Twenty adult patients presenting dermal cysticercosis without cerebral or ocular involvement were treated with praziquantel. The first eleven cases received 60 mg/kg/day and the last nine cases 30 mg/kg/day. In both groups the daily dose was split into three oral intakes 4 to 6 hours apart and the drug administration lasted for 6 consecutive days. The latter group of patients also got dexamethasone, 3 mg daily, from one day before until four days after the treatment period with praziquantel. The drug proved to be 100% efficacious as demonstrated histopathologically by the death of the cysticerci of Taenia solium (Cysticercus cellulosae) in serial biopsies taken from the 2nd week on after the end of treatment, as well as clinically by the steady disappearence of the dermal nodules during the 6 months following the therapy. Tolerance of praziquantel was good as the incidence and severity of side-effects were not relevant. The drug safety was confirmed through laboratory tests which failed to detect any abnormal findings related to the hematopoietic, liver and kidney functions.

INTRODUCTION: Human adenoviruses (HAdV) play an important role in the etiology of severe acute lower respiratory infection, especially in immunocompromised individuals. The aim of the present study was detect the HAdV through different methods: direct fluorescence assay (DFA) and nested-polymerase chain reaction (PCR-nested) from patients with acute respiratory infection (ARI) up to 7 days of symptoms onset.METHODS:Samples (n=643) were collected from different risk groups during from 2001 to 2010: 139 adults attended in an Emergency Room Patients (ERP); 205 health care workers (HCW); 69 from Renal Transplant Outpatients (RTO); 230 patients in hematopoietic stem cell transplantation (HSCT) program.RESULTS:Among all patients (n=643) adenovirus was detected on 13.2% by DFA and/or PCR: 6/139 (4.3%) adults from ERP, 7/205 (3.4%) from HCW samples, 4/69 (5.8%) from RTO and 68/230 (29.5%) from HSCT patients. Nested PCR showed higher detection (10%) compared to DFA test (3.8%) (p < 0.001). HSCT patients presented significantly higher prevalence of HAdV infection.CONCLUSIONS:Adenovirus detection through nested-PCR assay was higher. However the inclusion of molecular method in laboratorial routine diagnostic should be evaluated considering the reality of each specific health service.

Since 1958, we have studied experimental Chagas' disease (CD) by subcutaneous inoculation of 1,000 blood forms of Trypanosoma cruzi (Y strain) in Balb/C. mice. Evolution of parasitemia remained constant, beginning on the 5th and 6th day of the disease, increasing progressively, achieving a maximum on about the 30th day. After another month, only a few forms were present, and they disappeared from the circulation after the third month, as determined from direct examination of slides and the use of a Neubauer Counting Chamber. These events coincided with the appearance of amastigote nests in the tissues (especially the cardiac ones), starting the first week, and following the Gauss parasitemia curve, but they were not in parallel until the chronic stage. In 1997, we began to note the following changes: Parasites appeared in the circulation during the first week and disappeared starting on the 7th day, and there was a coincident absence of the amastigote nests in the tissues. A careful study verified that young forms in the evolutionary cycle of T. cruzi (epi + amastigotes) began to appear alongside the trypomastigotes in the circulation on the 5th and 7th post-inoculation day. At the same time, rounded, oval, and spindle shapes were seen circulating through the capillaries and sinusoids of the tissues, principally of the hematopoietic organs. Stasis occurs because the diameter of the circulating parasites is greater than the vessels, and this makes them more visible. Examination of the sternal bone marrow revealed young cells with elongated forms and others truncated in the shape of a "C" occupying the internal surface of the blood cells that had empty central portions (erythrocytes?). We hypothesize that there could be a loss of virulence or mutation of the Y strain of Trypanosoma cruzi.

B19 infection offers some general lessons about human viruses and their possible effects on the human host, as follows: (1) Ubiquitous apparently benign viruses may have severe effects on a compromissed host. The virus may be invariable but the host can have diverse susceptibilities. (2) B19 and some other human viruses (through for none is the evidence so clear as for B19) have narrowly targetted effects. The host cell of B19 is a specialised progenitor of mature red cells: impairment of the function of this cell by B19 may cause profound anaemia. (3) The 'normal'host response to B19 may also cause disease, though this is slef limiting. (4) The effects of malfunction of the virus'target cell are exacerbated when the immune response is impaired by congenital or acquired immunodeficiency, immunosupressive therapy or, in the case of the fetus, developmental immaturity that allows the virus to persist.

