Irrigation plays an important role for grape’s yield as well as on its quality for winemaking. Thus, the effects of deficit irrigation strategies on yield and quality of wine grapes cv. Syrah were evaluated in Petrolina, State of Pernambuco, Brazil. Evaluations were carried out throughout the second and third growing seasons, which were from November 2010 to February 2011 (rainy season) and from May to September 2011 (dry season), respectively. Vines were drip irrigated and the experimental design was completely randomized with three treatments and four replications. The treatments were full irrigation (FI), performed according crop evapotranspiration; regulated deficit irrigation (RDI), in which irrigation was interrupted in phenological growth stage of bunch closure, but was occasionally performed according soil water monitoring of the root zone; and deficit irrigation (DI), when irrigation was interrupted from bunch closure to harvesting. Differences on leaf water content among treatments were observed in both growing seasons and RDI and DI treatment plants presented moderate water stress. The number of bunches did not differ among treatments in both growing seasons; however, bunch weight per plant, average bunch weight and soluble solid content were higher in FI treatment during the dry season. Deficit irrigation strategies promoted water saving.

The flowers of this orchard tree presents six fleshy petals, fused two by two, and disposed propeller like. In the center is a cavity which harbors the cone shaped reproductive organs. An eriophyid mite, described by H. H. Keifer as Aculops flechtmann Keifer, 1972, causes considerable bronzing to the petals (Fig. 1-A). Large numbers of this mite attacking the basis of the reproductive organs causes them to dry and drop. On the young fruits, which are squamous, this mite causes some rusting and small necrotic areas (Fig. 1-B). As the fruit grows these small areas remain dark and barky. Very often the necrotic areas are invaded by the false spider mite Brevipalpus phoenicis (Geijskes, 1939), which enlarges the damage. The false spider mite also attacks leaves and green stems; the epidermis of the latter then assumes a barky appearance.

ABSTRACTMealybugs (Hemiptera: Pseudococcidae) are important pests of the grapevine Vitis spp. and are responsible for direct and indirect damage to production. The main mealybug species present in wine grapevine (Vitis vinifera L.) in Southern Brazil were identified and their incidence evaluated. Bunch-samples (n = 50) from 131 vineyards located in the Serra Gaúcha Region (RS) of Brazil were analyzed at harvest, and the occurrence of mealybugs in the roots was evaluated at the time of eradication of plants for replanting. Mealybugs were reared in laboratory until adulthood for species determination. The species Dysmicoccus brevipes (Cockerell, 1983), Dysmicoccus sp., Planococcus citri (Risso, 1813), Pl. minor(Maskell, 1897), Pseudococcus viburni (Signoret, 1875) and Pseudococcus sp. were identified in bunches. Dysmicoccus sp., D. umbambae Granara de Willink, 2009, Pl. citri and Pseudococcus sp. were found in the roots. Pl. citri (31.4%) and Dysmicoccus sp. (22.7%) were the most common species found in wine grape bunches in the Serra Gaúcha Region.

Phosphorus (P) applications to vineyards can cause P accumulation in the soil and maximize pollution risks. This study was carried out to quantify the accumulation of P fractions in sandy soils of vineyards in southern Brazil. Soil samples (layers 0-5, 6-10 and 11-20 cm) were collected from a native grassland area and two vineyards, after 14 years (vineyard 1) and 30 years (vineyard 2) of cultivation, in Santana do Livramento, southern Brazil, and subjected to chemical fractionation of P. Phosphorus application, especially to the 30-year-old vineyard 2, increased the inorganic P content down to a depth of 20 cm, mainly in the labile fractions extracted by anion-exchange resin and NaHCO3, in the moderately labile fraction extracted by 0.1 and 0.5 mol L-1 NaOH, and in the non-labile fraction extracted by 1 mol L-1 HCl, indicating the possibility of water eutrophication. Phosphorus application and grapevine cultivation time increased the P content in the organic fraction extracted by NaHCO3 from the 0-5 cm layer, and especially in the moderately labile fraction extracted by 0.1 mol L-1 NaOH, down to a depth of 20 cm.

