Foram conduzidos experimentos de competição entre cultivares de mandioca (Manihot esculentaCrantz) em ecossistema de terra firme (Manaus - Amazonas), com objetivo de obter melhores cultivares de mandioca com relação a produtividade e teor de caroteno pró-vitamínico A. Raízes de sete cultivares de mandioca amarela foram selecionadas para identificação e quantificação de carotenos com atividade de vitamina A, mediante o método descrito por Rodriguez. Observou-se, também, as perdas de pró-vitamina A pelo processamento e armazenamento de farinha. Constatou-se a presença de três isômeros do β - caroteno (o 13 - eis -β - caroteno, o β - caroteno todo trans e o 9 - eis -β - caroteno U). Quanto aos teores de vitamina A, expressos em equivalente de retinol, nas raízes variaram de 4,4 a 18,8. Com relação a perda de atividade da vitamina A pelo processamento variou de 25,0 a 40,0 %, enquanto que o armazenamento por 6 meses em sacos plásticos transparentes, à temperatura ambiente e à exposição de luz, resultou em degradação total de seus carotenóides. Os resultados permitem concluir que três cultivares (IM 232; IM 104 e BGM 021) apresentaram os maiores teores de pró-vitamina A.

Procedeu-se uma revisão de toda produção científica do Amazonas, da área da nutrição nas últimas décadas. A análise deste material, consubstanciada com as últimas pesquisas realizadas pelo grupo da Nutrição do INPA viabilizou a composição do diagnóstico da hipovitaminose A na região Amazônica. A investigação clínico-oftalmológica, envolveu 487 crianças de 2 a 5 anos, de ambos os sexos, oriundas das diferentes localidades amazônicas: Nhamundá, Alvarães, Anamã, Beruri e Benjamin Constant. Os níveis de retinol sérico foram determinados, exclusivamente, em 48 crianças do município de Nhamundá. A investigação clínica não evidenciou nenhum sinal e/ou sintoma pertinentes a carência de vitamina A. Níveis de ret-inol sérico, considerados normais e altos, foram registrados em 75,0% e 25,0%, respectivamente do universo estudado. O somatório destes resultados com as informações retrospectivas procedentes da literatura regional não permitem afirmar categoricamente a existência de hipovitaminose A como problema de Saúde Pública na região Amazônica.

Esse estudo teve como objetivo determinar os teores de vitamina A no Leite Humano Materno (LHM), de 50 mães doadoras do Banco de Leite Humano (BLH), da Maternidade Ana Braga, em Manaus-AM. As amostras foram coletadas semanalmente e mantidas congeladas até a pasteurização. As análises de retinol e β-caroteno no leite humano foram realizadas antes e após o tratamento (pasteurização), mediante o método cromatográfico. Foram retiradas alíquotas de 5 mL aproximadamente de leite de cada amostra. Os pesos dos bebês ao nascer das mães doadoras de leite incluídos no estudo foram > 2500g. A concentração de vitamina A no leite tanto antes quanto após o processamento, não foram suficientes para suprir o requerimento (46,75 ± 6,28 μg/dL e 31,55 ± 8,29 μg/dL, respectivamente), de crianças que se alimentam exclusivamente do leite do BLH, notadamente as pré-termo, cujos requerimentos de vitamina A são maiores e a capacidade de ingerir leite é menor do que os nascidos a termo. A perda de vitamina A pelo processamento, em nosso estudo foi de 32,51%. Para garantir a formação de reservas hepáticas e a proteção contra deficiência de vitamina A, seriam necessárias concentrações dessa vitamina no leite superiores a 80 μg/dL.

