The reduction of hepatic microsomal transfer protein (MTP) activity results in fatty liver, worsening hepatic steatosis and fibrosis in chronic hepatitis C (CHC). The G allele of the MTP gene promoter, -493G/T, has been associated with lower transcriptional activity than the T allele. We investigated this association with metabolic and histological variables in patients with CHC. A total of 174 untreated patients with CHC were genotyped for MTP -493G/T by direct sequencing using PCR. All patients were negative for markers of Wilson’s disease, hemochromatosis and autoimmune diseases and had current and past daily alcohol intake lower than 100 g/week. The sample distribution was in Hardy-Weinberg equilibrium. Among subjects with genotype 1, 56.8% of the patients with fibrosis grade 3+4 presented at least one G allele versus 34.3% of the patients with fibrosis grade 1+2 (OR = 1.8; 95%CI = 1.3-2.3). Logistic regression analysis with steatosis as the dependent variable identified genotypes GG+GT as independent protective factors against steatosis (OR = 0.4, 95%CI = 0.2-0.8; P = 0.01). The results suggest that the presence of the G allele of MTP -493G/T associated with lower hepatic MTP expression protects against steatosis in our CHC patients.

Esophageal cancer (EC) is a common malignancy worldwide. The X-ray repair cross-complementing 1 gene (XRCC1) is one of the most important candidate genes for influencing susceptibility to EC. This study aimed to investigate the effect of XRCC1 genetic variants on susceptibility to EC. A total of 383 EC patients (males: 239, females: 144, mean age: 56.62) and 387 cancer-free controls (males: 251, females: 136, mean age: 58.23) were enrolled in this study. The c.910A>G genetic variant of theXRCC1 gene was determined by polymerase chain reaction-restriction fragment length polymorphism and DNA sequencing methods. The allele and genotype frequencies indicated statistical differences between EC patients and cancer-free controls. The c.910A>G genetic variant was statistically associated with increased susceptibility to EC [GGvs AA: odds ratio (OR)=1.79, 95% confidence interval (CI)=1.12-2.86, P=0.014; GG vs AG/AA: OR=1.76, 95%CI=1.13-2.75, P=0.013; G vs A: OR=1.25, 95%CI=1.01-1.55, P=0.041]. The allele G and genotype GG could contribute to the increased susceptibility to EC. Our findings suggest that the c.910A>G genetic variant is associated with susceptibility to EC in the Chinese Han population, and might be used as a molecular marker for detecting susceptibility to EC.

The objective of this study was to evaluate cardiorespiratory fitness and pulmonary function and the relationship with metabolic variables and C-reactive protein (CRP) plasma levels in individuals with diabetes mellitus (DM). Nineteen men with diabetes and 19 age- and gender-matched control subjects were studied. All individuals were given incremental cardiopulmonary exercise and pulmonary function tests. In the exercise test, maximal workload (158.3±22.3vs 135.1±25.2, P=0.005), peak heart rate (HRpeak: 149±12 vs 139±10, P=0.009), peak oxygen uptake (VO2peak: 24.2±3.2 vs18.9±2.8, P<0.001), and anaerobic threshold (VO2VT: 14.1±3.4 vs 12.2±2.2, P=0.04) were significantly lower in individuals with diabetes than in control subjects. Pulmonary function test parameters, blood pressure, lipid profile (triglycerides, HDL, LDL, and total cholesterol), and CRP plasma levels were not different in control subjects and individuals with DM. No correlations were observed between hemoglobin A1C (HbA1c), CRP and pulmonary function test and cardiopulmonary exercise test performance. In conclusion, the results demonstrate that nonsmoking individuals with DM have decreased cardiorespiratory fitness that is not correlated with resting pulmonary function parameters, HbA1c, and CRP plasma levels.

High levels of low-density lipoprotein cholesterol (LDL-C) enhance platelet activation, whereas high levels of high-density lipoprotein cholesterol (HDL-C) exert a cardioprotective effect. However, the effects on platelet activation of high levels of LDL-C combined with low levels of HDL-C (HLC) have not yet been reported. We aimed to evaluate the platelet activation marker of HLC patients and investigate the antiplatelet effect of atorvastatin on this population. Forty-eight patients with high levels of LDL-C were enrolled. Among these, 23 had HLC and the other 25 had high levels of LDL-C combined with normal levels of HDL-C (HNC). A total of 35 normocholesterolemic (NOMC) volunteers were included as controls. Whole blood flow cytometry and platelet aggregation measurements were performed on all participants to detect the following platelet activation markers: CD62p (P-selectin), PAC-1 (GPIIb/IIIa), and maximal platelet aggregation (MPAG). A daily dose of 20 mg atorvastatin was administered to patients with high levels of LDL-C, and the above assessments were obtained at baseline and after 1 and 2 months of treatment. The expression of platelets CD62p and PAC-1 was increased in HNC patients compared to NOMC volunteers (P<0.01 and P<0.05). Furthermore, the surface expression of platelets CD62p and PAC-1 was greater among HLC patients than among HNC patients (P<0.01 and P<0.05). Although the expression of CD62p and PAC-1 decreased significantly after atorvastatin treatment, it remained higher in the HLC group than in the HNC group (P<0.05 and P=0.116). The reduction of HDL-C further increased platelet activation in patients with high levels of LDL-C. Platelet activation remained higher among HLC patients regardless of atorvastatin treatment.

