Com o objetivo de se estudar a aplicação do Sistema Integrado de Diagnose e Recomendação (DRIS) em cana-de-açucar, confrontou-se esta metodologia com a técnica da diagnose foliar baseada no nível crítico. Desse modo, observou-se o efeito de diferentes épocas de amostragem foliar de soqueiras cultivadas em 4 Grandes Grupos de Solos e, num deles, dois regimes hídricos: irrigado e não irrigado. Os ensaios se basearam no esquema fatorial N x P x K x L, onde L = localização dos fertilizantes em superfície e em profundidade, com as dosagens 0 - 60 - 120 -180 kg N/ha, 0-45 kg P(2)0(5)/ha e 0-90-180 kg K2O/ha. Os solos utilizados foram Latossol Roxo (LR) , Podzolico Vermelho Amarelo variação Laras (PVls), Latossol Vermelho Amarelo - fase arenosa (LVa) e o Latossol Vermelho Escuro-orto (LE) não controlado e sob irrigação por aspersão. A variedade empregada foi a CB 41-76 e coletou-se a folha +3 aos 3, 4, 5 e 6 meses de idade da cana-soca. As parcelas eram constituídas por 3 linhas úteis, 2 linhas de bordadura interna e 2 de bordadura externa, num total de 7 linhas com 10 m de comprimento e 1,5 m de espaçamento, ocupando uma área de 105 m². Os dados mostraram que a localização dos fertilizantes em profundidade não proporcionou efeito positivo nas produtividades agrícolas e de açúcar por área,com exceção ao solo PVls. De modo geral, o nitrogênio e o potássio aumentaram de forma linear os rendimentos da cana-de-açúcar, salvo para o potássio no solo LR. O fósforo elevou a produção agrícola para os solos LE, LR e PVls e provocou aumento da produção de açúcar por área para os dois primeiros solos. A irrigação não indicou benefício para a soqueira em solo LE. Foi possível o estabelecimento dos níveis críticos de N e K apenas para algumas épocas de amostragem foliar, enquanto para o fósforo os mesmos não foram viáveis para nenhuma época de coleta da folha +3. A época de coleta do tecido vegetal e o tipo de solo afetaram os níveis críticos de N e K. Por outro lado, o DRIS mostrou ser metodologia adequada para o diagnostico de N, P e K em soquei ras de cana-de-açucar e a interpretação dos índices transformados e praticamente inalterada com a época de amostragem foliar. Possuindo também, a propriedade de ter sua aplicação para diferentes tipos de solos.

In order to obtain the following informations: a) dry matter production and extraction of nutrients by the fruits at different ages; b) dry matter production and extraction of nutrient by the leaves and "trunk + branches" collected at the flowering stage; c) dry matter production and export of nutrients by pruning (leaves and branches) at the begining dormant stage; A trial was conducted on Latossolo Vermelho Escuro Orto group (Orthox) at Buri, São Paulo State, Brazil. The material was collected from 'Ohio Beauty' and 'Brazil' apples grafted on 'Doucin' 1-2; 3-4; 4-5 and 6-7 years old. The main conclusions were as follows: a) differences were observed on dry matter production by two varieties at the different stages of growth; b) differences were also observed between the two varieties on the matter production in the leaves and "trunk + branches" at the flowering stage, as well as by the leaves and branches pruned at the begining of dormant stages; c) differences were observed betwen the two varieties concerning to nutrient concentration (on dry matter basis) on the fruits collected at different stages of growth. Same results were observed on leaves and "trunk + branches" collected at flowering period; d) differences were observed on the exportation of the nutrients referring to growth period of fruit; e) at the flowering and dormant period, differences were observed on the contents of nutrients in the leaves, 'trunk + branches', on the two varieties; f) the nutrient exportation by the fruits obyed the following order: K>N>P>S>Ca>Mg>Fe>B > Cu > Mn > Zn > Mo; g) the nutrient extration by the aerial part the apple trees obyed the following order: N > K > Ca > Mg > P > S > Fe > B > Cu = Mn = Zn.

A presente pesquisa foi realizada no Centro de Energia Nuclear na Agricultura (CENA), Piracicaba, SP, Brasil, no período 1981-82, para verificar se o p32 absorvido pelas raízes do tolete da cana-de-açúcar se transloca para a gema germinante. Toletes + 10-Kuijper, de uma gema, do cv. CB-41-76, foram germinados em posição vertical, em bandejas contendo água destilada ao nível superior do nó. A estavam com mais de 150 cm de altura, adicionaram o P32 à solução nutritiva, para que fosse absorvido pelo sistema radicular normal, contando a radioatividade na folha do nó número 6, a partir do ápice. Não estudaram a absorção do P32 pelas raízes adventícias do tolete. RESNIK et alii (1976) estudaram a absorção do p32 por raízes do tolete, no cv. NCo 310 e sua translocação em toletes de 4nós. 0 P32 absorvido pelas raízes, durante 48 horas, translocou-se para os setores radiais e longitudinais do colmo. Acumulou-se em maior concentração nos nós do que nos internódios e no internódio superior do que no inferior. Não observaram a sua translocação para a gema. Este trabalho visa a determinar, autorradiograficamente, a translocação do P32 absorvido pelas raízes do tolete, e sua translocação para a gema germinante.