The use of cover crops in vineyards is a conservation practice with the purpose of reducing soil erosion and improving the soil physical quality. The objective of this study was to evaluate cover crop species and management systems on soil physical properties and grape yield. The experiment was carried out in Bento Gonçalves, RS, Southern Brazil, on a Haplic Cambisol, in a vineyard established in 1989, using White and Rose Niagara grape (Vitis labrusca L.) in a horizontal, overhead trellis system. The treatments were established in 2002, consisting of three cover crops: spontaneous species (SS), black oat (Avena strigosa Schreb) (BO), and a mixture of white clover (Trifolium repens L.), red clover (Trifolium pratense L.) and annual rye-grass (Lolium multiflorum L.) (MC). Two management systems were applied: desiccation with herbicide (D) and mechanical mowing (M). Soil under a native forest (NF) area was collected as a reference. The experimental design consisted of completely randomized blocks, with three replications. The soil physical properties in the vine rows were not influenced by cover crops and were similar to the native forest, with good quality of the soil structure. In the inter-rows, however, there was a reduction in biopores, macroporosity, total porosity and an increase in soil density, related to the compaction of the surface soil layer. The M system increased soil aggregate stability compared to the D system. The treatments affected grapevine yield only in years with excess or irregular rainfall.

In the subtropical regions of southern Brazil, rainfall distribution is uneven, which results in temporal variability of soil water storage. For grapes, water is generally available in excess and water deficiency occurs only occasionally. Furthermore, on the Southern Plateau of Santa Catarina, there are differences in soil properties, which results in high spatial variability. These two factors affect the composition of wine grapes. Spatio-temporal analyses are therefore useful in the selection of cultural practices as well as of adequate soils for vineyards. In this way, well-suited areas can produce grapes with a more appropriate composition for the production of quality wines. The aim of this study was to evaluate the spatio-temporal variability of water storage in a Cambisol during the growth cycle of a Cabernet Sauvignon vineyard and its relation to selected soil properties. The experimental area consisted of a commercial 8-year-old vineyard in São Joaquim, Santa Catarina, Brazil. A sampling grid with five rows and seven points per row, spaced 12 m apart, was outlined on an area of 3,456 m². Soil samples were collected with an auger at these points, 0.30 m away from the grapevines, in the 0.00-0.30 m layer, to determine gravimetric soil moisture. Measurements were taken once a week from December 2008 to April 2009, and every two weeks from December 2009 to March 2010. In December 2008, undisturbed soil samples were collected to determine bulk density, macro- and microporosity, and disturbed samples were used to quantify particle size distribution and organic carbon content. Results were subjected to descriptive analysis and semivariogram analysis, calculating the mean relative difference and the Pearson correlation. The average water storage in a Cambisol under grapevine on ridges had variable spatial dependence, i.e., the lower the average water storage, the higher the range of spatial dependence. Water storage had a stable spatial pattern during the trial period, indicating that the points with lower water storage or points with higher water storage during a certain period maintain these conditions throughout the experimental period. The relative difference is a simple method to identify positions that represent the average soil water storage more adequately at any time for a given area.

The objective of this work was to analyze the genetic diversity of 47 table grape accessions, from the grapevine germplasm bank of Embrapa Semiárido, using 20 RAPD and seven microsatellite markers. Genetic distances between pairs of accessions were obtained based on Jaccard's similarity index for RAPD data and on the arithmetic complement of the weighted index for microsatellite data. The groups were formed according to the Tocher's cluster analysis and to the unweighted pair‑group method with arithmetic mean (UPGMA). The microsatellite markers were more efficient than the RAPD ones in the identification of genetic relationships. Information on the genetic distance, based on molecular characteristics and coupled with the cultivar agronomic performance, allowed for the recommendation of parents for crossings, in order to obtain superior hybrids in segregating populations for the table grape breeding program of Embrapa Semiárido.

