The prevalence of Helicobacter pylori infection was evaluated by ELISA in 40 children and teenagers and in 164 adults from a rural area of the State of Mato Grosso, Brazil. Antibodies to H. pylori were detected in the serum of 31 (77.5%) children and teenagers and in 139 (84.7%) adults. The prevalence of infection increased with age (c2 for trend, p < 0.01) even though no variations occurred in the region in the present century in terms of living conditions or sanitation, economical development and migratory influx supporting the hypothesis that the infection is also acquired during later life in developing countries. An inverse correlation was observed between the prevalence of infection and annual family income (c2 for trend, p < 0.013). There was no correlation between type of system for sewage disposal and prevalence of infection (p = 0.8). In conclusion, the prevalence of H. pylori infection in Nossa Senhora do Livramento, a rural area from Brazil, is very high and similar to that observed in other developing countries. Furthermore, the increase in the prevalence of infection with age observed in this population seems to be due to both, cohort effect and acquisition of the infection during later life

Since the late 1970s pyrimethamine-sulfadoxine (PS; FansidarTM Hoffman-LaRoche, Basel) has been used as first line therapy for uncomplicated malaria in the Amazon basin. Unfortunately, resistance has developed over the last ten years in many regions of the Amazon and PS is no longer recommended for use in Brazil. In vitro resistance to pyrimethamine and cycloguanil (the active metabolite of proguanil) is caused by specific point mutations in Plasmodium falciparum dihydrofolate reductase (DHFR), and in vitro resistance to sulfadoxine has been associated with mutations in dihydropteroate synthase (DHPS). In association with a proguanil-sulfamethoxazole clinical trial in Brazil, we performed a nested mutation-specific polymerase chain reaction to measure the prevalence of DHFR mutations at codons 50, 51, 59, 108 and 164 and DHPS mutations at codons 436, 437, 540, 581 and 613 at three sites in the Brazilian Amazon. Samples from two isolated towns showed a high degree of homogeneity, with the DHFR Arg-50/Ile-51/Asn-108 and DHPS Gly-437/Glu-540/Gly-581 mutant genotype accounting for all infections in Peixoto de Azevedo (n = 15) and 60% of infections in Apiacás (n = 10), State of Mato Grosso. The remaining infections in Apiacás differed from this predominant genotype only by the addition of the Bolivia repeat at codon 30 and the Leu-164 mutation in DHFR. By contrast, 17 samples from Porto Velho, capital city of the State of Rondônia, with much in- and out-migration, showed a wide variety of DHFR and DHPS genotypes.

The zinc finger motifs (Cys2His2) are found in several proteins playing a role in the regulation of transcripton. SmZF1, a Schistosoma mansoni gene encoding a zinc finger protein was initially isolated from an adult worm cDNA library, as a partial cDNA. The full sequence of the gene was obtained by subcloning and sequencing cDNA and genomic fragments. The collated gene sequence is 2181 nt and the complete cDNA sequence is 705 bp containing the full open reading frame of the gene. Analysis of the genome sequence revealed the presence of three introns interrupting the coding region. The open reading frame theoretically encodes a protein of 164 amino acids, with a calculated molecular mass of 18,667Da. The predicted protein contains three zinc finger motifs, usually present in transcription regulatory proteins. PCR amplification with specific primers for the gene allowed for the detection of the target in egg, cercariae, schistosomulum and adult worm cDNA libraries indicating the expression of the mRNA in these life cycle stages of S. mansoni. This pattern of expression suggests the gene plays a role in vital functions of different life cycle stages of the parasite. Future research will be directed to elucidate the functional role of SmZF1.

