A leiteira (Euphorbia heterophylla) é uma planta daninha de ciclo anual encontrada com frequência em lavouras de soja na região Sul do Brasil, controlada em lavouras de soja geneticamente modificada (Roundup Ready® - RR) com uso do herbicida glyphosate. Entretanto, nos últimos anos, o controle dessa espécie não tem sido satisfatório em alguns locais, provocando a suspeita de que foram selecionados biótipos resistentes ao herbicida. Diante disso, o objetivo deste trabalho foi avaliar, por meio de curvas de dose-resposta, biótipos de leiteira com suspeita de resistência ao glyphosate. O experimento foi realizado em casa de vegetação, em delineamento experimental completamente casualizado, com quatro repetições. Os tratamentos constituíram-se de doses crescentes de glyphosate (0, 90, 180, 360, 720, 1.080, 1.440 e 2.160 g e.a. ha-1), aplicadas sobre cinco biótipos de leiteira, oriundos de lavouras de soja RR do Estado do Rio Grande do Sul. As variáveis avaliadas foram controle e matéria seca da parte aérea. Os resultados evidenciam que os biótipos de leiteira avaliados são suscetíveis ao glyphosate.

A utilização intensiva do glyphosate nas lavouras de soja Roundup Ready® (RR) no Rio Grande do Sul (RS), nos últimos anos, pode ter selecionado biótipos de leiteira (Euphorbia heterophylla) resistentes ao herbicida. Esse cenário dificultará ainda mais o manejo da espécie, já que permanecem indícios da presença de biótipos resistentes também em herbicidas inibidores da acetolactato sintase (ALS). Assim, os objetivos deste trabalho foram avaliar a sensibilidade da leiteira a herbicidas inibidores da ALS e ao glyphosate, verificar a distribuição dos biótipos resistentes no RS e determinar os principais fatores agronômicos associados a falhas de controle. Para isso, amostras de sementes de plantas de leiteira foram coletadas em lavouras de soja RR localizadas em 56 municípios do Estado do RS. Por ocasião das coletas, os agricultores responderam a questionário que abordava o manejo das plantas daninhas na área. Usando-se as sementes coletadas, foram conduzidos dois experimentos em casa de vegetação: no primeiro, avaliou-se a resposta de 86 biótipos ao herbicida glyphosate, aplicado na dose de 2.160 g e.a. ha-1; e, no segundo, a resposta de 73 biótipos ao herbicida imazethapyr, aplicado na dose de 200 g i.a. ha-1. Os resultados obtidos evidenciam que todos os biótipos de leiteira avaliados são suscetíveis ao glyphosate, porém existem biótipos resistentes aos inibidores da ALS. As respostas do questionário indicam que práticas de manejo como uso de subdoses e/ou utilização intensiva do glyphosate e a ausência de rotação de culturas favorecem falhas no controle de leiteira pelo herbicida glyphosate em soja.

Among the herbicides recommended for the dry season and registered to sugarcane crop, amicarbazone, isoxaflutole and the association diuron + hexazinone + sulfomethuron-methyl can be highlighted. These are pre-emergence herbicides efficient against broad-leaved weeds. Morning glory causes large losses in infested sugarcane fields by bending the stalks and interfering in harvesting. In this study the effectiveness of pre-emergence herbicides for two species of morning glory (Ipomoea hederifolia and Ipomoea grandifolia) was evaluated. Treatments were arranged in completely randomized factorial design (4 x 7). There were four periods of water restriction (0, 30, 60 and 90 days), seven chemical treatments [diuron + hexazinone + sulfometuron-methyl (1387 + 391 + 33.35 g a.i. ha-1), diuron + hexazinone + sulfometuron-methyl (1507.5 + 425 + 36.25 g a.i. ha-1), diuron + hexazinone + sulfometuron-methyl (1658.25 + 467.5 + 39.87 g a.i. ha-1), diuron + hexazinone + sulfometuron­methyl (1809 + 510 + 43.5 g a.i. ha-1), amicarbazone (1190 g a.i. ha-1), amicarbazone + isoxaflutole (840 + 82.5 g a.i. ha-1)] and a control with no application. At 7, 14, 21 and 28 days after the restoration of moisture, control was visually evaluated. After the final evaluation, the dry mass of morning glories was measured. At 90 days of water restriction, diuron + hexazinone + sulfometuron-methyl was more effective to control I. hederifolia than the amicarbazone + isoxaflutole tank mixture. The four diuron + hexazinone + sulfometuron­methyl doses have reduced morning glory dry mass to zero; whereas treatments with amicarbazone have not. The most effective treatment for morning glory control was diuron + hexazinone + sulfometuron-methyl. This result may be due to a possible synergistic interaction.

