Foram realizados experimentos em ambiente controlado, em câmaras de crescimento, com duas cultivares de feijoeiro (Phaseolus vulgaris), Rosinha e Carioca, para a quantificação de componentes monocíclicos da mancha angular. Os componentes monocíclicos avaliados foram: período de incubação, severidade da doença e taxa de crescimento das lesões, sob as temperaturas de 6, 10, 14, 17, 21, 25, 29 e 33 ºC. Nenhum sintoma da mancha angular ocorreu em 6 e 33 ºC. A severidade da mancha angular foi elevada na faixa de temperatura entre 20 e 25 ºC e o período de incubação foi favorecido pela temperatura de 25 ºC, para as duas cultivares testadas. Esta temperatura também proporcionou a maior taxa de crescimento das lesões e estes dados foram ajustados, em função da temperatura por uma equação do primeiro grau. O efeito do período de molhamento foliar (5, 6, 12, 18, 24, 30, 36, 42, 48 e 54 h) na severidade da doença também foi avaliado. A severidade da mancha angular do feijoeiro aumentou com o aumento da duração do molhamento foliar até o máximo de 54 h. As variações de severidade da mancha angular do feijoeiro, em relação ao número de horas de molhamento foliar, foram explicadas pelo modelo de Richards. A função Beta-Richards explicou o efeito combinado da temperatura e da duração do molhamento foliar na severidade da mancha angular do feijoeiro para ambas as cultivares.

Este trabalho objetivou estudar o efeito da temperatura e do molhamento foliar no desenvolvimento da ferrugem e da mancha angular do feijoeiro (Phaseolus vulgaris). Foram conduzidos ensaios em condições de campo durante os plantios das águas (outubro a dezembro), de inverno (maio a agosto) e da seca (fevereiro a maio). Os ensaios foram instalados em blocos casualizados com três repetições e dois tratamentos. Os tratamentos consistiram de: 1) inoculação com Uromyces appendiculatus e 2) inoculação com Phaeoisariopsis griseola. No inverno, a ferrugem ocorreu com maior intensidade, atingindo um valor máximo de severidade (Ymax) igual a 1,3 e as maiores taxas de crescimento (0,09, 0,04 e 0,07 aos 34, 55 e 62 dias, respectivamente), devido à predominância, nessa época, de temperaturas inferiores a 21,1 °C, e de maior número de horas diárias com temperaturas menores ou iguais a 16 °C, quando ocorreu molhamento foliar. Nos plantios das águas e da seca, a mancha angular ocorreu com maior intensidade, atingindo Ymax iguais a 2,6 e 3,5, respectivamente e as maiores taxas de crescimento (0,06, 0,02 e 0,15 aos 46, 60 e 67 dias, respectivamente, no plantio das águas, e 0,13 e 0,38 aos 51 e 58 dias, respectivamente, no plantio da seca), devido à predominância, nessa época, de temperaturas superiores a 21,1 °C, quando ocorreu molhamento foliar e não ocorrência de temperaturas menores ou iguais a 16 °C. Com esse estudo pôde-se caracterizar, em função das condições climáticas, o plantio de inverno como favorável à ocorrência da ferrugem e os plantios da água e da seca como favorável à ocorrência da mancha angular.

Vinte e oito genótipos, incluindo cultivares de algumas instituições de pesquisa e linhagens do programa de melhoramento da Embrapa Arroz e Feijão, foram avaliados quanto a resistência a oito patótipos do fungo Phaeoisariopsis griseola, agente causal da mancha angular do feijoeiro comum. Estes genótipos foram inoculados aos 14-16 dias após o plantio com uma suspensão contendo 2,0 x 10(4) conídios mL-1. Os sintomas foram avaliados de 14 a 18 dias após a inoculação, utilizando uma escala de nove graus onde foram consideradas resistentes (reação compatível) as plantas que apresentaram os graus de 1 a 3 e suscetíveis (reação incompatível), as que apresentaram os graus de 4 a 9. Os genótipos 'Ouro Negro' e 'LM 202202530' apresentaram reação de resistência a oito e a sete dos oito patótipos utilizados, respectivamente. A cultivar BRS Requinte foi resistente a cinco patótipos e as cultivares BRS Pontal e Cornell 49-242 foram resistentes a apenas quatro patótipos.

