Aminoalcohols have found important applications in synthetic and medicinal chemistry, being used as chiral building blocks for the synthesis of many biologically active compounds. This class of compounds has been also used as chiral auxiliaries and ligands in asymmetric synthesis. Due to the importance of aminoalcohols in the treatment of several diseases, such as tuberculosis, the aim of this article is the synthesis and preliminary evaluation against tuberculosis of six aminoalcohols in 5 or 6 steps using D-mannitol as starting material, which is a useful carbohydrate employed in many syntheses.

A tandem syn-selective conjugate addition - Nef reaction was observed when phenylnitromethane and oxygenated derivatives were allowed to react with an enoate derived from D-mannitol at rt in the presence of TBAF or DBU. While nitro-adducts predominate after 4h of reaction, the corresponding ketones were the main products after 12-24h of reaction. The Nef reaction occurred without racemization of the stereogenic center generated in the conjugate addition step.

In this work the separation of the chiral anesthetic compounds ketamine and bupivacaine was development using two chiral stationary phases (CSP). Ketamine enantiomers were well separate in the polysaccharide-based CSP (microcrystalline cellulose triacetate - MCTA) while bupivacaine in the tartardiamide-based CSP (Kromasil CHI-TBB). In both cases, the effect of temperature was investigated under analytical conditions. An improvement in the separation performance with temperature was observed. Thermodynamic parameters were evaluated by the van't Hoff plot. We concluded that enthalpic effects controlled the retention in these chiral columns. The enantiomers of ketamine and bupivacaine were separated under overloaded conditions with a good performance.

D-mannitol is used in several fields including food, pharmaceuticals, cosmetics and textiles being an important carbohydrate found widespread in nature. Due to its chirality, it is largely used in organic synthesis with several applications, such as ligands, polymers, chiral pool, for preparation of small chiral building blocks, key intermediates in total synthesis. In this context, the aim of this review is to highlights recent applications of D-mannitol, especially in total synthesis.

The philosophy of organocatalysis is based on the utilization of organic compounds to catalyze organic transformations without the intervention of metals. This area has attracted much attention of the synthetic chemistry community on the last years, which can be confirmed by the explosion of published papers dealing with this subject. Phosphorus compounds, urea and thiourea derivatives, alkaloids, guanidine derivatives, for example, have already been used as organocatalysts. In this review we have focused on the use of chiral amines as organocatalyst. We have also chosen some outstanding examples to demonstrate the potentiality of this strategy in the synthesis of natural products and biologically active compounds.

The synthesis of chiral acetylenic regioisomers was described by using an appropriate intermediate such as isopropylidene glycerol, a synthon widely used in the enantioselective syntheses. This intermediate was prepared from D-mannitol. The nine obtained compounds have been characterized by their respective spectral data. The mixture of chiral acetylenic alcohols showed activity against Escherichia coli when tested through the monitoring of CO2 released during microbial respiration by using a conductimetric system.

The volatile oil obtained from the leaves of Hydrocotyle bonariensis Lam (Araliaceae) was analyzed by GC, Chiral-GC and GC-MS. It was identified 14 compounds and the monoterpene (+)-limonene (53.6%) and sesquiterpene γ-muurolene (10.5%) were the main components. The allelopathic effects of the oil were evaluated against two seeds, Lactuca sativa and Allium cepa. The results show that the oil exhibited inhibition effects in the germination and seedling growth of plants species relative to the control.

This work describes an overview of our synthesis of pteridic acids A and B and discloses some interesting results related to the lithium enolate-mediated aldol reaction used as key step to set up the C5-C15 fragment of these natural products. This first example, as far we know, of an aldol reaction between a chiral enolate of a (Z) enone and a chiral aldehyde has driven us to a series of experiments showing the remarkable relation between enolization selectivity and reaction conditions.

The development of Chiral Stationary Phases (CSPs) for high performance liquid chromatography has been studied by various researches around the world, especially, since 1980. This simple interest has been transformed into a tool of great technological value for the industrial community and scholars in general providing the existence of several CSPs, which act through different mechanisms of chiral discrimination. This paper describes the main types of CSPs that are used for the resolution of the majority of chiral compounds.

