No sentido de aquilatar a extração dos macro e micronutrientes, com exceção do Cl e Mo, aliada ao crescimento da planta, amostragens de rainha margarida (Callestephus chinensis) foram executadas aos 0, 18, 34, 46, 59 e 77 dias após o transplante. As plantas foram divididas em raiz, caule, folhas, botões florais, flores analisadas para N, R, K, Ca, Mg, S, B, Cu, Fe, Mn e Zn. Observou-se que o crescimento da rainha margarida é contínuo, acentuando-se após os 59 dias de transplante. O teor porcentual dos nutrientes aos 34-46 dias, na matéria seca, oscilou em torno de 4,09% - 4,40% para N; 0,44% - 0,46% para P; 1,65% - 3,19% para K; 1,01% - 1,10% para Ca; 0,34% - 0,45% para Mg; 0,42% - 0,43% para S. Para os micronutrientes os valores encontrados, na mesma época, foram em ppm: B - 23-36; Cu - 18-20; Fe - 105-150; Mn - 115-135; Zn - 64-111. Uma planta de rainha-margarida aos 77 dias contem: 2.049,9 mg de N; 212,5 mg de P; 2.496,6 mg de K; 915,7 mg de Ca; 356,6 mg de Mg; 159,1 mg de S; 2.140 ug de B; 3.070 ug de Cu; 17.142 ug de Fe; 6.946 ug de Mn; 3.931 ug de Zn.

As leguminosas tropicais constituem atualmente uma fonte de proteína de alta qualidade além de outras qualidades como produção de madeira no caso das arbustivas e como proteção contra erosão nas demais. Sendo a leucena (Leucaena leucooephala (Lam.) de Wit cv. Peru) uma espécie bastante promissora para a pecuária brasileira, foi conduzido um ensaio de campo com a finalidade de se conhecer o hábito alimentar dessa leguminosa. O ensaio foi instalado visando obter dados para análise de crescimento, concentração e extração dos macronutrientes (N, P, K, Ca, Mg e S, Mn e Zn) e matéria seca digestível das folhs "in vivo", em bovino, em função da idade da planta. A partir dos dados obtidos, constatou-se que: a) a produção de matéria seca total é maxima aos 360 dias de idade e o maior incremento na produção de folhas, expresso em matéria seca, se dá dos 240 aos 360 dias; b) as concentrações de nitrogênio e potássio diminuem nas folhas e caules com a idade da planta, ao passo que a de cálcio no caule não é afetada pela idade; c) aos 360 dias, época de produção máxima de matéria secam as folhas contém em 535,46 g/planta: 16,36 g nitrogênio, 0,61 g de fósforo, 10,65 g de potássio, 8,08 g de cálcio, 1,58 g de magnésio, 0,51 g de enxofre; d) aos 360 dias, os caules contém em 1783 g/planta: 15,82 g de nitrogênio, 0,65 g de fósforo, 20,37 g de potássio, 7,01 g de cálcio, 0,44g de magnésio, 1,06 g de enxofre; e) o acúmulo de macronutrientes na planta aos 360 dias obedece a seguinte ordem: N > K > a > g > S > P; f) aos 360 dias, a matéria seca digestível das folhas é 51,05%.

Em uma plantação de cafeeiros (Coffea arabica cv. Catuai) com dois, três, quatro e cinco anos de idade no campo, situada em Latossolo Vermelho Amarelo, fase cerrado no município de Salto, SP, determinou-se o recrutamento de micronutrientes no caule, ramos, folhas e frutos, durante as fases fenológicas de repouso, granaçao e maturação. Concluiu-se que: A maior acumulação de cobre, manganês e zinco ocorre nos meses de julho, janeiro e junho em cafeeiros com cinco anos de idade. A acumulação de boro e ferro varia em função das épocas. Em janeiro e junho o acúmulo de ferro nao mostrou diferenças entre as idades. O cafeeiro aos cinco anos de campo exporta através da colheita, em função do conteúdo total da planta 30% de B, 46% de Cu, 26% de Fe, 14% de Mn e 25% de Zn.

