From August 1997 to August 1998, 334 specimens of Triatoma longipennis and 62 of T. picturata were collected in four groups of localities placed in the zone from Guadalajara, Jalisco to Tepic, Nayarit, in the West Coast of Mexico. Most T. longipennis were collected outdoors (69.2%) while most T. picturata (58.1%) were collected indoors. All collected specimens were examined for Trypanosoma cruzi infection, which was detected on 98 (29.3%) T. longipennis and 17 (27.4%) T. picturata. This study confirms the role of T. longipennis and T. picturata as some of the main T. cruzi vectors to humans in Mexico. Habitation Infestation Rate with T. longipennis was of 0.09 and with T. picturata was of 0.03 and the predominating ecotopes were pile of blocks, chicken coops, pigsties, wall crawls and beds.

Eight alien freshwater snail species were introduced into Martinique Island during the last 50 years. The introduced snails include four planorbids (Biomphalaria straminea, Helisoma duryi, Amerianna carinata and Gyraulus sp.), three thiarids (Melanoides tuberculata, M. amabilis and Tarebia granifera) and one ampullarid (Marisa cornuarietis). Four of these species rapidly colonized the whole Martinican hydrographic system whereas the other four remained restricted to some particular sites. The invasion processes were documented during the last 20 years and showed (i) a rapid invasion of the island by several morphs of M. tuberculata at the beginning of the 80's; (ii) the introduction of T. granifera in 1991 and M. amabilis in 1997; and (iii) the rapid spread of these last two species throughout the island. In the years following its introduction, M. tuberculata was used in biological control experiments against the snail hosts of schistosomiasis, B. glabrata and B. straminea. Experiments were conducted with success in several groups of water-cress beds which constituted the latest transmission sites for schistosomiasis at the beginning of the 80's. A malacological survey carried out in 2000 all over the island showed the absence of B. glabrata but the presence of some residual populations of B. straminea. Long-term studies carried out in Martinique have shown that the thiarids are able to maintain relatively stable populations over a long period of time, thus preventing recolonization by the snail hosts. Within this context the invasion of the hydrographic system of Martinique by thiarid snails has resulted in an efficient and sustainable control of the intermediate hosts of schistosomiasis.

A aplicação de silicato de Ca e Mg pode diminuir a acidez do solo e aumentar a disponibilidade de Ca, Mg, P e Si às plantas. O Si, mesmo não sendo essencial do ponto de vista fisiológico, traz inúmeros benefícios para o crescimento e o desenvolvimento vegetal, especialmente sob estresse. Este trabalho teve por objetivo avaliar a nutrição, a produtividade e a qualidade de tubérculos e batata cultivada em solo corrigido com calcário ou silicato sob dois níveis de umidade no solo. Os tratamentos foram constituídos pela aplicação de calcário dolomítico ou silicato de Ca e Mg, visando elevar a saturação por bases do solo a 60 %, e por duas tensões de água no solo: 0,020 MPa (sem deficiência hídrica) e 0,050 MPa (com deficiência hídrica). O experimento foi conduzido em casa de vegetação, em vasos contendo 50 kg dm-3 de Latossolo Vermelho de textura média. O delineamento foi inteiramente ao acaso em fatorial 2 x 2, com oito repetições. A deficiência hídrica teve pouca influência na nutrição da batateira, porém reduziu a produção de tubérculos comercializáveis. O silicato de Ca e Mg proporcionou os mesmos níveis de correção do solo e de fornecimento desses nutrientes que o calcário dolomítico, além de maior disponibilidade de P e Si no solo e maior absorção desses elementos pelas plantas de batata, podendo ser utilizado em substituição ao calcário. O fornecimento de Si à cultura da batata, mediante a aplicação de silicato, proporcionou maior altura de plantas, menor acamamento das hastes e maior produção de tubérculos comercializáveis.