Weed resistance to herbicides has been a major issue in Brazil, mainly due to the inefficiency of the herbicides used in no-till areas and to the high cost of these herbicide treatments. Failures in controlling the weed Conyza have been reported in Western and Northern grain crop areas in Paraná (Brazil). This work aimed to evaluate the potential occurrence of C. sumatrensis biotypes resistant to the herbicides chlorimuron-ethyl and glyphosate. Experiments were carried out under greenhouse conditions with four biotypes (Cascavel-2, Toledo-4, Tupãssi-6, and Assis Chateaubriand-7) possibly resistant to, as well as a population considered susceptible to chlorimuron-ethyl and glyphosate. To obtain dose-response curves, eight herbicide doses of chlorimuron-ethyl (0, 2.5, 5, 10, 20, 40, 80 and 160 g ha-1) and glyphosate (0, 90, 180, 360, 720, 1,440, 2,880 and 5,760 g e.a. ha-1) were applied and weed control and shoot biomass evaluations were made. Results provided evidence that two biotypes (Cascavel-2 and Tupãssi-6) were resistant to glyphosate and four biotypes (Cascavel-2, Toledo-4, Tupãssi-6 and Assis Chateaubriand-7) were resistant to chlorimuron­ethyl. Multiple resistance to glyphosate and chlorimuron was confirmed for biotypes Cascavel­2 and Tupãssi 6. This is the first report on multiple resistance in Conyza sumatrensis, worldwide.

Organismos epifílicos em C. macrophyllus foram encontrados em duas parcelas amostradas em julho de 1996, em duas áreas com diferentes graus de perturbação pelo fogo, no Parque Estadual do Rio Doce, MG. Quatorze espécies epifílicas foram encontradas incluindo: 11 líquens, uma alga, uma hepática e um fungo. Os líquens totalizaram 81,8% das espécies na área menos perturbada e 72,7% na mais perturbada. A espécie mais abundante foi Porina epiphylla, que ocorreu em 80,8% dos forófitos da primeira área e em 97% dos forófitos da segunda. O número de folhas cobertas em cada forófito, o grau de cobertura em cada folha e a altura dos forófitos foram significativamente diferentes entre as duas áreas, enquanto que a área foliar e o número de folhas por forófito não o foram. Embora diferenças significativas não tenham sido encontradas na composição específica do epifilo entre as duas áreas, a maior ocorrência de folhas cobertas pelo epifilo pode estar associada à perturbação. A penetração de luz, intensificada pelas clareiras, pode ter aumentado o crescimento em altura do forófito e a colonização das folhas pelos organismos epifílicos.

Nutrient impoverishment in mesocosms was carried out in a shallow eutrophic reservoir aiming to evaluate the nutrient removal technique as a method for eutrophication reduction. Garças Pond is located in the Parque Estadual das Fontes do Ipiranga Biological Reserve situated in the southeast region of the municipality of São Paulo. Three different treatments were designed, each consisting of two enclosures containing 360 liters of water each. Mesocosms were made of polyethylene bags and PVC pipes, and were attached to the lake bottom. Treatment dilutions followed Carlson's trophic state index modified by Toledo and collaborators, constituting the oligotrophic, mesotrophic, and eutrophic treatments. Ten abiotic and 9 biological samplings were carried out simultaneously. Trophic states previously calculated for the treatments were kept unaltered during the entire experiment period, except for the mesotrophic mesocosms in which TP reached oligotrophic concentrations on the 31st day of the experiment. In all three treatments a reduction of DO was observed during the study period. At the same time, NH4+ and free CO2 rose, indicating decomposition within the enclosures. Nutrient impoverishment caused P limitation in all three treatments during most of the experiment period. Reduction of algal density, chlorophyll a, and phaeophytin was observed in all treatments. Competition for nutrients led to changes in phytoplankton composition. Once isolated and diluted, the mesocosms' trophic state did not change. This led to the conclusion that isolation of the allochthonous sources of nutrients is the first step for the recovery of the Garças Pond.