The tocopherol content of Brazil nut oil from different Amazon regions (Manicoré-AM, Rio Preto da Eva-AM, São João da Baliza-RR, Caroebe-RR, Belém-PA, and Xapurí-AC) was investigated by normal-phase high-performance liquid chromatography. For all authentic oils, two isomers: α- and γ-tocopherols were observed (37.92-74.48 µg g-1, 106.88-171.80 µg g-1, respectively), and their levels were relatively constant among the oils having these geographic origins, which would enable to distinguish Brazil nut oil from other plant oils for authentication purposes. Commercial Brazil nut oils were also evaluated, and some of these oils demonstrated a tocopherol content that was very different from that of the authentic oils. Therefore, we suggest that the tocopherol profile of Brazil nut oil can be useful chemical marker for quality control and authentication.

Silymarin is the flavonoids extracted from the seeds of Silybum marianum (L) Gearth as a mixture of three structural isomers: silybin, silydianin and silychristin, the former being the most active component. Silymarin protects liver cell membrane against hepatotoxic agents and improves liver function in experimental animals and humans. It is generally accepted that silymarin exerts a membrane-stabilizing action preventing or inhibiting membrane peroxidation. The experiments with soybean lipoxygenase showed that the three components of silymarin brought about a concentration-dependent non-competitive inhibition of the lipoxygenase. The experiments also showed an analogous interaction with animal lipoxygenase, thus showing that an inhibition of the peroxidation of the fatty acid in vivo was self-evident. Silybin almost completely suppressed the formation of PG at the highest concentration (0.3 mM) and proved to be an inhibitor of PG synthesis in vitro. In our experiments, silybin at lower dose (65 mg/Kg) decreased liver lipoperoxide content and microsomal lipoperoxidation to 84.5% and 68.55% of those of the scalded control rats respectively, and prevented the decrease of liver microsomal cytochrome p-450 content and p-nitroanisole-0-demethylase activity 24 h post-scalding. Effects of silymarin on cardiovascular systen have been studied in this university since 1980. O. O silymarin 800 mg/Kg/d or silybin 600 mg/Kg/d reduced plasma total cholesterol, LDL-C and VLDL-C. They however, enhanced HDL-C in hyperlipenic rats. Further studies showed that silymarin enhanced HDL-C in hyperlipemic rats. Further studies showed that silymarin enhanced HDL-C but didn't affect HDL-C, a property of this component which is beneficial to treatment of atherosclerosis. The results showed silymarin 80 mg or silybin 60 mg decreased in vitro platelet aggregation (porcentagem) in rats. The maximal platelet aggregation induced by ADP declined significantly, and time to reach maximal platelet aggregation and five-minute disaggregation didn't change. In our experiments, iv silybin 22,4 mg/kg lowered the amplitude and duration of diastolic blood pressure (DBP) more than those of systolic (SBP), but the descending aortic blood flow, cardiac contractility and ECG did not change significantly in anesthetized open-chest cats. The results indicated a reduction of peripheral resistance and dilatatory action on the resistant blood vessels. These effects are beneficial to coronary heart disease. We also observed the effects of silybin on morphological change, the release of glutamic oxaloacetate aminotrasferase (GOT) and lactate dehydrogenase (LDH) as well as the radioactivity of 3H-TdR incorporated into DNA in normal cardiac cells and cells infected by coxsackie B5, virus os newborn rats. The results showed that silynin did not affect the morphology of normal cell, and that the pathological change of cells infected by virus was delayed and reduced as compared to control. We have investigated the effect of silybin on synthesis and release of LTs in the cultured porcine cerebral basilar arteries (PCBA). Silybin 100 and 500 µmol/L declined the amounts of LTs released from the PCBA incubsated in the presence of A 23187, AA and indomenthacin. The result suggests that silybin can inhibit the activity of 5-lipoxygenase of cerebral blood vessel and may protect the brain from ischemia.

The finding of Panstrongylus geniculatus nymphs inside a house in northeastern Antioquia, Colombia, and the reports related to their increasing presence in homes suggest the need for surveillance methods for monitoring the invasion processes. We analyzed the morphological differences between a wild population and its laboratory descendants, using the techniques of geometric morphometry, with the idea that such differences might parallel those between sylvatic and synanthropic populations. The analyses over five generations showed differences in size but not in shape. Head size and wing size were both reduced from sylvatic to laboratory populations, but the decrease in head size occurred only up to the second generation while the decrease in wing size proceeded up to the fifth generation. In contrast, although a decrease in sexual size dimorphism has been proposed as a marker of colonization in human dwellings, we did not detect any significant loss of dimorphism between sexes of P. geniculatus over the five generations studied. We conclude that size changes may have a physiological origin in response to a change of ecotopes, but more than five generations may be required for the expression of permanent morphological markers of human dwellings colonization.