O bagaço de cana-de-açúcar "in natura" (BIN) associado ou não ao bicarbonato de sódio foi testado como substituto do feno de gramínea como fonte de fibra longa para rações de ruminantes balanceadas com altas proporções de bagaço auto-hidrolisado (BAH). A ração básica (I) continha 54% BAH; 10% milho grão; 25% farelo de algodão; 8% feno de gramínea; 0,9% calcáreo; 0,5% uréia; e 1,5% premix mineral, base seca. As rações II e III continham BIN e BIN mais bicarbonato de sódio (1,1%, base seca) respectivamente em substituição ao feno de gramínea da ração I. Foram usados bovinos Nelore machos não castrados e fêmeas (18 de cada sexo) em crescimento com médias iniciais de peso vivo e idade de 199 kg e 11 meses. O delineamento estatístico usado foi um fatorial com 3 rações e dois sexos, com dois animais por parcela. O período de adaptação foi de 15 dias e o experimental de 87 dias. Os dados para GPV (kg/dia); ingestão de MS (% PV); conversão alimentar (kg MS/Kg GPV); e pH fecal foram de: 0,909; 2,79; 7,41; e 6,46 para a ração I; 0,867; 2,65; 7,24; e 6,57 para a ração II; e 1,019; 2,88; 7,03 e 6,73 para a ração III. A ração III foi superior rações I e II para ganho de peso (P < 0,05), e apresentou um pH fecal maior do que o da ração I (P < 0,05). Os machos foram superiores às fêmeas em ganho de peso (1,044 vs 0,820; P < 0,01) e conversão alimentar (6,7vs7,7kg MS/Kg GPV; P < 0,01). Foi observada uma correlação negativa signi ficativa (P < 0,05) entre conversão alimentar e pH fecal (r =-0,50). Os elevados níveis de consumo (2,8% PV), o baixo pH do BAH (2,9 a 3,4), e a aparente baixa atividade de ruminação observados sugerem que o pH, a nível de rume e de trato digestivo inferior, é um fator limitante em dietas com altas proporções de BAH.

This report belongs to the series of works carried out Oswaldo Cruz Ins¬titute, on the treatment of treponematosis with penicillin. The present report deals with investigations performed in order to ascer¬tain the following points: 1) the mininal curative dosis for the initial lesions of yaws; 2) the effect of reduction of the number of injections each day, to verifie the possibility of application of penicillin in the prophylaxis of yaws in rural zones; 3) reduction of the time of treatment by application of high dosis. 1) With dosis of 150 and 100 Oxford units each four hours, clinical reco¬very was obtained after 17 days of treatment. With 50 O.u. during 40 days clinical recovery was not obtained. 2) a) With 3 injections of 400 O.u. each day (6,12 and 18 hoórs clocks) clinical recovery was obtained after 14 to 16 days; b) with 2 injections of 400 O.u. each day (6 and 18 hoors clocks), clinical recovery was obtained after 16 to 23 days; c) with 1 injection of 1.600 and 3.200 each day, clinical recovery was obtained after 30 and 20 days. 3) With dosis of 33.3 and 46.7 O.u. by each kilo of weight each four hours, during 15 days, clinical recovery was obtained more or less in 25 days. The same result was obtained with the dosis of 61.5 and 166.7 O.u. by each kilo of weight, each four hours, during 4 days. But with 100.000 O.u. in fine dosis of 20.000 in a day ou by, clinical recovery was not obtained.