Cultivares de batata mais produtivas possivelmente exigem maior quantidade de micronutrientes, porém no Brasil há carência de informações sobre extração e exportação de micronutrientes pelas principais cultivares de batata utilizadas. O objetivo deste trabalho foi avaliar a produtividade de tubérculos, a extração e a exportação de micronutrientes nas cultivares de batata Ágata, Asterix, Atlantic, Markies e Mondial. O experimento foi conduzido durante a safra de inverno, em um Latossolo Vermelho, no município de Itaí (SP). As parcelas foram constituídas pelas cinco cultivares, e as subparcelas, por épocas de coletas, realizadas no momento do plantio e a cada sete dias após a emergência. As cultivares Mondial e Asterix, mais produtivas, apresentaram maior extração de micronutrientes, com quantidades médias por hectare de 71 g de B, 122 g de Cu, 2.228 g de Fe, 618 g de Mn e 405 g de Zn. As menores quantidades extraídas foram observadas na cultivar Atlantic, com valores de 50, 81, 1.960, 544 e 270 g ha-1 de B, Cu, Fe, Mn e Zn, respectivamente. A fase de maior demanda por B ocorre logo após o início da formação de tubérculos, aos 34 DAP, enquanto a maior demanda por Fe e Mn inicia-se a partir dos 42 DAP e vai até 63 DAP. O Cu e o Zn são absorvidos em maiores proporções a partir dos 64 DAP até o final do ciclo. A quantidade de B, Cu, Mn e Zn exportada foi dependente da cultivar, com valores por hectare variando de 48 a 22 g de B, 79 a 16 g de Cu, 65 a 37 g de Mn e 167 a 83 g de Zn. A quantidade de Fe exportada não variou entre as cultivares, sendo, em média, de 243 g ha-1. A quantidade de micronutrientes extraída e exportada pela batateira variou com as cultivares utilizadas, indicando necessidade de manejo diferencial da adubação.

Christ's thorn (Ziziphus spina-christi (L.) Desf.) is a cross-pollinated plant with a wide range of genetic variability in nature and, for this reason, vegetative propagation assumes importance for improvement programs. The objective of this work was to evaluate cutting, T budding and tissue culture methods for this species. Shoots of 22-25 cm length were treated by two culture media and three shoot diameters for cutting trial. The T budding treatments consisted of three and five collection dates in spring and autumn, respectively. Tissue culture nodal segments bearing axillary buds were removed from shoots of mature trees at different seasons. Experiments to determine the best disinfectant chemical, appropriate conditions and materials to prevent phenolic compound exudation, explant characteristics, media type and cytokinin-auxin ratios were carried out. Successful rooting happened only on the sand beds and with cuttings greater than 8 mm diameter. The effects of T budding seasons on budtake percentage were significantly different. The best time for explant harvesting was mid of summer. Amount of rooting on media containing IBA as well as activated charcoal and disinfection with Ca(OCl)2 at concentration of 5% for 20 minutes were the best treatments.

Botrytis blight caused by Botrytis cinerea is an important disease of rose (Rosa hybrida) grown in greenhouses in Brazil. As little is known regarding the disease epidemiology under greenhouse conditions, pathogen survival in crop debris and as sclerotia was evaluated. Polyethylene bags with petals, leaves, or stem sections artificially infected with B. cinerea were mixed with crop debris in rose beds, in a commercial plastic greenhouse. High percentage of plant parts with sporulation was detected until 60 days, then sporulation decreased on petals after 120 days, and sharply decreased on stems or leaves after 90 days. Sporulation on petals continued for 360 days, but was not observed on stems after 150 days or leaves after 240 days. Although the fungus survived longer on petals, stems and leaves are also important inoculum sources because high amounts of both are deposited on beds during cultivation. Survival of sclerotia produced on PDA was also quantified. Sclerotia germination was greater than 75% in the initial 210 days and 50% until 360 days. Sclerotia weight gradually declined but they remained viable for 360 days. Sclerotia were produced on the buried petals, mainly after 90 days of burial, but not on leaves or stems. Germination of these sclerotia gradually decreased after 120 days, but lasted until 360 days. Higher weight loss and lower viability were observed on sclerotia produced on petals than on sclerotia produced in vitro

Foram caracterizados 123 isolados de Phytophthora infestans obtidos de 21 lavouras de tomateiro e oito de batateira, em municípios do Estado de Goiás e Cidades Satélites de Brasília, no período de abril de 2001 a setembro de 2003. Os isolados foram caracterizados para os marcadores grupo de compatibilidade (123 isolados); isoenzima glucose 6-fosfato-isomerase (Gpi) (34 isolados) e resistência aos fungicidas mefenoxam (77 isolados) e metalaxyl (32 isolados de batateira), usando o método de disco de folhas. Todos os 78 isolados de tomateiro foram classificados no grupo de compatibilidade A1, enquanto os 45 de batateira foram do grupo A2. Os fenótipos para Gpi dos isolados de tomateiro (19) e de batateira (15) foram 86/100, típico da linhagem clonal US-1, e 100/100, típico da linhagen clonal BR-1, respectivamente. Quanto à resistência a mefenoxam, constataram-se isolados de tomateiro resistentes (36%), intermediários (48%) e sensíveis (16%). A maioria dos isolados de batateira foi classificada como sensível (82%) e apenas 9% de intermediários e resistentes. Dos isolados de batateira avaliados para resistência ao metalaxyl, 25% foram resistentes, 62% intermediários e 13% sensíveis. A população de P. infestans no Distrito Federal e no Estado de Goiás é constituída de duas linhagens clonais, com especificidade por hospedeiro.