A recombinant-antigen enzyme immunoassay (EIA), BioSCREEN TM anti-Treponema pallidum, was compared favorably with the T. pallidum hemagglutination test, in the detection of specific antibodies in different groups of sera from patients with primary (n = 38), secondary (n = 10), early latent (n = 28) and congenital syphilis (n = 2), patients with leptospirosis ( n= 8), infectious mononucleosis (n = 7), hepatitis (n = 9), diabetes mellitus (n = 11), rheumatoid arthritis (n = 13), leprosy (n = 11), tuberculosis (n = 9), HIV/Aids ( n= 12), systemic lupus erythematosus (n = 4), rheumatic fever (n = 3), old-persons (n = 9), pregnant women (n = 29) and blood donors (n = 164). The coincidence between them was 95.1%. The sensitivity and specificity of the EIA were 93.3% and 95.5%, respectively. Fifteen serum specimens belonging to old-persons, pregnant women, blood donors, and patients with human leptospirosis, hepatitis, diabetes mellitus, tuberculosis and rheumatic fever gave false-positive results by Venereal Disease Research Laboratory and/or Rapid Plasma Reagin. The EIA can be used as alternative method for the serological confirmation of syphilis.

In the last decade microsatellites have become one of the most useful genetic markers used in a large number of organisms due to their abundance and high level of polymorphism. Microsatellites have been used for individual identification, paternity tests, forensic studies and population genetics. Data on microsatellite abundance comes preferentially from microsatellite enriched libraries and DNA sequence databases. We have conducted a search in GenBank of more than 16,000 Schistosoma mansoni ESTs and 42,000 BAC sequences. In addition, we obtained 300 sequences from CA and AT microsatellite enriched genomic libraries. The sequences were searched for simple repeats using the RepeatMasker software. Of 16,022 ESTs, we detected 481 (3%) sequences that contained 622 microsatellites (434 perfect, 164 imperfect and 24 compounds). Of the 481 ESTs, 194 were grouped in 63 clusters containing 2 to 15 ESTs per cluster. Polymorphisms were observed in 16 clusters. The 287 remaining ESTs were orphan sequences. Of the 42,017 BAC end sequences, 1,598 (3.8%) contained microsatellites (2,335 perfect, 287 imperfect and 79 compounds). The 1,598 BAC end sequences 80 were grouped into 17 clusters containing 3 to 17 BAC end sequences per cluster. Microsatellites were present in 67 out of 300 sequences from microsatellite enriched libraries (55 perfect, 38 imperfect and 15 compounds). From all of the observed loci 55 were selected for having the longest perfect repeats and flanking regions that allowed the design of primers for PCR amplification. Additionally we describe two new polymorphic microsatellite loci.

We evaluated the performance of a commercial immunoblotting in the serodiagnosis of Helicobacter pylori infection in Brazilian patients. The presence of anti-H. pylori antibodies was also investigated in a group of 20 duodenal ulcer patients after successful treatment. One hundred and ninety one patients were studied. Among the 164 infected patients, 46 had gastric carcinoma. The duodenal ulcer patients were treated with antimicrobial drugs and the eradication of the microorganism was confirmed in all of them one month after the end of the treatment by the 13C-urea breath test. Sera were assayed for H. pylori antibodies using the Helicoblot 2.0 (Genelabs Diagnostics, Singapore). The sensitivity, specificity, positive, and negative predictive values of the test were 93.9%, 92.6%, 98.7%, and 71.4%, respectively. The sensitivity of the test was similar in patients with (93.5%) and without (95.7%) gastric carcinoma. Twenty-four months after the end of the treatment, the band of 116 kDa was still detected in one of the patients. In conclusion, the Helicoblot 2.0 is an accurate test to diagnose H. pylori infection and although it can not be employed to monitor the bacterium eradication, it may be useful for diagnosing past infection, especially in gastric carcinoma patients.

Studies on natural infection by Leishmania spp of sandflies collected in endemic and nonendemic areas can provide important information on the distribution and intensity of the transmission of these parasites. This study sought to investigate the natural infection by Leishmaniain wild female sandflies. The specimens were caught in the city of Corumbá, state of Mato Grosso do Sul (Brazil) between October 2012-March 2014, and dissected to investigate flagellates and/or submitted to molecular analysis to detect Leishmania DNA. A total of 1,164 females (77.56% of which were Lutzomyia cruzi) representing 11 species were investigated using molecular analysis; 126 specimens of Lu. cruziwere dissected and also submitted to molecular analysis. The infection rate based on the presence of Leishmania DNA considering all the sandfly species analysed was 0.69%; only Leishmania (Leishmania) amazonensis was identified in Lu. cruzi by the molecular analysis. The dissections were negative for flagellates. This is the first record of the presence of L. (L.) amazonensis DNA in Lu. cruzi, and the first record of this parasite in this area. These findings point to the need for further investigation into the possible role of this sandfly as vector of this parasite.