During Schistosoma mansoni infection, there is morphological evidence of involvement of various hematopoietic growth factors, which cause eosinophil, neutrophil, megakaryocytic and erythroid extramedullary foci in the liver, lymph nodes and omental and mesenteric milky spots. While the eosinophil metaplasia in the periphery of hepatic granulomas roughly reproduced the intensity of the medullary eosinopoiesis, the neutrophil metaplasia, on the contrary, was more intense during the period of neutrophil depression in the bone marrow. This fact suggests that extramedullary hematopoietic foci are locally regulated, and amplify and/or compensate the systemic hematopoietic response during the infection.

Allergen-induced bone marrow responses are observable in human allergic asthmatics, involving specific increases in eosinophil-basophil progenitors (Eo/B-CFU), measured either by hemopoietic assays or by flow cytometric analyses of CD34-positive, IL-3Ralpha-positive, and/or IL-5-responsive cell populations. The results are consistent with the upregulation of an IL-5-sensitive population of progenitors in allergen-induced late phase asthmatic responses. Studies in vitro on the phenotype of developing eosinophils and basophils suggest that the early acquisition of IL-5Ralpha, as well as the capacity to produce cytokines such as GM-CSF and IL-5, are features of the differentiation process. These observations are consistent with findings in animal models, indicating that allergen-induced increases in bone marrow progenitor formation depend on hemopoietic factor(s) released post-allergen. The possibility that there is constitutive marrow upregulation of eosinophilopoiesis in allergic airways disease is also an area for future investigation.

Um experimento de cruzamentos dialélicos 5 x 5 foi conduzido na Estação Experimental de Tietê, SP, com o objetivo de avaliar a capacidade combinatória de nogueira macadâmia (Macadamia integrifolia Maiden & Betche), cultivares IAC Campinas A, IAC 2-23 A, IAC 4-12 B, IAC 4-20 e IAC 5-10. A avaliação de frutos fixados aos 14 dias após polinização manual revelou que todas as cultivares apresentaram auto-incompatibilidade. Houve diferenças significativas quanto a capacidades geral (CGC) e específica de combinação (CEC) e efeito recíproco (ER) entre elas. A cultivar IAC Campinas A reduziu a frutificação inicial em todos os cruzamentos em que participou como progenitor feminino. Os melhores resultados de frutificação inicial foram apresentados pelos cruzamentos entre as cultivares: IAC 4-20 x IAC Campinas A, IAC 4-12 B x IAC 2-23 A, IAC 4-20 x IAC 4-12 B, IAC 5-10 x IAC 2-23 A, IAC 5-10 x IAC 4-20, IAC 4-20 x IAC 2-23 A e IAC 5-10 x IAC 4-12 B.

Se evaluó la variabilidad genética para la vida postcosecha y el peso de los frutos en veintiséis familias F3 de un híbrido interespecífico entre la cv. Caimanta de Lycopersicon esculentum Mill. y la línea LA722 de L. pimpinellifolium (Jusl.) Mill. con el objeto de obtener genotipos divergentes para ambos caracteres. Los promedios de días de vida postcosecha fueron 8, 15, 18 y 13 y de peso en gramos fueron 52,17, 0,94, 4,45 y 6,41 para cv. Caimanta, LA722, la F1 y la generación F3, respectivamente. Ninguna familia fue similar al progenitor `Caimanta' para el carácter días de vida postcosecha y sólo una familia tuvo una vida postcosecha como la F1. Para el peso de los frutos, cuatro familias fueron similares a la F1 y ninguna fue como los progenitores. La familia con peso de los frutos con 20,15 g y vida postcosecha de 21,5 días fue la que presentó mayores valores. El menor peso de los frutos fue en una familia con 2,81 g y el de menor vida postcosecha en una con 9,98 días. La amplia variabilidad genética encontrada para ambos caracteres entre las familias permitirá obtener genotipos divergentes.