A definition of Biomphalaria helophila (Orbigny, 1835) is presented, based on examination of the shell and reproductive system of topotypic specimens and extended to a number of samples from other localities. The following nominal species and subspecies, collected from type localities, proved junior synonyms of B. helophila: Planorbis albicans Pfeiffer, 1839; Planorbis dentatus Gould, 1844; Planorbis dentiferus CB Adams, 1845; Planorbis dentiferus edentatus CB Adams, 1851; Planorbis dentiens Morelet, 1849; Planorbula dentiens edentula Fischer & Crosse, 1880; Planorbis stagnicola Morelet, 1851; and Tropicorbis shimeki FC Baker, 1945. B. helophila was also identified in samples from Costa Rica, Guatemala, Haiti, Dominican Republic, Puerto Rico and Barbados.

Lutzomyia (Nyssomyia) intermedia (Lutz & Neiva) is redescribed, a neotype is designated, and a very similar species, L. (N.) neivai (Pinto, 1926), is resurrected; the male and the female are redescribed. The two species can be differentiated by differences in the spermathecae, common ducts and number of cibarial horizontal teeth of the females and in the size and proportions of several structures of both sexes. The known geographical distribution of both species is given.

The molecular karyotype of nine Trypanosoma rangeli strains was analyzed by contour-clamped homogeneous electric field electrophoresis, followed by the chromosomal localization of ß-tubulin, cysteine proteinase, 70 kDa heat shock protein (hsp 70) and actin genes. The T. rangeli strains were isolated from either insects or mammals from El Salvador, Honduras, Venezuela, Colombia, Panama and southern Brazil. Also, T. cruzi CL-Brener clone was included for comparison. Despite the great similarity observed among strains from Brazil, the molecular karyotype of all T. rangeli strains analyzed revealed extensive chromosome polymorphism. In addition, it was possible to distinguish T. rangeli from T. cruzi by the chromosomal DNA electrophoresis pattern. The localization of ß-tubulin genes revealed differences among T. rangeli strains and confirmed the similarity between the isolates from Brazil. Hybridization assays using probes directed to the cysteine proteinase, hsp 70 and actin genes discriminated T. rangeli from T. cruzi, proving that these genes are useful molecular markers for the differential diagnosis between these two species. Numerical analysis based on the molecular karyotype data revealed a high degree of polymorphism among T. rangeli strains isolated from southern Brazil and strains isolated from Central and the northern South America. The T. cruzi reference strain was not clustered with any T. rangeli strain.

Mast cells and eosinophils actively participate in tissue repair and are prominent components of Schistosoma mansoni granulomas. Since pentoxifillyne (PTX) is an immunomodulatory and antifibrotic substance, we aimed to characterize, by morphological techniques, the effect of this drug on fibrosis developed inside murine hepatic schistosomal granulomatous reaction, beyond the quantification of eosinophil and mast cell populations. The drug (1 mg/100 g animal weight) was administrated from 35 to 90 days post-infection, when the animals were killed. The intragranulomatous interstitial collagen network was analyzed by confocal laser scanning microscopy, the number of eosinophils and mast cells was quantified and the results were validated by t-student test. Treatment did not interfere on the granuloma evolution but caused a significant decrease in the total and involutive number of hepatic granulomas (p = 0.01 and 0.001, respectivelly), and in the intragranulomatous accumulation of eosinophils (p = 0.0001). Otherwise, the number of mast cells was not significantly altered (p = 0.9); however, it was positively correlated with the number of granulomatous structures (r = 0.955). In conclusion, PTX does not affect development and collagen deposition in S. mansoni murine granuloma, but decreases the intragranulomatous eosinophil accumulation possibly due to its immunomodulatory capability, interfering in cellular recruitment and/or differentiation.

Between 1985 and 2000, epidemiological surveys of the American tegumentary leishmaniasis (ATL) were carried out in several rural and urban communities in Espírito Santo, Brazil. A total of 100 stocks of Leishmania (comprising isolates from both human and canine hosts with ATL) were identified by two methods of molecular characterization, using specific monoclonal antibodies and multilocus enzyme electrophoresis. Parasite isolates from 19 municipalities were found to belong to the same zymodeme and serodeme type as of the Leishmania (Viannia) braziliensis reference strain. In contrast, our genotyping studies have shown intra-specific variation among these parasites (comparisons of the variability of the internal transcribed spacers between the small and large subunits of the rRNA genes of the 22 stocks studiedrevealed at least 11 genotypes). Two main clusters of L. (V.) braziliensis genotypes were observed, representing parasites collected from different endemic regions in the state, where transmission reflects distinct eco-epidemiological features. Infection with this pathogen was associated with the characteristic disease forms, but neither the clinical outcome nor the response to treatment could be related to the genetic polymorphism of the isolates, as defined by using the proposed methodology.