O objetivo deste estudo foi identificar as restrições causadas pela incontinência urinária (IU) à vida da mulher, considerando-se o tipo de incontinência, e verificar como manejam este problema. Trata-se de uma análise secundária de dados obtidos em estudo anterior, coletados por meio de entrevista com perguntas abertas e fechadas. A amostra foi composta por 164 mulheres com queixa de IU, internadas em clínicas de ginecologia e urologia de dois hospitais-escola da cidade de Campinas, SP. Apresentavam idade entre 25 e 85 anos e 104 (64%) referiram uma ou mais restrições. Citaram alterações nas atividades sexuais (40,9%), sociais (33,5%), domésticas (18,9%) e ocupacionais (15,2%). A incontinência urinária mista e a de urgência forem as que mais afetavam a vida das mulheres. A estratégia mais utilizada para minimizar a IU foi o uso de forro e/ou absorvente higiênico. Concluiu-se que a IU tem implicações negativas no cotidiano dessas mulheres.

Fruit flies are the major pests in fruit orchards and require a frequent insecticide aplication control, which increases production cost and chemical residues in fruits. Adults of Anastrepha fraterculus were sampled from twelve peach, plum, orange, tangerine and acid lime orchards in four counties in the Western Region of Santa Catarina. Modified McPhail plastic traps, baited with glucose 10%, were used to collect the flies from October 1998 to September 2000. Trap monitoring, bait replacement and fruit flies sorting by species and sex were done weekly. A total of 4,164 specimens of A. fraterculus was collected and highest population was registered in the county of Chapecó (64,8% of all sampled flies). Adults were collected all year long, with the highest population peaks occurring from December and January, although the fluctuation was different for each fruit species due to their particular phenology and in different years. Positive correlation among temperature, atmospheric humidity and population levels of adults of A. fraterculus was observed. According to the degree days obtained for each year, 4851.9, 4632.9 and 4983.7, respectively in 1998, 1999 and 2000, it was established that A. fraterculus could present an average of 11.2 generations a year.

Avaliar o potencial de crescimento de Lysiphlebus testaceipes (Cresson) é importante para seu uso em programas de controle biológico de pulgões. Este trabalho teve como objetivo determinar a tabela de vida de fertilidade de L. testaceipes em Rhopalosiphum maidis (Fitch) e Aphis gossypii Glover. Na avaliação da mortalidade de imaturos, do desenvolvimento e da razão sexual foram utilizadas 12 fêmeas do parasitóide e 480 ninfas de cada pulgão testado. Na avaliação da longevidade e da fertilidade foram utilizadas 15 fêmeas do parasitóide e uma colônia por dia de cada pulgão, até a morte da fêmea do parasitóide, sendo 300 (1º dia); 250 (2º dia); 200 (3º dia); 150 (4º dia); 100 (5º dia) e 50 ninfas nos demais dias. L. testaceipes apresentou taxas de mortalidade de imaturos de 5,6% em R. maidis e de 9,2% em A. gossypii, desenvolvimento de 10,2 e 10,1 dias e razão sexual de 0,71 e 0,66, respectivamente. L. testaceipes apresentou fecundidade de 498,2 ovos em R. maidis e de 327,8 ovos em A. gossypii. Os parâmetros de crescimento de L. testaceipes em R. maidis e A. gossypii foram, respectivamente, R O= 205,38 e 164,08 fêmeas; r m= 0,449 e 0,431 fêmeas/fêmeas/dia; lambda = 1,57 e 1,54 fêmeas/dia; T= 11,86 e 11,83 dias e TD= 10,78 e 11,27 dias. L. testaceipes apresenta alto potencial de crescimento em R. maidis e A. gossypii. R. maidis mostrou ser hospedeiro adequado aos propósitos de criação massal e utilização em sistema de criação aberta para L. testaceipes.

