Spermatogonial chromosomes of Pachylis laticornis and Pachylis pharaonis begin anaphasic movement with both ends turned toward the same pole, maintaining this form util they reach the poles. This is a proof that they are provided with one kinetochore at each end. Additional proof for a longitudinal division of each longitudinal half of the anaphase chromosomes of the primary sper- matocytes is presented against the idea of a previous end-toend pairing at metaphase. The longitudinal split of the chromosomes of the secondary spermatocytes which used to be considered as tertiary split is therefore a true secondary split. The heterochromosome in both species passes undivided to one pole in the first division of the spermatocyte. In Pachylis laticornis it appears connected with the poles by means of two fibrils detached from each extremity, what may be considered as indicating a rather premature longitudinal spliting. The behavior of the heterochromosome of Pachylis pharaonis is highly interesting and affords one of the most beautiful evidences in favour of the dicentricity of the chromosomes. Really, in metaphase the heterochromosome appears at the equator of the cell with a more or less round shape. In the beginning of anaphase it becomes fusiform. As anaphase proceeds it distends itself between the autosomal plates forming a long fusiform bridge or sends toward the plates a thick chromosomal thread. The bulky part of the heterochromosome as it passes to one side it reincorporates the substance of the thread in this side. The thread in the other side, which becomes generally thiner, is left with its kinetochore in the cell at this side. The heterochromosome therefore becomes terminally monocentric in the first division of the spermatocyte. Some figures, however, suggest that the heterochromossome from time to time may pass with both kinetochores to one of the cells, as ordinarily happens in the case of Pachylis laticornis. Summing up, other things apart the behavior of the heterochromosome in both species studied here puts out of doubt the question of the existence of two terminally located kinetochores.

We analysed species abundance and composition during one year of sampling at Itapuã State Park, a conservation unit in the State of Rio Grande do Sul, southern Brazil. Samples were collected monthly from May 2003 to April 2004 along a 4 km forest trail divided in three transects along the slopes of a small hill. Terrestrial isopods were hand searched by three people for 40 minutes at two sampling sites along each transect, summing 240 minutes of sampling effort per transect per month. Six species distributed in six families were found. The collector curve stabilised when half the samples were taken, demonstrating sampling sufficiency: analytical estimates did not predict more species to be found in the trail. Atlantoscia floridana (van Name, 1940) was dominant in all transects. Abundance varied along the year with a summer and a winter peak, but peaks were not consistent among transects. The first part of the trail, used for ecotourism, was less diverse, had less species (4 as compared to 6 in the other transects), and was thus also less similar in composition.

Toddia França, 1912 under the light microscope occurs as inclusion corpuscles in the cytoplasm of erythrocytes of cold-blooded vertebrates sometimes accompanied by crystalloid bodies. Its position among the protozoans or the viruses has been discussed by some authors, but remained unclear. To elucidate this problem we studied Toddia from a Brazilian frog (Leptodactylus ocellatus) by electron microscopy. In the cytoplasm of the infected cells we found no protozoan, but rather virus-like particles often hexagonal in outline, averaging 195 nm excluding their two involving membranes, and presenting a central area of variable electron density. Particles at different stages of development were generally found around or on area lighter density than the cytoplasm. which resembled a virus synthesis site. At high magnification, the nuclear or cytoplasmic crystals allied to Toddia resembled the crystalline lattice of the inclusion bodies associated with the polyhedrosis viruses and poxviruses from insects, of the capsules of granulosis viruses and of other protein crystals in ultrathin sections. Cytochemical tests in Toddia corpuscles displayed exclusively the presence of deoxyribonucleic acid. These findings indicate that Toddia is not a protozoan and demonstrate that it is in all probability a viral inclusion corpuscle. Taking into account the nucleic acid type found in its structure (DNA) and the hexagonal shape usually shown in ultrathin sections by its component particles, which have a cytoplasmic site of synthesis and assembly, we tentatively relate Toddia with the so-called "Icosahedral Cytoplasmic Deoxyriboviruses". We believe that the present paper gives the first report of virus-like particles in L. ocellatus.

Global left ventricular (LV) systolic dysfunction is the strongest predictor of morbidity and mortality in Chagas disease. Echocardiography is considered the gold standard for the detection of LV dysfunction, but not always available in endemic areas where chagasic cardiomyopathy is most common. Brain natriuretic peptide (BNP) is a neurohormone that has been recently described as a simple and inexpensive diagnostic and prognostic marker for patients with congestive heart failure. Chagasic patients (n = 63) and non-infected healthy individuals (n = 18) were recruited prospectively and underwent complete clinical examination, echocardiography and 24-h Holter monitoring. BNP was measured from thawed plasma samples using the Triage BNP test. We observed high levels of BNP in association with depression of LV ejection fraction, with increase of LV end-diastolic diameter and with LV premature complexes. An elevated concentration of BNP, defined as a concentration of 60 pg/ml or more, had a sensitivity of 91.7%, specificity of 82.8%, positive predictive value of 52.4%, and negative predictive value of 98% for detecting LV dysfunction (LV ejection fraction < 40%).BNP measurement using a simple, relatively inexpensive and rapid test has a promising role in identifying LV dysfunction associated with chagasic cardiomyopathy. Equally important, patients with Trypanosoma cruzi infection who have low levels of BNP level in plasma have a very low likelihood of severe cardiac involvement, and echocardiography is probably not necessary.

