Metabolic rates were determined by dissolved oxigen changes in light and dark bottles, filled with river water and after input of mixed effluent concentrations. In another experiment, dissolved inorganic nutrients, chlorophyll-alpha and other physico-chemical parameters were analyzed. Water column showed higher decomposition than production rates. Waste inputs increased primary production rates, but in higher concentrations forced the system to heterotrophy. The natural richness of macrophytes and macroalgae could be limiting the phytoplanktonic production by phosphorus assimilation. Observation of the nitrogenated inorganic nutrients suggest that the natural pelagic system is directed to nitrification. Mixed waste input inverted this trend, toward denitrification.

Sugar cane cultivation in the State of São Paulo has expanded in the last years, with an annual production of 20010(6) t in an area of 2.5 10(6) hectares. The use of herbicides, pesticides and fertilizers in sugar cane plantations, together with deforestation of riparian vegetation, have caused impacts on the hydric resources of the adjacent areas. The aim of this work is to evaluate the influence of sugar cane plantations on streams in the central region of the State of São Paulo, studying 16 organochlorinated compounds and 7 metals (Cu, Fe, Cd, Zn, Mn, Cr and Ni) found in the sediments of 11 streams. The results show that there is a higher concentration of metals and organochlorinated compounds in streams without riparian vegetation when compared to forested areas.

It is widely recognized that pharmacokinetic optimization needs to be addressed early in drug discovery to reduce the high failure rate in bringing drugs to market. Poor absorption, too short duration of action due to high elimination rate, or the presence of active metabolites are examples of properties that can potentially lead to unsuccessful clinical programmes. Here I describe a brief overview of advantages and molecular strategies for improving metabolic and pharmacokinetic properties applied to the discovery of fluconazol, beta-blockers, ritonavir and ezetimibe and to the development of the prodrugs enalapril and bambuterol.

Residues of herbicides from sugarcane were monitored in waters and sediments of Corumbataí River and tributaries. Ametryne, atrazine, simazine, hexazinone, glyphosate, and clomazone were detected in water samples, with negligible levels of ametryne and glyphosate in sediment samples. The area of recharge of the Guarani aquifer presented the highest triazine and clomazone levels. The triazines were detected at higher levels, with atrazine above Brazil's potability and quality standards. Total herbicide levels at some sampling points were 13 times higher than the European Community potability limit. There is no Brazilian standard for ametryne, although the risk is larger due to ametryne's higher toxicity for the aquatic biota.

Docosahexaenoic acid (C22:6, n-3, DHA) is a polyunsaturated fatty acid (PUFA) present in large concentrations in the brain and, due to the presence of six double bonds in its structure, is highly susceptible to oxidation by enzymes and reactive oxygen/nitrogen species. The peroxidation of PUFAs has been implicated in an increasing number of human disorders, including neurodegenerative diseases. Hence, a better understanding of the metabolism pathways of DHA should provide new insights about its role in neurodegenerative diseases. Here we review the main aspects related to DHA metabolism, as well as, the recent findings showing its association with neurodegenerative diseases.

The aim of the study was to synthesize the urea (13CO(NH2)2), with 99% 13C atoms, and to perform a quality analysis for the diagnosis (breath test) of Helicobacter pylori. Furthermore, the process was submitted to economic analysis. The reaction was performed in a stainless steel reactor, lined with polytetrafluoroethylene, under low pressure and temperature. The synthesis method was shown to be appropriate (2.35 g; 81.9% yield), evidenced by physico-chemical and microbiological results, according to Brazilian legislation. The production and diagnosis costs were competitive compared with national and international market values, rendering this a valuable tool in clinical medicine.

Arsenic is considered a semimetal, and its wide distribution in the Earth's crust in different chemical forms, including organic and inorganic species, has a great deal of influence on the mechanisms of toxicity. Exposure to arsenic can be either through occupational practice (use of pesticides) or by the consumption of water and food containing the element. Rice is considered a fundamental constituent of the basic diet of Brazilians and is usually cultivated in flooded conditions. Such a plantation system results in an increased amount of As in the soil and hence a greater accumulation of As in the plant, which is highlighted by the inorganic species' classification as highly toxic. Besides the use of mitigation techniques to reduce the toxicological risk, monitoring the concentration of As and its chemical species in rice and rice products is required through the establishment of legislation in the area. Thus, some world organizations are conducting improved research to determine and establish acceptable concentrations of As and its chemical species in rice, e. g., in 2012, FDA researchers described a chemical speciation methodology for As in rice and rice products. Hence, the application of existing chemical speciation methods and the establishment of parameters for ensuring food security and exposure risk assessment deserve particular consideration.

