Coherent vortices in turbulent mixing layers are investigated by means of Direct Numerical Simulation (DNS) and Large-Eddy Simulation (LES). Subgrid-scale models defined in spectral and physical spaces are reviewed. The new "spectral-dynamic viscosity model", that allows to account for non-developed turbulence in the subgrid-scales, is discussed. Pseudo-spectral methods, combined with sixth-order compact finite differences schemes (when periodic boundary conditions cannot be established), are used to solve the Navier- Stokes equations. Simulations in temporal and spatial mixing layers show two types of pairing of primary Kelvin-Helmholtz (KH) vortices depending on initial conditions (or upstream conditions): quasi-2D and helical pairings. In both cases, secondary streamwise vortices are stretched in between the KH vortices at an angle of 45° with the horizontal plane. These streamwise vortices are not only identified in the early transitional stage of the mixing layer but also in self-similar turbulence conditions. The Re dependence of the "diameter" of these vortices is analyzed. Results obtained in spatial growing mixing layers show some evidences of pairing of secondary vortices; after a pairing of the primary Kelvin-Helmholtz (KH) vortices, the streamwise vortices are less numerous and their diameter has increased than before the pairing of KH vortices.

Bidens gardneri is a very common herbaceous species in the cerrados of the state of São Paulo, whose seeds become light sensitive at 25°C only. Achenes of this species were stored in refrigerator at 4°C and in cerrado soil and in forest soil. The field experiments were carried out in the cerrado at the Reserva Biológica e Estação Experimental de Moji Guaçu, in Moji Guaçu and in the forest of the Instituto de Botânica, in São Paulo, Brazil. Achenes of B. gardneri vary in size and achenes from 7 to 12 mm long were used. Achenes stored for up to 6 months at 4°C showed light sensitivity; after 9 months storage, the difference in germination between light and darkness had disappeared for the smallest and the largest achenes used. Seeds of B. gardneri germinated during the period of storage in soil; the number of germinated seeds increased over the storage time, while the number of intact achenes decreased for the same period, no matter if the experiment was being carried out in the cerrado or in the forest. Therefore, the achenes germinated in soil in darkness. Light sensitivity was lost in intact achenes that had been stored in soil for three months.

Bidens gardneri is a herbaceous species of the cerrados, whose seeds are light sensitive at 25 °C, but they become indifferent to light when stored in soil. In this work the effects of moisture content, temperature and light (during storage) upon light sensitivity during germination were studied. Ripe achenes were collected in the cerrados of Itirapina and Moji Guaçu, State of São Paulo, Brazil. The storage conditions of the achenes varied in each experiment. Achenes were stored in darkness or light, in closed bottles, at 4 °C, 20/30 °C or 25 °C. Achenes were imbibed for 24 h at 4 °C, 25 °C or 20/30 °C (in darkness) and then stored for 1, 10, 20, 30 and 40 days (40 days only for 4 °C and 25 °C). Germination tests were conducted at 25 °C and 20/30 °C. The achenes not previously imbibed showed sensitivity to light during germination. High moisture content did not affect light sensitivity of the achenes during germination but high moisture content together with storage temperatures of 25 °C and 20/30 °C had a deleterious effect upon the longevity of the achenes. Alternate temperatures during germination did not change the light sensitivity of newly collected achenes from Itirapina but changed the light sensitivity of the achenes stored imbibed at 4 °C in darkness. Alternate temperatures during storage of achenes with low moisture content did not change their photoblastism when germination was carried out at 25 °C. Alternate temperatures during storage of achenes with high moisture content followed by alternate temperatures during germination changed the light sensitivity of the achenes.

Fructans of the inulin type are the major reserve carbohydrates in tuberous roots of Viguiera discolor, a perennial herb native to the cerrado. Changes in molecular mass of the polymer, followed by releasing free fructose suggested that hydrolysis could be related to the sprouting of the buds after the dormant period, when aerial parts of the plant are naturally absent. Excision of aerial parts resulted in the increase of fructan 1-exohydrolase (1-FEH) activity in tuberous roots after sprouting. 1-FEH was partially purified from this material by binding to ConA-Sepharose and the highest activity was detected at pH 5.4 and between 20 and 40 °C. Values of Km for V. discolor inulin could not be determined since no saturation was observed up to 10%. The study of the kinetics of the 1-FEH activity showed that it does not follow Michaelis-Menten and apparently presents allosteric behaviour, as data fits in the Hill equation. The 1-FEH from V. discolor is a glycoprotein, more active on low molecular mass fructans than on high molecular mass inulin from the same species.

Com o objetivo de colaborar na delimitação taxonômica de Aechmea, subgênero Lamprococcus, foram estudadas as folhas de 25 táxons morfologicamente relacionados entre si (13 de Aechmea, três de Ronnbergia E. Morren & André, quatro de Araeococcus Brongn. e cinco de Lymania Read). Apesar das folhas serem todas hipoestomáticas e possuírem mesofilo dorsiventral portador de hipoderme, elas exibem características que podem ser utilizadas na delimitação de cada representante. Essas características incluem: posição dos estômatos na superfície foliar, presença ou não de oclusão nas câmaras subestomáticas, ocorrência ou não de grupos de fibras dispersos no mesofilo, natureza da parede das células que compõem as bainhas e extensões dos feixes vasculares e morfologia das células dos diafragmas, que interrompem os canais de aeração. A distribuição desses caracteres, nos diferentes táxons, permitiu elaborar uma chave de identificação, com base apenas na estrutura foliar.

