OBJETIVO: comparar a efetividade de baixas doses de misoprostol vaginal (12,5 versus 25 mcg) para indução do trabalho de parto. MÉTODOS: ensaio clínico controlado, randomizado, duplo-cego, realizado entre maio de 2005 e abril de 2006. Foram incluídas 62 gestantes com gravidez a termo, membranas íntegras, que necessitaram de indução do parto. Foi administrado 25 mcg (32) ou 12,5 mcg de misoprostol (30), a cada quatro horas, até, no máximo, oito doses. Estudaram-se o tipo de parto, tempo entre o início da indução e o parto, complicações perinatais e efeitos maternos adversos. As variáveis de controle foram idades materna e gestacional, paridade e índice de Bishop. Os testes estatísticos utilizados foram cálculos de médias, desvios padrão e teste t de Student (variáveis numéricas contínuas), c² (variáveis categóricas) e Mann-Whitney (variáveis discretas). RESULTADOS: não houve diferença significante entre o Grupo de 12,5 e 25 mcg em relação ao intervalo de tempo entre a primeira dose e o parto (1.524 minutos versus 1.212 minutos, p=0,3), na freqüência de partos vaginais (70 versus 71,8%, p=0,7), no escore de Apgar inferior a sete ao quinto minuto (3,3 versus 6,25%, p=0,5) e na freqüência de taquissistolia (3,3 versus 6,2%, p=0,5). A média da dose total administrada de misoprostol foi significativamente maior no Grupo de 25 mcg (40 versus 61,2 mcg, p=0,03). CONCLUSÕES: misoprostol vaginal na dose de 12,5 mcg foi eficiente, com efeitos colaterais semelhante, à dose de 25 mcg de misoprostol vaginal, para indução do parto a termo.

OBJETIVO:Comparar as características clínicas e laboratoriais, os resultados maternos e perinatais de gestantes com pré-eclâmpsia versushipertensão gestacional.MÉTODOS:Análise retrospectiva dos prontuários médicos de pacientes com diagnóstico de pré-eclâmpsia e hipertensão gestacional, cujas gestações foram resolvidas em um período de cinco anos. Foram coletadas informações laboratoriais, resultados obstétricos e perinatais. As comparações entre os grupos foram realizadas com o uso do teste adequado para a variável analisada: testet não pareado, teste U de Mann-Whitney, ou teste do χ2. Consideramos p<0,05 como nível de significância estatística.RESULTADOS:Foram avaliadas 199 pacientes no grupo com hipertensão gestacional (HG) e 220 pacientes no grupo com pré-eclâmpsia (PE). No grupo HG o índice de massa corpórea médio foi 34,6 kg/m2 e no grupo PE, 32,7 kg/m2, com diferença significativa. O grupo PE apresentou valores de pressão arterial sistólica superiores ao grupo HG. Em relação aos exames laboratoriais, a média de valores denotou, de uma forma geral, maior gravidade no grupo PE. Pacientes submetidas à cesárea foram 59,1% dos casos no grupo PE e 47,5% no grupo HG. Em relação aos resultados perinatais, a idade gestacional e o peso ao nascer foram significativamente inferiores no grupo PE.CONCLUSÃO:As mulheres com hipertensão gestacional apresentam características epidemiológicas de pacientes com risco de doenças crônicas. As pacientes com pré-eclâmpsia apresentam parâmetros clínicos e laboratoriais de maior gravidade, taxas superiores de cesárea e piores resultados maternos e perinatais.

Avaliou-se a incidência de Vibrio spp. a partir de lesões superficiais em mamíferos marinhos encalhados ou capturados em redes de pesca nas regiões litorâneas do Sudeste (Rio de Janeiro) e Sul (RS) do Brasil. Foram coletadas 198 amostras, pelas instituições de pesquisa DEENSP, GEMARS e Ceclimar, as quais foram enviadas ao Labent/IOC/FIOCruz, onde foram submetidas ao enriquecimento em Água Peptonada Alcalina (APA) adicionada de 1% e 3% de NaCl e in-cubadas a 37ºC por 18-24 horas. Em seqüência foram semeadas em meio Agar Tiossulfato Citrato Bile Sacarose (TCBS) e as colônias suspeitas submetidas à caracterização bioquímica. Foram isoladas 108 cepas bacterianas, destacando-se Vibrio alginolyticus, V. parahaemolyticus, V. vulnificus e V. fluvialis como os principais patógenos isolados. Os resultados obtidos apontam para a necessidade de implementar atividades de vigilância e monitorização bacteriológica, particularmente de espécies selvagens, e reforçar os programas de proteção ambiental em casos de mamíferos marinhos ameaçados de extinção.