As atividades amilolítica e pectinolítica de 45 isolados de Alternaria solani, provenientes de diferentes hospedeiros, foram estimadas por meio da difusão enzimática em meio sólido específico e mensuração do halo de degradação do substrato. Todos os isolados degradaram pectina. Apenas 17 isolados apresentaram atividade amilolítica, sendo nove isolados provenientes de batateira. Somente o isolado AS18 se destacou como bom produtor de ambas as enzimas. Uma vez que a atividade pectinolítica foi mais evidente, avaliou-se a influência de pectinases na agressividade de A. solani ao tomateiro. Para isso, cinco isolados (2 de berinjela, 2 de tomateiro e 1 de batateira) contrastantes quanto à produção de pectinases foram selecionados para testes em folíolos destacados e plantas inteiras. Quatro isolados foram utilizados no teste em folíolos destacados (AS6, AS7, AS12 e AS26), e constatou-se haver variabilidade patogênica. A correlação obtida entre o tamanho das lesões e a atividade pectinolítica foi de r = 0,963 (P = 0,087). Cinco isolados (AS6, AS7, AS12, AS25 e AS26) foram inoculados em plantas inteiras de tomate. Os isolados não diferiram quanto ao número de lesões/cm² de área foliar, porém variaram em agressividade. Houve correlação (r = 0,916; P = 0,042) entre a atividade de pectinases e o índice de doença, sugerindo possível papel para as enzimas pécticas durante a infecção de A. solani em tomateiro. É provável que as diferenças no perfil enzimático dos isolados estejam associadas ao hospedeiro original de onde os mesmos foram obtidos. Os resultados reforçaram evidências de especificidade por hospedeiro em populações de A. solani.

This paper presents the experimental characterization of hydrodynamics and gas-liquid mass transfer in a three-phase fluidized bed containing polystyrene and nylon particles. The influence of gas and liquid velocities on phase holdups and volumetric gas-liquid mass transfer coefficient was investigated for flow conditions similar to those applied in biotechnological process. The phase holdups were obtained by the pressure profile technique. The volumetric gas-liquid mass transfer coefficient was obtained adjusting the experimental concentration profiles of dissolved oxygen in the liquid phase with the predictions of the axial dispersion model. According to experimental results the liquid holdup increases with the gas velocity, whereas the solid holdup decreases. The gas holdup increases significantly with the increase in gas velocity, and it shows for the three-phase fluidized bed comparable values or larger than those of bubble column. The volumetric gas-liquid mass transfer coefficient increases significantly with an increase in the air velocity for both bubble column and fluidized beds. In addition, in the operational condition of high liquid velocity, the presence of low-density particles in the bed increased the gas-liquid mass transfer, and thus the volumetric mass transfer coefficient values obtained in the fluidized bed were comparable or larger than those of bubble column.

Ouabain is an endogenous substance occurring in the plasma in the nanomolar range, that has been proposed to increase vascular resistance and induce hypertension. This substance acts on the a-subunit of Na+,K+-ATPase inhibiting the Na+-pump activity. In the vascular smooth muscle this effect leads to intracellular Na+ accumulation that reduces the activity of the Na+/Ca2+ exchanger and to an increased vascular tone. It was also suggested that circulating ouabain, even in the nanomolar range, sensitizes the vascular smooth muscle to vasopressor substances. We tested the latter hypothesis by studying the effects of ouabain in the micromolar and nanomolar range on phenylephrine (PE)-evoked pressor responses. The experiments were performed in normotensive and hypertensive rats in vivo, under anesthesia, and in perfused rat tail vascular beds. The results showed that ouabain pretreatment increased the vasopressor responses to PE in vitro and in vivo. This sensitization after ouabain treatment was also observed in hypertensive animals which presented an enhanced vasopressor response to PE in comparison to normotensive animals. It is suggested that ouabain at nanomolar concentrations can sensitize vascular smooth muscle to vasopressor stimuli possibly contributing to increased tone in hypertension