Com o objetivo específico de obter uma população segregante, visando à avaliação na frutificação efetiva, quantidade de sementes normais e peso e diâmetro de frutos oriundos de polinização aberta e dirigida, foram realizados cruzamentos entre dois porta-enxertos de macieira, Marubakaido (Malus prunifolia Borkh.) e M.9 (Malus pumila Mill.) na Estação Experimental de São Joaquim -- EPAGRI/SC. 'Marubakaido' apresentou florescimento mais precoce e mais prolongado que a cultivar M.9. A percentagem de frutificação efetiva, para o porta-enxerto 'M.9' usado como progenitor feminino, foi de 26% e 32%, e usado como doador de pólen, foi de 5% e 25%, no anos de 1999 e 2000, respectivamente. Foram encontrados frutos deformados nos cruzamentos de 'M.9' X 'Marubakaido' e com um maior número de sementes atípicas quando comparadas ao sistema de polinização aberta. O mesmo não ocorreu no cruzamento recíproco. O peso e o diâmetro dos frutos foram maiores quando houve a polinização dirigida em 'M.9', o mesmo não acontecendo com 'Marubakaido', comparativamente à polinização aberta. A análise via contrastes ortogonais revelou valores de F significativos também para número de sementes normais e anormais frente aos dois sistemas de polinização em 'M.9'.

No Brasil, foi desenvolvido um novo híbrido de mamão obtido através de melhoramento do cruzamento entre um progenitor do Grupo Formosa e um progenitor do Grupo Solo. Recentemente, os produtores de mamões do Rio Grande do Norte introduziram o cultivo desse híbrido em seus pomares: no entanto, os estudos acerca do potencial de conservação desses frutos restringem-se aos testes realizados nas próprias empresas. O presente trabalho propôs-se a avaliar a vida útil pós-colheita do mamão híbrido UENF/Caliman 01 cultivado no Estado do Rio Grande do Norte. Os frutos foram provenientes de um plantio comercial, localizado no município de Ceará-Mirim-RN. Os mamões foram colhidos no estádio de maturação I (menos de 15% da superfície da casca amarela). No galpão de embalagem, os frutos passaram por lavagem, seleção, tratamentos hidrotérmico, imersão em fungicida e cera. No laboratório da Universidade Federal Rural do Semi-Árido (Mossoró-RN), os frutos foram armazenados à temperatura de 25º C ± 2º C e umidade relativa de 55% ± 5%. Aos 0; 2; 4; 6; 8 e 10 dias de armazenamento, realizaram-se avaliações sobre as seguintes variáveis: aparência externa e interna, coloração da casca, perda de massa, firmeza, sólidos solúveis, acidez titulável, pH e açúcares. Esse híbrido apresenta características fenotípicas do Grupo Formosa, com frutos alongados, massa média de 1,28 kg, polpa com espessura de 2,74 cm, coloração vermelha e conteúdo elevado de sólidos solúveis (13,65%) e açúcares (10,53%). As variáveis de qualidade analisadas limitaram a vida útil pós-colheita do híbrido UENF/Caliman 01 em oito dias.

Chromosome studies were performed in two varieties of Mangifera indica L. (mango), 'IAC-140 Espadona' and in its progenitor 'Espada Stahl'. Both varieties showed 2n=40 chromosomes though the karyotype formulae were 8m + 10sm + 2sm s for 'Stahl' and 7m + 11sm + 2sm s for 'IAC-140'. The varieties showed moderate karyotype asymmetry which was estimated according to four different indices. Both varieties exhibited three chromosome pairs with silver impregnation after NOR-banding. The number of nucleoli within interphase cells varied from one, the commonest, to eight. The nucleolus persistent phenomenon was observed in more than 22% of metaphase cells of both varieties, seeing that in 'Stahl', up to two nucleoli were evidenced. This variety also showed one nucleolus in several anaphase cells. The studies were suitable for evidencing diversity at chromosomal level between these two varieties.

The main objective of this work was to evaluate the diversification of sorghum (Sorghum bicolor) populations as a way to manage resistance to the sorghum anthracnose fungus Colletotrichum graminicola. A total of 18 three-way hybrids were obtained by crossing six single cross male-sterile F1 hybrids, derived by crossing A (non restorer sterile cytoplasm) and B (non restorer normal cytoplasm) lines, with three fertile R (restorer) lines, previously evaluated for their differential reaction to the pathogen. Variation in the level of resistance was observed, as indicated by the values of the area under the disease progress curve (AUDPC) obtained for each hybrid. Lines contributed differently to the level of resistance of each hybrid. All hybrids in which CMSXS169R was the male progenitor were classified as highly resistant. Some hybrids had a level of resistance superior to the maximum levels of each line component individually.

