Despite the considerable environmental importance of mercury (Hg), given its high toxicity and ability to contaminate large areas via atmospheric deposition, little is known about its activity in soils, especially tropical soils, in comparison with other heavy metals. This lack of information about Hg arises because analytical methods for determination of Hg are more laborious and expensive compared to methods for other heavy metals. The situation is even more precarious regarding speciation of Hg in soils since sequential extraction methods are also inefficient for this metal. The aim of this paper is to present a technique of thermal desorption associated with atomic absorption spectrometry, TDAAS, as an efficient tool for quantitative determination of Hg in soils. The method consists of the release of Hg by heating, followed by its quantification by atomic absorption spectrometry. It was developed by constructing calibration curves in different soil samples based on increasing volumes of standard Hg2+ solutions. Performance, accuracy, precision, and quantification and detection limit parameters were evaluated. No matrix interference was detected. Certified reference samples and comparison with a Direct Mercury Analyzer, DMA (another highly recognized technique), were used in validation of the method, which proved to be accurate and precise.

Chromosome studies were performed in V. champinii, V. cinerea, V. girdiana, V. labrusca, V. rotundifolia, V. rupestris and V. vinifera with the purpose of species characterization using chromosome morphometric data and NOR banding. A median ideogram was obtained for each species. The karyotype formula obtained varied from 7m + 12sm to 9m + 11sm. The species showed moderate chromosome asymmetry values according to TF% form, Stebbins, Romero Zarco and Paszko indices. V. champinii and V. girdiana were apart from the other species by CVcl and CVci graphic representation and also formed a group apart in the dendrogram based on Euclidian distances. The chromosome pair number 3 harbors the secondary constriction and a satellite segment in all species analyzed with Giemsa staining and it may be the same observed after NOR banding technique. It seems that the process of speciation in the North American Euvitis species studied involved some discrete changes in chromosome morphometry which have been reflected in the asymmetry index.

The main obstacle to the use of compost from urban waste in agriculture is the presence of heavy metals. Once in the soil, their effect is accumulative and they may contaminate crops and water. The present study reports the evaluation of the chemical distributions of Cu, Pb, Mn and Zn in three different sized fractions (unsieved, < 1,18mm and > 1,18mm) of compost, by means of a sequencial extraction procedure and a chemometric analysis of the total content of all metals in each fraction. The pattern recognition methods showed significant differences in total heavy metal contents for the different fractions. The finest one was the most contaminated. Meanwhile, this fraction presented lower amounts of metals in avaliable forms. This behavior can be attributed to the presence of metal particles in their elemental states in this fraction.

In the selective reduction procedure proposed by Magos SnCl2 was used as reductant for inorganic mercury while total mercury was determined after reduction with a mixture of SnCl2 and CdCl2. The difference between total mercury and inorganic mercury determines the content of organic mercury. The procedure of the present work differs of Magos in that the mercury vapour is carried to the absorption cell after magnetic stirring of the solution in the reaction flask; in the Magos procedure, mercury vapour is carried by bubbling the gas in to the solution. In contrast to the Magos procedure this slight modification overcame the necessity of at calibration by analyte addition, saving time and gainning accuracy.

This paper provides a review on separation methods and analytical techniques for the determination of several species of organic and inorganic arsenic in different matrices. Arsenic is an element whose speciation is of particular interest due to the great variation of toxicity levels exhibited for its different chemical forms. Arsenic (III) and As (V) are the most toxic species while organic compounds such as arsenobetaine (AsB), produced by methylation of inorganics species (carcinogenics) are relatively less toxic, hence the great importance of arsenic speciation in the determination of the degree of contamination of an environmental or biological system.

Speciation studies of Fe, Cr, Co, Ni and Cu on reactive fraction (adsorved on oxides, hydroxides, carbonates and clay minerals) and pyrite were performed in Avicennia schaueriana and Rhizophora mangle sediments from Amapá shoreline-Brazil. The soil under Avicennia showed a higher heavy metal concentration in reactive fraction than under Rhizophora. The soil under Rhizophora showed low heavy metal bioavailability, having an increasing association with pyrite across sediment section.

