The objective of this work was to identify expressed simple sequence repeats (SSR) markers associated to leaf miner resistance in coffee progenies. Identification of SSR markers was accomplished by directed searches on the Brazilian Coffee Expressed Sequence Tags (EST) database. Sequence analysis of 32 selected SSR loci showed that 65% repeats are of tetra-, 21% of tri- and 14% of dinucleotides. Also, expressed SSR are localized frequently in the 5'-UTR of gene transcript. Moreover, most of the genes containing SSR are associated with defense mechanisms. Polymorphisms were analyzed in progenies segregating for resistance to the leaf miner and corresponding to advanced generations of a Coffea arabica x Coffea racemosa hybrid. Frequency of SSR alleles was 2.1 per locus. However, no polymorphism associated with leaf miner resistance was identified. These results suggest that marker-assisted selection in coffee breeding should be performed on the initial cross, in which genetic variability is still significant.

The objective of the present work was to characterize banana accessions from the Germplasm Bank at Embrapa Mandioca e Fruticultura Tropical (Brazil), using agronomical, physical and physicochemical characteristics of fruit and simple sequence repeats (SSR) markers. Twenty-six accessions were analyzed, in which high genetic variability was found, especially for the agronomical characters number of fruit and weight of bunch. Accessions with high contents of carotenoids (diploid 'Jaran'), polyphenols (triploid 'Caipira' and tetraploid 'Teparod') and vitamin C (diploid 'Tuugia' and an unknown triploid AAA) in the fruit were identified. Thirteen microsatellite primers revealed an average of 7.23 alleles, which showed high variability. A dendrogram was prepared using the Gower algorithm for the distance matrices obtained from the agronomical, physical and physicolchemical analysis of fruit and SSR markers. Adopting the average genetic divergence as the cut-off point, three clusters were found: G1, formed by the diploids 'Jaran', 028003-01 and M-48; G2, by the diploids 'Malbut' and 'Ido 110'; and G3, by 21 tri-and tetraploid accessions, including one diploid, 'Tuugia'. The triploids with the B genome 'Thap Maeo', 'Walha', 'Pacha Nadan' and 'Champa Madras' were grouped in G2. Results from this work can be used for breeding hybrids with good agronomical traits and fruit quality.

Os marcadores microssatélites são ferramentas úteis em diversas análises genéticas em plantas. No caso do mamoeiro (Carica papaya L.), poucos locos de microssatélites foram descritos até o momento. Assim, o objetivo deste trabalho foi explorar a base de dados do GenBank / NCBI (National Center of Biotechnoloy Information) à procura de microssatélites de mamoeiro, visando a seu futuro uso em estudos genéticos e moleculares aplicados ao melhoramento genético. As seqüências foram obtidas no GenBank / NCBI, no formato FASTA, e analisadas para a presença de microssatélites com um mínimo de 20; 7 e 5 repetições dos motivos de mono-, di- e trinucleotídeos, respectivamente, e acima de 4 repetições para tetra- e pentanucleotídeos. Seqüências com mais de 90% de similaridade foram consideradas redundantes e, portanto, eliminadas das análises. Foram analisadas 44.591 seqüências, das quais 3.180 foram não-redundantes e apresentaram 3.947 microssatélites. Desse total, 3.587 foram classificados como microssatélites perfeitos, 8 imperfeitos, 65 interrompidos, 239 compostos-perfeitos, 8 compostos-imperfeitos e 40 compostos-interrompidos. As repetições de di- e trinucleotídeos representaram 65,7 e 14,4% do total de seqüências analisadas, respectivamente. Somente os motivos do tipo AT/TA representaram 44,1% dos microssatélites encontrados. Os motivos mais comuns de tri-, tetra- e pentanucleotídeos foram AAT, AATT e TTTAA, respectivamente. Observou-se que, nas seqüências disponíveis, o genoma do mamoeiro apresenta, em média, um microssatélite a cada 5,65 kb.

