The data reported in this paper are in accordance with the hypothesis previously established (2) of the interaction of a main pair of alleles as controlling the inheritance of black, blue and blue-splashed plumage in chickens received from the State of Goias, Brazil. The black plumage is relatively uniform but the blue and blue splashed plumages indicate the action of other genes regulating the lack of uniformity.

Frutos de seis cultivares de manga, Oliveira-Neto, Bourbon, Brasil, Extrema, Haden e Imperial foram submetidos a tratamentos por imersão em água quente a 50°C por 30 minutos e a 55°C por 10 minutos visando-se verificar a aceitação destes tratamentos e consequente controle das podridões de antracnose, Colletotrichum gloeosporioides Penz. O experimento mostrou perfeita aceitação pelas cultivares aos níveis de temperatura e tempos de exposição, sendo que os dois tratamentos térmicos conferiram excelente controle das podridões de antracnose para os frutos das seis cultivares amadurecidos que foram à temperatura ambiente.

A chemical test previously described for the diagnosis of pregnancy was applied to the study of the excretion of gonadotropin in the urine during menstrual cycle. The chemical test is based on the selective adsorption by kaolim of the reducing substances biologically related to urinary gonadotropin. The active substance when acidified to pH 4.0 is adsorbed by the kolin and eluated with O.1N sodium hydroxide. The alkaline solution is treated by Somogyi's copper reagent and the excess not reduced is titrated by 0.005 N sodium thiosulfate. Gonadotropin is quantitatively addorbed by kaolin at pH 4.0 and eluated by alkaline solution as previously demonstrated by the A. (1). In the present paper the complete menstrual cycle was studied daily. It was observed that normally there are two distinct maxima of excretion. This study is based on 11 normal cycles (24-30 days) and 34 abnormal ones. Normal cycles showed a intramenstrual estrogens elimination from 200 to 260 mice units determinated by the Allen - Doisy full estrus smear test. The abnormal cycles belonging also to normal women showed much less estrogen excretion (14 to 25 mice units) Table II). In those cases with decreased estrogen excretion no fall in the curve after 14 th. day was observed. The A. suggest that the peaks of gonadotropin excretion is not related to the oculation but possibly due, the first one, to the follicle stimulating hormone and the second to the luteinizing hormone of hormone stimulating of the inerstitial tissue.

Twenty one cases of hepatoesplenic schistosomiasis patients without clinical and laboratory evidence of renal disease, were studied by surgical biopsies using light microscopy and immunofluorescence. The cases were classified histologically as: normal pattern (6 cases); minimal changes (6 cases); and mesangial proliferative glomerulonephritis (9 cases). By the immunofluorescence microscopy using anti IgM, IgG, IgA and C3, the predominant finding in all biopsies, except the normal cases, was granular deposits of IgM in the mesangium along with C3. On the other hand, IgG was present in all cases including normal biopsies along the capillary walls. However IgG was also present in the mesangium only in cases with glomerular lesions. This finding may well be similar to that recently described as IgM mesangial nephropathy. According to our cases a mesangial proliferative glomerulonephritis, characterized by segmental cell proliferation and deposition of IgM in the mesangium, is probably the entity found in the early stages of mansonic schistosomiasis.

Clinical and serological follow up examinations were performed on 203 persons, from three to twenty years of age, from the otolaryngology department of a hospital in the city of Rio de Janeiro, with no symptomatology suggesting toxoplasmosis, but suffering from chronic tonsillitis. According to results obtained during the first indirect immunofluorescence tests, the patients were divided into following groups: Group I (non-reactive IgG and IgM), 98 persons (48.3%); Group II (1:16 ≤ IgG ≤ 1:256 and non-reactive IgM), 74 persons (36.5%); Group III (IgM ≥ 1:1024 and non-reactive IgM), 18 persons (8.8%), and Group IV (IgG and IgM reactive), 13 persons (6.4%). One to two years later, 131 (64.5%) of the 203 persons were reexamined by a second indirect immunofluorescence test. In the case of 66 persons (Group I) whose serum was non-reactive in the IgG and IgM classes during the first indirect immunofluorescence test, serum conversion was observed in aproximately 21.2%. in 65 individuals (49.6%), (Groups II, III and IV),with reactive serum in the IgG classes during the first indirect immunofluorescence test, the second reaction showed an increase in titres in 20% of the cases, a decrease in 67.7% of the cases, or no alterations in 12.3 of the cases. In the IgM class, all 131 sera were non-reactive at 116 dilution the second immunofluorescence test, including the 13 cases that had previously been reactive in the immunoglobulin class, Symptomatology suggesting toxoplasmosis was only observed in one case during the second testing, this patient's principal physical sign being hypertrophied lymph nodes. during this period, the Toxoplasma antibodies showed titres of IgG 1:32000 and non-reactive IgM, whilst one year previously, during the first test, these titres were IgG 1:1024 and IgM 1:64. Differences in the age, sex and skin coloring of patients were not statistically significant as regards alterations in the indirect immunofluorescence test titres.

