O uso de extratos de plantas, em grande parte ainda inexplorada na Amazônia, constitui uma alternativa para o controle de insetos fitófagos, devido o baixo custo operacional, facilidade de preparação, utilização e segurança para o meio ambiente. O objetivo do presente trabalho foi avaliar a ação inseticida de Piper aduncum sobre Aetalion sp. Insetos adultos, coletados em Clitoria fairchildiana, foram separados em grupos de dez indivíduos, colocados em recipientes plásticos e expostos à aplicação tópica de extratos aquosos de folhas e raízes de P. aduncum. Os extratos foram aplicados nas concentrações de 10, 20 e 30 mg.ml-1. Os grupos de controle tratados com água destilada. Os testes tiveram 48 horas de duração e a cada 12 horas a porcentagem de mortalidade foi avaliada. O experimento se caracterizou num delineamento inteiramente ao acaso com três tratamentos em cinco repetições mais o grupo controle. O extrato de folhas de P. aduncum apresentou menor toxicidade (CL50 = 20,9 mg.ml-1) do que o extrato de raízes (CL50 = 20,2 mg.ml-1), mas não foram estatisticamente distintos. Tanto o extrato aquoso de raízes como o de folhas de P. aduncum apresentam atividade inseticida sobre adultos de Aetalion sp. Entretanto, por não causar impacto ambiental durante a coleta, recomenda-se o extrato de folhas em programas de controle alternativo desse inseto.

Piper L. (Piperaceae) é monofilético, com espécies de difícil distinção taxonômica, sendo necessários ferramentas que auxiliem a identificação interespecífica. Os objetivos do trabalho foram descrever os macropadrões e caracterizar in situ a composição química dos cristais caulinares e foliares de Piper arboreum Aubl. var. arboreum, P. callosum Ruiz & Pav. e P. tuberculatum Jacq., visando avaliar o uso dos cristais para a separação interespecífica. Para tanto, amostras foram submetidas à análise em microscopias de luz e eletrônica de varredura, microanálises espectroscópicas de raios-X por dispersão de energia e testes histoquímicos. Foram observados cristais prismáticos cuneiformes, tabulares, cúbicos e bipiramidais; ráfides; estiloides; drusas de cristais em bloco; rosetas cristalinas de estiloides e concrescências cristalinas. Foram descritos 15 macropadrões cristalinos reunidos em três categorias. Os cristais foram classificados quimicamente como: puros de oxalato de cálcio e misturas entre oxalatos e sulfatos e entre oxalatos, sulfatos e sílica. Rosetas cristalinas, concrescências cristalinas silicificadas e 14 dos 15 macropadrões cristalinos descritos são registros inéditos para inclusões biominerais de Piper e Piperaceae. Os macropadrões cristalinos caulinares e foliares observados são constitutivos e úteis à separação interespecífica das espécies estudadas. Ademais, os cristais desempenham funções relevantes para a autoecologia dos taxa de Piper analisados.

RESUMO Piper arboreum Aubl. var. arboreum e P. tuberculatum Jacq. (Piperaceae), conhecidas como pimenta-longa, são comercializadas como produtos tradicionais fitoterápicos e, frequentemente, identificadas erroneamente, não havendo relatos sobre controle de qualidade dessas espécies. O objetivo do trabalho foi preparar e caracterizar drogas vegetais foliares e caulinares dessas espécies para estabelecer-lhes parâmetros de autenticidade botânica e grau de pureza para o controle de qualidade. Foram utilizados métodos padrões para o preparo e análises estrutural e físico-química das drogas vegetais. São caracteres diagnósticos macroscópicos para distinguir as drogas vegetais íntegras: tamanho, forma, cor, sabor, textura, características da superfície de fratura e da secção transversal e os padrões de venação foliar. São caracteres diagnósticos para distinguir as drogas vegetais pulverizadas: padrões de cutícula e cera epicuticular, tipos de tricoma e espessamento parietal dos elementos traqueais e a forma do hilo dos grãos de amido. Emergências são marcadores morfológicos chave para as drogas vegetais de P. tuberculatum. Os valores dos parâmetros físico-químicos foram similares nas drogas vegetais das espécies, exceto o de cinzas ácido-insolúveis que foi maior nas drogas vegetais caulinares de P. arboreum var. arboreum. As maiores concentrações de metais ocorreram nas drogas vegetais de P. arboreum var. arboreum. As concentrações mais elevadas foram de Al, Cu, Mn e V e as mais baixas de Cd, Co e Hg. Os resultados obtidos poderão ser utilizados como parâmetros de controle de qualidade para as drogas vegetais de ambas as espécies.

Parametasphondylia piperis (Diptera, Cecidomyiidae, Asphondyliini, Schizomyiina), um novo gênero e espécie galhadora associada com Piper sp. (Piperaceae) é descrita e ilustrada (larva, pupa, macho e fêmea) com base em material obtido em Minas Gerais, Brasil.