We investigated the angiotensin II (Ang II)-generating system by analyzing the vasoconstrictor effect of Ang II, angiotensin I (Ang I), and tetradecapeptide (TDP) renin substrate in the absence and presence of inhibitors of the renin-angiotensin system in isolated rat aortic rings and mesenteric arterial beds with and without functional endothelium. Ang II, Ang I, and TDP elicited a dose-dependent vasoconstrictor effect in both vascular preparations that was completely blocked by the Ang II receptor antagonist saralasin (50 nM). The angiotensin converting enzyme (ACE) inhibitor captopril (36 µM) completely inhibited the vasoconstrictor effect elicited by Ang I and TDP in aortic rings without affecting that of Ang II. In contrast, captopril (36 µM) significantly reduced (80-90%) the response to bolus injection of Ang I, without affecting those to Ang II and TDP in mesenteric arteries. Mechanical removal of the endothelium greatly potentiated (70-95%) the vasoconstrictor response to Ang II, Ang I, and TDP in aortic rings while these responses were unaffected by the removal of the endothelium of mesenteric arteries with sodium deoxycholate infusion. In addition, endothelium disruption did not change the pattern of response elicited by these peptides in the presence of captopril. These findings indicate that the endothelium may not be essential for Ang II formation in rat mesenteric arteries and aorta, but it may modulate the response to Ang II. Although Ang II formation from Ang I is essentially dependent on ACE in both vessels, our results suggest the existence of an alternative pathway in the mesenteric arterial bed that may play an important role in Ang II generation from TDP in resistance but not in large vessels during ACE inhibition

The most conspicuous effect of bradykinin following its administration into the systemic circulation is a transient hypotension due to vasodilation. In the present study most of the available evidence regarding the mechanisms involved in bradykinin-induced arterial vasodilation is reviewed. It has become firmly established that in most species vasodilation in response to bradykinin is mediated by the release of endothelial relaxing factors following the activation of B2-receptors. Although in some cases the action of bradykinin is entirely mediated by the endothelial release of nitric oxide (NO) and/or prostacyclin (PGI2), a large amount of evidence has been accumulated during the last 10 years indicating that a non-NO/PGI2 factor accounts for bradykinin-induced vasodilation in a wide variety of perfused vascular beds and isolated small arteries from several species including humans. Since the effect of the non-NO/PGI2 endothelium-derived relaxing factor is practically abolished by disrupting the K+ electrochemical gradient together with the fact that bradykinin causes endothelium-dependent hyperpolarization of vascular smooth muscle cells, the action of such factor has been attributed to the opening of K+ channels in these cells. The pharmacological characteristics of these channels are not uniform among the different blood vessels in which they have been examined. Although there is some evidence indicating a role for KCa or KV channels, our findings in the mesenteric bed together with other reports indicate that the K+ channels involved do not correspond exactly to any of those already described. In addition, the chemical identity of such hyperpolarizing factor is still a matter of controversy. The postulated main contenders are epoxyeicosatrienoic acids or endocannabinoid agonists for the CB1-receptors. Based on the available reports and on data from our laboratory in the rat mesenteric bed, we conclude that the NO/PGI2-independent endothelium-dependent vasodilation induced by BK is unlikely to involve a cytochrome P450 arachidonic acid metabolite or an endocannabinoid agonist.

Oxytocin (OT), a nonapeptide, was the first hormone to have its biological activities established and chemical structure determined. It was believed that OT is released from hypothalamic nerve terminals of the posterior hypophysis into the circulation where it stimulates uterine contractions during parturition, and milk ejection during lactation. However, equivalent concentrations of OT were found in the male hypophysis, and similar stimuli of OT release were determined for both sexes, suggesting other physiological functions. Indeed, recent studies indicate that OT is involved in cognition, tolerance, adaptation and complex sexual and maternal behaviour, as well as in the regulation of cardiovascular functions. It has long been known that OT induces natriuresis and causes a fall in mean arterial pressure, both after acute and chronic treatment, but the mechanism was not clear. The discovery of the natriuretic family shed new light on this matter. Atrial natriuretic peptide (ANP), a potent natriuretic and vasorelaxant hormone, originally isolated from rat atria, has been found at other sites, including the brain. Blood volume expansion causes ANP release that is believed to be important in the induction of natriuresis and diuresis, which in turn act to reduce the increase in blood volume. Neurohypophysectomy totally abolishes the ANP response to volume expansion. This indicates that one of the major hypophyseal peptides is responsible for ANP release. The role of ANP in OT-induced natriuresis was evaluated, and we hypothesized that the cardio-renal effects of OT are mediated by the release of ANP from the heart. To support this hypothesis, we have demonstrated the presence and synthesis of OT receptors in all heart compartments and the vasculature. The functionality of these receptors has been established by the ability of OT to induce ANP release from perfused heart or atrial slices. Furthermore, we have shown that the heart and large vessels like the aorta and vena cava are sites of OT synthesis. Therefore, locally produced OT may have important regulatory functions within the heart and vascular beds. Such functions may include slowing down of the heart or the regulation of local vascular tone.