Mastitis has been recognized for some time as the most costly disease in dairy herds. From February to November 2004, 670 samples of bovine mastitic milk from which 231 Escherichia coli strains were isolated, were collected from two Brazilian states. The strains were screened for the presence of Shiga toxin-producing (stx 1 and stx 2) and intimin (eae) genes. Twenty (8.6%) strains were detected by PCR to harbor the Shiga toxin genes (8 the stx 1 gene, 12 the stx 2 gene and none both of them). Two (0.8%) of the Escherichia coli strains studied were eae positive non Shiga toxin-producing. The strains were also examined for resistance to 12 antimicrobial agents. The predominantly observed resistance was to tetracycline (92.2%), streptomycin (90.4%), nalidixic acid (88.3%), amikacin (86.5%) and cephalothin (84.8%). Multidrug resistance was found among 152 isolates (65.8%).

Pythiosis is caused by Pythium insidiosum and the occurrence of disease in horses was described in the North and Northwest State of Rio de Janeiro, Brazil. The disease was described in cattle, sheep, humans, and horses in different states and regions across the country. This paper describes the development of IgY and IgG polyclonal antibodies, in chicken and rabbits, respectively against proteins extracted from kunkers and hyphae of P. insidiosum from affected horses. The proteins were recognized by chicken, rabbit and horse antibodies by immunodiffusion and Western blot against majority bands of 27 and 43 KDa, and titrated by ELISA. The antibodies IgY developed by the first time against Brazilian strains of P. insidiosum may represent a valuable tool in the detection of antigens of the pathogen and contribute to further studies aimed at immunotherapy and knowledge about this disease in endemic areas in Rio de Janeiro and in Brazil.

This paper deals with the use of the conjugate gradient method of function estimation for the simultaneous identification of two unknown boundary heat fluxes in parallel plate channels. The fluid flow is assumed to be laminar and hydrodynamically developed. Temperature measurements taken inside the channel are used in the inverse analysis. The accuracy of the present solution approach is examined by using simulated measurements containing random errors, for strict cases involving functional forms with discontinuities and sharp-corners for the unknown functions. Three different types of inverse problems are addressed in the paper, involving the estimation of: (i) Spatially dependent heat fluxes; (ii) Time-dependent heat fluxes; and (iii) Time and spatially dependent heat fluxes.

For several years it was believed that angiotensin II (Ang II) alone mediated the effects of the renin-angiotensin system. However, it has been observed that other peptides of this system, such as angiotensin-(1-7) (Ang-(1-7)), present biological activity. The effect of Ang II and Ang-(1-7) on renal sodium excretion has been associated, at least in part, with modulation of proximal tubule sodium reabsorption. In the present review, we discuss the evidence for the involvement of Na+-ATPase, called the second sodium pump, as a target for the actions of these compounds in the regulation of proximal tubule sodium reabsorption.

The distribution, morphology and morphometry of microglial cells in the chick cerebral hemispheres from embryonic day 4 (E4) to the first neonatal day (P1) were studied by histochemical labeling with a tomato (Lycopersicon esculentum) lectin. The histochemical analysis revealed lectin-reactive cells in the nervous parenchyma on day E4. Between E4 (5.7 ± 1.35 mm length) and E17 (8.25 ± 1.2 mm length), the lectin-reactive cells were identified as ameboid microglia and observed starting from the subventricular layer, distributed throughout the mantle layer and in the proximity of the blood vessels. After day E13, the lectin-reactive cells exhibited elongated forms with small branched processes, and were considered primitive ramified microglia. Later, between E18 (5.85 ± 1.5 mm cell body length) and P1 (3.25 ± 0.6 mm cell body length), cells with more elongated branched processes were observed, constituting the ramified microglia. Our findings provide additional information on the migration and differentiation of microglial cells, whose ramified form is observed at the end of embryonic development. The present paper focused on the arrangement of microglial cells in developing cerebral hemispheres of embryonic and neonatal chicks, which are little studied in the literature. Details of morphology, morphometry and spatial distribution of microglial cells contributed to the understanding of bird and mammal central nervous system ontogeny. Furthermore, the identification and localization of microglial cells during the normal development could be used as a morphological guide for embryonic brain injury researches.

Limitations on tissue proliferation capacity determined by telomerase/apoptosis balance have been implicated in pathogenesis of idiopathic pulmonary fibrosis. In addition, collagen V shows promise as an inductor of apoptosis. We evaluated the quantitative relationship between the telomerase/apoptosis index, collagen V synthesis, and epithelial/fibroblast replication in mice exposed to butylated hydroxytoluene (BHT) at high oxygen concentration. Two groups of mice were analyzed: 20 mice received BHT, and 10 control mice received corn oil. Telomerase expression, apoptosis, collagen I, III, and V fibers, and hydroxyproline were evaluated by immunohistochemistry, in situ detection of apoptosis, electron microscopy, immunofluorescence, and histomorphometry. Electron microscopy confirmed the presence of increased alveolar epithelial cells type 1 (AEC1) in apoptosis. Immunostaining showed increased nuclear expression of telomerase in AEC type 2 (AEC2) between normal and chronic scarring areas of usual interstitial pneumonia (UIP). Control lungs and normal areas from UIP lungs showed weak green birefringence of type I and III collagens in the alveolar wall and type V collagen in the basement membrane of alveolar capillaries. The increase in collagen V was greater than collagens I and III in scarring areas of UIP. A significant direct association was found between collagen V and AEC2 apoptosis. We concluded that telomerase, collagen V fiber density, and apoptosis evaluation in experimental UIP offers the potential to control reepithelization of alveolar septa and fibroblast proliferation. Strategies aimed at preventing high rates of collagen V synthesis, or local responses to high rates of cell apoptosis, may have a significant impact in pulmonary fibrosis.

