The objective of this work was to evaluate the total and thermotolerant coliform densities in the oyster culture water of Cananeia, SP, Brazil, correlating these densities with environmental variables and tidal variations. Superficial water samples were collected in two tide conditions (spring and neap) from three areas of Cananéia municipality (Mandira, Itapitangui and Cooperostra). The three studied areas showed good conditions for the culture regarding coliform densities. The two tidal conditions differed significantly as to total coliform concentration; however, the same procedure was not performed for thermotolerant coliforms. No correlation was observed between water temperature, pH, and concentrations of total and thermotolerant coliforms. Coliform density was positively correlated with rainfall and negatively correlated with salinity. Spring and neap tides differed significantly as to coliform number. Simple diagnosis of environmental conditions of the crop fields is insufficient to assess water quality of shellfish cultivation. A continuous monitoring program of planted areas is necessary both for the assessment of water quality potential for marine culture and for ensuring safe consumption of seafood, besides constituting an important tool to understand the relationships between contamination and the involved environmental variables.

The objective of this work was to evaluate the protective effect of different forms of insecticide application on the transmission of yellow dwarf disease in barley cultivars, as well as to determine the production costs and the net profit of these managements. The experiments were carried out during 2011 and 2012 growing seasons, using the following managements at main plots: T1, seed treatment with insecticide (ST) + insecticide on shoots at 15-day interval; T2, just ST; T3, insecticide applied on shoots, when aphid control level (CL) was reached; T4, without insecticide; and T5, ST + insecticide on shoots when CL was reached. Different barley cultivars - BRS Cauê, BRS Brau and MN 6021 - were arranged in the subplots. Insecticides lambda cyhalothrin (pyrethroid) and thiamethoxam (neonicotinoid) were used. There were differences on yellow dwarf disease index in both seasons for the different treatments, while damage to grain yield was influenced by year and aphid population. Production costs and net profit were different among treatments. Seed treatment with insecticide is sufficient to reduce the transmission of yellow dwarf disease in years with low aphid population pressure, while in years with larger populations, the application of insecticide on shoots is also required.

El cultivo de la variedad de platanera Gruesa, selección local de Dwarf Cavendish, ha experimentado un importante aumento en los últimos años en las Islas Canarias, tanto al aire libre como bajo invernadero. La eliminación de hojas, tras la floración, es una práctica habitual en los cultivos bajo invernadero. Asimismo es frecuente la pérdida de hojas por el efecto de los vientos en los cultivos al aire libre. El objetivo de este trabajo es evaluar, mediante simulación de pérdida foliar por daños mecánicos, la influencia que tiene la disminución de superficie foliar sobre el llenado y cosecha de la fruta en dicho cultivar. Para ello, cuatro meses antes de la cosecha se efectuaron cinco niveles de defoliación: 0%, 25%, 50%, 75% y 100%. Se valoran dos métodos diferentes de defoliación, eliminación de limbo foliar y tronchado de hojas con posterior corte de éstas. Se analizan y presentan datos morfológicos, fenológicos y productivos, así como valoración de la metodología empleada en este trabajo para la simulación de daños. A partir de un 25% de defoliado, equivalente a 7.5 hojas funcionales por planta, se detectaron diferencias significativas con las plantas testigos.

This article surveys the birth and development of the chemical community in Brazil in the last 50 years. Starting from humble beginnings, a vigorous community developed and rapidly expanded the scope of its activities across the country. Many problems remain unsolved, however, and to these have now been added dismal government policies which threaten to dwarf many of the accomplishments obtained thus far. Brazilian Chemistry is at the threshold of a new age which will be far different from the previous half-century.

Garlic viruses often occur in complex infections in nature. In this study, a garlic virus complex, collected in fields in Brazil, was purified. RT-PCR was performed using specific primers designed from the consensus regions of the coat protein genes of Onion yellow dwarf virus, a garlic strain (OYDV-G) and Leek yellow stripe virus (LYSV). cDNA of Garlic common latent virus (GCLV) was synthesized using oligo-dT and random primers. By these procedures individual garlic virus genomes were isolated and sequenced. The nucleotide sequence analysis associated with serological data reveals the presence of two Potyvirus OYDV-G and LYSV, and GCLV, a Carlavirus, simultaneously infecting garlic plants. Deduced amino acid sequences of the Brazilian isolates were compared with related viruses reported in different geographical regions of the world. The analysis showed closed relations considering the Brazilian isolates of OYDV-G and GCLV, and large divergence considering LYSV isolate. The detection of these virus species was confirmed by specific reactions observed when coat protein genes of the Brazilian isolates were used as probes in dot-blot and Southern blot hybridization assays. In field natural viral re-infection of virus-free garlic was evaluated.