O subgênero Chevaliera está representado por 21 espécies, das quais grande parte ocorre no Brasil, habitando diferentes ecossistemas. Com o objetivo de auxiliar a taxonomia das espécies, são apresentados dados da anatomia foliar de 11 espécies do subgênero. Entre os caracteres mais relevantes destacam-se: posição dos estômatos, câmara subestomática com células espessadas, hipoderme, número de camadas do parênquima aqüífero, o formato das células dos canais de aeração e distribuição das fibras no mesofilo.

Zephyranthes Herb. is a taxonomically complex and cytologically variable group, with about 65 species of Neotropical distribution. Chromosome number variability in 32 individuals of a Zephyranthes sylvatica population from Northeast Brazil was investigated. Three cytotypes were found: 2n = 12 (one metacentric, four submetacentric and one acrocentric pairs), in 24 individuals; 2n = 12 + 1B, in five and three individuals with 2n = 18, a triploid cytotype. All diploid individuals showed chromosomes with polymorphism in pair one and two, while in triploids this polymorphism was observed in all chromosome triplets, generally with two homomorphic chromosomes and a higher or lower heteromorphic chromosome. All individuals had reticulated interfasic nucleus and a slightly asymmetric chromosome complement, with one metacentric chromosome pair and the others more submetacentric to acrocentric. These data confirm the cytological variability previously registered for the genus. Mechanisms involved in karyotypic evolution in this population are discussed.

Este trabalho descreve a morfologia, anatomia e ontogênese do pericarpo e pápus de Bidens gardneri Baker e B. pilosa L., espécies de Heliantheae (Asteraceae). Verificou-se que as duas espécies são muito similares, possuem ovário ínfero, bicarpelar, sincárpico e unilocular. O pericarpo se desenvolve a partir do alongamento das células, sem fase multiplicativa. O exocarpo é unisseriado e recoberto por cutícula; ocorrem tricomas tectores multicelulares, que persistem até a maturidade. O mesocarpo externo possui de uma a três camadas de células parenquimáticas em B. pilosa e de uma a cinco células em B. gardneri. A região mediana do mesocarpo possui de quatro a oito camadas em ambas as espécies e, entre a região mediana do mesocarpo e o mesocarpo interno, estão imersos feixes vasculares colaterais. O endocarpo é unisseriado, mas nas regiões de fusão dos carpelos, é bi a trisseriado. Ocorre deposição de fitomelanina entre o mesocarpo externo e o mediano, mais precoce em B. pilosa. No ápice do pericarpo, observa-se a protuberância onde se insere o pápus aristado, formado por três a quatro cerdas rígidas, que apresentam tricomas voltados para a base. Na base do fruto, ocorre uma pequena projeção, o carpopódio, onde o fruto insere-se no eixo da inflorescência. Na maturidade, o pericarpo das duas espécies é desidratado, e, em B. pilosa, as camadas do pericarpo ficam mais colapsadas; em B. gardneri, também ocorre a desidratação, mas algumas células do exocarpo e mesocarpo externo permanecem distintas.

A espécie Dyckia brevifolia Baker apresenta populações disjuntas e distribuição restrita. Dyckia brevifolia é uma espécie policárpica com propagação clonal. A biologia reprodutiva desta espécie foi estudada nas margens do Rio Itajaí-Açu, Santa Catarina, Brasil. As características florais, a produção de néctar e os visitantes florais foram estudados. Para caracterizar o sistema reprodutivo foram conduzidos cinco tratamentos: agamospermia, autopolinização espontânea, autopolinização manual, polinização cruzada e controle. Cada inflorescência apresentou 60,4 ± 14,5 flores e 58,3 ± 13,3 frutos e a razão fruto/flor foi de 0,97. O número médio de sementes por fruto foi de 129,6 ± 24,3. As flores abrem da base para o ápice da inflorescência e o número de flores abertas por dia por inflorescência foi em média de 6,8 ± 1,2. A antese floral ocorre ao longo do dia e a flor tem duração de um dia e meio. O volume e a concentração médios do néctar foram de 30,5 µL e 25,7%, respectivamente. Os principais visitantes florais foram abelhas, beija-flores e borboletas, sendo o beija-flor Amazilia versicolor Vieillot o principal polinizador. Esta espécie também foi polinizada por abelhas dos gêneros Xylocopa e Bombus. Com relação ao sistema reprodutivo, os resultados indicam que D. brevifolia é autocompatível e que a agamospermia pode ocorrer. A autocompatibilidade apresentada pela espécie, bem como o comportamento dos visitantes florais indicam que as principais formas de polinização promovidas são a autopolinização e a geitonogamia.
Genetic engineering of baker's and wine yeasts using formaldehyde hyperresistance-mediating plasmids

Yeast multi-copy vectors carrying the formaldehyde-resistance marker gene SFA have proved to be a valuable tool for research on industrially used strains of Saccharomyces cerevisiae. The genetics of these strains is often poorly understood, and for various reasons it is not possible to simply subject these strains to protocols of genetic engineering that have been established for laboratory strains of S. cerevisiae. We tested our vectors and protocols using 10 randomly picked baker's and wine yeasts all of which could be transformed by a simple protocol with vectors conferring hyperresistance to formaldehyde. The application of formaldehyde as a selecting agent also offers the advantage of its biodegradation to CO2 during fermentation, i.e., the selecting agent will be consumed and therefore its removal during down-stream processing is not necessary. Thus, this vector provides an expression system which is simple to apply and inexpensive to use