Hyla claresignata Lutz &amp; Lutz, 1939, is a large species apparently not closely allied to the other known Brazilian hylas. It is characterized by the very small tympanum; the head is short and the snout rounded; the legs are long, the hands and feet unusually large, the latter extensively webbbed. The specific name is derived from the insular, irregular, or roughly triangular, dark spots, with a light halo, found mostly in the dorso-lateral region and on the legs. It belongs to the rain-forest fauna of the Marítime Range. The adult is a bromeliad-dweller and the tadpole rhyacophilous. DESCRIPTION. Vomerine teeth in two separate, oblique, groups, behind the large choanae, parallel to the posterior half of their inner border. Tongue entire, short, very broad and hardly free behind. Snout short, rounded, with distinct canthus rostralis and gradually sloping loreal region. Eye very large and prominent, its horizontal diameter almost equal to the distance between its anterior corner and the tip of the snout. Tympanum very small, less than one third of the diameter of the eye, but distinct, partly covered by a short, heavy ridge. Lateral fingers less than one third webbed; fourth finger slightly longer than the second, just reaching the base of the disk of the third; subarticular tubercles well developed; an angular pollex rudiment, more noticeable in the males. Toes almost completely webbed, the edge of the web inserted at the base of the disk on the third and the fifth; an inner metatarsal tubercle. Skin smooth above, granular beneath, on the throat minutely so. No dermal appendage on the hell. Habit robust, head broader than long, body rather heavy, slightly narrowed in the postaxillary region. Legs long, the tibiotarsal articulation reaching beyond the tip of the snout when adpressed. Type (female): 61 mm. (Fig. 1.) DIAGNOSIS of TADPOLE (by G. Orton). "A large specialized, mountain-stream tadpole, with wide head an elongated, flattened snout, greatly enlarged lips and high tooth formula. Eyes dorsal. Spiracle sinistral, projecting, situated far back on side. Anus dextral. Tooth formula 8/12 to 9/14 in fully grown larvae. Tail with a prominent, vertical dark band across musculature and fins; a second concentration of dark pigment near tip of tail, may or may not form a similar but narrower band. Gastro-intestinal parasitism of 24 buffalo cows before parturition, and post-parturition, their infection and that of their respective calves during the following 30 weeks were studied. Willis, Hoffmann and whenever possible, the modified Gordon &amp; Whitlock techniques were used for fecal examinations. Toxocara vitulorum eggs were the earliest forms encountered in calves feces, as follows: during the 1st week after birth, 58.33% of the calves were positive, and in the 4th week, 100% of these animals were positive. Eggs of Strongyloides sp were in the 1st week after birth in two of the calves and in the 5th week, all for them were positive. The next parasites to appear were the Coccidia of which oocysts were detected in the feces of two calves in the 2nd week after birth, and 58.33% of the calves were positive for these in the 3rd week, and in the 6th week, all calves shed oocysts in their feces. On the other hand, eggs of Strongylids were the last forms to appear in calves feces. However, despite their sporadic appearance in the feces, eggs of these parasites were observed continuously from the 11th week onwards, and at this point, the percentage of positive samples began to increase to reach its peak. Relatively to adult animals, eggs of T. vitulorum were observed in the feces of 11 cows, one or twice at most; eggs of Strongyloides sp were seen only once in the feces of four buffalo cows and eggs of Strongylids in 21 out of 24 cows. Oocysts of Coccidia were observed in 16 cows. Mechanisms of infestation of calves with these parasites are discussed.

Strategies to construct the physical map of the Trypanosoma cruzi nuclear genome have to capitalize on three main advantages of the parasite genome, namely (a) its small size, (b) the fact that all chromosomes can be defined, and many of them can be isolated by pulse field gel electrophoresis, and (c) the fact that simple Southern blots of electrophoretic karyotypes can be used to map sequence tagged sites and expressed sequence tags to chromosomal bands. A major drawback to cope with is the complexity of T. cruzi genetics, that hinders the construction of a comprehensive genetic map. As a first step towards physical mapping, we report the construction and partial characterization of a T. cruzi CL-Brener genomic library in yeast artificial chromosomes (YACs) that consists of 2,770 individual YACs with a mean insert size of 365 kb encompassing around 10 genomic equivalents. Two libraries in bacterial artificial chromosomes (BACs) have been constructed, BACI and BACII. Both libraries represent about three genome equivalents. A third BAC library (BAC III) is being constructed. YACs and BACs are invaluable tools for physical mapping. More generally, they have to be considered as a common resource for research in Chagas disease

