A survey of the carrion fauna was made at two sites in Curitiba, State of Paraná, with the objective of describing the insects associated with carrion and setting up a preliminary data-base for medico-legal purposes in south Brazil. Vertebrate exclusion experiments were carried out in each season between 1994 and 1995 with a 250 g laboratory-bred rat (Rattus norvegicus). Five stages of decomposition were identified: fresh, bloated, decaying, dry and adipocere-like. Some species showed seasonal and site preference and so could be used to identify the probable place and season where death took place. Sarconesia chlorogaster (Diptera, Calliphoridae) was restricted to an open field site and to cooler months. Hemilucilia semidiaphana (Diptera, Calliphoridae) and Pattonella resona (Diptera, Sarcophagidae) were restricted to the forest site and warmer months. Phaenicia eximia (Diptera, Calliphoridae) and Oxyletrum discicolle (Coleoptera, Silphidae) were present at both sites throughout the year and could be useful for population level analysis. Dissochaetus murray (Coleoptera, Cholevidae) was present throughout the year at the forest site and was associated with the adipocere-like stage. Ants played an important role producing post-mortem injuries to the carcasses. Insects of 32 species are reported as being useful in community level approaches

The parasitic specificity of larval, nymph and adult Amblyomma cajennense on six different host species: Oryctolagus cuniculus, Rattus norvegicus, Gallus gallus domesticus, Anas platyrhynchus, Coturnix coturnix and Streptopelia decorata is described. In terms of the numbers of larvae and nymphs recovered, O. cuniculus was the best host species. The modal day for drop-off of larvae and nymphs was day three for the mammal hosts, but variable in the birds. We conclude that adult A. cajennense have a strong degree of specificity due to the fact that the tick failed to complete its life cycle on any of the evaluated hosts. The immature stages, on the other hand, showed a low level of specificity, most especially in the larval stage, indicating the existence of secondary hosts which probably serve as dispersers in the wild. The results also indicated a variable drop-off rhythm for larvae and nymphs in two periods, diurnal (6-18 hr) and nocturnal (18-6 hr), which differed depending upon the host.

The present work aims at learning the period of resistance to starvation (molting/death) of Triatoma rubrofasciata in different stages of development and the respective loss of weight until death. Eggs of specimens from the greater area of the city of São Luis in the State of Maranhão, Brazil, yielded approximately 300 nymphs. These nymphs were placed in labelled Borrel glasses, in which they were weekly fed on rats (Rattus norvegicus), until reaching the stage to be observed. The experiments were conducted in a climatic chamber regulated at 29 ± 1° C, 70% relative humidity and 12 hr photoperiod. The resistance to starvation increased according to the stage of development, except for adult bugs, whose results were similar to the 3rd stage nymphs. In all these development stages there was an abrupt loss of weight in the first week, followed by a gradual loss until death. Comparing this work with those of other authors, it was observed that T. rubrofasciata is among the less resistant triatomine species.

Eimeria motelo sp. n. is described from faeces of the yellow-footed tortoise, Geochelone denticulata (L.). Oocysts are irregularly ellipsoidal or cylindrical, with slightly expressed lobed protrusions and irregularities at the poles, possibly caused by wrinkling of the oocyst wall, 17 (15-19) × 9.4 (8.5-11) µm, shape index (length/width) being 1.81 (1.45-2). The oocyst wall is smooth, single-layered, 0.5 µm thick with no micropyle. There are no polar bodies. Sporocysts are ellipsoidal, 8.9 (7.5-10) × 4.4 (4-5) µm, shape index 2.03 (1.7-2.5). A sporocyst residuum is present, composed of many granules of irregular size. The sporozoites are elongate, lying lengthwise in the sporocysts. Comparison with other species of the genus Eimeria parasitising members of family Testudinidae indicates that the presently described coccidium represents a new species. The name of Eimeria carinii Lainson, Costa & Shaw, 1990 is found to be preoccupied by a homonym, Eimeria carinii Pinto 1928 given to a coccidium from Rattus norvegicus. Therefore, it is replaced by Eimeria lainsoni nom. nov.

Syphacia muris worm burdens were evaluated in the rat Rattus norvegicus of the strains Wistar (outbred), Low/M and AM/2/Torr (inbred), maintained conventionally in institutional animal houses in Brazil. Morphometrics and illustration data for S. muris recovered from Brazilian laboratory rats are provided for the first time since its proposition in 1935.