Foi avaliada a reação de 23 genótipos de feijoeiro comum (Phaseolus vulgaris) à mancha angular causada por Phaeoisariopsis griseola, utilizando os isolados monospóricos Ig 664 e Ig 669, caracterizados como os patótipos 63.23 e 63.19, respectivamente. Quatorze dias após a semeadura (estádio V3), as plantas foram inoculadas com uma suspensão contendo 2,5 x 10(4) conídios/mL, nas faces adaxial e abaxial da primeira folha trifoliolada. A avaliação dos sintomas foi efetuada aos 14 e 18 dias após a inoculação, utilizando-se uma escala variando de 1 (sem sintomas) a 9 ( > 25% da área foliar com lesões de mancha angular). Das cultivares analisadas, BRS Valente, CNFC 10281, CNFP 10138, BRS Grafite, BRS Requinte, BRS Pontal, MAR 02, Cornell 49-242 e AND 277 foram resistentes aos dois isolados. As cultivares Carioca Rubi, CNFC 9504, CNFC 10150 e Soberano foram suscetíveis aos dois patótipos testados. O patótipo 63.23 foi o mais virulento. As novas fontes de resistência à mancha angular identificadas neste trabalho podem ser utilizadas em programas de melhoramento do feijoeiro comum, que visem incorporar resistência em novas cultivares.

O feijoeiro (Phaseolus vulgaris L.) está entre as principais culturas da agricultura nacional e pode ser afetada por várias doenças, como a mancha angular causada por Pseudocercospora griseola. Com o objetivo de desenvolver métodos alternativos ao químico para o controle desta doença, verificou-se o potencial de extratos de micélio de Pycnoporus sanguineus para atividade antimicrobiana in vitro contra P. griseola e para indução de resistência e ativação de enzimas de defesa em feijoeiro, como peroxidase, polifenoloxidase e β-1,3-glucanase, além de sua influência nos teores de proteínas e clorofilas. Os experimentos in vitro e em casa de vegetação foram constituídos pelos extratos de micélio de P. sanguineus e dos controles água, acibenzolar-S-metil (ASM: 75 mg i.a./L) e fungicida azoxystrobin (40 mg i.a./L). In vitro o extrato de micélio apresentou efeito sobre o crescimento micelial, a esporulação e a germinação de esporos de P. griseola. In vivo a severidade foi reduzida em 93% e 50% em casa de vegetação e a campo respectivamente, em relação ao controle água. A atividade das enzimas peroxidase e polifenoloxidase e os teores de proteínas e clorofilas foram maiores nas plantas tratadas com o extrato. Estes resultados indicam a eficiência de P. sanguineus para o controle alternativo da mancha angular do feijoeiro.

Data available in the literature were used to develop a warning system for bean angular leaf spot and anthracnose, caused by Phaeoisariopsis griseola and Colletotrichum lindemuthianum, respectively. The model is based on favorable environmental conditions for the infectious process such as continuous leaf wetness duration and mean air temperature during this subphase of the pathogen-host relationship cycle. Equations published by DALLA PRIA (1977) showing the interactions of those two factors on the disease severity were used. Excell spreadsheet was used to calculate the leaf wetness period needed to cause different infection probabilities at different temperature ranges. These data were employed to elaborate critical period tables used to program a computerized electronic device that records leaf wetness duration and mean temperature and automatically shows the daily disease severity value (DDSV) for each disease. The model should be validated in field experiments under natural infection for which the daily disease severity sum (DDSS) should be identified as a criterion to indicate the beginning and the interval of fungicide applications to control both diseases.

Uma condição com aumento da fenda bucal de eqüinos por lesão na comissura labial foi estudada. Este aumento tinha extensão variável e era uni ou bilateral. Na mucosa da bochecha da comissura labial exposta havia pequenas erosões. Durante a mastigação havia perda de pequena quantidade de capim e saliva pela fenda bucal aumentada. Os animais apresentavam bom estado nutricional. O exame histopatológico de tecido retirado da comissura labial revelou epidermite superficial. Nas quatro propriedades onde se verificou o problema, constatou-se que os eqüinos eram mantidos em sistema extensivo de criação em pastagem de Panicum maximum (variedades Tanzânia, Mombaça, Tobiatã e Colonião), com folhas maduras, altas, lignificadas e de bordos cortantes. De acordo com os dados epidemiológicos, com os achados clínicos e histopatológicos, concluí-se que essas lesões foram causadas pela ação cortante das folhas de Panicum maximum, associada à forma de apreensão da pastagem alta e mastigação pelos eqüinos.

