OBJECTIVE: The aim of this study was to verify whether HDL particles isolated from patients with coronary artery disease (CAD) and low HDL-C had diminished ability to promote cholesterol efflux from cultured cells compared with HDL isolated from subjects without CAD and with normal HDL-C. METHODS: Smooth muscle cells isolated from human aortas cultured and radiolabeled with ³H-cholesterol were loaded with cholesterol and incubated with increasing concentrations of HDL isolated from 13 CAD patients with low HDL-C (CAD group) or from 5 controls without CAD (C group). Efflux of cellular cholesterol was measured by cellular depletion of radiolabeled cholesterol and by the appearance of ³H-cholesterol into experimental medium expressed as a percentage of total labeled cholesterol. RESULTS: Cholesterol efflux increased with the amount of HDL present in the medium, and no difference was found between groups at various HDL protein concentrations: efflux was 28 ± 6.3% (C) and 25.5 ± 8.9% (CAD) with 25 mg/mL; 34 ± 4.3% (C) and 31.9 ± 6.6% (CD) with 50 mg/mL and 39.5 ± 3.5% (C) and 37.1 ± 4.4% (CAD) with 100 mg/mL, HDL. CONCLUSION: Because the HDL fraction of CAD patients with low HDL-C have normal ability to extract cholesterol from cells of the vessel wall, it is suggested that low HDL-C atherogenicity should be ascribed to diminished concentrations of HDL particles rather than to the qualitative properties of the HDL fraction.

OBJETIVO: Determinar a incidência de fístulas arteriovenosas pulmonares (FAVPs) pós-operação de Glenn bidirecional; as possíveis variáveis independentes que poderiam influenciar no seu aparecimento; comprovar o uso do ecocardiograma contrastado com microbolhas na sua detecção; e testar a sensibilidade e especificidade da angiografia pulmonar. MÉTODOS: Operados 59 pacientes, entre março 1990 e dezembro 1995, com idades entre 2 e 132 meses (média 32,7±33,6). Todos submetidos a exames clínico, laboratorial, ecocardiograma contrastado com microbolhas e cateterismo cardíaco. RESULTADOS: Nos 54 pacientes sobreviventes as FAVPs ocorreram em 20 (37,0%). A idade desses pacientes variou de 2 a 132 meses (média 29,6± 29,7). A atresia tricúspide em 10 (50,0%) e o coração univentricular em 8 (40,0%), foram as cardiopatias prevalentes. Em 13 (65%) a operação de Glenn bidirecional foi realizada à direita, em 2 (10,%), à esquerda e em 5 (25%) foi bicaval. O tempo de seguimento dos pacientes FAVPs, variou de 4 a 84 meses (média 32,4±21,65), e nos sem fístulas de 1 a 77 meses (média de 23,4±18,8), com valor de p=0,04, com significância estatística. O diagnóstico de FAVPs foi feito pelo ecocardiograma contrastado com microbolhas nos 20 casos, sendo considerado positivo, ao se detectar retorno de microbolhas pelas veias pulmonares. A angiografia pulmonar mostrou alterações compatíveis com FAVPs em 16, demonstrando sensibilidade de 80%. CONCLUSÃO: A incidência de FAVPs pós-operação de Glenn bidirecional foi alta (37%), e o intervalo de tempo decorrido após a operação de Glenn bidirecional, foi a única variável independente que se correlacionou de forma significativa com o aparecimento das FAVPs (p=0,04); o ecocardiograma com microbolhas foi o método padrão de diagnóstico; a angiografia pulmonar se revelou um método com sensibilidade de 80,0%.