The effects of blood components, nerve-cord severance, and ecdysone therapy on the posterior midgut epithelial cells of 5th-instar Rhodnius prolixus nymphs 10 days after feeding were analyzed by transmission electron microscopy. Cutting the nerve-cord of the blood-fed insects partially reduced the development of microvilli and perimicrovillar membranes (PMM), and produced large vacuoles and small electrondense granules; insects fed on Ringer's saline diet exhibited well developed microvilli and low PMM production; swolled rough endoplasmatic reticulum and electrondense granules; Ringer's saline meal with ecdysone led to PMM development, glycogen particles, and several mitochondria in the cytoplasm; epithelial cells of the insects fed on Ringer's saline meal whose nerve-cord was severed showed heterogeneously distributed microvilli with reduced PMM production and a great quantity of mitochondria and glycogen in the cytoplasm; well developed microvilli and PMM were observed in nerve-cord severed insects fed on Ringer's saline meal with ecdysone; Ringer's saline diet containing hemoglobin recovered the release of PMM; and insects fed on human plasma showed slightly reduced PMM production, although the addition of ecdysone in the plasma led to a normal midgut ultrastructural organization. We suggest that the full development of microvilli and PMM in the epithelial cells depends on the abdominal distension in addition to ingestion of hemoglobin, and the release of ecdysone.

Complex physalin metabolites present in the capsules of the fruit of Physalis angulata L. have been isolated and submitted to a series of assays of antimicrobial activity against Pseudomonas aeruginosa ATCC 27853, Staphylococcus aureus ATCC 29213, S. aureus ATCC 25923, S. aureus ATCC 6538P, Neisseria gonorrhoeae ATCC 49226, Escherichia coli ATCC 8739; E. coli ATCC 25922, Candida albicans ATCC 10231 applying different methodologies such as: bioautography, dilution broth, dilution agar, and agar diffusion techniques. A mixture of physalins (pool) containing physalins B, D, F, G inhibit S. aureus ATCC 29213, S. aureus ATCC 25923, S. aureus ATCC 6538P, and N. gonorrhoeae ATCC 49226 at a concentration of 200 mg/µl, using agar dilution assays. The mixture was inactive against P. aeruginosa ATCC27853, E. coli ATCC 8739; E. coli ATCC 25922, C. albicans ATCC 10231 when applying bioautography assays. Physalin B (200 µg/ml) by the agar diffusion assay inhibited S. aureus ATCC 6538P by ± 85%; and may be considered responsible for the antimicrobial activity.

The susceptibility of Anopheles aquasalis (F3 generation) and An. darlingi (F1 generation) to Plasmodium vivax circumsporozoite protein phenotypes from a limited number of blood samples of malaria patients in Belém, state of Pará, Brazil, was examined. A polymerase chain reaction was used to determine the P. vivax phenotypes in blood samples and the blood-fed infected mosquitoes were dissected and tested by ELISA. In all patient infections, more infected An. aquasalis and An. darlingi were positive for VK210 compared with VK247.

Studies were carried out to evaluate the efficacy of the growth regulator, triflumuron (TFM) (Starycide® sc 480 Bayer), for disrupting the development of Rhodnius prolixus fifth-instar nymph by oral, topical or continuous treatment. All treatments were able to induce high levels of mortality, delay development and molt inhibition. Oral treatment induced molt inhibition in all insects that survived at doses of 0.25, 0.50 and 5.0 mg/mL of a blood meal. The highest levels of both mortality in 24 h and molt inhibition were always observed after topical treatment. The lowest doses needed to obtain considerable biological effects were always observed after continuous treatment. In this way, the highest levels of mortality within 30 days were detected after continuous treatment, which also induced an extended inter-molting period, a lower number of over-aged nymphs and the highest level of molting in nymphs that survived. Moreover, the effects of TFM on insects were often displayed in a dose response manner. These results indicate that TFM acts as a potent growth inhibitor of R. prolixus nymphs and has the potential to be used in integrated vector control programs against hematophagous triatomine species.

Mycobacterium tuberculosis (Mtb) has acquired resistance and consequently the antibiotic therapeutic options available against this microorganism are limited. In this scenario, the use of usnic acid (UA), a natural compound, encapsulated into liposomes is proposed as a new approach in multidrug-resistant tuberculosis (MDR-TB) therapy. Thus the aim of this study was to evaluate the effect of the encapsulation of UA into liposomes, as well as its combination with antituberculous agents such as rifampicin (RIF) and isoniazid (INH) against MDR-TB clinical isolates. The in vitro antimycobacterial activity of UA-loaded liposomes (UA-Lipo) against MDR-TB was assessed by the microdilution method. The in vitro interaction of UA with antituberculous agents was carried out using checkerboard method. Minimal inhibitory concentration values were 31.25 and 0.98 µg/mL for UA and UA-Lipo, respectively. The results exhibited a synergistic interaction between RIF and UA [fractional inhibitory concentration index (FICI) = 0.31] or UA-Lipo (FICI = 0.28). Regarding INH, the combination of UA or UA-Lipo revealed no marked effect (FICI = 1.30-2.50). The UA-Lipo may be used as a dosage form to improve the antimycobacterial activity of RIF, a first-line drug for the treatment of infections caused by Mtb.