This work describes the construction of a home-made low-cost reactor, using easily available materials, capable of destroying efficiently dissolved organic matter. Just 30 minutes of irradiation were sufficient to destroy more than 99% of the humic acids present in a solution of 4 mg C L-1. Copper speciation was evaluated in natural waters of different salinities to test the reactor's efficiency in destroying organically complexed metal species. The effect of the organic matter concentration, salinity, dissolved oxygen and temperature in the photo-oxidation process is discussed.

The sequential extraction procedure proposed by Tessier and total digestion were applied for the analysis of sediment samples from Tubarão River. The recoveries were between 93.5 and 102.5% for Cu and 99.2 and 111% for Pb. The precision was tipically better than 6% for Cu and 3% for Pb. Comparison of the values obtained for the total digest with the sum of the extracted fractions showed that there were no significant losses in the extraction steps so that the method can be used for the monitoring availability and mobility of these analytes. According to ten points of sampling, was possible to determined the average of labiles phases (fractions 1 and 4) for Pb and Cu: 33.70% and 18.18%, respectively; and the inert phases (fraction 5- residual): 66.30% for Pb and 81.82% for Cu.

The determination of the physical-chemical mercury speciation in tropical coastal waters using the derivatization technique with CVAFS detection was optimized. This methodology is able to separate the elementary, reactive, organic and total mercury phases with low operational costs. The importance of bromide chloride solution, the diluted aqua regia, used as the oxidative solution, and different compositions of the reductive solution of stannous chloride were tested and the better results for coastal waters are presented. The recuperation of the sum of the different phases ranges from 80 to 94% of the total Hg content in duplicates of different marine samples. The optimized methodology permits important studies on the mercury cycle in the coastal environment.

Probably one of the most difficult and challenging aspects of measuring trace metals in natural waters is to avoid contamination during sampling, manipulation and analysis. This work discusses how to avoid contamination using simple procedures, and considers alternative methods to purify deionised water and low grade reagents to enable accurate determination of trace metals in natural waters in a common laboratory. Measurements were performed by cathodic stripping voltammetry and copper was used as a model metal to test the procedures. It was possible to evaluate copper speciation in natural waters even when total dissolved copper concentration was as low as 1.5 nmol L-1. The methods' accuracy was confirmed by analysis of certified seawater.

Lead absorption is influenced by the species that are formed and the physicochemical characteristics of lead, among others. Lead plasma concentration is < 5% of total blood lead and represents the biologically active fraction able to cross the cell membranes. Health risks mainly depend on a specific metal and its species. Speciation analysis is the analytical activity of identifying and determining different metal species. Chromatographic methods are very useful in the identification of species and the techniques most used to determine metals in biological fluids are ICP OES/MS and AAS. Lead speciation analysis in blood plasma is fundamental for understanding and evaluating the interaction mechanisms between that analyte and its biological targets.

The efficiency of a new procedure for the digestion of natural waters, based on a microwave-activated photochemical reactor was evaluated in this work. Fluorescence spectra showed a 99% reduction in the emission of a 40 mg L-1 humic acid solution after 15 min of UV irradiation. In the presence of H2O2, only 3 min were necessary to accomplish a reduction of almost 100% in the emission and 6 min to reduce the concentration of dissolved organic carbon by 95%. The copper recovery from synthetic samples containing commercial humic acid, from soil suspensions, as well as from natural waters varied between 91.5 and 106.6%. The digestion of dissolved and unfiltered samples was successfully accomplished in 6 and 12 min, respectively. No contaminations or sample losses were observed. Results of copper speciation in natural waters showed that this metal is predominantly bound to natural ligands. Only 3-6% of the total recoverable copper is present in the labile form.