Distilia é um tipo de sistema reprodutivo que utiliza características morfológicas e fisiológicas como forma de potencializar a xenogamia. Dentre as 25 famílias de Angiospermas que apresentam esse sistema, Rubiaceae é a que tem a maior riqueza. Este estudo teve como objetivo investigar os aspectos relacionados ao sistema distílico de quatro espécies de Palicourea distribuídas no Cerrado do Brasil Central: Palicourea marcgravii A. St.-Hil., P. officinalis Mart., P. coriacea (Cham.) K. Schum. e P. macrobotrys (Ruiz & Pav.) Roem. & Schult. As áreas de coleta no Distrito Federal foram: Parque Nacional de Brasília, Estação Ecológica de Águas Emendadas e Reseva Ecológica do IBGE. As espécies Palicourea marcgravii e P. officinalis apresentaram-se como distílicas em função da presença dos dois morfos florais, da sua razão equilibrada e do sistema de incompatibilidade auto e intramorfo. As espécies P. coriacea e P. macrobotrys apresentaram-se como variantes pela ocorrência de indivíduos com morfo homostilo, além da distilia na primeira espécie e pelo morfo homostilo ser dominante na segunda espécie. Não se sabe ainda as forças evolutivas que determinam essas variações, contudo acredita-se que as características genéticas intrínsecas de cada táxon têm uma considerável influência e que cada espécie pode responder de forma diferente, ou até mesmo não responder, perante as mesmas pressões seletivas a que estão submetidas.

O presente trabalho teve como objetivo utilizar marcadores RAPD para conhecer a variabilidade genética de populações de Tibouchina papyrus (Pohl) Toledo, provenientes da região de Serra Dourada e Serra dos Pirineus, no Estado de Goiás. Os seis iniciadores RAPD produziram um total de 147 locos, variando entre 23 e 26 por iniciador. A avaliação hierárquica da estruturação da variabilidade genética, realizada pela a AMOVA, considerando a existência de duas regiões (Serra dos Pirineus e Serra Dourada) apresentou uma estimativa de F ST = 0,3439. O valor do componente entre regiões (F CT) foi igual a 18,96% e a variação entre populações dentro de regiões igual a 15,43%. As estimativas de fluxo gênico sugerem a existência de uma baixa proporção de migrantes entre populações. As análises multivariadas (UPGMA e NMDS) indicam que existe uma relação entre distância genética e espaço geográfico, hipótese esta que foi confirmada por uma análise de padrão espacial utilizando o teste de Mantel (r = 0,71; P = 0,015 com 1000 permutações aleatórias). Os resultados indicam assim que esta estrutura tenha se originado seguindo um modelo de diferenciação estocástica (neutro), ou seja, por um balanço entre fluxo gênico a curtas distâncias e deriva genética nas populações. Os valores de diversidade genética obtidos apóiam a hipótese de que a espécie T. papyrus é uma espécie xenógama facultativa e o fato de não serem encontrados altos níveis de homozigose, indica que devem existir mecanismos relacionados à biologia reprodutiva da espécie, que previnem, de alguma maneira, a ocorrência de elevadas taxas de endogamia, que poderia ter um efeito deletério em médio e longo prazo.

