The immune response expressed by IgG antibodies in BALB/c mice experimentally infected with Toxocara canis, was studied with the aim of verifying the possible in vivo cross-reactivity between antigens of T. canis and other parasites (Ascaris suum, Taenia crassiceps, Schistosoma mansoni, Strongyloides venezuelensis and Toxoplasma gondii). Experiments included three groups of mice: one infected only by T. canis, another with one of the other species of parasites and a third concomitantly infected with T. canis and the other species in question. Animals were bled by orbital plexus at 23, 38 and 70 days post infection (p.i.). Sera were analyzed for anti-Toxocara antibodies by ELISA and Immunoblotting, using excretion-secretion antigens (ES), obtained from culture of third-stage larvae of T. canis. For all experiments a control group comprised by ten non-infected mice was used. Only in the case of A. suum infection, in these experimental conditions, the occurrence of cross-reactivity with T. canis was observed. However, in the case of co-infection of T. canis - S. mansoni, T. canis - S. venezuelensis and T. canis - T. crassiceps the production of anti-Toxocara antibodies was found at levels significantly lower than those found in mice infected with T. canis only. Co-infection with S. mansoni or S. venezuelensis showed lower mortality rates compared to what occurred in the animals with single infections. Results obtained in mice infected with T. canis and T. gondii showed significant differences between the mean levels of the optical densities of animals infected with T. canis and concomitantly infected with the protozoan only in the 23rd day p.i.

We have studied the cardiac chronotropic responses to the Valsalva maneuver and to dynamic exercise of twenty chronic chagasic patients with normal left ventricular function and no segmental wall abnormalities by two-dimensional echocardiogram. The absolute increase in heart rate of the patients (Δ = 21.5 ± 10 bpm, M±SD) during the maneuver was significantly diminished when compared to controls (Δ = 31.30 ± 70, M±SD, p = 0.03). The minimum heart rate (58.24 ± 8.90 vs. 62.80 ± 10, p = 0.68) and the absolute decrease in heart rate at the end of the maneuver (Δ = 38.30 ± 13 vs. Δ = 31.47 ± 17, p = 0.10) were not different from controls. The initial heart rate acceleration during dynamic exercise (Δ = 12 ± 7.55 vs. Δ = 19 ± 7.27, M±SD, p = 0.01) was also diminished, but the heart rate recovery during the first ten seconds was more prominent in the sero-positive patients (Median: 14, Interquartile range: (9.75-17.50 vs. 5(0-8.75, p = 0.001). The serum levels of muscarinic cardiac auto-antibodies were significantly higher in the chagasic patients (Median: 34.58, Interquartile Range: 17-46.5, Optical Density) than in controls (Median: 0, Interquartile Range: 0-22.25, p = 0.001) and correlated significantly and directly (r = 0.68, p = 0.002) with early heart rate recovery during dynamic exercise. The results of this investigation indirectly suggest that, the cardiac muscarinic auto-antibodies may have positive agonist effects on parasympathetic heart rate control of chagasic patients.

Studies of immune responses as they occur in patients with schistosomiasis appear to progress relative to corrent technological advances, and to advance despite the understandable inability to pursue in vivo manipulations in this host/parasite system. Emphasis is most often placed on making immunological comparisons between such patient groups as reinfected/non-reinfected, intestinals/hepatosplenic, high/low intensities of infection, infected/uninfected within endemic areas, and those born of infected/uninfected mothers. Based on these types of comparisons, reasonable conjectures can be made regarding the immunological occurrences during this chronic exposure condition. Some consideration is now being given to the immune mechanisms of some of the observations made, and while some of these must then be carried back to experimental models for further manipulation-based analysis, new technological developments continue to assist in the field/bench ability to ask questions that might assist our understanding to a point where this knowledge can be applied to shaping developmental approaches to vaccine development and the goal of alleviating morbidity.

Artemisinin-based combination therapy-resistant malaria is rare in Sub-Saharan Africa. The World Health Organization identifies monitoring and surveillance using day-3 parasitaemia post-treatment as the standard test for identifying suspected artemisinin resistance. We report three cases of early treatment failure due to possible artemisinin-based combination therapy-resistant Plasmodium falciparum malaria. All cases showed adequate clinical and parasitological responses to quinine. This study reveals a need to re-evaluate the quality and efficacy of artemisinin-based combination therapy agents in Nigeria and Sub-Saharan Africa.