A falha ou perda da eficácia relativa dos resíduos culturais compromete o controle da erosão hídrica por meio da cobertura do solo em sistemas conservacionistas de preparo do solo. Com o objetivo de investigar a falha de resíduos culturais em diferentes situações no sistema de semeadura direta, foi instalado um experimento na Estação Experimental Agronômica da UFRGS, em Eldorado do Sul (RS), sobre um Argissolo Vermelho distrófico típico com 0,105 m m-1 de declividade média. Os tratamentos consistiram de resíduos recém-colhidos de aveia preta (out/97) e soja (maio/98) e resíduos semidecompostos de milho e soja (150 e 175 dias após a colheita, out/97 e 98, respectivamente), em diferentes quantidades e formas de manejo. Aplicou-se chuva simulada nas parcelas experimentais com intensidade constante de 63,5 mm h-1, até obtenção da taxa constante de enxurrada e, então, iniciou-se, concomitantemente à aplicação de chuva, a adição de fluxos extras de água limpa na cabeceira das parcelas, para simular comprimentos de rampa mais longos. Os dez níveis de fluxos extras de água planejados variaram de 16 a 197 (10-5) m³ s-1 m-1, com duração de 7 min cada nível. A falha do resíduo foi identificada e o equivalente comprimento crítico de rampa calculado, a partir da observação visual no campo e do uso de critério teórico baseado na relação da taxa de erosão com a taxa de descarga da enxurrada. Os intervalos de ocorrência dos comprimentos críticos de rampa obtidos variaram de 29-58 m (3.950 kg ha-1 de resíduos semidecompostos de soja, soltos) a 152-164 m (6.200 kg ha-1 de resíduos semidecompostos de milho, semi-ancorados). No tratamento com 5.600 kg ha-1 de resíduos de aveia preta, semi-ancorados, não foi observada falha do resíduo. Os resultados comprovaram que existem limites de comprimento de rampa no sistema de semeadura direta, dependendo do regime de chuva, solo, declividade e condições de manejo.

Os sistemas de manejo do solo modificam a dinâmica do fósforo alterando o conteúdo das diferentes formas de P. Objetivou-se avaliar o efeito de sistemas de cultivo em longo prazo (16 anos de plantio) sobre as diferentes formas de P no solo. Os tratamentos constaram de combinações entre dois sistemas de cultivo: milho exclusivo (M) e milho consorciado com feijão (MF), com duas doses (0 e 40 m³ ha-1 ano-1) de adubo orgânico (AO), e três doses (0, 250 e 500 kg ha-1) de N-P-K, 4-14-8 (AM). Solo sob um fragmento de Floresta Atlântica foi utilizado como referência de um estado em equilíbrio. Os valores de P orgânico total (Pot) variaram de 184,2 a 280,2 e de 147,9 a 282,9 mg kg-1, em amostras de solo das camadas de 0-10 e 10-20 cm, respectivamente, sendo os maiores valores observados para combinação 500 kg ha-1 + adubação orgânica, correspondendo, em média, a 26,4 % do P total no solo. Houve tendência da relação C/Pot manter-se constante, entre os tratamentos, constatando-se aumento dos valores de Pot com o aumento do teor de carbono orgânico total no solo. O adubo mineral promoveu incremento do P na biomassa microbiana (Pbm) apenas no sistema de milho exclusivo. Em média, o aumento do Pbm foi de 262 e 164 % para o sistema que recebeu o composto orgânico no sistema de milho exclusivo e consorciado com feijão, respectivamente. Em média, a fração de P orgânico solúvel em meio ácido correspondeu a 90 % do Pot predominando sobre a fração solúvel em base. Nos tratamentos com 500 kg ha-1 de 4-14-8 e 500 kg ha-1 + composto orgânico, no sistema de consórcio, foram obtidos aumentos nos valores de P total lábil de 53 e 157 %, respectivamente, comparados aos da testemunha. O P orgânico lábil (Pol) correspondeu, em média, a 3,7 % do Pot para os sistemas de cultivo, já para a Floresta Atlântica, esta relação foi de 10,7 %, nas duas profundidades. Os aumentos nos teores das formas mais lábeis de P, proporcionados pela adubação orgânica, evidenciam a importância deste sistema de manejo no favorecimento da ciclagem de P.

