibilitou a obtenção de brotos de abacaxizeiro Pérola, em quantidade suficiente e ao mesmo tempo de fácil individualização, seguida da regeneração de plântulas que foram cultivadas em hidroponia.

Mauritia vinifera (buriti) is a palm tree that grows wild in different areas of Brazil, particularly in the Amazonian region. The buriti oil is rich in carotenoids, especially in β-carotene. The growing interest in other natural sources of β-carotene has stimulated the industrial use of buriti as a raw material for pulp oil extraction. Most processes are based on the conventional technologies, involving drying and pressing the pulp for oil recovery and further separation of carotenoids in a liquid phase using organics solvents. In the present work, the ethanol-based process was evaluated for simultaneous carotenoids recovering and fractionating from buriti pulp. The raw material and ethanol, 1:4 ratio, were placed in an erlenmeyer flask and maintained at 30rpm for 1 hour in a temperature-controlled bath at 65ºC. The mixture was filtered under vacuum and cooling at 10ºC to allow for the separation of the solvent in two phases. Carotenoids composition, determined by HPLC, has indicated a β-carotene concentration about 12 times greater in the lower phase than in the upper phase. The profile of the carotenoids in the denser phase is quite similar to that of raw buriti oil, and the concentration of total carotenoids is 40% higher than that of the original raw oil, making the ethanol-based process particularly attractive for industrial applications.

The sublethal effect of extracts of Azadirachta indica on Ceratitis capitata was evaluated. Two pairs of flies were treated in plastic tubes with cotton placed in plastic cages. An artificial diet (hydrolyzed protein + sugar) was provided ad libitum. The extracts affected significantly the longevity of C. capitata. The pre-oviposition period were not significantly affected by the extracts. The A. indica branches extracted with dichloromethane (888 ppm) affected significantly the fecundity and fertility, reducing the number of eggs laid to approximately 80 % and the egg hatching by 30 % at the 8th day. Therefore, the neem branches extracted with dichloromethane affected the reproduction of C. capitata.

O objetivo deste trabalho foi determinar o comportamento reológico do suco de abacaxi- pérola natural e tratado com enzimas pectinolíticas. As condições de tratamento enzimático foram otimizadas através de um planejamento experimental do tipo fatorial completo 2k, com três repetições do ponto central. Na avaliação do comportamento reológico foram utilizadas duas amostras submetidas a peneiramento (N e D), analisadas em quatro diferentes temperaturas (10; 25; 50 e 65 °C). As análises reológicas foram realizadas utilizando um viscosímetro de cilindros concêntricos Brookfield e os dados experimentais foram ajustados ao modelo de Mizrahi-Berk. A relação entre temperatura e viscosidade aparente foi descrita por uma equação tipo Arrhenius. A otimização da atividade enzimática indicou, através da análise de variância e da metodologia de superfície de resposta, que as variáveis temperatura e tempo de tratamento exerceram efeito estatisticamente significativo (p<0,05) sobre a concentração de pectina presente na amostra. O modelo utilizado mostrou-se adequado para descrever o comportamento reológico dos sucos de acordo com os parâmetros R², χ² e Bf. Os baixos valores obtidos para o índice de comportamento indicaram um comportamento pseudoplástico (n<1). A temperatura exerceu influência sobre a tensão de cisalhamento e a viscosidade aparente dos sucos analisados, sendo os menores valores observados nas amostras analisadas a 65 °C. A equação tipo Arrhenius descreveu de modo satisfatório o efeito da temperatura sobre a viscosidade aparente. Os valores da energia de ativação (Eat) foram de 4,54 Kcal.g.mol-1 e 4,89 Kcal.g.mol-1, respectivamente, para as amostras do suco de abacaxi natural e despectinizado, aumentando com o tratamento enzimático. A atividade enzimática proporcionou uma redução nos valores dos parâmetros de comportamento reológico das amostras, bem como na viscosidade aparente, em todas as temperaturas utilizadas, sendo o maior percentual de redução observado a 65 °C (41,66%).

The aim of this study was to evaluate the effect of sucrose concentration in the culture medium on growth and on the establishment of mycorrhizas during the acclimatization of pineapple cv. Pérola. The plantlets were micropropagated in MS culture medium with 0, 10, 20 and 30 g L-1 of sucrose and then they were acclimatized during 12 weeks under greenhouse conditions, in a sandy soil - compost mixture, uninoculated or inoculated with a Rhizophagus clarus isolate. Plantlets from the culture medium with 20 g and 30 g of sucrose L-1 showed higher shoot and root biomass than those from sugar-free medium. Mycorrhizal colonization was lower in plantlets micropropagated in sucrose-free medium, but the intensity of arbuscules did not differ among treatments. In the 12-week period of acclimatization, mycorrhizal colonization had no effect on plant biomass.