Objetivou-se com este trabalho avaliar o desempenho agronômico de genótipos tri- e tetraploides de bananeira, em dois ciclos de produção, visando a selecionar os mais promissores para recomendação de cultivo na região do Recôncavo da Bahia. O delineamento utilizado foi o de blocos casualizados, com 11 genótipos distribuídos em 3 blocos, com quatro plantas úteis por parcela, conduzidos em espaçamento de 3 m x 2 m. Analisaram-se os seguintes caracteres agronômicos: altura da planta (m); diâmetro do pseudocaule (cm); número de folhas vivas na floração; número de folhas vivas na colheita; comprimento do engaço (cm); diâmetro do engaço (cm); peso do cacho (kg); peso das pencas (kg); peso individual da penca (g); número de pencas; número de frutos por cacho; espessura da casca (mm); comprimento do pedicelo (cm); diâmetro do pedicelo (mm); suscetibilidade ao despencamento (Lb); firmeza da polpa com casca; peso do fruto (g); comprimento do fruto(cm); diâmetro do fruto (mm); índice de alongamento; peso da polpa (g); rendimento polpa/casca; rendimento da polpa (%); diâmetro da polpa (mm); espessura da casca (mm); firmeza da polpa (Lb); acidez titulável (%); sólidos solúveis (ºBrix); ratio (SS/AT) e pH. Das trinta variáveis mensuradas, doze foram significativas para a fonte de variação 'ciclos'. Em relação à interação 'genótipos x ciclos', é possível afirmar que não houve comportamento diferenciado dos genótipos do primeiro para o segundo ciclos de produção, excluindo-se a 'altura de planta', o 'número de folhas vivas na floração', o 'diâmetro do fruto' e a 'espessura da casca'. Em relação ao efeito dos 'genótipos', os resultados mostraram-se significativos para todas as características, exceto para suscetibilidade ao despencamento. Considerando os dados agronômicos, os genótipos da série YB e as cultivares BRS Princesa e BRS Garantida mostram-se promissoras para o cultivo na região do Recôncavo da Bahia, pois apresentaram bom desempenho agronômico.

The preparation of 2', 3'-di-O-hexanoyluridine (2) by a Candida antarctica B lipase-catalysed alcoholysis of 2', 3', 5'-tri-O-hexanoyluridine (1) was optimised using an experimental design. At 25 ºC better experimental conditions allowed an increase in the yield of 2 from 80% to 96%. In addition to the yield improvement, the volume reaction could be diminished in a factor of 5 and the reaction time significantly shortened.

In the present work three ferroin reagents were studied for the simultaneous spectrophotometric determination of iron and copper: 1,10-phenanthroline, 2,2'-bipyridine and 2,4,6-tri(2-pyridyl)-1,3,5-triazine. Effect of pH, conditions, order reagent addition, interferences, amount of reagents, lineal range, sensitivity and stability of each system were compared. The 2,4,6-tri(2-pyridyl)-1,3,5-triazine can be used for determination of iron in the presence of copper with a detection limit of 5 µg L-1 and coefficient of variation of 2.0%; However it was not possible to determine directly copper in the presence of iron with this reagent. 1,10-phenanthroline can be used for simultaneous determination of the metallic ions with detection limits of 7 and 8 mg L-1 and coefficients of variation of 1.8 and 2.3% in the determination of iron and copper, respectively. The results showed also that 2,2'-bipyridine can be used for simultaneous determination of the metallic ions with detection limits of 11 and 32 µg L-1 and coefficients of variation of 1.9 and 2.5% in the determination of iron and copper, respectively. The reagents were used for spectrophotometric determination of iron and copper in ethanol fuel.

This paper reports an HPLC-ESI-MS/MS investigation on the oxidation of 3,5- and 4,5- dicaffeoylquinic acid using iron(III) tetraphenylporphyrin chloride as catalyst. Two major mono-oxidised products of the quinic acid moiety have been identified for both compounds. However, only the 4,5-derivative afforded two different tri-oxo products. Thus, it seems that the oxidation pattern depends on the number and positions of the caffeic acid moieties present in caffeoylquinic acid molecules.