Tannery residues and coal mine waste are heavily polluting sources in Brazil, mainly in the Southern States of Rio Grande do Sul and Santa Catarina. In order to study the effects of residues of chrome leather tanning (sludge and leather shavings) and coal waste on soybean and maize crops, a field experiment is in progress since 1996, at the Federal University of Rio Grande do Sul Experimental Station, county of Eldorado do Sul, Brazil. The residues were applied twice (growing seasons 1996/97 and 1999/00). The amounts of tannery residues were applied according to their neutralizing value, at rates of up to 86.8 t ha-1, supplying from 671 to 1.342 kg ha-1 Cr(III); coal waste was applied at a total rate of 164 t ha-1. Crop yield and dry matter production were evaluated, as well as the nutrients (N, P, K, Ca, Mg, Cu and Zn) and Cr contents. Crop yields with tannery sludge application were similar to those obtained with N and lime supplied with mineral amendments. Plant Cr absorption did not increase significantly with the residue application. Tannery sludge can be used also to neutralize the high acidity developed in the soil by coal mine waste.

Nitrous oxide (N2O) is the most important non-CO2 greenhouse gas and soil management systems should be evaluated for their N2O mitigation potential. This research evaluated a long-term (22 years) experiment testing the effect of soil management systems on N2O emissions in the postharvest period (autumn) from a subtropical Rhodic Hapludox at the research center FUNDACEP, in Cruz Alta, state of Rio Grande do Sul. Three treatments were evaluated, one under conventional tillage with soybean residues (CTsoybean) and two under no-tillage with soybean (NTsoybean) and maize residues (NTmaize). N2O emissions were measured eight times within 24 days (May 2007) using closed static chambers. Gas flows were obtained based on the relations between gas concentrations in the chamber at regular intervals (0, 15, 30, 45 min) analyzed by gas chromatography. After soybean harvest, accumulated N2O emissions in the period were approximately three times higher in the untilled soil (164 mg m-2 N) than under CT (51 mg m-2 N), with a short-lived N2O peak of 670 mg m-2 h-1 N. In contrast, soil N2O emissions in NT were lower after maize than after soybean, with a N2O peak of 127 g m-2 h-1 N. The multivariate analysis of N2O fluxes and soil variables, which were determined simultaneously with air sampling, demonstrated that the main driving variables of soil N2O emissions were soil microbial activity, temperature, water-filled pore space, and NO3- content. To replace soybean monoculture, crop rotation including maize must be considered as a strategy to decrease soil N2O emissions from NT soils in Southern Brazil in a Autumn.

The species Salix x rubens is being grown on the Southern Plateau of Santa Catarina since the 1940s, but so far the soil fertility requirements of the crop have not been assessed. This study is the first to evaluate the production profile of willow plantations in this region, based on the modified method of Summer & Farina (1986), for the recommendation of fertility levels for willow. By this method, based on the law of Minimum and of Maximum for willow production for the conditions on the Southern Plateau of Santa Catarina, the following ranges could be recommended: pH: 5.0-6.5; P: 12-89 mg dm-3; Mg: 3.2-7.5 mg; Zn: 5.0-8.3 mg dm-3; Cu: 0.8-4.6 mg dm-3; and Mn; 20-164 mg dm-3. The Ca/Mg ratio should be between 1.2 and 2.9. For K and Ca only the lower (sufficiency level), but not the upper threshold (excess) was established, with respectively 114 mg dm-3 and 5.3 cmol c dm-3. It was also possible to determine the upper threshold for Al and the Al/Ca ratio, i.e., 1.7 cmol c dm-3 and 0.28, respectively. For maximum yields, the clay in the soil surface layer should be below 320 g dm-3.