Colombian strain of Trypanosoma cruzi, biodeme Type III (T. cruzi I), has been cloned by micromanipulation at two phases of the acute infection: early (10 days ) and advanced (30 days). Twelve clones were obtained therefrom. Characterization by their biological and biochemical behavior showed an identity among the several clones and their parental strain, albeit with different degrees of virulence. Molecular characterization of the kinetoplast DNA (kDNA) after amplification by polymerase chain reaction revealed identical profiles of the bands from the kDNA minicircle by the analysis of restriction fragment lenght polymorphism for the isolated clones, their parental strain, and to the clones isolated at two different phases of the infection. Results suggest the predominance of a "principal clone", in the composition of the Colombian strain, responsible for the biological and biochemical behavior. However, no relationship was detected between the molecular profile of kDNA and the degree of virulence presented by the several clones.

It has been shown previously that the laticifer fluid of Calotropis procera (Ait.) R.Br. is highly toxic to the egg hatching and larval development of Aedes aegypti L. In the present study, the larvicidal potential of other laticifer fluids obtained from Cryptostegia grandiflora R.Br., Plumeria rubra L. and Euphorbia tirucalli L. was evaluated. We attempted to correlate larvicidal activity with the presence of endogenous proteolytic activity in the protein fraction of the fluids. After collection, the fluids were processed by centrifugation and dialysis to obtain the soluble laticifer protein (LP) fractions and eliminate water insoluble and low molecular mass molecules. LP did not visibly affect egg hatching at the doses assayed. LP from Cr. grandiflora exhibited the highest larval toxicity, while P. rubra was almost inactive. E. tirucalli was slightly active, but its activity could not be correlated to proteins since no protein was detected in the fluid. The larvicidal effects of LP from C. procera and Cr. grandiflora showed a significant relationship with the proteolytic activity of cysteine proteinases, which are present in both materials. A purified cysteine proteinase (papain) from the latex of Carica papaya (obtained from Sigma) was similarly effective, whereas trypsin and chymotrypsin (both serine proteinases) were ineffective. The results provide evidence for the involvement of cysteine proteinase activity in the larvicidal action of some laticifer fluids. C. procera is an invasive species found in areas infested with Ae. aegypti and thus could prove useful for combating mosquito proliferation. This is the first report to present evidence for the use of proteolytic enzymes as chemical agents to destroy Ae. aegypti larvae.

In this study, we describe the fate of fatty acids that are incorporated from the lumen by the posterior midgut epithelium of Rhodnius prolixus and the biosynthesis of lipids. We also demonstrate that neutral lipids (NL) are transferred to the haemolymphatic lipophorin (Lp) and that phospholipids remain in the tissue in which they are organised into perimicrovillar membranes (PMMs). 3H-palmitic acid added at the luminal side of isolated midguts of R. prolixus females was readily absorbed and was used to synthesise phospholipids (80%) and NL (20%). The highest incorporation of 3H-palmitic acid was on the first day after a blood meal. The amounts of diacylglycerol (DG) and triacylglycerol synthesised by the tissue decreased in the presence of Lp in the incubation medium. The metabolic fates of 3H-lipids synthesised by the posterior midgut were followed and it was observed that DG was the major lipid released to Lp particles. However, the majority of phospholipids were not transferred to Lp, but remained in the tissue. The phospholipids that were synthesised and accumulated in the posterior midgut were found to be associated with Rhodnius luminal contents as structural components of PMMs.

Lipophorin (Lp) is the main haemolymphatic lipoprotein in insects and transports lipids between different organs. In adult females, lipophorin delivers lipids to growing oocytes. In this study, the interaction of this lipoprotein with the ovaries of Rhodnius prolixus was characterised using an oocyte membrane preparation and purified radiolabelled Lp (125I-Lp). Lp-specific binding to the oocyte membrane reached equilibrium after 40-60 min and when 125I-Lp was incubated with increasing amounts of membrane protein, corresponding increases in Lp binding were observed. The specific binding of Lp to the membrane preparation was a saturable process, with a Kdof 7.1 ± 0.9 x 10-8M and a maximal binding capacity of 430 ± 40 ng 125I-Lp/µg of membrane protein. The binding was calcium independent and pH sensitive, reaching its maximum at pH 5.2-5.7. Suramin inhibited the binding interaction between Lp and the oocyte membranes, which was completely abolished at 0.5 mM suramin. The oocyte membrane preparation from R. prolixus also showed binding to Lp from Manduca sexta. When Lp was fluorescently labelled and injected into vitellogenic females, the level of Lp-oocyte binding was much higher in females that were fed whole blood than in those fed blood plasma.