A total of 128 ticks of the genus Amblyomma were recovered from 5 marsupials (Didelphis albiventris) - with 4 recaptures - and 17 rodents (16 Bolomys lasiurus and 1 Rattus norvegicus) captured in an urban forest reserve in Campo Grande, State of Mato Grosso do Sul, Brazil. Of the ticks collected, 95 (78.9%) were in larval form and 22 (21.1%) were nymphs; the only adult (0.8%) was identified as A. cajennense. Viewed under dark-field microscopy in the fourth month after seeding, 9 cultures prepared from spleens and livers of the rodents, blood of the marsupials, and macerates of Amblyomma sp. nymphs revealed spiral-shaped, spirochete-like structures resembling those of Borrelia sp. Some of them showed little motility, while others were non-motile. No such structures could be found either in positive Giemsa-stained culture smears or under electron microscopy. No PCR amplification of DNA from those cultures could be obtained by employing Leptospira sp., B. burgdorferi, and Borrelia sp. primers. These aspects suggest that the spirochete-like structures found in this study do not fit into the genera Borrelia or Leptospira, requiring instead to be isolated for proper identification.

Seeking the identification of Angiostrongylus cantonensis as a potential etiological agent of three clinical cases of eosinophilic meningitis, mollusc specimens were collected in the state of Espírito Santo, Brazil. The snails were identified as Sarasinula marginata (45 specimens), Subulina octona (157), Achatina fulica (45) and Bradybaena similaris (23). Larvae obtained were submitted to polymerase chain reaction and restriction fragment length polymorphism diagnosis. Their genetic profile were corresponded to A. cantonensis. Rattus norvegicus experimentally infected with third-stage larvae, developed menigoencephalitis, and parasites became sexually mature in the lungs. Additionally, larvae obtained from A. fulica snails, from São Vicente, state of São Paulo, also showed genetic profiles of this nematode. This is the first record of Brazilian molluscs infected with this nematode species.

Cutaneous leishmaniasis (CL) is a neglected clinical form of public health importance that is quite prevalent in the northern and eastern parts of Egypt. A comprehensive study over seven years (January 2005-December 2011) was conducted to track CL transmission with respect to both sandfly vectors and animal reservoirs. The study identified six sandfly species collected from different districts in North Sinai: Phlebotomus papatasi, Phlebotomus kazeruni, Phlebotomus sergenti, Phlebotomus alexandri, Sergentomyia antennata and Sergentomyia clydei. Leishmania (-)-like flagellates were identified in 15 P. papatasi individuals (0.5% of 3,008 dissected females). Rodent populations were sampled in the same districts where sandflies were collected and eight species were identified: Rattus norvegicus (n = 39), Rattus rattus frugivorous (n = 13), Rattus rattus alexandrinus (n = 4), Gerbillus pyramidum floweri (n = 38), Gerbillus andersoni (n = 28), Mus musculus (n = 5), Meriones sacramenti (n = 22) and Meriones crassus (n = 10). Thirty-two rodents were found to be positive for Leishmania infection (20.12% of 159 examined rodents). Only Leishmania major was isolated and identified in 100% of the parasite samples. The diversity of both the vector and rodent populations was examined using diversity indices and clustering approaches.

Angiostrongylus cantonensis is the etiologic agent of eosinophilic meningoencephalitis in humans. Cases have been recorded in many parts of the world, including Brazil. The aim of this study was to compare the differences in the biology and morphology of two different Brazilian haplotypes of A. : ac8 and ac9. A significantly larger number of L1 larvae eliminated in the faeces of rodents at the beginning of the patent period was observed for ac9 haplotype and compared to the total of L1 larvae eliminated, there was a significant difference between the two haplotypes. The ac9 haplotype showed a significant difference in the proportion of female and male specimens (0.6:1), but the same was not observed for ac8 (1.2:1). The morphometric analysis showed that male and female specimens isolated from ac8 haplotype were significantly larger with respect to body length, oesophagus length, spicule length (male) and distance from the anus to the rear end (female) compared to specimens from ac9. The morphological analysis by light microscopy showed little variation in the level of bifurcations at the lateral rays in the right lobe of the copulatory bursa between the two haplotypes. The biological, morphological and morphometric variations observed between the two haplotypes agree with the observed variation at the molecular level using the cytochrome oxidase subunit I marker and reinforce the possible influence of geographical isolation on the development of these haplotypes.