Presently glycerol is considered a co-product of biodiesel industry. As the biodiesel production is exponentially increasing, glycerol generated from the transesterification of vegetable oils and fats is also being produced on a large scale, and turned out to be essential seeking for novel alternatives to the consumption of the extra volume, in crude and/or as derivatives high added value. This review mainly deals with chemical and enzymatic transformations of glycerol to obtain chiral building blocks for synthesis of pharmaceuticals and natural products.

A practical and didactic sequence of experiments was proposed to illustrate the stereochemistry concept, optically active compounds, resolution of racemates, and use of the NMR technique, including 2D-COSY for identification of organic compounds, on a laboratory course for undergraduate students. The sequence was: extractions of racemic ibuprofen and chiral naproxen from commercial tablets; syntheses of diastereoisomeric amides reacting chiral (S)-(-)-α-methylbenzylamine with (±)-ibuprofen; separation and determination of absolute configuration of amides by ¹H NMR spectroscopy and GC analysis, and hydrolysis of amides to obtain (+)- and (-)-ibuprofen.

Optical activity is the ability of chiral substances to rotate the plane of plane-polarized light and is measured using an instrument called a polarimeter. An educational software application to explore, both interactively and visually, the concepts related to polarimetry to facilitate their understanding was developed. The software was field-tested and a questionnaire evaluating the graphics interface, usability and the software as an educational tool, was answered by students. The results characterized the computer application developed as an auxiliary tool for assisting teachers in lectures and students in the learning process.

The preparation of enantiomerically pure or enriched substances is of fundamental importance to pharmaceutical, food, agrochemical, and cosmetics industries and involves a growing market of hundreds of billions of dollars. However, most chemical processes used for their production are not environmentally friendly because in most cases, stoichiometric amounts of chiral inductors are used and substantial waste is produced. In this context, asymmetric catalysis has emerged as an efficient tool for the synthesis of enantiomerically enriched compounds using chiral catalysts. More specifically, considering the current scenario in the Brazilian chemical industry, especially that of pharmaceuticals, the immediate prospect for the use of synthetic routes developed in Brazil in an enantioselective fashion or even the discovery of new drugs is practically null. Currently, the industrial production of drugs in Brazil is primarily focused on the production of generic drugs and is basically supported by imports of intermediates from China and India. In order to change this panorama and move forward toward the gradual incorporation of genuinely Brazilian synthetic routes, strong incentive policies, especially those related to continuous funding, will be needed. These incentives could be a breakthrough once we establish several research groups working in the area of organic synthesis and on the development and application of chiral organocatalysts and ligands in asymmetric catalysis, thus contributing to boost the development of the Brazilian chemical industry. Considering these circumstances, Brazil can benefit from this opportunity because we have a wide biodiversity and a large pool of natural resources that can be used as starting materials for the production of new chiral catalysts and are creating competence in asymmetric catalysis and related areas. This may decisively contribute to the growth of chemistry in our country.

Separations using supercritical fluid chromatography (SFC) with packed columns have been re-discovered and explored in recent years. SFC enables fast and efficient separations and, in some cases, gives better results than high performance liquid chromatography (HPLC). This paper provides an overview of recent advances in SFC separations using packed columns for both achiral and chiral separations. The most important types of stationary phases used in SFC are discussed as well as the most critical parameters involved in the separations and some recent applications.

This work addresses the use of chiral gas chromatography in resolving optically active stereoisomers and racemates found in fruit flavours. It presents the types of chiral selectors applied to terpene-derived metal coordination compounds, polysiloxane-linked α-amino acid and mixed chiral stationary phases, and focuses on derivatized cyclodextrins, the most popular chiral stationary phases presently used in chromatographic analysis. Knowledge about the techniques involved in chiral recognition and enantiomer identification in the fruit flavour field is given along with examples from the latest studies.