Capillary gas-liquid chromatography was used to analyse the cuticular hydrocarbons of three triatomine species, Triatoma dimidiata, T. barberi and Dipetalogaster maxima, domestic vectors of Chagas disease in Mexico. Mixtures of saturated hydrocarbons of straight and methyl-branched chains were characteristic of the three species, but quantitatively different. Major methylbranched components mostly corresponded to different saturated isomers of monomethyl, dimethyl and trimethyl branched hydrocarbons ranging from 29 to 39 carbon backbones. Sex-dependant, quantitative differences in certain hydrocarbons were apparent in T. dimidiata.

The purpose of this study was to examine the circulating filarial antigen (CFA) detected by the monoclonal antibody (mAb) Og4C3-ELISA in paired samples of serum and hydrocele fluid from 104 men with hydrocele, living in an endemic area of Wuchereria bancrofti. Nocturnal blood specimens were filtered and examined for microfilariae (MF) and ultrasound was used in order to identify the presence of adult worms (the filaria dance sign - FDS) in the lymphatic vessels of the scrotal area. Four groups were selected according to their parasitological status: group I - 71 MF- and FDS-; group II - 21 MF+ and FDS+; group III - 10 MF- and FDS+ and group IV- 2 MF+ and FDS-. CFA was identified simultaneously (fluid and serum) in 11 (15.5%), 21 (100%), 3 (30%), and 1 (50%) in groups I, II, III, and IV, respectively. In despite of high CFA+ level (antigen Og4C3) units/ml, the Geometrical Mean (GM) = 2696) in the sera of these 36/104 paired samples, when compared to the hydrocele fluid, (GM = 1079), showed a very good correlation between the CFA level in the serum and CFA level in the fluid (r = 0.731). CFA level in the serum of the 23 microfilaremics (groups II and IV) was extremely high (GM = 4189) and was correlated with MF density (r = 0.442). These findings report for the first time the potential alternative use of the hydrocele fluid to investigate CFA using the mAb Og4C3-ELISA.

A susceptible strain of Aedes albopictus derived from the Gainesville strain (Florida, USA) was established in our laboratory. The larvicidal efficacies of the neurotoxic insecticides temephos, permethrin and the pure cis and trans-permethrin isomers and the microbial insecticide Bacillus thuringiensis israelensis (Bti) against Ae. albopictus were estimated and compared to a susceptible strain of Aedes aegypti. The larvicidal effect of insect growth regulator pyriproxyfen was also evaluated in both mosquito strains. The median lethal concentration/median emergency inhibition values for Ae. aegypti and Ae. albopictus, respectively, were: temephos, 3.058 and 6.632 ppb, permethrin, 3.143 and 4.933 ppb, cis-permethrin, 4.457 and 10.068 ppb, trans-permethrin, 1.510 and 3.883 ppb, Bti, 0.655 and 0.880 ppb and pyriproxyfen, 0.00774 and 0.01642 ppb. Ae. albopictus was more tolerant than Ae. aegypti to all six larvicides evaluated. The order of susceptibility for Ae. aegypti was pyriproxyfen > Bti > trans-permethrin > temephos > permethrin > cis-permethrin and for Ae. albopictus was pyriproxyfen > Bti > trans-permethrin > permethrin > temephos > cis-permethrin. Because both species can be found together in common urban, suburban and rural breeding sites, the results of this work provide baseline data on the susceptibility of Ae. albopictus to insecticides commonly used for controlling Ae. aegypti in the field.