The antioxidant effects of Caryocar brasiliense Camb, commonly known as the pequi fruit, have not been evaluated to determine their protective effects against oxidative damage in lung carcinogenesis. In the present study, we evaluated the role of pequi fruit against urethane-induced DNA damage and oxidative stress in forty 8-12 week old male BALB/C mice. An in vivo comet assay was performed to assess DNA damage in lung tissues and changes in lipid peroxidation and redox cycle antioxidants were monitored for oxidative stress. Prior supplementation with pequi oil or its extract (15 µL, 60 days) significantly reduced urethane-induced oxidative stress. A protective effect against DNA damage was associated with the modulation of lipid peroxidation and low protein and gene expression of nitric oxide synthase. These findings suggest that the intake of pequi fruit might protect against in vivo genotoxicity and oxidative stress.

Avaliou-se a produção de aflatoxina B1 pelo Aspergillus flavus IMI 190443, inoculado em sementes de genótipos de amendoim: Tatu Vermelho, VRR-245, 2117 e 2155, in natura e autoclavado a 1210 C por 20 minutos, previamente cultivados no Centro Experimental do Instituto Agronômico de Campinas, em 1995/96. A aflatoxina B1 foi quantificada por cromatografia em camada delgada, utilizando comparação visual com padrões. Os níveis de aflatoxina B1 das amostras autoclavadas e in natura dos genótipos Tatu Vermelho, VRR-245 e 2155 foram significativamente diferentes (p <0,05, Teste de Duncan) quando comparados aos níveis do genótipo 2117. O genótipo 2117, originário da Índia, apresentou independente do processo com e sem aquecimento os menores níveis de aflatoxina B1 em relação aos dos outros genótipos. Os resultados obtidos mostram perspectivas de exploração da resistência varietal no controle da aflatoxina.

No presente estudo foi determinado o perfil sensorial e a aceitação de três marcas comerciais de chá light sabor pêssego, denominadas A, B e C, acondicionadas em embalagem PET e adquiridas no comércio local. O perfil sensorial foi determinado por Análise Descritiva Quantitativa (ADQ) utilizando-se uma equipe de 11 provadores rigorosamente selecionados e treinados. A aceitação dos produtos foi avaliada por 33 consumidores representativos do público alvo. Os resultados da ADQ foram submetidos à Análise de Variância (ANOVA), Teste de Média de Tukey e Análise de Componentes Principais. As amostras comerciais apresentaram perfis semelhantes em alguns termos descritores e diferenças significativas em outros. A amostra A caracterizou-se principalmente pelos atributos aroma e sabor de banana passa; a amostra B por aroma artificial de pêssego e refrescância; e a amostra C por aroma natural de pêssego e doçura. O Teste Afetivo, analisado por ANOVA e Teste de Média de Tukey, indicou maior aceitação para a amostra C. A cor das amostras foi avaliada pelo método Cielab L*a*b*, sendo que a amostra C se destacou das demais pela cor vermelha.

In view of the interest in analyzing volatile compounds by SPME, the following five microfibers were tested, polydimethylsiloxane; polyacrylate; polydimethylsiloxane/divinylbenzene; carboxen/polydimethylsiloxane, and carbowax/divinylbenzene, to select the one which presents the best performance for the adsorption of the volatile compounds present in the headspace of acid lime juice samples. Sample stabilization time variations (30 and 60 minutes) were assessed as well the addition of NaCl to the samples. It was verified that the chromatogram with the most adsorbed volatile compounds was obtained with PDMS/DVB microfiber at 30 minutes and the addition of 0.2 g NaCl.

The aim of this study was to evaluate the effects of pH, dextrose and yeast extract on the cadmium toxicity on Saccharomyces cerevisiae PE-2. In the first assay, the YED mediums with different pH (2, 3, 4, 5, 6, 7, and 8) containing 0.0 and 0.05 mmol Cd L-1 were inoculated with yeast suspension and incubated at 30 °C for 18 hours. During the anaerobic growth, the biomass concentration was determined. The yeast trehalose content, cell viability, and the growth rate were assessed at the beginning and at the end of the growth stages. In the second assay the YED mediums were diluted to the total, ½, and ¼ content of dextrose and yeast and 0.0 and 0.05 mmol Cd L-1 were added. The pH of the mediums was adjusted to 5. The culture mediums were inoculated and incubated at 30 °C for 18 hours. The yeast growth was not affected by cadmium at high pH, but at low pH the yeast becomes more sensitive to the toxic effect. The yeast susceptibility to cadmium was enhanced by the decrease of yeast extract strength and the increase of dextrose strength.