Biofilm formed by Staphylococcus aureus is considered an important virulence trait in the pathogenesis of infections associated with implantable medical devices. Gene expression analyses are important strategies for determining the mechanisms involved in production and regulation of biofilm. Obtaining intact RNA preparations is the first and most critical step for these studies. In this article, we describe an optimized protocol for obtaining total RNA from sessile cells of S. aureus using the RNeasy Mini Kit. This method essentially consists of a few steps, as follows: 1) addition of acetone-ethanol to sessile cells, 2) lysis with lysostaphin at 37°C/10 min, 3) vigorous mixing, 4) three cycles of freezing and thawing, and 5) purification of the lysate in the RNeasy column. This simple pre-kit procedure yields high-quality total RNA from planktonic and sessile cells of S. aureus.

This study aimed to determine whether psychological factors affect health-related quality of life (HRQL) and recovery of knee function in total knee replacement (TKR) patients. A total of 119 TKR patients (male: 38; female: 81) completed the Beck Anxiety Inventory (BAI), Beck Depression Inventory (BDI), State Trait Anxiety Inventory (STAI), Eysenck Personality Questionnaire-revised (EPQR-S), Knee Society Score (KSS), and HRQL (SF-36). At 1 and 6 months after surgery, anxiety, depression, and KSS scores in TKR patients were significantly better compared with those preoperatively (P<0.05). SF-36 scores at the sixth month after surgery were significantly improved compared with preoperative scores (P<0.001). Preoperative Physical Component Summary Scale (PCS) and Mental Component Summary Scale (MCS) scores were negatively associated with extraversion (E score) (B=-0.986 and -0.967, respectively, both P<0.05). Postoperative PCS and State Anxiety Inventory (SAI) scores were negatively associated with neuroticism (N score; B=-0.137 and -0.991, respectively, both P<0.05). Postoperative MCS, SAI, Trait Anxiety Inventory (TAI), and BAI scores were also negatively associated with the N score (B=-0.367, -0.107, -0.281, and -0.851, respectively, all P<0.05). The KSS function score at the sixth month after surgery was negatively associated with TAI and N scores (B=-0.315 and -0.532, respectively, both P<0.05), but positively associated with the E score (B=0.215, P<0.05). The postoperative KSS joint score was positively associated with postoperative PCS (B=0.356, P<0.05). In conclusion, for TKR patients, the scores used for evaluating recovery of knee function and HRQL after 6 months are inversely associated with the presence of negative emotions.

A descafeinação do café canephora é vantajosa tanto para a obtenção da cafeína quanto para valorização desta espécie. A cafeína, subproduto da descafeinação, atua como estimulante e diurético no organismo e é usado pelas indústrias de bebidas de refrigerante e farmacêutica. Existem patentes industriais aplicando CO2 supercrítico na extração da cafeína do café. Porém pouco é divulgado sobre a extração da trigonelina (alcalóide importante na formação da vitamina‘ niacina’), que atua no sistema nervoso central, na secreção da bile e no intestino. O objetivo deste trabalho foi levantar subsídios experimentais e teóricos para a extração de alcalóides dos grãos de café canephora, variedade robusta, usando CO2 supercrítico. Os dados foram obtidos num aparelho de extração a altas pressões onde as variáveis termodinâmicas são independentemente controladas. Os alcalóides foram analisados por Cromatografia Liquida de Alta Eficiência (CLAE). As solubilidades da cafeína pura e a obtida dos grãos de café no CO2 supercrítico foram determinadas a 313, 323 e 343 K, de 9,5 a 23,5 MPa. Os resultados revelam a existência de um comportamento retrógrado para a solubilidade da cafeína pura e a obtida dos grãos de café; também mostram a seletividade do CO2 supercrítico pela cafeína quando comparada a trigonelina. A modelagem termodinâmica do equilíbrio sólido-fluido usando uma equação de estado de quarta ordem correlacionou satisfatoriamente as solubilidades da cafeína pura no solvente supercrítico.