Solanum glaucophyllum (Sg) [= S. malacoxylon] is a calcinogenic plant inducing "Enzootic Calcinosis" in cattle. The 1,25-dihydroxyvitamin D3, its main toxic principle, regulates bone and calcium metabolism and also exerts immunomodulatory effects. Thymocyte precursors from bone marrow-derived progenitor cells differentiate into mature T-cells. Differentiation of most T lymphocytes is characterized not only by the variable expression of CD4/CD8 receptor molecules and increased surface density of the T cell antigen receptor, but also by changes in the glycosylation pattern of cell surface glycolipids or glycoproteins. Thymocytes exert a feedback influence on thymic non-lymphoid cells. Sg-induced modifications on cattle thymus T-lymphocytes and on non-lymphoid cells were analysed. Heifers were divided into 5 groups (control, intoxicated with Sg during 15, 30 or 60 days, and probably recovered group). Histochemical, immunohistochemical, lectinhistochemical and morphometric techniques were used to characterize different cell populations of the experimental heifers. Sg-poisoned heifers showed a progressive cortical atrophy that was characterized using the peanut agglutinin (PNA) lectin that recognizes immature thymocytes. These animals also increased the amount of non-lymphoid cells per unit area detected with the Picrosirius technique, WGA and DBA lectins, and pancytokeratin and S-100 antibodies. The thymus atrophy found in intoxicated animals resembled that of the physiological aging process. A reversal effect on these changes was observed after suppression of the intoxication. These findings suggest that Sg-intoxication induces either directly, through the 1,25-dihydroxyvitamin D3 itself, or indirectly through the hypercalcemia, the observed alteration of the thymus.

A morphological and cell culture study from nasal mucosa of dogs was performed in order to establish a protocol to obtain a cell population committed to neuronal lineage, as a proposal for the treatment of traumatic and degenerative lesions in these animals, so that in the future these results could be applied to the human species. Twelve mongrel dogs of 60-day aged pregnancy were collected from urban pound dogs in São Paulo. Tissue from cribriform ethmoidal lamina of the fetuses was collected at necropsy under sterile conditions around 1h to 2h postmortem by uterine sections and sections from the fetal regions described above. Isolated cells of this tissue were added in DMEM/F-12 medium under standard conditions of incubation (5% CO², >37ºC). Cell culture based on isolated cells from biopsies of the olfactory epithelium showed rapid growth when cultured for 24 hours, showing phase-bright sphere cells found floating around the fragments, attached on culture flasks. After 20 days, a specific type of cells, predominantly ellipsoids or fusiform cells was characterized in vitro. The indirect immunofluorescence examination showed cells expressing markers of neuronal precursors (GFAP, neurofilament, oligodendrocyte, and III â-tubulin). The cell proliferation index showed Ki67 immunostaining with a trend to label cell groups throughout the apical region, while PCNA immunostaining label predominantly cell groups lying above the basal lamina. The transmission electron microscopy from the olfactory epithelium of dogs revealed cells with electron-dense cytoplasm and preserving the same distribution as those of positive cell staining for PCNA. Metabolic activity was confirmed by presence of euchromatin in the greatest part of cells. All these aspects give subsidies to support the hypothesis about resident progenitor cells among the basal cells of the olfactory epithelium, committed to renewal of these cell populations, especially neurons.

The umbilical cord blood (UCB) is an important source of pluripotent stem cells, which motivated researches on ontogeny and transplantation. The morphological characterization of umbilical cord cells is the first step to establish subsequent experiments on these areas. Although some information on humans can be found, no data on UCB is available for bovines. Therefore, this work is the first attempt to conduct an ultrastructural characterization of bovine umbilical cord blood. Blood was collected from the umbilical cord of twenty fetuses by punction of the umbilical vein. Samples were processed for whole leucocytes observation by centrifugation and the buffy coat was collected. Cells were washed and pelleted and prepared according to the standard protocol of the transmission electron microscopy. The presence of cells with morphologic characteristics compatible with the precursors from the erythrocytic, neutrophilic, eosinophilic, basophilic, and lymphocytic lineages was observed. Atypical cells with peculiar morphological features, strongly similar to apoptotic cells, were seen. Bovine neutrophils with three types of cytoplasmic granules were also found in the blood. The ultrastructural characteristics of observed bovine UCB cells where similar to those found in other species, suggesting that bovines could possibly constitute an experimental model for approaches on UCB cells research.