OBJETIVO: Avaliar freqüência, correlações clínicas e influência prognóstica do potencial tardio no eletrocardiograma de alta resolução, em portadores de insuficiência cardíaca de diferentes etiologias. MÉTODOS: Foi estudado o eletrocardiograma de alta resolução, durante 42 meses, em 288 portadores de insuficiência cardíaca de diferentes etiologias, 215 homens (74,65%) e 73 mulheres (25,35), de idades entre 16 e 70 anos (média 51,5, desvio-padrão 11,24). As etiologias da insuficiência cardíaca foram: cardiomiopatia hipertensiva, 78(27,1%); cardiomiopatia dilatada idiopática, 73(25,4%); cardiomiopatia isquêmica, 65(22,6%); cardiomiopatia da doença de Chagas, 42(14,6%); cardiomiopatia alcoólica, 9(3,1%); cardiomiopatia periparto, 6(2,1%); valvopatias em 2(4,2%) e miocardite viral, 3(1,04%). Foram avaliadas a duração do QRS Standard, duração do QRS filtrado, duração do sinal abaixo de 40µV e a raiz quadrada nos últimos 40ms quanto a idade, sexo, etiologia, achados do eletrocardiograma de repouso de 12 derivações, do ecocardiograma, do eletrocardiograma de longa duração e mortalidade. Para a análise estatística, foram utilizados os testes: exato de Fisher, t de Student, de Man-Whitney, análise de variância, Log-HanK e o método de Kaplan-Meyer. RESULTADOS: O potencial tardio foi diagnosticado em 90 (31,3%) pacientes e não houve correlação com as etiologias. Sua presença associou-se a: menor consumo máximo de oxigênio a cicloergoespirometria (p=0,001); taquicardia ventricular sustentada e não sustentada ao Holter (p=0,001), morte súbita e mortalidade (p<0,05). Houve uma maior sobrevida nos pacientes sem potencial tardio. CONCLUSÃO: A presença de potencial tardio não se associou às etiologias e mostrou-se um marcador de pior prognóstico.

FUNDAMENTO: A detecção do limiar anaeróbico (LA) pela análise da variabilidade da frequência cardíaca (LiVFC) pode significar uma nova maneira de avaliação da capacidade funcional cardiorrespiratória (CFCR) em pré-adolescentes. OBJETIVO: Testar o método de LiVFC para detecção do LA em pré-adolescentes não obesos (NO), obesos (O) e obesos mórbidas (OM), a fim de determinar diferenças em sua CFCR. MÉTODOS: Foram estudados 30 pré-adolescentes, com idades entre 9 e 11 anos, divididos em três grupos de 10: a) grupo NO - índice de massa corpórea (IMC) com percentil do National Center for Chronic Disease Prevention and Health Promotion entre 5 e 85; b) grupo O - IMC de percentil entre 95 e 97 e c) grupo OM - IMC com percentil acima de 97. Todos foram submetidos a um protocolo incremental realizado em esteira rolante e registraram-se os batimentos cardíacos para detecção do LiVFC, que foi determinado pelo valor de 3,0 ms do índice do desvio-padrão 1 (SD1), extraído dos intervalos RR. RESULTADOS: Os valores médios no momento do LiVFC mostraram maiores valores para o grupo NO, destacando-se: a) VO2 (ml/kg/min) NO = 27,4 ± 9,2; O = 13,1 ± 7,6 e OM = 11,0 ± 1,7; b) FC (bpm): NO = 156,3 ± 18,0; O =141,7 ± 11,4 e OM = 137,7 ± 10,4; e c) distância percorrida (metros): NO = 1.194,9 ± 427,7; O = 503,2 ± 437,5 e OM = 399,9 ± 185,1. CONCLUSÃO: O LiVFC se mostrou efetivo para avaliação da CFCR e poderá vir a ser aplicado como método alternativo à ergoespirometria em determinadas situações.