OBJETIVO: Este trabalho analisa as conseqüências da irradiação-X no desenvolvimento do sistema nervoso de fetos de ratos. MATERIAIS E MÉTODOS: O trabalho foi constituído de 10 Rattus norvegicus albinos, Wistar, fêmeas, grávidas, com idade de oito semanas. Cinco ratas fêmeas constituíram o grupo controle e outras cinco tiveram suas regiões abdominais expostas por 30 segundos a uma dose de 0,3 Gy proveniente de um aparelho odontológico Gnatus de 70 kV e 10 mA. No 17º dia gestacional, ambos os grupos foram submetidos a histerectomia. As seções selecionadas foram examinadas para análise cerebral comparativa entre os grupos. RESULTADOS: O exame clínico revelou não haver diferenças morfológicas entre os grupos controle e experimental e nenhum dos animais apresentou anormalidades grosseiras. Vinte e sete por cento dos animais do grupo experimental apresentaram hemorragia cerebral moderada e 73% apresentaram hemorragia severa e danos no tecido nervoso. Nenhum animal do grupo controle apresentou hemorragia cerebral ou danificações de tecido nervoso. CONCLUSÃO: Estas evidências demonstram que pequenas doses de radiação-X podem causar hemorragias cerebrais e, conseqüentemente, lesão tecidual nervosa.

OBJETIVO: A obesidade mórbida é uma doença crônica que se acompanha de várias complicações médicas e de uma menor esperança de vida, prejudicando o aspecto físico, emocional, econômico e social. As pesquisas clínicas e experimentais são amplamente justificadas nesta doença, considerando-se que a opção de tratamento operatório é recente. As gastroplastias e as derivações gástricas são algumas das modalidades terapêuticas propostas e produzem uma grande câmara gástrica remanescente, que em algumas técnicas fica totalmente excluída do trânsito alimentar. As possíveis alterações deste segmento, transformado em apêndice do sistema digestório, representam um vasto campo para pesquisa. Tivemos como objetivo avaliar o peso e as alterações da dosagem de gastrina sérica do rato, após denervação vagal troncular subdiafragmática, com exclusão da câmara gástrica e duodeno do trânsito alimentar do animal. MÉTODO: Foi realizada vagotomia troncular e exclusão gastroduodenal em cem ratos, dos quais trinta puderam ser estudados. Eram ratos machos adultos (Rattus Norvegicus Albinus), da linhagem Wistar, com peso médio de 378,67 g. Este estudo foi complementado pela dosagem sérica de gastrina por radioimunoensaio com I125 e duplo anticorpo para gastrina. RESULTADOS: O peso de todos os ratos estudados diminuiu em média de 378,67 g para 285,83 g. Os trinta animais perderam em média 92,83 g (-24,52%). Houve diferenças significativas entre o peso inicial dos animais e o peso final (p< 0,001). A gastrina sérica também diminuiu em vinte e um dos trinta ratos operados, caindo de 59,68 pg/ml para 46,77 pg/ml em média, correspondendo a uma diminuição de 12,91 pg (-19,73%) entre as duas médias (p< 0,001). A análise estatística teve como objetivo avaliar se houve diminuição nas médias das variáveis: peso e gastrina, medidas antes e depois da operação proposta. Para avaliar as diferenças entre as médias, utilizou-se o teste estatístico do T-Student. CONCLUSÕES: Houve uma diminuição significativa do peso e da dosagem de gastrina sérica do animal.

OBJETIVO: Avaliar as alterações anatomopatológicas e histopatológicas da pleura e do parênquima pulmonar após a injeção de óleo de copaíba, extrato aquoso de crajiru e polivinilpirrolidona iodado (PVPI) no espaço pleural de ratos. MÉTODO: Foram utilizados 128 Rattus norvegicus var. Wistar, machos, com peso médio 198,9g (± 24,9g), randomizados em quatro grupos: copaíba, PVPI, crajiru e simulação. As substâncias foram injetadas no espaço pleural direito dos animais, os quais foram mortos em 24 h, 48 h, 72 h e 504 h, para análise macro e microscópica da pleura visceral e pulmão direito. RESULTADOS: Macroscopicamente, observou-se intensa reação pleuro-pulmonar no grupo copaíba com significância estatística (p= 0,001) em relação aos outros grupos e entre os diferentes momentos. Microscopicamente, a espessura pleural apresentou maior aumento no grupo copaíba com significância estatística nos tempos 72 h e 504 h. O PVPI provocou reação inflamatória aguda em 24 h e 48 h com melhora em 72 h, porém, na última observação, evidenciou-se lesão crônica pulmonar. O crajiru apresentou-se pouco irritativo e sem significância em relação aos demais. CONCLUSÃO: A copaíba mostrou-se muito irritante; o PVPI, moderadamente irritante, e o extrato aquoso de crajiru apresentou pouca reação inflamatória na pleura e parênquima pulmonar dos animais de experimentação.

