Schistosoma mansoni eggs are classified, according to morphological characteristics, as follows: viable mature and immature eggs; dead mature and immature eggs, shells and granulomas. The scope of this study was to compare the staining characteristics of different morphological types of eggs in the presence of fluorescent labels and vital dyes, aiming at differentiating live and dead eggs. The eggs were obtained from the intestines of infected mice, and put into saline 0.85%. The fluorescent labels were Hoechst 33258 and Acridine Orange + Ethidium Bromide and vital dyes (Trypan Blue 0.4% and Neutral Red 1%). When labelled with the probe Hoechst 33258, some immature eggs, morphologically considered viable, presented fluorescence (a staining characteristic detected only in dead eggs); mature eggs did not present fluorescence, and the other types of dead eggs, morphologically defined, showed fluorescence. As far as Acridine Orange + Ethidium Bromide are concerned, either the eggs considered to be live, or the dead ones, presented staining with green color, and only the hatched and motionless miracidium was stained with an orange color. Trypan Blue was not able to stain the eggs, considered to be dead but only dead miracidia which had emerged out of the shell. Neutral Red stained both live and dead eggs. Only the fluorescent Hoechst 33258 can be considered a useful tool for differentiation between dead and live eggs.

The aim of the study was to characterize the cell damage mechanisms involved in the pathophysiology of cytotoxicity of polymyxin B in proximal tubular cells (LLC - PK1) and discuss about the nurses interventions to identify at risk patients and consider prevention or treatment of nephrotoxicity acute kidney injury. This is a quantitative experimental in vitro study, in which the cells were exposed to 375μM polymyxin B sulfate concentration. Cell viability was determined by exclusion of fluorescent dyes and morphological method with visualization of apoptotic bodies for fluorescence microscopy. Cells exposed to polymyxin B showed reduced viability, increased number of apoptotic cells and a higher concentration of the enzyme lactate dehydrogenase. The administration of polymyxin B in vitro showed the need for actions to minimize adverse effects such as nephrotoxicity.

The use of Passiflora species for ornamental purposes has been recently developed, but little is known about pollen viability and the potential for crossing different species. The objective of this study was to evaluate the pollen viability of six Passiflora species collected from different physiological stages of development through in vitro germination and histochemical analysis using dyes. The pollen was collected in three stages (pre-anthesis, anthesis and post-anthesis). Three compositions of culture medium were used to evaluate the in vitro germination, and two dyes (2,3,5-triphenyltetrazolium chloride, or TTC, and Lugol's solution) were used for the histochemical analysis. The culture medium containing 0.03% Ca(NO3) 4H2O, 0.02% of Mg(SO4 ).7H2O, 0.01% of KNO3, 0,01% of H3BO3, 15% sucrose, and 0.8% agar, pH 7.0, showed a higher percentage of pollen grains germinated. Anthesis is the best time to collect pollen because it promotes high viability and germination. The Lugol's solution and TTC dye overestimated the viability of pollen, as all accessions showed high viability indices when compared with the results obtained in vitro.

The electrochemistry field has increased in recent years, specially in the search for new sensors to monitor specific analyte in complex samples. In order to improve electrodes, many rhodium compounds have been used as electron mediators for novel sensors development. The most used compounds for this purpose are metal, metal complexes and some organic dyes. Rhodium complexes are known by their good catalytic properties and it could be useful in the sensor field. However, there are only a few reports, on the use of rhodium complexes in sensors and biosensors. A brief review of the electrochemistry of rhodium complexes and some discription of their properties which make those compounds suitable for development of sensor and biosensor.

The initiation step of the light-induced polymerization kinetics of vinyl monomers using dye-sensitized photoinitiators to generate active radicals is discussed. The photoredox processes of basic dyes with amines and sulfinates are described as photochemical systems capable of starting free-radical polymerization of vinyl monomers in homogeneous and microheterogeneous media. Photophysical techniques like laser flash photolysis and time-correlated single photon counting are used to investigate the excited-state kinetics of the dyes.

Adsorption of cationic dyes on clays can be used as a model for the interactions between organic compounds and these minerals. Cationic dyes like methylene blue are used to study these interactions because of the spectroscopic changes observed when their molecules are adsorbed on clay surfaces. Depending on the structure and layer charge of the clay particles several processes may occur, like adsorption of dye monomers and aggregates on the external and internal surfaces of the clay tactoids, migration towards internal surfaces, protonation, etc. Under certain conditions the deaggregation-aggregation of the clay particles are accelerated trapping dye species during these processes. A general scheme is proposed for the processes occurring between clays and dyes in aqueous suspensions, which can be used to explain the behaviour of specific systems.