Kikuio grass (Pennisetum clandestinum Hochst) is beyond any doubt, a pasture very important for farm animals; since its chemical composition is very similar to that of alfalfa, the present field trial was carried out; a randomized block design with 8 treatments was selected as follows: 1 N - P - K - Ca - Mg (complete manuring) 2 N - P - K - Ca----- (without Mg) 3 N - P - K-------Mg (without Ca) 4 ----P - K - Ca - Mg (without N) 5 N------K - Ca Mg (without P) 6 N - P - Ca - Mg (without K) 7 organic matter (without mineral fertilizers) 8 control Nitrogen was applied as NaN03 (topdressed) and as ammonium sulfate; P2O5 was given as superphosphate associated to bonemeal; K2O was applied as muriate, CaO as "sambaquis" (oyster shells); MgO was given as MgSO4 (topdressed). The source of organic matter was farmyard manure. As far yields are concerned the following observations were made: 1. treatment n. 7 was superior to all others; 2. considering the mineral fertilizers, good responses were due to N and P2O5; 3. the control yield was exceedingly poor, being inferior to all the others treatments; The chemical analyses revealed that: 1. the protein content decreased accordingly to this order: 7, 6, 5 and 1; treatment 4 (without N) gave the lowest protein content; 2. treatment n. 4 produced the highest fat content; treatment no. 7 ranked second; no. 8 gave the lowest fat content; 3. crude fiber: highest - treatment 7; lowest - 8; 4. ashes: the ashes content was higher in treatment 5; proprobably because the most abundant element in the ashes is K, the ash content of treatment 6 (no K) was very low; 5. non nitrogenous substances (determined by difference) - high in treatment 8 and low in treatment 7; 6. mineral elements in the ashes - the element omitted from a given treatment was very low in the grasses therein obtained; this shows the relative poverty of the soil in that element. As general remark the Authors suggest the use of farmyard manure in the fertilization of Kikuio grass; farmyard manure could probably substitute wither green manure or compost.

Plantas de Anthurium andraeanum foram cultivadas em soluções nutritivas carentes em N, P, K, Ca, Mg, S e B, afim de se obter o quadro sintomatológico das carências, assim como os níveis analíticos dos elementos nas folhas, caule e raiz. Os sintomas de carência foram obtidos para os nutrientes na seguinte ordem decrescente: N, Ca, Mg, S, B, P e K. Os teores porcentuais dos nutrientes na matéria seca foram, em folhas sadias e afetadas, respectivamente; N% -1,56-1,25; P % 0,37-0,25; K % - 3,37-0,53; Ca %-1,44-0,65; Mg % - 0,37-0,28; S % - 0,18-0,16. Para B, os valores foram de 86 e 47 ppm respectivamente. A fim de aquilatar as quantidades de nutrientes extraídas pelo antúrio, plantas com um, dois e três anos foram colhidas e separadas em folhas, caule, raize e flor e analisadas para os macro e macronutrientes, com exceção do Mo. Observou-se que a extração e sensivelmente aumentada na passagem do segundo para o terceiro ano. Plantas com três anos contém: N-434 mg; P-61 mg; Ca-327 mg; Mg-224 mg; S-45 mg; B-2434 µg; Cu-204 µg; Fe-7851 µg;Mn-7842 µg; e Zn-237 µg.

The author gives the clinical records of 20 patients, 14 of which were treated during 17 months, and the rest much less. The method used for treatment was the ecclectical preferred by the A. since many years , in which he associates electricity with minor surgery, chaulmestrol (pure or with 0.5 p. c. iodine or 4 p. c. creosote) and other auxilliary curative agents. The chaulmoogra derivatives were used daily as nostrils tamponage (2 p.c. mentholated or thymolated ester), periodically by injections inside the enlarged lymph nodes and nerve abscesses, and twice weekly by subcutaneous infiltrations (MUIR method) 5 c.c. each, and chaulmoogra soap tablets per os. The galvanocauterisation session was once per week, on active leprotic lesions, followed by painting with 30 p. c. trichloracetic acid solution. All 20 patients were bacilliferous before treatment, and became stronger positive after some months treatment. At the end 14 negativated and 6 remained positive and sometimes bacilli being very scanty. 16 out of 20 gave interesting serological reactions, viz.: Wassermann, Stern, Kahn. Rubino, Witebsky and Gaté (Formol-gel) positive in 5; Stern, Rubino. Witebsky and Gaté positive and Wassermann and Kahn negative in 2; Stern. Kahn, Witebsky and Gaté positive and Wassermann anticomplementary in 1; Wassermann. Stern, Kahn, Rubino and Gaté negative in 1. in the beginning, and a few months later Stern. Witebsky and Gaté becoming positive; Stern. Rubino. Witebsky and Gaté positive and anticomplementary W. in 1; Stern, Witebsky and Gaté positive and Wassermann. Kahn and Rubino negative in 1; Witebsky and Gaté positive and Wassermann, Stern, Kahn and Rubino negative in 1; Witebsky 3 times anticomplemantary and strongly positive Gaté in 1; Wassermann and Gaté positive in 1; Stern and Rubino positive in 1 and Stern test negative in one. In 7 cases high Formol-gel were associated with a high sedimentation index. Many cases had very high S.I. being a false measure of the severity of the disease; others remained very high notwithstanding the great improvement of the disease. All patients with more than 12 months treatment became practically symptom free. Lepra reaction amongst them was rare and always started by embolic rash, being controlled by destruction of such skin lesions by galvanocauterisation. In a few cases the lepromata infiltrated with "Subintrol" (a 3 p. c. special chaulmoogra soap prepared by Dr. ASTROGILDO MACHADO) were completely destroyd and never relapsed. In October 1946 Dr. ERNEST MUIR saw here a few cases treated by the author's method and suggested the combination of Diasone with galvanocauterisation which is being done now with satifactory results. The second part of this paper, reporting many leprosy cases treated by the so-called ecclectical method, which are symptom free and negativated since five to ten years, will be published as soon as the sulfone-therapy be summarised in some reliable scientific report to be compared with.