Resumo: O objetivo deste trabalho foi avaliar os efeitos de viroses em videiras sintomáticas e assintomáticas sobre as variáveis agronômicas relacionadas ao vigor das plantas e à qualidade enológica da uva, e comparar os isolados virais obtidos nessas duas condições. Realizaram-se dois experimentos com quatro cultivares. Todas as plantas foram indexadas, por meio da reação em cadeia da polimerase via transcrição reversa (RT-PCR) em tempo real, quanto à provável ocorrência dos seguintes vírus: Grapevine virus A (GVA), Grapevine virus B (GVB), Grapevine virus D (GVD), Grapevine leafroll-associated virus (GLRaV-1 ao -4, GLRaV-4 estirpe 5), Grapevine rupestris stem pitting-associated virus (GRSPaV) e Grapevine fleck virus (GFkV). As variáveis avaliadas foram: número de gemas brotadas e não brotadas, número de ramos com ou sem cachos, número total de gemas, número de cachos, massa de cachos frescos, massa total de bagas, massa do engaço, número de bagas por cacho, massa média de baga, sólidos solúveis totais, acidez total titulável, pH, massa de ramos podados ou diâmetros do tronco do porta-enxerto e da copa. Os efeitos negativos foram mais pronunciados nas plantas com sintomas de viroses; no entanto, constatou-se frequentemente que plantas sem sintomas também estavam infectadas. A análise molecular de GRSPaV, GVA e GLRaV-2, isolados de plantas sintomáticas e assintomáticas, resultou em alta percentagem de identidade de nucleotídeos entre isolados homólogos.

The first phytopathogenic bacterium with its DNA entirely sequenced is being detected and isolated from different host plants in several geographic regions. Although it causes diseases in cultures of economic importance, such as citrus, coffee, and grapevine little is known about the genetic relationships among different strains. Actually, all strains are grouped as a single species, Xylella fastidiosa, despite colonizing different hosts, developing symptoms, and different physiological and microbiological observed conditions. The existence of genetic diversity among X. fastidiosa strains was detected by different methodological techniques, since cultural to molecular methods. However, little is know about the phylogenetic relationships developed by Brazilian strains obtained from coffee and citrus plants. In order to evaluate it, fAFLP markers were used to verify genetic diversity and phylogenetic relationships developed by Brazilian and strange strains. fAFLP is an efficient technique, with high reproducibility that is currently used for bacterial typing and classification. The obtained results showed that Brazilian strains present genetic diversity and that the strains from this study were grouped distinctly according host and geographical origin like citrus-coffee, temecula-grapevine-mulberry and plum-elm.

The objectives of this study were to evaluate the performance of cultivars, to quantify the variability and to estimate the genetic distances of 66 wine grape accessions in the Grape Germplasm Bank of the EMBRAPA Semi-Arid, in Juazeiro, BA, Brazil, through the characterization of discrete and continuous phenotypic variables. Multivariate statistics, such as, principal components, Tocher's optimization procedure, and the graphic of the distance, were efficient in grouping more similar genotypes, according to their phenotypic characteristics. There was no agreement in the formation of groups between continuous and discrete morpho-agronomic traits, when Tocher's optimization procedure was used. Discrete variables allowed the separation of Vitis vinifera and hybrids in different groups. Significant positive correlations were observed between weight, length and width of bunches, and a negative correlation between titratable acidity and TSS/TTA. The major part (84.12%) of the total variation present in the original data was explained by the four principal components. The results revealed little variability between wine grape accessions in the Grape Germplasm Bank of Embrapa Semi-Arid.

The objective of this study was to evaluate agronomic and molecular traits of the 'Italia Muscat' clone and compare it with the cv. 'Italia', providing information to support the cultivation of 'Italia Muscat' this cultivar in the São Francisco River Valley. Agronomic characteristics of both clones were evaluated for two seasons in 2004. The characteristics were phenology, bud break (%), bud fertility (%), yield (kg) mass of bunches (g), length and width of bunches (cm), mass of berries (g), length and diameter of berries (mm), TSS (ºBrix), ATT (% titratable acidity) and TSS/TTA. Molecular analysis of seven SSR markers were carried out. The clone 'Italia Muscat' showed larger berries, mass of bunches and better TSS/TA ratio than 'Italia'. The molecular analysis resulted in the same allelic profile in both clones, highlighting the need to use a larger number of microsatellite markers or other molecular technique to allow their discrimination. Based on their morpho-agronomic characteristics, 'Italia Muscat' seems to be a good table grape cultivar alternative for grape growers of São Francisco River Valley.

This work presents an evolution profile of labels and containers of commercial chemicals employed in laboratories since 1870. Most chemicals were made in Germany before the Second World War, after which many other manufacturers arrived on the Brazilian market. North-american products were dominant in the 1940s, but Brazilian chemicals have increased their participation along time. Labels presented increasingly more information, from originally simple names of the compounds at the beginning of the XXth century to the data presented today such as chemical formulae, safety regards and detailed chemical analysis. The raw material for container manufacturing also changed: glass was dominant until the 1950s, but nowadays plastic flasks are preferred whenever possible. Cork covers were replaced by screw caps. The diversity of commercial products also sharply increased with time, especially after the 1950s, following the many new and specific applications of chemicals for research and commercial purposes.