Introduction Schistosomiasis is endemic in 76 countries and territories. Several studies have found an inverse correlation between parasitic disease and the development of allergies. The purpose of the present study was to determine whether infection with Schistosoma mansoni in subjects with a low parasite load is protective against allergy. The final sample consisted of 39 S. mansoni-positive and 52 S. mansoni-negative residents of a small community in northeastern Brazil. Methods All subjects were submitted to the Kato-Katz test, anti-S. mansoni IgG measurement, the prick test for aeroallergens, eosinophil counts and serum IgE measurement. Results Subjects who reacted to one or more antigens in the prick test were considered allergic. Only 7 S. mansoni-positive subjects (17.9%) reacted to one or more antigens, whereas 20 S. mansoni-negative subjects (38.5%) tested positive for allergy. Conclusions Our findings suggest that, in areas of low endemicity, infection with S. mansoni significantly reduces the risk of the development of allergy in subjects with a low parasite load.

Objetivou-se neste trabalho avaliar a dinâmica florestal, sobretudo da regeneração natural, em um fragmento de floresta tropical primária, entre 1998 c 1999, em Peixe-Boi (PA). Foram demarcadas três parcelas permanentes (1 ha cada) onde todos os indivíduos com DAP ≥ 10 cm foram registrados; os indivíduos com 10cm ≥ DAP ≥ 5cm foram amostrados em 6.000 m2, aqueles entre 5cm ≥ DAP ≥ 2cm em 2.400 m2 e com DAP ≤ 2cm em 240 m2. Foram estimados 143.000 indivíduos, desde plântulas até árvores pertencentes a 337 espécies e 76 famílias. Mimosaceae foi a família de maior riqueza (44 espécies); 14 famílias ocorreram com uma única espécie sendo que metade delas apresentaram também um único indivíduo. Independentemente da classe diamétrica verificou-se o egresso de 56 espécies versus o ingresso de 68, gerando um ganho líquido de 12 espécies. A dinâmica da composição e da abundância da regeneração natural foi muito intensa. Observou-se a saída de uma família face ao ingresso de outras 14, aumentando cm quase 30% o número de espécies. A maior mortalidade foi verificada em Bauhinia cf. rutilans e Mabea aff. speciosa (300 e 21 indivíduos). Rinorea negleta e Leçythis idatimon recrutaram 171 e 89 espécimes. A razão recrutamento/ mortalidade foi, em quaisquer das classes diamétricas, sempre superior a unidade. O estoque de mudas para se obter uma árvore, uma arvoreta e uma vara foi, respectivamente de 297, 160 e 48 mudas. O número de espécies e a abundância aumentaram no período, assim como a área basal e a biomassa.

Foi encontrada uma riqueza de 15 espécies de pseudoscorpiões, de 12 gêneros e 5 famílias (Chthoniidae, Geogarypidae, Olpiidae, Atemnidae e Chernetidae), habitando diferentes plantas da vegetação do sub-bosque, em floresta primária de terra firme, no alto rio Urucu, Coari, Amazonas, no período de 1991 a 1996. As plantas foram examinadas pelo método de "bateção". Apolpium aff. vastum foi à espécie mais freqüente e abundante sobre as plantas. Dentre os tipos de plantas avaliados, as maiores diversidades de espécies de pseudoscorpiões foram registradas nas pequenas palmeiras, tanto com fronde junto ao chão, como elevada acima do chão. A análise da composição das espécies que ocorreram sobre os diferentes tipos de plantas avaliadas foi realizada pelo "modo-Q", tendo como base a matriz de coeficientes de similaridade de "Jaccard", o que demonstrou maior similaridade entre a fauna das palmeiras e outras plantas que acumulam detritos acima do chão, entre as bromélias e entre aráceas de chão e outras plantas que acumulam detritos junto ao chão.

Os ecossistemas amazônicos caracterizam-se pela alta diversidade de espécies. Foram realizadas as análises florísticas e fitossociológicas de 0,05 ha de um sub-bosque na Amazônia Central (60º12'40" W e 2º35'45" S), adotando-se o critério de distribuição em classes de altura: C1 (altura < 0,5 m), C2 (0,5 < altura < 1,5 m), C3 (1,5 < altura < 3,0 m) e C4 (altura > 3 m e CAP < 0,3 m). Foram levantados 2.434 indivíduos, pertencentes a 67 famílias, 163 gêneros e 355 espécies. Fabaceae (27), Mimosaceae (22), Lauraceae (21), Caesalpiniaceae e Rubiaceae (18) constituem as cinco famílias mais ricas em espécies. Quanto ao número de indivíduos, as famílias Marantaceae (209), Chrysobalanaceae (198), Mimosaceae (191), Burseraceae (175), Annonaceae (172) e Arecaceae (137) foram as mais representativas. No que tange às categorias de altura, os dados mostraram maior número de indivíduos, diversidade e grau de similaridade florística entre as classes 1 e 2. Licania caudata Prance, Duguetia flagellaris Huber, Monotagma tuberosum Hagberg, Protium apiculatum Swart e Pariana cf. campestris Aubl. foram as espécies de maior participação nos parâmetros verticais da regeneração natural e posição sociológica. O padrão agregado de distribuição espacial foi o predominante para as 30 espécies de maior participação na estrutura vertical da fitocenose.