Reduction of five substituted octalones employing lithium tri-sec-butylborohydride (L-selectride®) in THF and ethyl ether led to allylic alcohols with moderate diastereoselectivity. The stereoselectivity of addition of bulky hydrides showed to be different from most examples in the literature and was strongly influenced by substitution on the octalone ring.

The chemical investigation of the hexane and ethanol extracts from aerial parts of Vernonia chalybaea conducted to the isolation and characterization of a new aliphatic tetrahydroxyl ether, along with a series of known compounds such as 4 α,10 α-epoxyaromadendrane, friedelin, taraxasteryl acetate, pseudotaraxasteryl acetate, lupeyl acetate, lupeol, α-amiryn, β-amiryn, β-sitosterol, stigmasterol, 2,3-dimethyl-1,2,3-tri-hydroxybuthanol, angophorol, angophorol-7-O-glucoside, angophorol-7-O-rutinoside, 3,7-dimethoxy-5,3',4'-trihydroxyflavone and acacetin. The structures of all compounds were determined by spectroscopic analysis and comparison with published spectral data.

The NMR (RMN¹H, 13C, COSY, HMQC, HMBC, NOE-DIFF, NOESY) and mass spectra data analysis of sixteen flavonoids, including nine natural, 7-O-methylkanferol (ramnocitrin), 3,7-di-O-methylkanferol (kumatakenin), 3-O-methylquercetin, 3,7,3',4'-tetra-O-methylquercetin (retusin), 3,7,8,4'-tetra-O-methylgossipetin, 3,7,8,3',4'-penta-O-methylgossipetin, 7-O-methylapigenin (genkwanin), 3,7,8-tri-O-methylherbacetin, 7,4'- di-O-methylquercetin (ombuine), isolated from Solanum paludosum and S. jabrense, and seven prepared methyl and acetyl derivatives, are discussed according the substitution on the rings A, B and C.

The physicochemical properties (solubilization, structural organization and stability) of meso-tetrakis(p-methoxyphenyl)porphyrin (TMPP), a promising photosensitizer for photodynamic therapy, solubilized in polymeric micelles of tri-block copolymers PluronicTM P-123 and F-127, were studied. The formulations obtained by the solid dispersion method led to monomerization of TMPP in these copolymers. Solubility studies showed that P-123 solubilizes double the photosensitizer than F-127. The self-aggregation phenomenon was affected by the [TMPP]/[poloxamer] ratio and medium temperature. The decrease in the temperature of these systems promoted the formation of different kinds of TMPP aggregates intrinsically connected with the structural changes occurring in the micelles.

In the last ten years, the interest in natural and semi-synthetic cucurbitacin derivatives has increased, primarily due their cytotoxic and anti-tumoral activities. However, the isolation of glycosylated cucurbitacins has been difficult due the presence of β-glucosidase enzyme. With the aim of obtaining new glycosylated derivatives, the glycosylation of dihydrocucurbitacin B under Köenigs-Knorr and imidate reaction conditions was studied. Novel glycoside derivatives 16-(1,2-orthoacetate-3,4,6-tri-O-acetyl-α-D-glucopyranosyl)-dihydrocucurbitacin B (2), 2-O-β-D-2,3,4,6-tetra-O-acetyl-galactopyranosyl dihydrocucurbitacin B (3) and 2-O-β-D-galactopyranosyl dihydrocucurbitacin B (4) were synthesized for the first time in 17% (2 and 3) and 48% (4) yields.

Environmentally friendly acid carbon (CG) catalysts, containing a high amount of sulfonated and oxygenated groups, were prepared from glycerin, a biodiesel waste. CGs were produced by glycerin carbonization in the presence of H2SO4 at 1:3 m:m ratio in a closed autoclave at 180ºC for different times: 0.25; 1; 3 and 6 hours (CG-0.25h; CG-1h; CG-3h and CG-6h, respectively). The catalyst properties for all carbons were evaluated in the glycerol etherification reaction with tert-butyl alcohol (TBA). The yield for mono-tert-butyl glycerol (MTBG), di-tert-butyl glycerol (DTBG) and tri-tert-butyl-glycerol (TTBG) was high and very similar for all CGs, of about 43% and 20% for the MTBG and DTBG + TTBG, respectively. Furthermore, the activity of these catalysts were close to those obtained using a commercial resin, Amberlyst-15, of about 50% and 27% for MTBG and DTBG + TTBG, respectively.