This study compares contrast thresholds for sinewave gratings, or spatial frequencies (1/CSF) with contrast thresholds for angular frequencies (1/aCSF) and for radial frequencies, or J0 targets (1/rCSF). Observers had to differentiate between one of these frequency stimuli and a stimulus at mean luminance within a forced-choice procedure. All measurements were made with the same equipment, methods and subjects. Our results show higher sensitivity to, or lower thresholds for, angular frequencies when compared to either sinewave gratings or J0 targets. Contrast values in arbitrary units, in the lower threshold range for angular frequencies, were about half those required to differentiate sinewave gratings from mean luminance in its most sensitive range

Independence among channels processing different aspects of spatial information, including orthogonal stimuli, has been generally assumed in the literature. We tested independence between the processing of jo targets and the processing of either vertical sinusoidal gratings or angular frequency stimuli with suprathreshold summation. We found the detection of a jo target at 1 cpd to be affected in an inhibitory fashion by either background angular frequencies in the range of 3-96 cycles or sinewave gratings in the range of 0.8-3.0 cpd. These results demonstrate interactions both among orthogonal stimuli and among channels processing vertical sinewave gratings and jo target stimuli. Our discussion focuses on the hypothesis of frequency decomposition in polar coordinates

The influence of voltage on the conductance of toad skin was studied to identify the time course of the activation/deactivation dynamics of voltage-dependent Cl- channels located in the apical membrane of mitochondrion-rich cells in this tissue. Positive apical voltage induced an important conductance inhibition which took a few seconds to fully develop and was instantaneously released by pulse inversion to negative voltage, indicating a short-duration memory of the inhibiting factors. Sinusoidal stimulation at 23.4 mM [Cl-] showed hysteresis in the current versus voltage curves, even at very low frequency, suggesting that the rate of voltage application was also relevant for the inhibition/releasing effect to develop. We conclude that the voltage modulation of apical Cl- permeability is essentially a fast process and the apparent slow components of activation/deactivation obtained in the whole skin are a consequence of a gradual voltage build-up across the apical membrane due to voltage sharing between apical and basolateral membranes

The effect of several ions (Cl-, Na+, K+, Ca2+) on the rate of plasminogen (Pg) activation by recombinant staphylokinase (rSTA) is reported. Both monovalent and divalent ions affect the rate at which Pg is activated by rSTA, in a concentration-dependent manner (range 0-100 mM). In almost all cases, a decrease of the initial velocity of activation was observed. Cl- showed the most striking inhibitory effect at low concentrations (64% at 10 mM). However, in the presence of a fibrin surface, this inhibition was attenuated to 38%. Surprisingly, 10 mM Ca2+ enhanced the Pg activation rate 21% when a polymerized fibrin matrix was present. These data support the idea that ions can modulate the rate of Pg activation through a mechanism that may be associated with changes in the molecular conformation of the zymogen. This effect is strongly dependent on the presence of a fibrin clot.

Fibronectin (FN), a large family of plasma and extracellular matrix (ECM) glycoproteins, plays an important role in leukocyte migration. In normal central nervous system (CNS), a fine and delicate mesh of FN is virtually restricted to the basal membrane of cerebral blood vessels and to the glial limitans externa. Experimental autoimmune encephalomyelitis (EAE), an inflammatory CNS demyelinating disease, was induced in Lewis rats with a spinal cord homogenate. During the preclinical phase and the onset of the disease, marked immunolabelling was observed on the endothelial luminal surface and basal lamina of spinal cord and brainstem microvasculature. In the paralytic phase, a discrete labelling was evident in blood vessels of spinal cord and brainstem associated or not with an inflammatory infiltrate. Conversely, intense immunolabelling was present in cerebral and cerebellar blood vessels, which were still free from inflammatory cuffs. Shortly after clinical recovery minimal labelling was observed in a few blood vessels. Brainstem and spinal cord returned to normal, but numerous inflammatory foci and demyelination were still evident near the ventricle walls, in the cerebral cortex and in the cerebellum. Intense expression of FN in brain vessels ascending from the spinal cord towards the encephalon preceded the appearance of inflammatory cells but faded away after the establishment of the inflammatory cuff. These results indicate an important role for FN in the pathogenesis of CNS inflammatory demyelinating events occurring during EAE.