A incidência de doenças causadas por molicutes e por vírus foi avaliada em lavouras de milho (Zea mays) nas Províncias de Tucumán e de Córdoba, na Argentina, em fevereiro de 2000. Na Província de Tucumán verificou-se que 44% das lavouras apresentaram altos níveis de incidência de plantas com sintomas de enfezamentos causados por molicutes (50 a 100%), em altitudes variando de 300 a 2.000 m. A presença de fitoplasma e de espiroplasma foi confirmada em amostras de folhas de plantas com sintomas de enfezamentos, através dos testes de PCR e de "Western blotting". Constatou-se, porém, que a eficiência desses testes para detecção destes patógenos, quando os sintomas apresentados pelas plantas eram muito acentuados, foi da ordem de 70%, e de apenas 30% quando os sintomas eram menos acentuados. Na localidade Jesus Maria, foram encontradas plantas apresentando acentuado nanismo, folhas estreitas e com deformações. Dentre quatro amostras destas plantas, submetidas a testes de PCR, em duas foi detectada a presença de fitoplasma, possivelmente d istinto do "Maize Bushy Stunt Phytoplasma". A cigarrinha Dalbulus maidis, inseto vetor dos molicutes, foi encontrada apenas em Tucumán, estando ausente em Córdoba. O Mal de Rio Cuarto virus foi detectado em seis lavouras em Córdoba, e em três em Tucumán. A cigarrinha Delphacodes kuscheli foi detectada em todas as lavouras em Córdoba, e em apenas três lavouras em Tucumán. O Maize dwarf mosaic virus foi detectado em cerca de 60% das lavouras amostradas nas duas Províncias e o Maize rayado fino virus em apenas uma localidade em Tucumán.

Os vírus pertencentes ao subgrupo do Sugarcane mosaic virus (SCMV, gênero Potyvirus, família Potyviridae) infectam e causam mosaico em diferentes espécies botânicas da subfamília Panicoideae (família Poaceae), porém apenas o SCMV e o Sorghum mosaic virus (SrMV) infectam naturalmente cana-de-açúcar. No Brasil, a espécie SCMV parece ser o único agente causal da doença. Embora a maioria das variedades comerciais de cana-de-açúcar seja considerada resistente ou tolerante ao SCMV, relatos de incidência de mosaico em tais variedades têm ocorrido no campo. Amostras com sintomas, de diversos clones e variedades, foram coletadas em campos experimentais e comerciais de cana-de-açúcar. Dentre as variedades, a RB72-454, uma das mais plantadas no país e considerada resistente à doença, também apresentou plantas com sintomas de mosaico. As amostras foram testadas por DAS-ELISA, com anti-soros policlonais para as espécies SCMV, Johnsongrass mosaic virus (JGMV) e Maize dwarf mosaic virus (MDMV), apresentando resultados negativos. Porém, sintomas de mosaico foram observados em mudas de sorgo "Rio" e "TX2786" quando inoculadas mecanicamente com os isolados, indicando tratar-se de infecção pelo SCMV. RNA total foi extraído das folhas de cana e submetido a RT-PCR com oligonucleotídeos específicos para SCMV e SrMV. Fragmentos específicos de aproximadamente 880 pares de bases foram amplificados com os oligonucleotídeos para o SCMV, confirmando os resultados da inoculação mecânica. Os produtos de PCR foram clonados e seqüenciados. Um dos isolados de SCMV encontrado constitui uma nova estirpe, mais severa, capaz de infectar plantas da variedade RB72-454 e de outras variedades, consideradas tolerantes, no campo.

OBJECTIVE: To standardize a technique for ventilating rat fetuses with Congenital Diaphragmatic Hernia (CDH) using a volume-controlled ventilator.METHODS: Pregnant rats were divided into the following groups: a) control (C); b) exposed to nitrofen with CDH (CDH); and c) exposed to nitrofen without CDH (N-). Fetuses of the three groups were randomly divided into the subgroups ventilated (V) and non-ventilated (N-V). Fetuses were collected on day 21.5 of gestation, weighed and ventilated for 30 minutes using a volume-controlled ventilator. Then the lungs were collected for histological study. We evaluated: body weight (BW), total lung weight (TLW), left lung weight (LLW), ratios TLW / BW and LLW / BW, morphological histology of the airways and causes of failures of ventilation.RESULTS: BW, TLW, LLW, TLW / BW and LLW / BW were higher in C compared with N- (p <0.05) and CDH (p <0.05), but no differences were found between the subgroups V and N-V (p> 0.05). The morphology of the pulmonary airways showed hypoplasia in groups N- and CDH, with no difference between V and N-V (p <0.05). The C and N- groups could be successfully ventilated using a tidal volume of 75 ìl, but the failure of ventilation in the CDH group decreased only when ventilated with 50 ìl.CONCLUSION: Volume ventilation is possible in rats with CDH for a short period and does not alter fetal or lung morphology.