The role of vitamin C on physiological responses of matrinxã (Brycon amazonicus) submitted to air exposure was analyzed. Nine hundred fish (70.15 g) were distributed in fifteen 500 l boxes (60 fish.box-1) and fed five rations (treatments): Control (no vitamin C); T100 (100 mg); T200 (200 mg); T400 (400 mg) and T800 (800 mg of vitamin C kg.ration-1). Each ration was offered to fish of three boxes during 60 days before the stress challenge that consisted of exposing fish to air for two minutes. Samplings were carried out for 5, 15, 30 and 60 minutes after the air exposure. Blood was collected for glucose, cortisol, total protein, sodium, chloride, hematocrit, hemoglobin determination, and white and red cell count. Liver was removed for hepatosomatic index (HSI) calculation and glycogen determination. Vitamin C did not affect the levels of cortisol, chloride, total protein, hemoglobin, leukocytes, hepatic glycogen or HSI in air exposed fish. Blood glucose levels elevation observed 60 minutes after the challenge did not depend on the levels of vitamin C, nor did the drop in serum sodium levels verified 60 minutes after stressor. In general, hematocrit did not change by effect of vitamin C but it was lower at 15 and 30 minutes after the challenge. The number of erythrocytes decreased in fish after 5 minute sampling in all treatments, especially at 30 and 60 minutes. The air exposure evoked alterations in stress indicators of matrinxã, and the vitamin C did not alter the responses.

The addition of salt to the water has been used to mitigate stress and improve survival in fishes. This study investigated the effects of sodium chloride (0.0, 1.0, 3.0 and 6.0 g/l) on levels of plasma cortisol, glucose, tryacilglycerol, total protein, hematocrit, hemoglobin, erythrocyte number, liver glycogen and lipid, and muscle lipid in adult matrinxã(Brycon amazonicum) after a 4-h transport and during a 96-h recovery period. Fish were sampled before and after transport, and 24 and 96 h of the recovery period. Plasma cortisol was higher than initial condition immediately after transportation, except in fish transported in 3.0 and 6.0 g/l of salt. A similar pattern was observed for blood glucose but fish transported in water with 0.0, 1.0 and 3.0 g/l of salt needed more than 24 h to return to the initial condition. Liver glycogen was lower after transport in fish not exposed to salt. Hemoglobin, erythrocyte number, total plasma protein and liver lipid did not change during the experiment but hematocrit was lower after transport in all treatments and returned to pre-transport values in 24 h. Reductions of muscle lipid and plasma tryacilglycerol were observed during the recovery period in fish from all treatments. The results show that 6.0 g/l NaCl added to the transport water reduce the stress responses and a 96-h recovery period is needed if no salt is used to mitigate the stress.

Pirarucu (Arapaima gigas) is an obligatory air-breathing fish from the Amazon basin. Previous study showed that pirarucu juveniles present a latency period in their response to moderate stress (transportation). Therefore the objective of this study was to verify the effects of a prolonged air exposure stress in lactate, glucose, cortisol, haematocrit, haemoglobin, and liver glycogen in pirarucu. Thirty-six fish were handled by netting and subjected to air exposure for 75-min. Six fish were sampled before handling and at 0, 6, 24, 48, and 96h after handling. Fish cortisol, lactate and haematocrit rose after handling, returning to previous unstressed values on the following sampling (6h after handling). Glucose increased significantly after handling and that was maintained for 24 h. There were no changes in haemoglobin and liver glycogen as a consequence of handling. The results demonstrate a quick response when exposed to an acute stressor and a fast recovery, suggesting that pirarucu does not use their glycogen reserves during an acute stress. The results suggest that pirarucu exhibit physiological stress responses to handling similar in magnitude to those previously documented for many teleostean fishes, including salmonids.

Este estudo compara a resposta de uma comunidade aves à exploração madeireira de baixo impacto através da distribuição das espécies em floresta controle e manejada ao longo de cinco anos com amostragem pré e pós-exploração. O procedimento de ordenação mostrou que a similaridade da comunidade de aves após a exploração madeireira foi menor que a similaridade entre as amostras do período de pré-exploração. Além disso, a ordem das abundâncias das 43 espécies mais comuns foi alterada entre o período anterior à exploração e 3 a 4 anos após a exploração na floresta manejada. A exploração afetou as taxas de captura de 20 espécies, diretamente ou através de uma interação com o tempo. Os insetívoros terrícolas e os insetívoros que seguem bandos mistos foram as guildas mais afetadas pela exploração madeireira. Ambas as guildas mostraram declínio, correlacionado com o tempo, em suas abundâncias em floresta manejada. Insetívoros seguidores de formigas e insetívoros arborícolas não apresentaram diferenças entre a floresta manejada e controle, apesar de algumas espécies dessas guildas terem apresentado efeitos relacionados ao manejo florestal, diretamente ou através de uma interação entre o tempo e o manejo. Frugívoros também não apresentaram efeitos significativos relacionados ao manejo florestal. Nectarívoros aumentaram tanto na floresta explorada como na floresta controle. Efeitos temporais de aumento ou declínio observados em guildas e em 12 espécies ocorreram tanto na floresta controle como na floresta explorada. Este resultado sugere que mudanças temporais na área explorada podem estar relacionadas à sucessão enquanto que mudanças temporais na floresta controle podem estar relacionados à exploração com intensidade de 40m³/ha de parcelas adjacentes.