This study evaluates whether blood collected on filter paper kept at 4 degrees C and tested at different intervals of time (1, 7, 15, 30 and 60 days after collection) would present similar results when compared to the serum samples and whether the type of filter paper influences the results. Eluates from filter paper samples were tested for Trypanosoma cruzi antibodies using indirect immunofluorescence antibody test (IFAT), indirect haemagglutination (IHA) and enzyme-linked immunosorbent assay (ELISA) as reference, the antibody titer in sera. Analysis of data showed that results obtained with IFAT, IHA (cut off point = 1:40) and ELISA in sera had similar sensitivity and good concordance among reactions. The use of a multiple linear regression model indicated that titer fall in eluates occurs up to the 7th day after the collection, and it is more marked for samples with lower antibodies titers. However, no significant differences were observed by IFAT, IHA (cut off point = 1:20) and ELISA in the proportion of positive reactions between sera and eluates. The results also showed that Melitta, Klabin or Whatman (reference) filter papers could be indicated for surveys, since they have shown similar capacity of maintenance of anti-T. cruzi immunoglobulins.

Cytological studies were made on larvae of Gigantodax marginalis, G. chilensis, G. fulvescens and Cnesia dissimilis from four creeks in Lanin National Park, Neuquen province, Argentina. Chromosome maps and idiograms of these species are presented. The following inversions were observed: G. marginalis: IL-1 (X-linked inversion), IL-2 (Y-linked inversion), IIS-1.2, IIL-1, IIIL-4,5; G. chilensis: IL-4 (X-linked inversion), IIS-1.2, IIIL-4,5; G. fulvescens:IL-1 (X-linked inversion), IL-3 (Y-linked inversion), IIS-1.2, IIL-1, IIIL-4,5; C. dissimilis: IL-1, IL-5, IIIL-1. Karyological information was used to construct a cladogram and Cnesia sp. Was found to show close resemblance to the three Gigantodax spp.

A study was conducted to investigate the thermal constant in an experimental population of Muscina stabulans (Fallén 1817). A natural population was obtained from a livestock and their F1 generations were maintained at four constant temperatures (16°C, 20°C, 26°C and 31°C). The thermal constant was calculated by the hyperbole method using the base temperature, tb = 4.4°C and the thermal constant K = 35.3 GD. Ricker's geometric regression, considering tb = 7.8°C and K = 28.9 GD was also calculated.