OBJETIVO: Descreve-se a técnica de um novo um modelo de pseudoartrose em Rattus norvegicus albinus livre de espécies patogênicas (SPF). MÉTODOS: Os animais foram aleatoriamente distribuídos em dois Grupos: Grupo 1 - Controle (n =6), Grupo 2 - Experimental (n =6). No grupo Controle realizou-se um acesso ao fêmur, seguido da síntese por planos do músculo e da pele. O grupo Experimental foi submetido à osteotomia do fêmur aduzida da confecção de um retalho pediculado de fáscia lata para interposição entre os segmentos resultantes da fratura induzida cirurgicamente. Prosseguiu-se com o alinhamento e aproximação dos segmentos ósseos, mediante uso de fios de náilon introduzidos em orifícios efetuados na extremidade proximal e distal. RESULTADOS: Estudos radiológico e anatomopatológico evidenciaram a ausência de consolidação óssea com persistência de tecido fibro-osteóide decorridos 120 dias de evolução pós-operatória. CONCLUSÃO: O modelo experimental é adequado para a formação de pseudoartrose em ratos.

Os "timbós verdadeiros" (plantas do gênero Derris), originários da Amazônia Brasileira, tem demonstrado importância crescente por produzirem uma classe de compostos flavonoídicos relacionados à rotenona, que possuem atividade tóxica para peixes e mamíferos. Neste estudo foi determinado a dose letal 50% (DL50) do extrato alcoólico do pó de Derris spp para três espécies de peixes filogeneticamente diferentes e um mamífero roedor (rato). As DL50 de 2,6 microgramas/ml para Collosoma macropomum (tambaqui), 4,8 microgramas/ml para Oreochromis niloticus (tilápia), 14,2 microgramas/ml para Plecostomus sp (cascudo) e DL50 de 100,0 mg/kg para Rattus norvegicus (rato) denotam acentuadas diferenças entre os valores de DL50, principalmente entre os peixes e o rato. Isto possivelmente é devido a fatores farmaco-cinéticos que se relacionam com as diferentes barreiras teciduais encontradas pelos rotenóides quando administrados pela via oral em mamíferos.

The relation between hyperglycemia and diabetic neuropathy has already been demonstrated in some studies. Among the theories proposed for its etiology the oxidative stress stands out. The performance of nitric oxide as a link between the metabolic and vascular neuropathogenic factors that triggers the diabetic neuropathy has already been put forward. This study aimed to assess the quantification and measurements of the cell body profile area (CBPA) of NADPH-diaphorase reactive (NADPH-dp) myenteric neurons of the jejunum of diabetic rats (induced by streptozotocin) supplemented with Ascorbic Acid (AA). These changes in the myenteric neurons seem to be related to the gastrointestinal disturbances observed in diabetes mellitus (DM). Twenty male Wistar rats (Rattus norvegicus) were distributed in 4 groups (n=5): controls (C), control supplemented (CS), diabetic (D), and diabetic suplemented (DS). DM was induced by estreptozotocin (50mg/kg body wt). One week after the induction and confirmation of the DM (glycemia exam), animals of the groups CS and DS received 50mg of AA three times a week by gavage. After 90 days of experiment, the animals were anesthetized with lethal thiopental dose (40mg/kg) and the collected jejunum processed for the histochemistry NADPH-diaphorase technique. Whole-mount preparations were obtained for quantitative and morphometric analysis of the myenteric neurons. A quantity of jejunum neurons in the Group D (96±7.5) was not different (P>0.05) from Group DS (116±8.08), C (92±9.7), and CS (81±5.4), but in Group DS the quantity was higher (P<0.05) than in Group C and CS. The CBPA of neurons from Group D (189.50±2.68µm²) and DS (195.92±3.75µm²) were lower (P<0.05) than from Group C (225.13±4.37µm²) and CS (210.23±3.15µm²). The streptozotocin-induced DM did not change the jejunum-ileum area, the jejunum myenteric plexus space organization and the density of NADPH-dp neurons. The 50g AA-supplementation, three times a week, during 90 days, did not decrease hyperglycemia; however, it had a neuroprotective effect on the myenteric neurons, minimizing the increase on the CBPA of NADPH-dp neurons and increasing the amount of NADPD-dp neurons.