Foram determinados os teores de mercúrio total em espécies de cação - anjo, cação-azul, cambeva, caçoa, machote e anequim - comercializadas em São Paulo - SP, Brasil. Os níveis encontrados variaram de 0,04 a 4,71mg Hg/kg, sendo que 54% estavam acima de 1mg Hg/kg, que é o limite aceito pela legislação brasileira especificamente para peixes predadores, para o propósito de consumo. Os resultados mostram que espécies de cação têm sido comercializadas em desacordo com a legislação, expondo os consumidores aos riscos tóxicos provocados pelo mercúrio. Foram feitas considerações em relação aos elevados níveis de mercúrio observados e o consumo correspondente, baseado na Ingestão Semanal Tolerável Provisional de 0,3mg de mercúrio total por pessoa, que no máximo não deverá ser superior a 0,2mg na forma de metilmercúrio, recomendada pela FAO/OMS. Considerações também foram feitas em relação à necessidade de inspeção e monitoramento das espécies brasileiras de cação usadas como alimento.

Avaliou-se os níveis de mercúrio total em amostras de peixes de água doce, procedentes de pesqueiros e pisciculturas de 39 regiões do estado de São Paulo. As espécies de peixes estudadas foram: pacu (Piaractus mesopotamicus), tilápia (Oreochromis niloticus), piauçu (Leoporinum sp), matrinxã (Brycon cephalus), tambaqui (Colossoma macropomum) e carpa (Cyprinus carpis). Foram avaliadas três metodologias de digestão de amostras para a determinação de mercúrio total, variando-se o volume de mistura sulfonítrica utilizada e a concentração do permanganato de potássio, sendo que o uso de 10mL de mistura sulfonítrica e solução de permanganato de potássio a 7% (m/v) na digestão da amostra foi a que forneceu as recuperações mais altas (96%), precisão de 1% e limite de quantificação de 0,3µg/kg. Após a validação do método, foram realizadas as determinações de teor de mercúrio total em 293 amostras de pescado, usando a técnica de espectrometria de emissão com fonte de plasma acoplado em gerador de hidretos. Os níveis médios de mercúrio encontrados variaram entre: tambaqui (0,0003-0,012mg/kg), carpa (0,063mg/kg), matrinxã (0,0003-0,074mg/kg), pacu (0,0003-0,078mg/kg), piauçu (0,0003-0,183mg/kg) e tilápia (0,0003-0,217mg/kg). Os resultados mostraram que nenhuma das amostras procedentes das diferentes regiões e diferentes pesque-pagues, apresentaram níveis de mercúrio total acima do permitido pela legislação brasileira, que é de 0,5mg/kg para pescado não-predador.

Este trabalho objetivou avaliar a estabilidade do suco tropical de acerola adoçado, elaborado pelos processos hot fill (garrafas de vidro) e asséptico (embalagens cartonadas), com relação às alterações químicas e físico-químicas (pH, sólidos solúveis totais, acidez total titulável, cor, açúcares redutores, não redutores e totais), sensoriais e microbiológicas, durante 350 dias de armazenamento em condições similares às de comercialização (28 °C ± 2 °C). Ao final do experimento, constatou-se que as amostras de suco de ambos os processos mantiveram uma adequada estabilidade microbiológica. O suco do processo hot fill teve maior aceitação global, enquanto o do processo asséptico manteve, ao final dos 350 dias, a aceitação inicial. As amostras do processo asséptico apresentaram inicialmente melhor sabor em comparação com as do processo hot fill, no entanto, as do processo hot fill mantiveram o sabor estável, enquanto o sabor do suco do processo asséptico teve menor aceitação ao longo do armazenamento. Ainda foram observadas, alterações químicas e físico-químicas nos sucos de ambos os processos. Em geral, o processo hot fill foi o mais eficiente em manter a estabilidade do suco.