One of the main opportunistic fungal infections amongst immunocompromised individuals is oral candidosis, which has been found in up to 90% of human immunodeficiency virus (HIV)-infected patients. This study employed yeasts isolated from the saliva and oral cavities of 114 HIV-infected patients living in Campinas, São Paulo. Of the isolates, 57.8% were identified as Candida albicans and 42.1% as non-C. albicans. The latter isolates were subsequently identified as C. krusei (7.5%), C. lusitaniae (5.2%), C. tropicalis (4.6%), C. parapsilosis (4.6%), C. glabrata (2.8%), C. kefyr (1.7%), C. guilliermondii (1.7%), C. intermedia (1.1%), C. norvegensis (0.5%), and Rhodotorula rubra (1.7%). Susceptibility of the isolates to amphotericin B, fluconazole, miconazole, and itraconazole was also determined by a microdilution method adopted by the National Committee for Clinical Laboratory Standards. The isolates demonstrated various susceptibilities to the antifungal agents. In particular 29 C. albicans and 13 non-C. albicans isolates showed low susceptibility to FLCZ (> 64 µg/ml). This study revealed huge diversity of Candida species, in particular the increasing emergence of non-C. albicans associated with the oral flora of HIV-infected patients.

The present study evaluated rickettsial infection in Amblyomma spp. ticks collected in a farm in Coronel Pacheco, a Brazilian spotted fever (BSF) endemic area. A total of 78 A. cajennense and 78 A. dubitatum free-living adult ticks were collected and tested by polymerase chain reaction (PCR) targeting a fragment of the rickettsial gene gltA. Only one pool of three A. cajennense ticks showed the expected product by PCR. This pool was further tested by PCR using sets of primers targeting the rickettsial genes gltA, ompA, and ompB. All reactions yielded the expected bands that by sequencing, showed 100% identity to the corresponding sequences of the Rickettsia rickettsii gene fragments gltA (1063-bp), ompA (457-bp), and ompB (720-bp). The minimal infection rate of R. rickettii in the A. cajennense population was 1.28% (at least one infected tick within 78 ticks).The present study showed molecular evidence for the presence of R. rickettsii in A. cajennense from a BSF-endemic area in Coronel Pacheco, state of Minas Gerais. Although R. rickettsii has been previously reported infecting A. cajennense ticks in Brazil and other Latin American countries, the present study performed the first molecular characterization of R. rickettsii from the tick A. cajennense.

Nitroimidazoles exhibit high microbicidal activity, but mutagenic, genotoxic and cytotoxic properties have been attributed to the presence of the nitro group. However, we synthesised nitroimidazoles with activity against the trypomastigotes of Trypanosoma cruzi, but that were not genotoxic. Herein, nitroimidazoles (11-19) bearing different substituent groups were investigated for their potential induction of genotoxicity (comet assay) and mutagenicity (Salmonella/Microsome assay) and the correlations of these effects with their trypanocidal effect and with megazol were investigated. The compounds were designed to analyse the role played by the position of the nitro group in the imidazole nucleus (C-4 or C-5) and the presence of oxidisable groups at N-1 as an anion receptor group and the role of a methyl group at C-2. Nitroimidazoles bearing NO2 at C-4 and CH3 at C-2 were not genotoxic compared to those bearing NO2 at C-5. However, when there was a CH3 at C-2, the position of the NO2 group had no influence on the genotoxic activity. Fluorinated compounds exhibited higher genotoxicity regardless of the presence of CH3 at C-2 or NO2 at C-4 or C-5. However, in compounds 11 (2-CH3; 4-NO2; N-CH2OHCH2Cl) and 12 (2-CH3; 4-NO2; N-CH2OHCH2F), the fluorine atom had no influence on genotoxicity. O presente trabalho teve por objetivo determinar o carbono e o nitrogênio presentes na biomassa microbiana, usando a irradiação do forno de microondas em substituição ao clorofórmio nos métodos fumigação-incubação e fumigação-extração. Foram utilizadas duas amostras de um Podzólico Vermelho-Escuro, submetidas aos procedimentos de incubação e de extração, após serem fumigadas com clorofórmio e irradiadas com microondas por diferentes períodos (2, 5, 10, 15, 20, 30, 40 e 50 min). Observou-se que a irradiação durante 2 min foi suficiente para estimar o C e o N presentes na biomassa microbiana nos procedimentos de incubação e extração, com valores semelhantes aos verificados pela fumigação com clorofórmio. A irradiação com microondas reduziu os coeficientes de variação nas amostras de solo submetidas ao procedimento de extração, comprovando ser o método irradiação-extração o mais adequado para a estimativa do C e do N microbianos.