Analysis of zymograms with SDS-polyacrilamide gel electrophoresis containing gelatin as substrate, and performed on samples of haemolymph or fat body taken from Rhodnius prolixus inoculated or not with Enterobacter cloacae, demonstrated distinct patterns of protease activities: (i) in the haemolymph two proteases were induced in insects inoculated with bacteria; (ii) two proteases were detected in the fat bodies derived from non-inoculated controls or insect inoculated with sterile culture medium; (iii) haemolymph and fat body had both the same apparent molecular weights proteases (46 and 56 kDa); and (iv) these enzymes were characterized as metallo-proteases. The association of these enzymes in Rhodnius infected with bacteria was discussed.

The Palha district, municipality of Bananal, State of São Paulo, Brazil, had 10.3% cases of Schistosoma mansoni diagnosed from 1994 to 2000 by coproscopy: about three times the municipality average. The immunofluorescent antibody test was used to assess gut-associated IgM antibody titers of samples from 452 inhabitants. It disclosed 129 (28.5%) positive cases. Subjects were classified according to age, sex, birthplace, and period of residence. Titers varied from 8 to 4,096 (geometric mean: 170.2). Seropositives were aged from 6 to 69 years (average: 24.5), 75% of them aged 34 or less, predominantly males (78 or 60.5%). Of all subjects, 65.7% were born and had been living in Bananal since; 24.2% came from neighboring municipalities and are residing in Bananal from two months to 89 years (average: 22.7 years). Further Kato-Katz coproscopy from 97 seropositives (geometric mean titer, 619) revealed S. mansoni eggs in 11 subjects (11.3%). Serology was deemed useful in screening subjects to be further investigated by coproscopy, considering that blood collection had better acceptance than supplying fecal samples. Higher than average serological titers may indicate new cases in endemic areas. Longitudinal studies associated with epidemiological investigation, including titer evolution are advised, as isolated data are difficult to interpret.

This study estimated the prevalence and distribution of human papillomavirus (HPV) types among women with cervical intraepithelial neoplasia (CIN) grade III and invasive cervical cancer from Goiás (Brazil Central Region). Seventy-four cases were analyzed and consisted of 18 CIN III, 48 squamous cell carcinomas, 4 adenocarcinomas, 1 adenosquamous carcinoma and 3 undifferentiated carcinomas. HPV-DNA sequences were examined in formalin-fixed and paraffin-embedded tissues using primers from L1 region GP5+/GP6+. Polymerase chain reaction products were typed with dot blot hybridization using probes for HPV 16, 18, 31, 33, 45, 54, 6/11, 42/43/44, 51/52, 56/58. The prevalence of HPV was estimated to be 76% (56/74). HPV 16 was the most frequently found type, followed by HPV 33, 18 and 31. The prevalence of untyped HPV was 6%; 79% percent of the squamous cell carcinoma cases and 61% percent of the CIN III were positive for HPV and the prevalence rate of HPV types was the same for the total number of cases. According to other studies, HPV type 16 is the most prevalent virus in all Brazilian regions, but there is variation regarding to other types. Type 18 is the second most prevalent HPV in North, Southeast and South Brazil regions and types 31 and 33 are the second most prevalent HPV in Northeast and Central Brazil, respectively.