A reação imunoenzimática - ELISA foi empregada na sorologia da Wuchereriose, utilizando como antígeno extrato bruto de verme adulto de Setaria equina. Foram estudados soros de 139 indivíduos em três grupos: grupo 1 - de pacientes com diagnóstico parasitológico de Wuchereriose; grupo 2 - de pacientes com diversas patologias; grupo 3 - de pessoas clinicamente normais, de área endêmica e não endêmica. O antígeno utilizado mostrou alta comunidade antigênica coma W. bancrofti. As reações cruzadas, obtidas em particular com soros de pacientes com parasitoses intestinais (áscaris, ancilóstoma) recomendam a investigação de frações antigênicas de Setaria equina que possam fornecer testes de maior especificidade.

Estudou-se o efeito da dieta láctea, por um período de 150 dias em camundongos infectados com diferentes números das formas sangüíneas de Plasmodium berghei, e observou-se o desenvolvimento da imunidade humoral nestes animais pela dosagem das imunoglobulinas das classes IgG e IgM no soro, usando o teste de imunofluorescência indireta. Os resultados indicam que a administração do leite, como único alimento em camundongos, protege-os cotnra infecção malárica fatal, independentemente do número de parasitas inoculados. Os animais desenvolveram altos níveis de anticorpos IgG, os quais persistiram no soro por longo período de tempo. Contudo, os anticorpos IgM somente foram detectáveis no soro durante as primeiras duas semanas de infecção. O P. berghei continua presente na circulação periférica, após dois meses de infecção, uma vez que o sangue destes animasi inoculados em camundongos mantidos em dieta norma, produziu infecção fatal nos recipientes. No entanto, ao exame microscópico não foi possível detectar o parasita da malária no sangue periférico destes animais. O protozoário esteve presente no baço e fígado dos camundongos durante todo o tempo de duração da pesquisa. A presença contínua do P. berghei nestes animais, em nível de infecção subclínica, ofereceu ao hospedeiro o desenvolvimento de uma imunidade sólida contra subseqüente infecção. Esta imunidade adquirida esteve presente, nestes animais, até cinco meses após a infecção.

Conjugates of goat anti-HBs IgG and horseradish peroxidase (HRP) prepared by two different methods, one using NaIO4 and the other SPDP, were compared. Anti-HBs antibodies obtained from goat, rabbit and guinea-pig were tested as capture serum. The ELISA showed a sensitivity similar to RIA and a level of antigen captation ranging from 4.37 to 8.75 nanograms/ml was obtained when rabbit or guinea-pig captures were used combined with both NaIO4 or SPDP conjugates.

A virus antigenic characterization methodology using an indirect method of antibody detection ELISA with virus-infected cultured cells as antigen and a micro virus neutralisation test using EIA (NT-EIA) as an aid to reading were used for antigenic characterization of Jatobal (BeAn 423380). Jatobal virus was characterized as a Bunyaviridae, Bunyavirus genus, Simbu serogroup virus. ELISA using infected cultured cells as antigen is a sensitive and reliable method for identification of viruses and has many advantages over conventional antibody capture ELISA's and other tests: it eliminates solid phase coating with virus and laborious antigen preparation; it permits screening of large numbers of virus antisera faster and more easily than by CF, HAI, or plaque reduction NT. ELISA and NT using EIA as an aid to reading can be applicable to viruses which do not produce cytopathogenic effect. Both techniques are applicable to identification of viruses which grow in mosquito cells.

A dengue outbreak started in March, 1986 in Rio de Janeiro and spread very rapidly to other parts of the country. The great majority of cases presented classical dengue fever but there was one fatal case, confirmed by virus isolation. Dengue type 1 strains were isolated from patients and vectors (Aedes aegypti) in the area by cultivation in A. albopictus C6/36 cell line. The cytopathic effect (CPE) was studied by electron microscopy. An IgM capture test (MAC-ELISA) was applied with clear and reproducible results for diagnosis and evaluation of virus circulation; IgM antibodies appeared soon after start of clinical disease, and persisted for about 90 days in most patients. The test was type-specific in about 50% of the patients but high levels of heterologous response for type 3 were observed. An overall isolation rate of 46,8% (813 virus strains out of 1734 specimens) was recorded. The IgM test increased the number of confirmed cases to 58,2% (1479 out of 2451 suspected cases). The importance of laboratory diagnosis in all regions where the vectors are present is emphasized.