Phenols are widely used in many areas and commonly found as industrial by-products. A great number of agricultural and industrial activities realise phenolic compounds in the environmental. Waste phenols are produced mainly by the wood-pulp industry and during production of synthetic polymers, drugs, plastics, dyes, pesticides and others. Phenols are also released into the environmental by the degradation of pesticides with phenolic skeleton. The phenols level control is very important for the environmental protection. Amperometric biosensor has shown the feasibility to complement laboratory-based analytical methods for the determination of phenolic compounds, providing alternatives to conventional methods which have many disadvantages. This brief review considers the evolution of an approach to amperometric measurement using the catalytic properties of some enzymes for phenolic compounds monitoring.

The self-assembly technique is a powerful tool to fabricate ultrathin films from organic compounds aiming at technological applications in molecular electronics. This relatively new approach allows molecularly flat films to be obtained on a simple and cheap fashion from various types of material, including polyelectrolytes, conducting polymers, dyes and proteins. The resulting multilayer films may be fabricated according to specific requirements since their structural and physical properties may be controlled at the molecular level. In this review we shall comment upon the evolution of preparation methods for ultrathin films, the process of adsorption and their main properties, as well as some examples of technological applications of layer-by-layer or self-assembled films.

The use of natural dyes to demonstrate principles of paper chromatography is proposed. Extraction of the coloring compounds were performed in order to obtain the aglycone form of the anthocyanins present in the crude extracts. Separations were carried out on chromatographic paper with BAW (butanol/acetic acid/water) as mobile phase and the results compared with literature data. The crude extracts were obtained from Tibouchina granulosa, Rododhendron simsii, Impatiens walleriana flowers which are wildely found in Brazil and Phaseolus vulgaris L. grains skin which is the principal ingredient of the world famous "feijoada". Such species were chosen in order to attract the students attention since they are present in their quotidian, in agreement with the new proposals for Brazilian education.

Chemically synthesized surfactants are widely used for many purposes in almost every sector of modern industry. Surface-active compounds of biological origin (biosurfactants) have been gaining attention in recent years because of some advantages such as biodegradability, low toxicity, diversity of applications and functionality under extreme conditions. Microbial biosurfactants are useful in bioremediation of water and soil, enhanced oil recovery, and in many formulations of petrochemical, chemical, pharmaceutical, food, cosmetic and textile industries. The importance of biosurfactants, their characteristics and industrial applications are discussed.

In this work, the energy transfer by dipole-dipole interaction between cationic dyes in n-alcohols (methanol, ethanol, 1-propanol and 1-butanol) is studied by time resolved and steady state fluorescence measurements. The critical radii of energy transfer were determined by three independent methods; the spectral overlap, fluorescence decay profiles, and relative intensity measurements. In all solvents, R0 values of the dye pairs obtained from spectral overlap were between 40 to 90 Å. Steady state and time resolved fluorescence measurements resulted in values of R0 in the range of 50 - 80 Å, with good correlation of values.

Synthetic dyes were determined by high performance liquid chromatography in 76 samples of regular jelly and diet jelly powders, of several flavors produced by seven different manufacturers. Three sample of each product, from different batches, were analyzed. The same qualitative composition was observed for products of the same flavor from different manufacturers, but the quantitative composition varied markedly from one manufacturer to the other. There were no or few significant differences between batches from a given manufacturer, demonstrating good quality control in the use of these additives by the food industry. None of the samples exceeded the limit stipulated by Brazilian legislation.

Procion Green HE-4BD is a reactive dye currently used in affinity purification, and commonly present as a contaminant in the final biological preparation. An assay method is described to determine trace amounts of the dye in the presence of human serum albumin(HSA) and leakage from agarose as affinity sorbent by cathodic stripping voltammetry. The proposed method is based on the reductive peak at -0.55V in B-R buffer pH 3 (E=0V and t= 240s), obtained when samples of HSA 2% (m/v) containing dye concentrations in sodium hydroxide pH 12 are submitted to a heating time of 330 min at 80 ºC. Linear calibration curves can be obtained for RG19 dye concentrations from 5x10-9 mol L-1 to 8 x10-8 mol L-1. The detection limit (3sigma) is 1x10-9 mol L-1.

A large variety of organic and inorganic compounds may be found in wastewater which can contribute to environmental contamination. Oxidation processes with ozone (O3; O3/UV; O3/H2O2; O3/TiO2; O3/Mn+2) and the use of ozone in the pre- or post-treatment of wastewater combined with biological processes has been extensively studied for the treatment of effluents. The aim of this work was to evaluate the potential of the ozonation process in the treatment of several industrial wastewaters, namely effluents from paper mills, and textile, whey (dairy industry), pharmaceutic sand pesticides plants.

Anthropogenic pollution of groundwater and surface water has become a very serious environmental problem around the world. A wide range of toxic pollutants is recalcitrant to the conventional treatment methods, thus there is much interest in the development of more efficient remediation processes. Degradation of organic pollutants by zero-valent iron is one of the most promising approaches for water treatment, mainly because it is of low cost, easy to obtain and effective. After a general introduction to water pollution and current treatments, this work highlights the advances, applications and future trends of water remediation by zero-valent iron. Special attention is given to degradation of organochloride and nitroaromatic compounds, which are commonly found in textile and paper mill effluents.