Foi estudada a morfologia polínica da única espécie conhecida de CHLORANTHACEAE do Sul do Brasil e de quatro espécies de PIPERACEAE. As famílias são distintas palinologicamente, entretanto, as espécies desta última família são muito semelhantes quanto aos seus grãos de pólen, pertencendo todos ao mesmo tipo polínico.

The immunogenic and protective activity of an extract of S. mansoni, obtained by incubation of viable adult worms in buffered saline, was evaluated in rabbits and mice. Animal immunization with this extract resulted in the development of both humoral and cellular immune response. All immunized rabbits developed high levels (91 to 100%) of cytotoxic antibodies as determined by in vitro assays of cytotoxic activity of their sera against viable schistosomules. Immunized animals challenged with S. mansoni cercariae showed a lower parasite load than that of normal controls. Protective activity was 88.6% and 54.0% in immunized rabbits and mice, respectively.

Baccharus triptera Mart, is a widespread Compositae used in Brazilian folk medicine to treat gastrointestinal disturbances, rheumatic disease, mild fever, diabetes and as an anti-helminthic. Water extract of small branches of the plant (WE) administered to mice and rats (0.1 to 2 g/Kg, p.o) did not alter spontaneous motor activity, sleeping time induced by barbiturates or the tailflick response in mice. The extract decreased by 40 por cento the number of writhings induced by 0.8 por cento scetic acid, i.p., but did not influence paw edema induced by carrageenan or dextran in rats WE (2g/Kg, p.o.) decreased the intestinal transit of charcoal in mice by 20//. Gastric secretion in pylorus ligated rats was reduced after treatment with WE (1 and 2 g/Kg. i.p. or intraduodenal and the gastric pH was raised. The extract (1 g/Kg, p.o.) prevented gastric ulcers induced in rats by immobilization at 4ºC, but not those induced by indomethacin (10 mg/Kg, s.c.). The results indicate that WE may relieve gastrointestinal disorders by reducing acid secretion and gastrointestinal hiperactivity. Neither analgesic nor anti-inflammatory activities were detectable.

Coleus barbatus (Labiatae) Benth is popularly used in Brazil "for the healing of liver and stomach diseases". The water extract (WE 1 to 10 g/Kg, p.o.) of stem and leaves given to rats and mice did not induce signs of intoxication. Preveious treatment of mice with WE (1 g/kg, p.o.) shortened the sleeping time induced by pentobarbital (50 mg/Kg, i.p.) by 37 por cento, althoyugh the extract alone did not increase the spontaneous activity nor did it induce hyperexcitability. In mice WE (2 g/Kg, p.o.) increased the intestinal transit of charcoal by 30 por cento, while reduced gastric secretions ion rats treated with WE (2g/Kg intraduodenal) 3,9 ± 1.0 to 0.5 ± 0.2 ml/4h, respectively). The treatment also reduced the total acid secretion from 34.4 ± 11.0 to 2.7 ± 0.5 mEq/l and raisedgastric pH from 2.2 ± 0.3 to 6.5 ± 0.8. Treatment with WE (2g/Kg, p.o.) protected against gastric ulcers induced by stress (5.3 ± 1.6 and 1.5 ± 0.5 ulcers/cm²), but did nor protect against indonethacin induced ulcers. The results show that the water extract of C barbatus Benth produces mild stimulation of thecentral nervous system and increases intestinal movements. The extract also reduces gastric secretion indicating an antidyspeptic activity, and protects against gastric ulcers induced by stress.

The repellent effect of the molluscicides Niclosamide (Bayluscide WP 70 (R)), Anacardium occidentale and the latex of Euphorbia splendens on Biomphalaria glabrata was observed through the investigation of the occurrence of escape behavior among molluscs that were exposed to dosages lower than the LD 50. The total number of individuals out of water among the surviving snails in the control group provided a "Natural Escape Index". The comparison between this total and the total number of surviving snails in each group exposed to the different dosages of the molluscicides after 24 hr provided the "Molluscicide Escape Index" and the detection of a "Repellency Range" to these snails. The escape indexes for Niclosamide, A. occidentale and E. splendens were 10, 6.22 and 6.44 respectively. Repellency occurred at the following concentration ranges: 0.01, 0.02 and 0.03 ppm Bayluscide, 0.1, 0.2 and 0.3 ppm A. occidentale and 0.05, 0.10, 0.15 and 0.20 ppm E. splendens. The Natural Escape Index obtained in the control group was zero.

In previous studies it was shown that the recombinant molecule, r-Sm14, induces high levels of protection against Schistosoma mansoni infection in two outbred animal models and immune crossprotection against infection by Fasciola hepatica in Swiss outbred mice. r-Sm14 was derived from a living worm extract, called SE, and is being developed as the molecular basis of an anti-helminth bivalent vaccine against the two parasites, for medical and veterinary application. Present data refer to SDS-PAGE and Western Blotting analysis of four different preparations of S. mansoni adult worms focusing Sm14 identification. The extracts correspond to the initial fraction of the SE extraction process, containing products released by living worms (SEi); SE2, reextraction of adult worms in PBS; and SE of separated male and female adult worms. In all extracts it was possible to detect the component of 14 kDa, that was recognized by specific anti-rSm14 antibody raised in rabbits.