Supporting patients with acute respiratory distress syndrome (ARDS), using a protective mechanical ventilation strategy characterized by low tidal volume and limitation of positive end-expiratory pressure (PEEP) is a standard practice in the intensive care unit. However, these strategies can promote lung de-recruitment, leading to the cyclic closing and reopening of collapsed alveoli and small airways. Recruitment maneuvers (RM) can be used to augment other methods, like positive end-expiratory pressure and positioning, to improve aerated lung volume. Clinical practice varies widely, and the optimal method and patient selection for recruitment maneuvers have not been determined, considerable uncertainty remaining regarding the appropriateness of RM. This review aims to discuss recent findings about the available types of RM, and compare the effectiveness, indications and adverse effects among them, as well as their impact on morbidity and mortality in ARDS patients. Recent developments include experimental and clinical evidence that a stepwise extended recruitment maneuver may cause an improvement in aerated lung volume and decrease the biological impact seen with the traditionally used sustained inflation, with less adverse effects. Prone positioning can reduce mortality in severe ARDS patients and may be an useful adjunct to recruitment maneuvers and advanced ventilatory strategies, such noisy ventilation and BIVENT, which have been useful in providing lung recruitment.

Abstract: The aim of this study was to assess the cardiopulmonary effects, the onset time after the administration of a detomidine/ketamine combination, and the recovery from anesthesia of cougars (Puma concolor) anesthetized with detomidine/ketamine and isoflurane or sevoflurane for abdominal ultrasound imaging. Fourteen animals were randomly allocated into two experimental groups: GISO (n=7) and GSEVO (n=7). Chemical restraint was performed using 0.15mg/kg detomidine combined with 5mg/kg ketamine intramuscularly; anesthesia induction was achieved using 2mg/kg propofol intravenously and maintenance with isoflurane (GISO) or sevoflurane (GSEVO). The following parameters were assessed: heart rate, respiratory rate, systolic and diastolic arterial blood pressure, mean arterial blood pressure, oxyhemoglobin saturation, rectal temperature, central venous pressure, and end-tidal carbon dioxide. The time to sternal recumbency (TSR) and time to standing position (TSP) were also determined. There was not statistically significant difference for the cardiopulmonary variables or TSP whereas TSR was significantly shorter in GSEVO. The time to onset of anesthesia was 11.1±1.2 minutes and 11.3±1.8 minutes for GISO and GSEVO, respectively. The anesthesia of cougars with detomidine/ketamine and isoflurane or sevoflurane was conducted with safety, cardiopulmonary stability, and increased time to sternal recumbency in the GISO group.

To investigate the allelopathic effect of Marsilea minuta against the germination and seedling growths of rice (Oryza sativa) and wheat (Triticum aestivum), germination bioassays were conducted in both Petri dish and soil cultures in laboratory conditions. Rice and wheat seeds were allowed to germinate in a 1, 2, 3, 4, and 5% (w/v) aqueous extract of whole plant and 2, 4, 6, and 8% (w/w) plant residue-incorporated soils of M. minuta. In Petri dish experiments, 5% (w/v) an aqueous extract of M. mimuta showed significantly lower germination percentages (18.8% and 56.3%), root lengths (0.9 and 4.5 cm), shoot lengths (3.3 and 12.4 cm), seedling lengths (4.1 and 25.0 cm), root dry weights (1.4 and 5.6 g), shoot dry weights (1.1 and 9.0 g), seedling biomasses (2.5 and 14.6 g), and seedling vigor indices (77.4 and 957.3) in rice and wheat, respectively. In pot experiments, the M. minuta residue infested soil, with 8% concentration, produced significantly lower germination percentages (25.3 and 37.5%), root lengths (2.7 and 6.1 cm), shoot lengths (6.2 and 16.5 cm), seedling lengths (8.9 and 22.6 cm), root dry weights (2.4 and 5.5 g), shoot dry weights (4.0 and 2.8 g), seedling biomasses (6.4 and 8.3 g), and seedling vigor indices (224.1 and 855.3) in rice and wheat, respectively. The highest phytotoxic action of 5% aqueous whole plant extract of M. minuta against test crops seem to be due to the presence of two potent phenolic compounds, namely p-coumaric acid (2.91 mg L-1) and m-coumaric acid (1.59 mg L-1) as determined by HPLC analysis.