Intermediate filament (IF) proteins constitute an extremely large multigene family of developmentally and tissue-regulated cytoskeleton proteins abundant in most vertebrate cell types. Astrocyte precursors of the CNS usually express vimentin as the major IF. Astrocyte maturation is followed by a switch between vimentin and glial fibrillary acidic protein (GFAP) expression, with the latter being recognized as an astrocyte maturation marker. Levels of GFAP are regulated under developmental and pathological conditions. Upregulation of GFAP expression is one of the main characteristics of the astrocytic reaction commonly observed after CNS lesion. In this way, studies on GFAP regulation have been shown to be useful to understand not only brain physiology but also neurological disease. Modulators of GFAP expression include several hormones such as thyroid hormone, glucocorticoids and several growth factors such as FGF, CNTF and TGFß, among others. Studies of the GFAP gene have already identified several putative growth factor binding domains in its promoter region. Data obtained from transgenic and knockout mice have provided new insights into IF protein functions. This review highlights the most recent studies on the regulation of IF function by growth factors and hormones.

Melatonin, the pineal hormone produced during the dark phase of the light-dark cycle, modulates neuronal acetylcholine receptors located presynaptically on nerve terminals of the rat vas deferens. Recently we showed the presence of high affinity nicotine-binding sites during the light phase, and low and high affinity binding sites during the dark phase. The appearance of the low affinity binding sites was due to the nocturnal melatonin surge and could be mimicked by exposure to melatonin in vitro. The aim of the present research was to identify the receptor subtypes responsible for the functional response during the light and the dark phase. The rank order of potency of agonists was dimethylphenylpiperazinium (DMPP) = cytisine > nicotine > carbachol and DMPP = nicotine = cytisine > carbachol, during the light and dark phase, respectively, due to an increase in apparent affinity for nicotine. Mecamylamine similarly blocked the DMPP response during the light and the dark phase, while the response to nicotine was more efficiently blocked during the light phase. In contrast, methyllycaconitine inhibited the nicotine-induced response only at 21:00 h. Since a7 nicotinic acetylcholine receptors (nAChRs) have low affinity for nicotine in binding assays, we suggest that a mixed population composed of a3ß4 - plus a7-bearing nAChR subtypes is present at night. This plasticity in receptor subtypes is probably driven by melatonin since nicotine-induced contraction in organs from animals sacrificed at 15:00 h and incubated with melatonin (100 pg/ml, 4 h) is not totally blocked by mecamylamine. Thus melatonin, by acting directly on the short adrenergic neurons that innervate the rat vas deferens, induces the appearance of the low affinity binding site, probably an a7 nAChR subtype.

Intercellular communication may be regulated by the differential expression of subunit gap junction proteins (connexins) which form channels with differing gating and permeability properties. Endothelial cells express three different connexins (connexin37, connexin40, and connexin43) in vivo. To study the differential regulation of expression and synthesis of connexin37 and connexin43, we used cultured bovine aortic endothelial cells which contain these two connexins in vitro. RNA blots demonstrated discordant expression of these two connexins during growth to confluency. RNA blots and immunoblots showed that levels of these connexins were modulated by treatment of cultures with transforming growth factor-ß1. To examine the potential ability of these connexins to form heteromeric channels (containing different connexins within the same hemi-channel), we stably transfected connexin43-containing normal rat kidney (NRK) cells with connexin37 or connexin40. In the transfected cells, both connexin proteins were abundantly produced and localized in identical distributions as detected by immunofluorescence. Double whole-cell patch-clamp studies showed that co-expressing cells exhibited unitary channel conductances and gating characteristics that could not be explained by hemi-channels formed of either connexin alone. These observations suggest that these connexins can readily mix with connexin43 to form heteromeric channels and that the intercellular communication between cells is determined not only by the properties of individual connexins, but also by the interactions of those connexins to form heteromeric channels with novel properties. Furthermore, modulation of levels of the co-expressed connexins during cell proliferation or by cytokines may alter the relative abundance of different heteromeric combinations.