Crude extract and fractions of Buddleja thyrsoides were investigated regarding antioxidant activities by DPPH, total phenolic contents by Folin-Ciocalteau and antimicrobial activity by the broth microdilution method. Total phenolics varied from 214.07 ± 3.6 to 438.4 ± 0.3 mg g-1. Crude extract, ethyl acetate, dichloromethane and butanolic fractions exhibited a weak scavenging activity (SC50=186.04 ± 10.8, 137.70 ± 8.5, 146.89 ± 9.0 and 165.71 ± 3.2 µg mL-1, respectively). A correlation between the antioxidant activities and total phenolic contents could be shown (r=0.857, p<0.01). The lowest value of MIC was observed with butanolic fraction against Saccharomyces cerevisiae (MIC and MFC at 62.5 µg mL-1). Dichloromethane and ethyl acetate fractions were effective against Staphylococcus aureus with MIC value at 250 and 500 µg mL-1 respectively.

A chromatographic technique for determination of rutin and narcissin in marigold extract and topical formulations was developed and validated. The method shows linearity over the concentration range of 0.2 - 6.0 μg/mL of rutin (r = 0.9986) and 0.8 - 12.0 μg/mL of narcissin (r = 0.9951). The values obtained for precision and accuracy are in agreement with ICH guidelines. Both the formulation excipients and the porcine ear skin samples did not interfere with the flavonoids determination. The recovery of rutin and narcissin in skin samples added with marigold extract was 81.41% and 83.35%, respectively, which demonstrate the applicability of this method to perform skin penetration studies.

The species Eremanthus mattogrossensis, known as "veludo do cerrado" (cerrado velvet), is native to the Brazilian Cerrado. Because the amount of metabolites present in plants may be influenced by biological and environmental factors, here we conducted an HPLC-DAD-MS/MS investigation of the metabolite concentrations found in the MeOH/H2O extract of the leaves of this species. The main compounds were identified and quantified, and the metabolites were grouped by chemical class (caffeoylquinic acids, flavonoids, and sesquiterpene lactone). Statistical analysis indicated a straight correlation between the quantity of metabolites and seasonality, suggesting that environmental properties elicit important metabolic responses.

A statistical mixture-design technique was used to study the effects of different solvents and their mixtures on the yield, total polyphenol content, and antioxidant capacity of the crude extracts from the bark of Schinus terebinthifolius Raddi (Anacardiaceae). The experimental results and their response-surface models showed that ternary mixtures with equal portions of all the three solvents (water, ethanol and acetone) were better than the binary mixtures in generating crude extracts with the highest yield (22.04 ± 0.48%), total polyphenol content (29.39 ± 0.39%), and antioxidant capacity (6.38 ± 0.21). An analytical method was developed and validated for the determination of total polyphenols in the extracts. Optimal conditions for the various parameters in this analytical method, namely, the time for the chromophoric reaction to stabilize, wavelength of the absorption maxima to be monitored, the reference standard and the concentration of sodium carbonate were determined to be 5 min, 780 nm, pyrogallol, and 14.06% w v-1, respectively. UV-Vis spectrophotometric monitoring of the reaction under these conditions proved the method to be linear, specific, precise, accurate, reproducible, robust, and easy to perform.

This article describes the isolation and identification of flavonoids in the hydroethanolic extract of the aerial parts from Tonina fluviatilis and evaluation of their antiradical activity. A method based on HPLC-DAD was developed and validated for detecting and quantifying flavonoids in hydroethanolic extracts. The flavonoids identified and quantified in the extract were 6,7-dimethoxyquercetin-3-O-β-D-glucopyranoside (1), 6-hydroxy-7-methoxyquercetin-3-O-β-D-glucopyranoside (2), and 6-methoxyquercetin-3-O-β-D-glucopyranoside (3). The developed method presented good validation parameters, showing that the results obtained are consistent and can be used in ensuring the quantification of these constituents in the extracts. Compounds 2 and 3 showed strong antiradical activity when compared with the positive controls (quercetin and gallic acid).