Objetivando o diagnóstico e quantificação das doenças da acerola (Malpighia emarginata) no Estado da Paraíba, realizou-se um levantamento nas microrregiões do Sertão, Curimatau, Brejo e Litoral. As doenças identificadas foram a cercosporiose (Cercospora sp.), fusariose (Fusarium oxysporum), fumagina (Capnodium sp.), antracnose (Colletotrichum gloeosporioides) e mancha de Alternaria (Alternaria sp.). Além dessas, no teste de patogenicidade, verificou-se o desenvolvimento de lesões circulares, secamento e queda de folhas que foram previamente feridas e inoculadas com um fungo do gênero Pestalotiopsis,isolado de material doente coletado em áreas de plantio. Nas microrregiões do Brejo e Curimatau verificaram-se, respectivamente, o maior e o menor número de doenças. A cercosporiose ocorreu em todas as microrregiões, enquanto a antracnose e a fumagina foram verificadas no Brejo e Litoral, e a fusariose nas áreas irrigadas do Sertão e Curimataú. No Sertão e no Brejo, ocorreu, ainda, a mancha de Alternaria e pestalotiose, respectivamente. Os maiores valores de incidência e severidade da fumagina, antracnose e cercosporiose foram associados a elevações da precipitação pluviométrica nas microrregiões em que foram assinaladas, como também a presença de insetos sugadores para a primeira doença.

Com o objetivo de conhecer as alterações metabólicas promovidas pelo Soil-borne wheat mosaic virus (SBWMV), um dos vírus economicamente mais importantes da cultura do trigo (Triticum aestivum), foram analisados os níveis de proteínas solúveis e determinadas as atividades da peroxidase e da protease em quatro cultivares (BRS Guabiju, BRS 194, BRS 179, BR 23) e uma linhagem (PF 980524) de trigo com diferentes níveis de resistência ao vírus. Os dados obtidos foram submetidos à análise de variância, comparando-se as médias, pelo Teste de Duncan a 5%. Os níveis de proteínas solúveis foram mais elevados nas plantas sem sintomas, enquanto que as atividades da peroxidase e da protease foram maiores em plantas com sintoma de mosaico do que em plantas assintomáticas. Além disso, pode-se constatar que quanto maior a suscetibilidade do genótipo, maior o nível de atividade da protease. Estes resultados são promissores para estudos de inibição da protease para controle de viroses.

O Apple stem pitting virus (ASPV) foi detectado por RT-PCR em amostras de cultivares de pereiras européias (Pyrus communis L.) cvs. Starkrimson e Abate Fetel, e asiáticas (P. pyrifolia var. culta) cvs. Kousui e Housui coletadas no início do outono de 2003 em pomar da Estação Experimental da Embrapa Uva e Vinho, Vacaria, RS. Utilizando várias combinações de oligonucleotídeos, foram amplificados fragmentos de DNA de 269 e 1554 pb, este último contendo o gene completo (1131 nt) da proteína capsidial do ASPV. Outro fragmento amplificado de 291 pb compreende parte do gene da polimerase viral. Estes fragmentos constituem-se em um excelente instrumento de diagnóstico do ASPV em pereiras. A comparação das seqüências de nucleotídeos do gene da proteína capsidial do ASPV com seqüências do banco de dados GenBank, revelou identidades de 89% com seqüências de um isolado alemão de macieira e de 85 a 88% com isolados poloneses, de pereiras. A indicadora herbácea Nicotiana occidentalis cv. 37B, inoculada mecanicamente com extrato foliar da cv. Housui, apresentou lesões locais necróticas, necrose foliar marginal e das nervuras. O ASPV também foi detectado por dot-ELISA nas cvs. Abate Fetel e Kousui, na cv. Starkrimson por imunoblot, e em Pyronia veitchii (Trabut) Guill. por enxertia de borbulhas da cv. Abate Fetel infetada.

Soil-borne wheat mosaic virus - SBWMV, agente causal da virose que se caracteriza, em termos econômicos, como uma das mais importantes enfermidades da cultura de trigo, pode também infectar uma vasta gama de gramíneas. Com o objetivo de conhecer as alterações metabólicas promovidas pelo mosaico do trigo, foram analisados os teores de açúcares totais e a concentração de prolina e determinou-se a atividade da nitrato redutase. O experimento foi conduzido na área experimental da Embrapa Trigo, usando cinco genótipos de trigo (BRS Guabiju, BRS 194, BRS 179, BR 23 e PF 980524) com diferentes níveis de resistência ao SBWMV. As determinações bioquímicas foram realizadas 45 dias após a emergência de plantas. A atividade da nitrato redutase foi mais elevada em plantas sem sintomas, quando comparada às com sintomas. Os níveis de açúcares e de prolina foram mais elevados em plantas com sintomas do que nas sem sintomas. Os resultados encontrados comprovam as alterações metabólicas promovidas pelo SBWMV nos cinco genótipos de trigo testados.