The present work evaluated several aspects of the generalized stress response [endocrine (cortisol), metabolic (glucose), hematologic (hematocrit and hemoglobin) and cellular (HSP70)] in the Amazonian warm-water fish matrinxã (Brycon amazonicus ) subjected to an acute cold shock. This species farming has been done in South America, and growth and feed conversion rates have been interesting. However, in subtropical areas of Brazil, where the water temperature can rapidly change, high rates of matrinxã mortality have been associated with abrupt decrease in the water temperature. Thus, we subjected matrinxã to a sudden cold shock by transferring the fish directly to tanks in which the water temperature was 10ºC below the initial conditions (cold shock from 28ºC to 18ºC). After 1h the fish were returned to the original tanks (28ºC). The handling associated with tank transfer was also imposed on control groups (not exposed to cold shock). While exposure to cold shock did not alter the measured physiological conditions within 1h, fish returned to the ambient condition (water at 28º C) significantly increased plasma cortisol and glucose levels. Exposure to cold shock and return to the warm water did not affect HSP70 levels. The increased plasma cortisol and glucose levels after returning the fish to warm water suggest that matrinxã requires cortisol and glucose for adaptation to increased temperature.

Na Amazônia, as taxas de desmatamento crescem desde 1991 e as previsões não são otimistas quanto à desaceleração desse processo. A devastação da floresta é acompanhada de uma expansão de florestas secundárias (FS) que se estabelecem nas áreas abandonadas. A tendência é um aumento de florestas secundárias, resultando num mosaico de floresta contínua e fragmentos separados por uma matriz de FS. Nesse cenário, autores acreditam que a Amazônia pode passar por um processo massivo de extinção de espécies. Por outro lado, a previsão de um processo massivo de extinção pode ser equivocada, pois muitas espécies florestais poderiam sobreviver nas florestas secundárias. Para avaliar o valor das florestas secundárias para espécies florestais amostramos por oito meses com redes de neblina uma capoeira (FS) em regeneração e uma floresta primária (FP) de uma paisagem fragmentada. Algumas espécies não foram capturadas na capoeira e aparentemente evitam esse tipo de hábitat. No entanto, a maioria das espécies do grupo focal não apresentou diferença na sobrevivência aparente entre os ambientes, o que nos indica que estão habitando a capoeira e a floresta primária da mesma forma. Na realidade amazônica, onde grande parte da matriz é composta por floresta secundária, a matriz tem valor para conservação e deve ser analisada como um elemento dinâmico que não apenas permite a movimentação de indivíduos, mas também serve de hábitat para muitas espécies de floresta primária. Mas ressaltamos que é fundamental a preservação de áreas de floresta primária que servirão de fonte às florestas secundárias adjacentes.

ABSTRACTIn fish farmings, diseases can be reduced by using immunostimulants. The aim of this study was to evaluate the immunostimulant potential of Mentha piperita in tambaqui fed with 0, 0.5, 1.0 and 1.5% of oil per kg of commercial fish feed. The fish were inoculated with Aeromonas hydrophila to challenge them. Hematological and biochemical parameters were determined after 30 days of feeding and seven days after the challenge. There was no mortality and M. piperita oil did not influence fish production parameters. However, blood hemoglobin concentration (Hb) increased in the fish fed with 0.5 and 1.5% of oil per kg of diet; albumin increased in those fed with 1.0%; cholesterol increased in all groups with oil; and triglycerides increased in those fed with 0.5%. After the bacterial challenge, the fish showed decreases in Hb when fed with diet enriched with 1.5% oil per kg of diet, in mean corpuscular volume with 1.0% and in mean corpuscular hemoglobin concentration with 0 and 1.5%. Protein levels increased in groups with 0 and 1.5% of oil and albumin when fed with 0 and 1.0%; cholesterol levels increased in the control group; and high levels of triglycerides were observed in the groups with 0, 0.5 and 1.5%. Thus, M. piperita essential oil promoted hematological alterations in tambaqui and can be recommended in diets containing up to 1.0% per kg, because of the minimal physiological modifications caused. However, additional studies are necessary to obtain more information regarding to the physiological effects of this immunostimulant.