Temporal and spatial variations in species composition and vertical distribution of macroalgal communities growing on mangrove trees were analyzed bimonthly in the Ilha do Cardoso State Park, São Paulo state (25°03'S and 47°55'W), Southeastern Brazil. The macroalgal communities from mangroves of Perequê and Sítio Grande rivers comprised 10 and 18 taxa respectively. Bostrychia radicans (Mont.) Mont. and B. calliptera (Mont.) Mont. were the predominant taxa, present almost throughout the year and in all the sites studied. The species composition of macroalgal communities from both mangroves presented temporal and spatial variations related to environmental factors. The highest number of taxa was observed during colder, drier months, coinciding with the highest means of high water neap and short periods of continuous emersion (April to August). Some mangrove algae such as B. calliptera, Rhizoclonium spp., Caloglossa spp., and Boodleopsis pusilla (Collins) W. Taylor, Joly et Bernatowicz showed a high degree of tolerance to desiccation, being able to tolerate continuous emersion up to six days. The spatial variations in species composition were related to light, as observed in Catenella caespitosa (Withering) L. Irvine, which occurred in well-lit sites. No pattern of vertical zonation was observed, since Rhizoclonium spp., B. radicans, and B. calliptera occur over the entire vertical range. Variations in the range of vertical distribution of macroalgae of Perequê mangrove were mainly related to the variations in the tidal levels (mean high water neap and/or mean high water spring) while those observed in Sítio Grande mangrove were related to salinity variations, except for B. calliptera and Caloglossa spp. related to tidal levels and high irradiance, respectively.

Systemic blood flow (Q) was measured by echodopplercardiography in 5 normal young adult males during apnea, eupnea and tachypnea. Measurements were made in a recumbent posture at 3-min intervals. Tachypnea was attained by doubling the respiratory frequency at eupnea at a constant tidal volume. An open glottis was maintained during apnea at the resting respiratory level. The Q values were positively correlated with the respiratory frequency, decreasing from eupnea to apnea and increasing from eupnea to tachypnea (P<0.05). These data demonstrate that echodopplercardiography, a better qualified tool for this purpose, confirms the positive and progressive effects of ventilation on systemic blood flow, as suggested by previous studies based on diverse technical approaches

Pulmonary dysfunction represents the most important cause of death in patients with paracoccidioidomycosis (PBM). In order to investigate the functional changes of the lungs in the early stages of PBM, a model of benign disease was developed by intratracheal challenge of 12-week old isogenic Wistar rats with 1 x 106 yeast forms of Paracoccidioides brasiliensis. Animals were studied 30 and 60 days after infection, when fully developed granulomas were demonstrable in the lungs. Measurements of airway resistance, lung elastance and tissue hysteresis were made during sinusoidal deformations (100 breaths/min, tidal volume = 2 ml) with direct measurement of alveolar pressure using the alveolar capsule technique. Infection caused a significant increase in hysteresis (infected: 1.69, N = 13; control: 1.13, N = 12, P = 0.024, ANOVA), with no alterations in airway resistance or lung elastance. Histopathological analysis revealed the presence of fully developed granulomas located in the axial compartment of the lung interstitial space. These results suggest that alterations of tissue mechanics represent an early event in experimental PBM

In awake rats a single recurrent larger tidal volume (deep breaths) occurs at regular intervals, followed by oscillations in arterial pressure and heart rate. In the present study we recorded the changes in blood pressure, heart rate and ventilation during the wakefulness-sleep cycle identified by electrocorticographic records in order to determine whether the deep breaths and cardiovascular oscillations were associated with changes in the electrocorticogram. During several episodes of slow-wave sleep (SWS) in 7 rats the deep breaths and oscillations in arterial pressure and heart rate were preceded by SWS desynchronization. The interval between deep breaths during SWS was 71 ± 4 s, the period between initial desynchronization and the generation of deep breaths was 3.98 ± 0.45 s and the duration of SWS desynchronization was 11 ± 0.65 s. Hypotension (-16 ± 1 mmHg) and tachycardia (+15 ± 5 bpm) were observed during deep breaths in the SWS state. These data indicate that the oscillations in arterial pressure and heart rate during SWS are associated with deep breaths, which in turn are preceded by desynchronization of the electrocorticogram in this state of sleep