We examined plant population structure and interspecific associations for juveniles and adults of four woody species (Andira legalis (Vell.) Toledo, Clusia hilariana Schltdl., Protium icicariba (DC.) Marchand and Vernonia crotonoides Sch. Bip. ex Baker) in a patchy vegetation on a sandy coastal plain (restinga) in SE - Brazil. We found 101 vegetation patches in a 0.5 ha grid and these were divided into two distinct size classes, with large patches (> 20 m²) containing the majority of adult individuals of the species studied. The most abundant species, P. icicariba (465 individuals) and C. hilariana (312), had actively regenerating populations, whereas A. legalis (20) and V. crotonoides (338) showed evidence of intermittent regeneration. The regeneration niches of the four species differed as did their investment in vegetative reproduction: for instance, 81% of C. hilariana seedlings were found growing inside tank-bromeliads contrasting with only 3% of P. icicariba in this habitat. Additionally, 28% of regenerants of C. hilariana originated vegetatively, contrasting with only 6% for P. icicariba. All significant associations between species found in the study were positive. There was a positive association between adults of C. hilariana and P. icicariba, as well as between adults of C. hilariana and juveniles of both. This suggests that P. icicariba is successfully establishing under the canopy of C. hilariana and highlights the role of C. hilariana in generating vegetation cover that will be later dominated by other woody plant species, as an important process for maintenance of plant diversity in this restinga vegetation.

Measurement of telomerase activity in clinically obtained tumor samples may provide important information for use as both a diagnostic marker and a prognostic indicator for patient outcome. In order to evaluate telomerase activity in tumor tissue without radiolabeling the product, we developed a simple telomeric repeat amplification protocol-silver-staining assay that is less time-consuming, is safe and requires minimal equipment. In addition, we determined the sensitivity of the silver-staining method by using extracts of telomerase-positive thyroid carcinoma cell lines which were serially diluted from 5,000 to 10 cells. Telomerase activity was also assayed in 19 thyroid tumors, 2 normal controls and 27 bone marrow aspirates. The results indicate that the technique permits the detection of telomerase activity from 5000 to as few as 10 cells. We propose that it could be immediately applicable in many laboratories due to the minimal amount of equipment required.

Cancer cachexia induces host protein wastage but the mechanisms are poorly understood. Branched-chain amino acids play a regulatory role in the modulation of both protein synthesis and degradation in host tissues. Leucine, an important amino acid in skeletal muscle, is higher oxidized in tumor-bearing animals. A leucine-supplemented diet was used to analyze the effects of Walker 256 tumor growth on body composition in young weanling Wistar rats divided into two main dietary groups: normal diet (N, 18% protein) and leucine-rich diet (L, 15% protein plus 3% leucine), which were further subdivided into control (N or L) or tumor-bearing (W or LW) subgroups. After 12 days, the animals were sacrificed and their carcass analyzed. The tumor-bearing groups showed a decrease in body weight and fat content. Lean carcass mass was lower in the W and LW groups (W = 19.9 ± 0.6, LW = 23.1 ± 1.0 g vs N = 29.4 ± 1.3, L = 28.1 ± 1.9 g, P < 0.05). Tumor weight was similar in both tumor-bearing groups fed either diet. Western blot analysis showed that myosin protein content in gastrocnemius muscle was reduced in tumor-bearing animals (W = 0.234 ± 0.033 vs LW = 0.598 ± 0.036, N = 0.623 ± 0.062, L = 0.697 ± 0.065 arbitrary intensity, P < 0.05). Despite accelerated tumor growth, LW animals exhibited a smaller reduction in lean carcass mass and muscle myosin maintenance, suggesting that excess leucine in the diet could counteract, at least in part, the high host protein wasting in weanling tumor-bearing rats.

The Edinger-Westphal nucleus (EWN) is a central preganglionic parasympathetic cell group that gives rise to cholinergic input to the ciliary ganglion, thereby regulating several neurovegetative ocular functions. Recently, the supposed presence of the neuropeptide urocortin (UCN) has been reported in EWN neurons in rodent brain. The purpose of the present study was to examine the distribution of UCN in avian brain and to investigate by immunohistochemical analysis the possible use of this substance as an EWN marker in a non-mammalian class of vertebrates. Brain tissue of pigeons was incubated with a specific antibody against UCN and the results showed labeling of many small neurons, forming a double wing in the dorsal mesodiencephalic transition area. Their size and shape, however, differed from those of EWN neurons, and they were preferentially located rostral to the EWN. Double-label experiments employing an antibody against the enzyme choline acetyltransferase (ChAT) showed that UCN is not localized to the cholinergic cells of the EWN and confirmed the rostral distributionof UCN never overlapping the ChAT+ EWN cells. Taken together, these results suggest that, at least in pigeons, the UCN+ population does not belong to the traditionally defined EWN.