An enzymatic spectrophotometric method for the determination of methyldopa in a dissolution test of tablets was developed using peroxidase from radish (Raphanus sativus). The enzyme was extracted from radish roots using a phosphate buffer of pH 6.5 and partially purified through centrifugation. The supernatant was used as a source of peroxidase. The methyldopachrome resulting from the oxidation of methyldopa catalyzed by peroxidase was monitored at 480 nm. The enzymatic activity was stable for a period of at least 25 days when the extract was stored at 4 or -20 ºC. The method was validated according to RDC 899 and ICH guidelines. The calibration graph was linear in the range 200-800 µg mL-1, with a correlation coefficient of 0.9992. The limits of detection and quantification in the dissolution medium were 36 and 120 µg mL-1, respectively. Recovery was greater than 98.9%. This method can be applied for the determination of methyldopa in dissolution tests of tablets without interference from the excipients.

Mudas das cultivares de abacaxi (Ananas comosus) 'Pérola' (suscetível) e 'Primavera' (resistente) foram inoculadas em casa de vegetação com isolados de Fusarium subglutinans f. sp ananas, resistentes e sensíveis ao benomyl, de diferentes áreas produtoras de abacaxi do Espírito Santo, visando estudar a virulência do patógeno. Foi utilizada suspensão de conídios obtidos pela raspagem da superfície das colônias desenvolvidas em meio BDA, cuja concentração final foi 10(5) conídios/ml. A inoculação de F. subglutinans f. sp ananas foi efetuada pela imersão de mudas injuriadas mecanicamente na base, com três meses de idade, na suspensão de inóculo. Dos 22 isolados testados preliminarmente, selecionou-se oito de F. subglutinans f. sp ananas, sendo quatro representativos do grupo dos sensíveis ao benomyl (E285, E277, E278 e E290) e quatro representativos do grupo dos resistentes ao benomyl (E272, E274, E279 e E283) para realização do teste final. Os resultados comprovaram a resistência da cv. Primavera a todos os isolados testados. Plantas da cv. Pérola apresentaram sintomas da doença aos 15 dias após a inoculação com isolados resistentes ao benomyl; os isolados sensíveis ao benomyl só foram capazes de causar sintomas severos da doença aos 45 e 60 dias após a inoculação. O isolado E272, resistente ao benomyl, foi o mais virulento, tendo causado a maior lesão e a morte das plantas aos 30 dias após a inoculação; os isolados E277 e E278 foram os mais virulentos. Houve diferenças em virulência de isolados de F. subglutinans f. sp. ananas resistentes e sensíveis ao benomyl ao abacaxizeiro.

This study aimed to evaluate the effect of Moringa oleifera Lam extract on the removal of total solids (TS), total suspended solids (TSS) and chemical oxygen demand (COD), in different filter media for treating wastewater of dairy cattle breeding (DCW). The moringa seed extract was obtained by grinding 50 g of seeds in one liter of distilled water and, after passing the solution through a quantitative paper filter of 25 microns, 60 mL of the extract were added to wastewater from cattle breeding before the filtration process in organic filters made of thin coal, bamboo leaves, eucalyptus leaves, gliricidia branches and sawdust. This was followed by the completely randomized experimental design, adopting a factorial of 5 x 2. Aliquots of filtered effluent were collected and the total solids (TS) concentrations, total suspended solids (TSS) and chemical oxygen demand (COD) were determined. It was found that the increase in the efficiency of removal of COD and total solids can be attributed to the coagulating power of the moringa seed extract, wherein the filter medium with bamboo leaves presented the best performance, showing potential for use as alternative filter material in the primary treatment of DCW.

Objective: to evaluate the effect of the Euphorbia tirucalli hydroalcoholic extract (ETHE) on the development of Ehrlich Tumor, in its ascitic form. Methods: we intraperitoneally inoculated 15 Swiss mice with 10.44 x 107 cells of Ehrlich Tumor and divided them in two groups one day after: ETHE Group (eight mice), treated with a dosage of 125 mg/kg/day of EHTE for five days; and Control Group (seven mice), treated only with 0.9% isotonic saline solution over the same period. The treatment was done by gavage. Ten days after inoculation, four mice from each group were sacrificed for quantification of tumor cell number, ascitic fluid volume and bone marrow cell number. The remaining animals were maintained to evaluate survival. Results: The ascitic fluid volume and the tumor cell number were decreased in the ETHE group when compared with the control group, but with no statistical significance. On the other hand, survival was higher in the ETHE group, as well as the number of bone marrow cells. Conclusion: Treatment with ETHE after inoculation of Ehrlich Tumor decreases its development and increases survival and the bone marrow cellularity, thus reducing the myelosuppression present in the Ehrlich Tumor bearing mice.