Se estudia el Ensayo Inmunoenzimático en Microgotas sobre Nitrocelulosa (Dot-ELISA)comparando dos preparados antigénicos de formas epimastigotas de cultivo de T. cruzi: 1) la fracción citoplasmática (antígeno citoplasmático y 2) el parásito total fijado previamente con formaldehido (antígeno integral). Se usaron sueros de: 95 pacientes chagásicos con serología convencional positiva, cardiopatía crónica y algunos con xenodiagnóstico positivo; 42 personas sanas y 32 con miocardipatía crónica con serología negativa y 74 pacientes con diferentes patologías incluyendo: sífilis, toxoplasmosis, lupus eritematoso diseminado, con factor reumatoide, leishmaniasis visceral, y leishmaniasis cutánea. Definidos los títulos diagnósticos (cut-off) de 1:512 con antígeno citoplasmático y de 1: 128 con antígeno integral, la especificidad fue 96% para el primero y de 100% para el segundo; mientras que la sensibilidad fue de 100% para ambas. En el estudio comparativo con las pruebas serológicas convencionales examinando 147 sueros tomados de personas referidas al laboratório, Dot-ELISA con antígeno citoplasmático presentó índices deco-positividad de 1,0, co-negatividad de 0,989 y eficiencia 0,993. Dot-ELIS con antígeno integral dió 1,0, 0,979 y 0,986 respectivamente. De acuerdo con esta evaluación, la técnica Dot-ELISA con antígeno integral se presenta como una alternativa práctica para el diagnóstico serológico de la enfermedad de Chagas.

American visceral leishmaniasis (AVL) is an important disease among children of northeast Brazil. In order to characterize antibody responses during AVL, sera of hospitalized patients were analyzed by ELISA and Western blot using a Leishmania chagasi antigen preparation. The ELISA was positive (asorbance [greater than or equals to] 0.196) at a serum dilution of 1:1024 in all patients at presentation, and fell to ward control levels over the following year. Only one of 72 control subjects tested positive, and that donor had a sibling with AVL. Immunoblots of the patients' sera recognized multiple bands, the most frequent of which were at approximately 116 kDa, 70 kDa, and 26 kDa. Less frenquently observed were bands at approximately 93 kDa, 74 kDa, 62 kDa, 46 kDa and 32 kDa. The ELISA responses and patterns of banding were distinctive for AVL, and could be used to differentiate patients with AVL from those with Chagas' disease of cutaneous leishmaniasis. Sera from six AVL patients followed for up to six weeks after treatment identified no new bands. Sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) of surface iodinated parasite proteins showed one major band and four minor bands, whereas SDS-PAGE of biotinylated prarasite proteins revealed a banding pattern similar to those of patient sera. AVL appears to produce characteristic immunoblot patterns which can be used along with a sensitive screening ELISA to diagnose AVL.

This paper presents the evaluation of an enzyme immunoassay in which Mayaro virus-infected cultured cells ara used as antigen (EIA-ICC) and an IgM antibody capture ELISA (MAC-ELISA) for Mayaro serologic diagnosis using 114 human sera obtained during a Mayaro outbreak occurred in Bolivia, in 1987. Results were compared with those obtained by haemagglutination-inhibition test (HAI). MAC-ELISA was the most sensitive technique for anti-Mayaro IgM detection. MAC-ELISA was twice sensitive as IgM EIA-ICC. The data shows that MAC-ELISA is a practical and valid technique for diagnosis of recent mayaro infection. IgG-ICC showed hight sensitivity and high specificity compared to HAI. The combination of anti-Mayaro IgG and IgM EIA-ICC results presented the highest sensitivity of the study. Anti-Mayaro IgG and IgM simultaneous detection by ELISA-ICC can be used for recent infection diagnosis (in spite of a less sensitive IgM detection than by MAC-ELISA), for surveillance and sero-epidemiologic studies, and for studies of IgG and IgM responses to Mayaro infection.

Se realizó estudio seroepidemiológico sobre la infección por Trypanosoma cruzi en habitantes de cuatro comunidades rurales de Venezuela, que presentan diferentes situaciones epidemiológicas en relación a la Enfermedad de Chagas, con la finalidad de evaluar la técnica de Dot-ELISA y compararla con las pruebas serológicas convencionales. En los caseríos de Kamana y caño Hondo del estado Zulia, donde no existe transmisión, la seropositividad fue 15,7% en adultos solamente. En las comunidades de las Rosas y Solano del estado Cojedes, área de alta endemicidad, la seropositividad en los menores de 14 años fue 8,9% y en los mayores de 15,51,6%. En el estudio comparativo con las pruebas serolgicas convencionlaes, Dot-ELISA presentó altos índices de co-positividad, co-negatividad y eficiencia. El valor predictivo positivo de la prueba fue de 66% y 60% con al antígeno citoplasmático e integral respectivamente, en el estado Zulia y 100% y 95% en el estado Cojedes. Estos resultados sugieren que Dot-ELISA puede ser una alternativa práctica para estudios seroepidemiológicos sobre la infección por Trypanosoma cruzi en los paises en desarrollo.