We report herein a study on the glycosylation of cyclohexanol with four D-glucosamine-based peracetylated glycosyl chlorides bearing different substituents at C-2 and three glycosylation promoters, silver carbonate, silver triflate and mercury II chloride/mercury II oxide, by the Koenigs-Knorr method. Under the conditions studied, glycosylation was successful only when 3,4,6-tri-O -acetyl-2-deoxy-2-phthalimido-α-D-glucopyranosyl chloride was used as the glycosyl donor, with silver carbonate proving the best promoter. In order to investigate the influence of the nature of the halogen at C-1, we also carried out the glycosylation of cyclohexanol with 3,4,6-tri-O -acetyl-2-deoxy-2-phthalimido-α-D-glucopyranosyl bromide, a more reactive glycosyl donor. As expected, the yield with the bromide derivative was higher with the three promoters and, again, silver carbonate was the most efficient promoter. Finally, to illustrate the well-known efficient procedure for conversion of the phtalimido group at C-2 to the corresponding acetamido group, cyclohexyl 3,4,6-tri-O -acetyl-2-deoxy-2-phtalimido-β-D-glucopyranoside was converted into cyclohexyl 2-deoxy-2-acetamido-β-D-glucopyranoside in two steps, namely, hydrazinolysis of the phtalimido group followed by chemoselective acetylation of the free amino group by treatment with acetic anhydride in methanol, at 77% overall yield.

Várias espécies de Cylindrocladium podem causar doenças em mudas de eucalipto, na propagação clonal. Apesar de, em algumas situações, fungicidas serem imprescindíveis no controle desses patógenos, faltam estudos para avaliar a efetividade desses produtos. Por isso, objetivou-se selecionar fungicidas sistêmicos para controlar C. candelabrum, determinar o efeito protetor, curativo e anti-esporulante dos fungicidas, sua translocação e persistência em plantas de eucalipto. Nos experimentos in vitro foram testados: azoxystrobin (AZO), triadimenol (TRI), boscalid (BOS), pyraclostrobin (PYR), carbendazim (CAR), tetraconazole (TET), tebuconazole (TEB), epoxiconazole+pyraclostrobin (EPO-PYR) e epoxiconazole (EPO) nas concentrações 0,1, 1, 5, 10, 50, 100 e 1000 µg de i.a/mL. Nos experimentos in vivo, utilizou-se um clone suscetível a C. candelabrum (Eucalyptus grandis x E. urophylla). Excetuando-se o estudo da atividade translaminar, aplicaram-se os fungicidas, em ambas as faces do limbo foliar. Após 24 h, as plantas foram inoculadas com suspensão de 10(5) conídios/mL de C. candelabrum. Testou-se o efeito protetor dos fungicidas, em diferentes concentrações: AZO (0,2; 0,25; 0,4; 0,65 e 0,8 g/L), EPO (0,75; 1,125; 1,5; 1,875 e 2,25 mL/L), EPO-PYR (1,25; 1,875; 2,5; 3,125 e 3,75 mL/L), PYR (0,75; 1,125; 1,5; 1,875 e 2,25 mL/L) e TEB (0,75; 1,125; 1,5; 1,875 e 2,25 mL/L), selecionados nos testes in vitro. Para avaliar o efeito curativo, anti-esporulante, a persistência e a atividade translaminar, utilizaram-se os fungicidas: AZO (0,4 g/L), EPO (1,5 mL/L), EPO-PYR (2,5 mL/L) e TEB (1,5 mL/L). Obteve-se efeito protetor, curativo e anti-esporulante com EPO, EPO-PYR e TEB, os quais tiveram atividade translaminar e persistência nos tecidos foliares.