A desordem nutricional mais comum no mundo é a deficiência de ferro. Sua disponibilidade é influenciada por outras substâncias. Os objetivos da pesquisa foram: avaliar a disponibilidade de ferro em alimentos fonte deste e de vitamina A, ovo (o), cenoura (c) e couve (cv), e verificar interação do ferro com alfa e beta-caroteno, e com outros interferentes. As misturas foram de ovo com cenoura e ovo com couve nas proporções (1:1), (1:1,5), (1:2), (1:2,5) e (1:3) e 3 alimentos sozinhos, totalizando 13 amostras. As amostras, após preparo, foram submetidas às análises de composição centesimal, ferro total, taninos, ácido fítico, ácido oxálico, alfa-caroteno e CE=Symbol>b-caroteno e diálise de ferro. Os dados foram analisados por teste de Tukey e correlação. Foi observada correlação positiva do ferro dialisável em relação à umidade e beta-caroteno e negativa para extrato etéreo, proteínas e cinza. A amostra 1,5 c apresentou teores mais elevados de taninos. As amostras 1 cv e 1,5 cv maiores teores de oxalato e ferro dialisável mais baixo. Pode ser concluído que misturas com maiores proporções de cenoura e couve aumentaram a disponibilidade do ferro e não houve interferência dos fatores antinutricionais. O beta-caroteno apresentou correlação positiva com ferro dialisável, exercendo influência na diminuição do efeito dos fatores antinutricionais.

The antioxidant capacity of beet is associated with non-nutritive constituents, such as phenolic compounds. The purpose of this research was to evaluate the effect of two different heat-processing techniques (drying and canned) on the antioxidant potential (ABTS) and phenolics content of beets. A forced air circulation dehydrator was used for the drying. Drying at high temperatures (100 + 90 °C/5.6 hours; 90 °C/6 hours) increased the antioxidant potential of the processed products while mild drying conditions decreased it (80 °C/6 hours; 100 + 70 °C/6 hours) or had no effect on it (70 °C/7 hours; 100 + 80 °C/6 hours). For the canned products, the antioxidant potential did not differ according to the pH (4.2 to 3.8) for any of the four acids tested. Some processing methods influenced the antioxidant potential of the processed products, and this was also dependent on changes in the total phenolics content.

The physiological state of a fruit is closely related to ripening and climatic conditions during the growing period when the fruit undergo changes in color, texture, and flavor. The ripening of the fruit can involve a complex series of biochemical reactions with alteration in enzymes activities, phenols, tannins, and ascorbic acid. The activity of enzymes (carboximethylcellulase, polygalacturonase, and pectinlyase), the total concentration of phenolic compounds, condensed tannins, and vitamin C in five stages of maturation were studied. Significant changes were observed between the maturity stages. The phenolic compounds were higher at green stage (705.01 ± 7.41); tannins were higher at green/purple stage (699.45 ± 0.22). The results showed that the ascorbic acid levels of the pulp varied significantly from 50.81 ± 1.43 to 6.61 ± 1.04 mg.100 g-1 during maturation. The specific activity of pectin lyase was higher at green stage (1531.90 ± 5.83). The specific activity of polygalacturonase was higher at mature stage (1.83 ± 0.0018). The specific activity of carboximetilcelulose was higher at ripe mature stage (4.61 ± 0.0024). The low ascorbic acid content found in jambolan fruit indicates that this fruit is not a rich source of this nutrient; however, other characteristics can make jambolan products fit for human consumption.

This study assessed the antioxidant, total phenolic, and physicochemical properties of in vitro Terminalia Catappa Linn (locally called castanhola) using the DPPH assay. The castanhola fruits had an average weight of 19.60 ± 0.00 g, combining shell, pulp, and seed weight, and a soluble solids content of 8 °Brix. The chemical composition was determined with predominance of carbohydrates (76,88 ± 0,58%).The titration method was used to determine Vitamin C content using 2,6-dichlorophenolindophenol (DCFI), known as reactive Tillmans resulting in no significant levels. Aqueous extracts of castanhola pulp showed a higher concentration of phenolics, 244.33 ± 18.86 GAE.g-1 of fruit, and alcoholic extracts, 142.84 ± 2.09 GAE.g-1 of fruit. EC50 values of the aqueous extract showed a greater ability to scavenge free radicals than the alcoholic extracts. The fruit had a significant content of phenolic compounds and high antioxidant capacity.

The importance of minimally processed commodities in the retail groceries of most developed countries has been rising continuously during the last decades. Cantaloupe melon is used more than any other fruit in fresh-cut processing. Ultraviolet (UV) light has been extensively used to simulate biological stres in plants and for determining resistance mechanisms of plant tissues. In this study the effect of ultraviolet irradiation on some properties of fresh-cut cantalope melon was determined during storage. Freshly cut cantalope melons cubes treated with ultraviolet irradiation at the doses of 1, 2 or 3 min before storage, and then placed in a cold room at 5±1°C temperature and 85-90% RH. Hue angle values of control group is low compared to UV-C treated samples, whereas L values of is high. EL of UV treated samples higher than those of control group. Total soluble solids of fresh-cut melon samples in UC3 treatment increased during storage. The results indicate that UV-C treatments on fresh-cut cantaloupe melon cubes increased total soluble solids independently from water loss.