Ouabain increases vascular resistance and may induce hypertension by inhibiting the Na+ pump. The effects of 0.18 and 18 µg/kg, and 1.8 mg/kg ouabain pretreatment on the phenylephrine (PHE; 0.1, 0.25 and 0.5 µg, in bolus)-evoked pressor responses were investigated using anesthetized normotensive (control and uninephrectomized) and hypertensive (1K1C and DOCA-salt treated) rats. Treatment with 18 µg/kg ouabain increased systolic and diastolic blood pressure in all groups studied. However, the magnitude of this increase was larger for the hypertensive 1K1C and DOCA-salt rats than for normotensive animals, while the pressor effect of 0.18 µg/kg ouabain was greater only in DOCA-salt rats. A very large dose (1.8 mg/kg) produced toxic effects on the normotensive control but not on uninephrectomized or 1K1C rats. Rat tail vascular beds were perfused to analyze the effects of 10 nM ouabain on the pressor response to PHE. In all animals, 10 nM ouabain increased the PHE pressor response, but this increase was larger in hypertensive DOCA-salt rats than in normotensive and 1K1C rats. Results suggested that a) increases in diastolic blood pressure induced by 18 µg/kg ouabain were larger in hypertensive than normotensive rats; b) in DOCA-salt rats, smaller ouabain doses had a stronger effect than in other groups; c) hypertensive and uninephrectomized rats were less sensitive to toxic doses of ouabain, and d) after treatment with 10 nM ouabain isolated tail vascular beds from DOCA-salt rats were more sensitive to the pressor effect of PHE than those from normotensive and 1K1C hypertensive rats. These data suggest that very small doses of ouabain, which might produce nanomolar plasma concentrations, enhance pressor reactivity in DOCA-salt hypertensive rats, supporting the idea that endogenous ouabain may contribute to the increase and maintenance of vascular tone in hypertension.

The present study was designed to determine relaxation in response to 17ß-estradiol by isolated perfused hearts from intact normotensive male and female rats as well as the contribution of endothelium and its relaxing factors to this action. Baseline coronary perfusion pressure was determined and the vasoactive effects of 17ß-estradiol (10 µM) were assessed by in bolus administration before and after endothelium denudation by infusion of 0.25 µM sodium deoxycholate or perfusion with 100 µM L-NAME, 2.8 µM indomethacin, 0.75 µM clotrimazole, 100 µM L-NAME plus 2.8 µM indomethacin, and 100 µM L-NAME plus 0.75 µM clotrimazole. Baseline coronary perfusion pressure differed significantly between males (84 ± 2 mmHg, N = 61) and females (102 ± 2 mmHg, N = 61). Bolus injection of 10 µM 17ß-estradiol elicited a transient relaxing response in all groups, which was greater in coronary beds from females. For both sexes, the relaxing response to 17ß-estradiol was at least in part endothelium-dependent. In the presence of the nitric oxide synthase inhibitor L-NAME, the relaxing response to 17ß-estradiol was reduced only in females. Nevertheless, in the presence of indomethacin, a cyclooxygenase inhibitor, or clotrimazole, a cytochrome P450 inhibitor, the 17ß-estradiol response was significantly reduced in both groups. In addition, combined treatment with L-NAME plus indomethacin or L-NAME plus clotrimazole also reduced the 17ß-estradiol response in both groups. These results indicate the importance of prostacyclin and endothelium-derived hyperpolarizing factor in the relaxing response to 17ß-estradiol. 17ß-estradiol-induced relaxation may play an important role in the regulation of coronary tone and this may be one of the reasons why estrogen replacement therapy reduces the risk of coronary heart disease in postmenopausal women.