Griscelli syndrome (GS) is a rare autosomal recessive disorder caused by mutation in the MYO5A (GS1, Elejalde), RAB27A (GS2) or MLPH (GS3) genes. Typical features of all three subtypes of this disease include pigmentary dilution of the hair and skin and silvery-gray hair. Whereas the GS3 phenotype is restricted to the pigmentation dysfunction, GS1 patients also show primary neurological impairment and GS2 patients have severe immunological deficiencies that lead to recurrent infections and hemophagocytic syndrome. We report here the diagnosis of GS2 in 3-year-old twin siblings, with silvery-gray hair, immunodeficiency, hepatosplenomegaly and secondary severe neurological symptoms that culminated in multiple organ failure and death. Light microscopy examination of the hair showed large, irregular clumps of pigments characteristic of GS. A homozygous nonsense mutation, C-T transition (c.550C>T), in the coding region of the RAB27A gene, which leads to a premature stop codon and prediction of a truncated protein (R184X), was found. In patient mononuclear cells, RAB27A mRNA levels were the same as in cells from the parents, but no protein was detected. In addition to the case report, we also present an updated summary on the exon/intron organization of the human RAB27A gene, a literature review of GS2 cases, and a complete list of the human mutations currently reported in this gene. Finally, we propose a flow chart to guide the early diagnosis of the GS subtypes and Chédiak-Higashi syndrome.

Multiple cell membrane alterations have been reported to be the cause of various forms of hypertension. The present study focuses on the lipid portion of the membranes, characterizing the microviscosity of membranes reconstituted with lipids extracted from the aorta and mesenteric arteries of spontaneously hypertensive (SHR) and normotensive control rat strains (WKY and NWR). Membrane-incorporated phospholipid spin labels were used to monitor the bilayer structure at different depths. The packing of lipids extracted from both aorta and mesenteric arteries of normotensive and hypertensive rats was similar. Lipid extract analysis showed similar phospholipid composition for all membranes. However, cholesterol content was lower in SHR arteries than in normotensive animal arteries. These findings contrast with the fact that the SHR aorta is hyporeactive while the SHR mesenteric artery is hyperreactive to vasopressor agents when compared to the vessels of normotensive animal strains. Hence, factors other than microviscosity of bulk lipids contribute to the vascular smooth muscle reactivity and hypertension of SHR. The excess cholesterol in the arteries of normotensive animal strains apparently is not dissolved in bulk lipids and is not directly related to vascular reactivity since it is present in both the aorta and mesenteric arteries. The lower cholesterol concentrations in SHR arteries may in fact result from metabolic differences due to the hypertensive state or to genes that co-segregate with those that determine hypertension during the process of strain selection.

Chronic obstructive pulmonary disease (COPD) is associated with inflammatory cell reactions, tissue destruction and lung remodeling. Many signaling pathways for these phenomena are still to be identified. We developed a mouse model of COPD to evaluate some pathophysiological mechanisms acting during the initial stage of the disease. Forty-seven 6- to 8-week-old female C57/BL6 mice (approximately 22 g) were exposed for 2 months to cigarette smoke and/or residual oil fly ash (ROFA), a concentrate of air pollution. We measured lung mechanics, airspace enlargement, airway wall thickness, epithelial cell profile, elastic and collagen fiber deposition, and by immunohistochemistry transforming growth factor-β1 (TGF-β1), macrophage elastase (MMP12), neutrophils and macrophages. We observed regional airspace enlargements near terminal bronchioles associated with the exposure to smoke or ROFA. There were also increases in airway resistance and thickening of airway walls in animals exposed to smoke. In the epithelium, we noted a decrease in the ciliated cell area of animals exposed to smoke and an increase in the total cell area associated with exposure to both smoke and ROFA. There was also an increase in the expression of TGF-β1 both in the airways and parenchyma of animals exposed to smoke. However, we could not detect inflammatory cell recruitment, increases in MMP12 or elastic and collagen fiber deposition. After 2 months of exposure to cigarette smoke and/or ROFA, mice developed regional airspace enlargements and airway epithelium remodeling, although no inflammation or increases in fiber deposition were detected. Some of these phenomena may have been mediated by TGF-β1.