O objetivo deste trabalho foi avaliar o efeito da suplementação de vitamina A na alimentação de juvenis de pacu cultivados em tanques-rede. Foram utilizados 2.000 peixes, com peso inicial médio de 66,93±15,03 g, distribuídos em delineamento inteiramente casualizado, em 20 tanques-rede de 5,0 m³, com cinco tratamentos e quatro repetições. As rações foram formuladas com 0, 3.000, 6.000, 9.000 e 12.000 UI de vitamina A por quilograma de dieta, por meio da suplementação com acetato de retinol. O arraçoamento foi realizado às 8:30, 13:30 e 17:30 h, até a saciedade aparente dos animais. Não foram observadas diferenças quanto aos parâmetros de desempenho produtivo, rendimento e composição química da carcaça, e aos parâmetros hematológicos e lipídios no fígado dos peixes que foram alimentados com diferentes níveis de vitamina A. A suplementação de vitamina A em dietas para juvenis de pacu criados em tanques-rede não influencia o desempenho produtivo dos peixes.

Pimaradienes, including isopimaradienes, with an endocyclic double bond between C-9 and C-11 are uncommon compounds in nature. The diterpenoid pimar-9(11),15-dien-19-oic acid (1) was isolated from Mikania triangularis (Asteraceae) and the correct stereochemistry of 1was established by ¹H and 13C NMR studies of several oxidative products, mainly epoxides, of this compound and its double bond isomers.

1,3,4-thiadiazolium-2-aminides and their isomers 1,3,4-triazolium-2-thiolates have been synthesized via anhydroacylation reactions. This work presents a study by infrared monitoring of the reaction between substituted aroyl acid chlorides and 1,4-diphenylthiosemcarbazide. The intermediates and products were isolated, purified and charaterized by IR and 13C NMR spectroscopy. The increasing or decreasing in intensity of characteristic stretching bands indicated the rate dependence on the electronic nature of substituents. The results also demonstrate that 1,3,4-triazolium-2-thiolates are obtained in anhydrous conditions whereas presence of water leads to a mixture of the isomers.

The presence of low carbonyl compounds (C1-C4) and glutaraldehyde was observed in 14 work-locations in São Paulo city, Brazil, during January to July 1997 period. The quantification of other carbonyls was not possible due to a sampling artifact caused probably by undesirable reactions between the ambient ozone and the organic substract of C18 cartridge used to collect carbonyls. High indoor concentrations, compared to outdoor levels, were observed for all compounds. Formaldehyde was the most abundant species (29 ppb) followed by acetone and acetaldehyde (25 and 17 ppb) with a small contribution of propanal, crotonaldehyde and C4 isomers (0.7 to 1.5 ppb) when related to the total indoor carbonyls level. Glutaraldehyde was observed only in two different occupational locations in a hospital at high concentrations (121 ppb). In general, high individual carbonyl levels as well as total carbonyls levels found in several locations indicated an unpropitious air quality for the occupants of these non-industrial sites.

A procedure for the nitration of phenol in a semi-micro scale, followed by separation of the formed orto- and para-nitrophenol isomers by column chromatography, is described. All the experiment, including determination of the melting point of the isolated products, require a period of 4 hours, and it is suitable for organic chemistry laboratory undergraduate courses.

A sodium mordenite zeolite (Na-MOR) was synthesized and modified by dealumination with chloridric acid solution (H-MOR). X-Ray Diffraction (XRD), Inductive Coupled Plasm (ICP) and Scanning Electron Micrograph (SEM) techniques were used for sample characterization. The zeolite catalytic behavior was evaluated through toluene disproportionation at 435°C. It was verified that mordenites were very selective for the disproportionation reaction and the samples with higher aluminum content showed larger initial activity, however, these samples showed too a higher deactivation velocity due to a blockage of the unidimensional porous system of the zeolite by coke deposits. The selectivity to xylene isomers was practically not influenced by the Si/Al ratio and changed with the time on stream, due to coke formation. Transition state shape selectivity inside the mordenite pores is also discussed.