Em São Paulo, ocorrem quatro isolados do vírus que induz o mosaico das nervuras da videira (Vitis spp.), os quais são diferenciados pelos sintomas que provocam em algumas variedades. Para confirmar a identidade desse vírus e o relacionamento existente entre os quatro isolados, foram aplicados os testes DAS-ELISA e TAS-ELISA usando anti-soros comerciais contra o Grapevine fleckvirus (GFkV). As fontes de antígeno foram tecidos de floema de ramos dormentes e de folhas jovens da brotação de primavera de videiras sabidamente infetadas. As reações nos testes imuno-enzimáticos envolvendo os quatro isolados do vírus foram positivas para o anti-soro contra o GFkV. Os resultados foram também positivos para amostras de 66 plantas infetadas de 26 variedades de videira procedentes de 11 regiões vitícolas de São Paulo e de 24 plantas de 12 variedades provenientes dos estados de Minas Gerais, Paraná, Pernambuco, Rio Grande do Norte e Santa Catarina. Extratos de folhas novas e tenras apresentaram valores de absorbância mais consistentes do que extratos de ramos dormentes. Plantas não infetadas foram empregadas como controle negativo. Os resultados obtidos confirmaram que os quatro isolados virais possuem relacionamento sorológico com o GFkV, sugerindo que os mesmos pertencem ao complexo viral que causa o "grapevine fleck disease".

Bacterial canker of grapevine (Vitis vinifera), caused by Xanthomonas campestris pv. viticola was first detected in Brazil in 1998, affecting grapevines in the São Francisco river basin, state of Pernambuco. The disease was also reported in Juazeiro, Bahia and later in Piauí and Ceará. Due to its limited geographical distribution and relatively recent detection in Brazil, very little is known about the pathogen's biology and diversity. Repetitive DNA based-PCR (rep-PCR) profiles were generated from purified bacterial DNA of 40 field strains of X. campestris pv. viticola, collected between 1998 and 2001 in the states of Pernambuco, Bahia and Piauí. Combined analysis of the PCR patterns obtained with primers REP, ERIC and BOX, showed a high degree of similarity among Brazilian strains and the Indian type strain NCPPB 2475. Similar genomic patterns with several diagnostic bands, present in all strains, could be detected. Fingerprints were distinct from those of strains representing other pathovars and from a yellow non-pathogenic isolate from grape leaves. The polymorphism observed among the Brazilian strains allowed their separation into five subgroups, although with no correlation with cultivar of origin, geographic location or year collected.

Viroids, non-protein-coding small (246-401 nt) circular single-stranded RNAs with autonomous replication, are currently classified into two families. Within the family Pospiviroidae, Citrus exocortis viroid (CEVd) belongs to the genus Pospiviroid while Hop stunt viroid (HSVd) is the single member of the genus Hostuviroid. These pathogens are distributed worldwide and infect a large number of hosts. In Brazil, isolates of CEVd and HSVd have been detected in both citrus and grapevine. To characterize and study the genetic variability of these viroids, total RNA from leaves of grapevine Vitis vinifera 'Cabernet Sauvignon' and V. labrusca 'Niagara Rosada' from Bento Gonçalves, RS, was used as a template for RT-PCR amplification with specific primers for the five viroids described infecting grapevines [HSVd, CEVd, Grapevine yellow speckle viroid 1 (GYSVd-1), Grapevine yellow speckle viroid 2 (GYSVd-2) and Australian grapevine viroid (AGVd)]. Leaf samples of Citrus medica infected with CEVd from São Paulo were also analyzed. The resulting products were separated by agarose gel electrophoresis and DNA fragments of the expected size were eluted, cloned and sequenced. The grapevine samples analyzed were doubly infected by CEVd and HSVd. A phylogenetic analysis showed that the Brazilian grapevine HSVd variants clustered with other grapevine HSVd variants, forming a specific group separated from citrus variants, whereas the Brazilian CEVd variants clustered with other citrus and grapevine variants.