We assessed the distribution of Trypanosoma cruzi infection in peridomestic triatomines collected manually at a district-wide scale in rural villages around Olta, Western Argentina, and typed the isolated strains according to their pathogenicity to laboratory mice. Of 1623 triatomines examined, only 14 (0.9%) were infected with T. cruzi based on microscopical examination of feces. The prevalence of T. cruzi infection was 0.8% in Triatoma infestans, 2.3% in T. guasayana, and nil in T. garciabesi, T. platensis, and T. eratyrusiformis. Local transmission occurred in kitchens, store-rooms and goat corrals or nearby, though at very low levels. T. cruzi was detected by at least one parasitological method in 11 (79%) of 14 microscope-positive bugs. Hemoculture was the most sensitive method (67%) followed by culture of organ homogenates, histopathology or xenodiagnosis of inoculated suckling mice (55-58%), and culture of microscope-positive bug feces (46%). The evidence suggests that most of the isolated T. cruzi strains would be myotropic type III. Our study establishes for the first time that peridomestic, microscope-positive T. guasayana nymphs were actually infected with T. cruzi, and may be implicated as a putative secondary vector of T. cruzi in domestic or peridomestic sites.

Susceptibility and compatibility experiments were carried out with 700 Biomphalaria tenagophila from the Paraná River basin exposed to infection with Schistosoma mansoni. Individual infection was performed with 10 miracidia of SJ2 strain from the Paraiba valley (Brazil) originally infective to B. tenagophila. These snails were laboratory-breed progeny of B. tenagophila collected from six localities of Argentina and one from Paraguay. From Argentina: Rincón de Vences (7%) and Posadas (11%) became infected with S. mansoni and the calculation of Frandsen's index (TCP/100) shows that they were Class II poorly compatible. Those snails from Goya (22%), Maloyas (5%), and Berón de Astrada (3%) were Class III compatible to the S. mansoni. None of the 100 snails exposed from Caá-Catí became infected (Class 0 incompatible). Tested samples from Paraguay (Encarnación) were infected (20%) and compatible (Class III). It was also studied the persistence of the infection in 244 snails of the first generation (F1) of those that were susceptible from three places. It was demonstrated an increment of the susceptibility in the F1 from Maloyas (chi2 = 27.22; p = 0.0001) and Posadas (chi2 = 4.24; p = 0.04). The results point out the possibility that schistosomiasis might be able to spread into the Paraná River basin where B. tenagophila exists.

The presence of enterotoxigenic Bacteroides fragilis and nontoxigenic B. fragilis (NTBF) among 109 strains isolated from 1980-2008 in Brazil were investigated by PCR. One strain, representing 0.9% of the total analyzed strains, harbored the bft gene which was identified as bft-1 isoform based on PCR-RFLP and sequencing. Forty-nine strains (44.9%) exhibited the NTBF pattern III which possesses the flanking region required for pathogenicity island acquisition in which the bftgene is codified. These data reinforce the potential of B. fragilis as an emerging enteropathogen in our country.

Malaria diagnoses has traditionally been made using thick blood smears, but more sensitive and faster techniques are required to process large numbers of samples in clinical and epidemiological studies and in blood donor screening. Here, we evaluated molecular and serological tools to build a screening platform for pooled samples aimed at reducing both the time and the cost of these diagnoses. Positive and negative samples were analysed in individual and pooled experiments using real-time polymerase chain reaction (PCR), nested PCR and an immunochromatographic test. For the individual tests, 46/49 samples were positive by real-time PCR, 46/49 were positive by nested PCR and 32/46 were positive by immunochromatographic test. For the assays performed using pooled samples, 13/15 samples were positive by real-time PCR and nested PCR and 11/15 were positive by immunochromatographic test. These molecular methods demonstrated sensitivity and specificity for both the individual and pooled samples. Due to the advantages of the real-time PCR, such as the fast processing and the closed system, this method should be indicated as the first choice for use in large-scale diagnosis and the nested PCR should be used for species differentiation. However, additional field isolates should be tested to confirm the results achieved using cultured parasites and the serological test should only be adopted as a complementary method for malaria diagnosis.