Aqueous suspension of ethanol extracts of Derris (Lonchocarpus) urucu (Leguminosae), collected in the state of Amazonas, Brazil, were tested for larvicidal activity against the mosquito Aedes aegypti (Diptera:Culicidae). The aim of this study was to observe the alterations of peritrophic matrix in Ae. aegypti larvae treated with an aqueous suspension of D. urucu extract. Different concentrations of D. urucu root extract were tested against fourth instar larvae. One hundred percent mortality was observed at 150 µg/ml (LC50 17.6 µg/ml) 24 h following treatment. In response to D. urucu feeding, larvae excreted a large amount of amorphous feces, while control larvae did not produce feces during the assay period. Ultrastructural studies showed that larvae fed with 150 µg/ml of D. urucu extract for 4 h have an imperfect peritrophic matrix and extensive damage of the midgut epithelium. Data indicate a protective role for the peritrophic matrix. The structural modification of the peritrophic matrix is intrinsically associated with larval mortality.

The present study was conducted to detected IgG antibodies using Strongyloides venezuelensis alkaline extract for the diagnosis of human strongyloidiasis by the enzyme-linked immunosorbent assay (ELISA). Sera from 90 subjects were analyzed (30 with strongyloidiasis, 30 with other parasites and 30 healthy individuals). Results were expressed in antibody titers, which were considered as positive when titer was > 80. Sensibility and specificity of the assay were 100% and 96.7%, respectively. It can be concluded that the heterologous alkaline extract could be employed in ELISA as a diagnostic aid in human strongyloidiasis, due to its advantages as easiness of obtaining, practicability in preparing, and high indexes of sensitivity and specificity.

Methanolic extracts of the leaves of Atlantia monophylla (Rutaceae) were evaluated for mosquitocidal activity against immature stages of three mosquito species, Culex quinquefasciatus, Anopheles stephensi, and Aedes aegypti in the laboratory.Larvae of Cx. quinquefasciatus and pupae of An. stephensi were found more susceptible, with LC50 values of 0.14 mg/l and 0.05 mg/l, respectively. Insect growth regulating activity of this extract was more pronounced against Ae. aegypti, with EI50 value 0.002 mg/l. The extract was found safe to aquatic mosquito predators Gambusia affinis, Poecilia reticulata, and Diplonychus indicus, with the respective LC50 values of 23.4, 21.3, and 5.7 mg/l. The results indicate that the mosquitocidal effects of the extract of this plant were comparable to neem extract and certain synthetic chemical larvicides like fenthion, methoprene, etc.

Infections due to protozoa of the genus Leishmania are a major worldwide health problem, with high endemicity in developing countries. The drugs of choice for the treatment of leishmaniasis are the pentavalent antimonials (SbV), which present renal and cardiac toxicity. Besides, the precise chemical structure and mechanism of action of these drugs are unknown up to date. In order to find new drugs against leishmaniasis, we have been studying extracts of Brazilian trees. In the present study, we have evaluated the effectiveness of an alkaloid extract of Aspidosperma ramiflorum Muell. Arg. (Apocynaceae), against the extracellular forms promastigotes of L. (L.) amazonensis and L. (V.) braziliensis. The alkaloid extract of A. ramiflorum was much more effective against L. (L.) amazonensis (LD50 < 47 µg/ml) than L. (V.) braziliensis. Based on these in vitro results against L. (L.) amazonensis new studies should be made to find the compounds with anti-leishmanial activity.

Searches for substances with antimicrobial activity are frequent, and medicinal plants have been considered interesting by some researchers since they are frequently used in popular medicine as remedies for many infectious diseases. The aim of this study was to verify the synergism between 13 antimicrobial drugs and 8 plant extracts - "guaco" (Mikania glomerata), guava (Psidium guajava), clove (Syzygium aromaticum), garlic (Allium sativum), lemongrass (Cymbopogon citratus), ginger (Zingiber officinale), "carqueja" (Baccharis trimera), and mint (Mentha piperita) - against Staphylococcus aureus strains, and for this purpose, the disk method was the antimicrobial susceptibility test performed. Petri dishes were prepared with or without dilution of plant extracts at sub-inhibitory concentrations in Mueller-Hinton Agar (MHA), and the inhibitory zones were recorded in millimeters. In vitro anti-Staphylococcus aureus activities of the extracts were confirmed, and synergism was verified for all the extracts; clove, guava, and lemongrass presented the highest synergism rate with antimicrobial drugs, while ginger and garlic showed limited synergistic capacity.