The diagnosis of single-lesion paucibacillary leprosy remains a challenge. Reviews by expert dermatopathologists and quantitative polymerase chain reaction (qPCR) results obtained from 66 single-plaque biopsy samples were compared. Histological findings were graded as high (HP), medium (MP) or low (LP) probability of leprosy or other dermatopathy (OD). Mycobacterium leprae-specific genes were detected using qPCR. The biopsies of 47 out of 57 clinically diagnosed patients who received multidrug therapy were classified as HP/MP, eight of which were qPCR negative. In the LP/OD (n = 19), two out of eight untreated patients showed positive qPCR results. In the absence of typical histopathological features, qPCR may be utilised to aid in final patient diagnosis, thus reducing overtreatment and delay in diagnosis.

Ant species (Hymenoptera, Formicidae) from the seasonally dry tropical forest of northeastern Brazil: a compilation from field surveys in Bahia and literature records. The Caatingas occur predominantly in northeastern Brazil and comparatively it is the biome that received less attention than any other ecosystem in Brazil, representing the region where invertebrate groups are less known. We present here the first list of ant species of the Caatingas, compiling information from the literature, from a study of samples preserved in alcohol in the Laboratory of Entomology (Universidade Estadual de Feira de Santana), and from a field survey conducted in Milagres, Bahia, submitting standardized 1-m² samples of the leaf-litter to Winkler extractors. Summing all information, 11 subfamilies, 61 genera and 173 species (plus one subspecies) of ants are recognized in the biome. This species number does not consider morphospecies that could not be named due to the lack of reliable recent taxonomic information for some Neotropical ant genera. The list presented here for ant species of the Caatingas is therefore underestimated, but it is relevant because it allows the identification of areas to be sampled in order to improve our knowledge of the diversity of ants in this biome.

Por meio do ensaio de compressibilidade, foram estudados os efeitos do manejo e da umidade na pressão de preconsolidação (σp) de três solos: Latossolo Vermelho-Amarelo, Latossolo Roxo e Latossolo Vermelho-Escuro sob cultura anual, mata natural e pastagem, na região de Lavras (MG), em duas profundidades (0-0,03 e 0,27-0,30 m). Para cada condição, foram coletadas cinco amostras indeformadas e uma deformada, com três repetições. As amostras, indeformadas e com diferentes umidades, foram utilizadas no ensaio de compressão uniaxial, obtendo-se as curvas de compressão, das quais foram extraídas as respectivas pressões de preconsolidação. Com as amostras deformadas, determinaram-se os limites de plasticidade e de contração, textura e matéria orgânica. Os modelos de compactação testados foram baseados na pressão de preconsolidação e na umidade do solo. Para uma mesma condição e profundidade, houve diferença significativa entre os valores dos teores de argila e areia nos três solos. Os valores da densidade do solo inicial (Ds i) foram estatisticamente diferentes para todas as condições na mesma profundidade, exceto na camada de 0-0,03 m para a cultura anual. À medida que a umidade do solo aumentou, a pressão de preconsolidação decresceu exponencialmente, indicando uma redução na capacidade de suporte de carga do solo. O LR apresentou, em geral, maior capacidade de suporte de carga do que o LE e LV. Essa maior capacidade de suporte de carga pode estar associada com o seu maior teor de argila e menor teor de areia. A capacidade de suporte de carga na zona de friabilidade variou de 154 a 167 kPa, para o LV; de 77 a 183 kPa, para o LR, e de 77 a 132 kPa, para o LE. As umidades 0,33 a 0,30 kg kg-1, 0,42 a 0,27 kg kg-1, e 0,35 a 0,33 kg kg-1 correspondem à faixa de friabilidade (LP - LC) do LV, LR e LE, respectivamente. O modelo baseado na história de tensão do solo evidenciou o efeito da compactação causada pelas máquinas de preparo do solo na camada de 0,27-0,30 m, para a cultura anual, enquanto, para a pastagem, ocorreu o efeito do pisoteio do gado na camada de 0-0,03 m.

Maghemite (γFe2O3) from tuffite is exceptionally rich in Mg, relatively to most of those reportedly found in other mafic lithosystems. To investigate in detail the compositional and structural variabilities of this natural magnetic iron oxide, sets of crystals were isolated from samples collected at different positions in a tuffite weathering mantle. These sets of crystal were individually powdered and studied by X-ray diffractometry, Mössbauer spectroscopy, magnetization measurements and chemical analysis. Lattice parameter of the cubic cell (a0) was found to vary from 0.834(1) to 0.8412(1) nm. Lower a0-values are characteristic of maghemite whereas higher ones are related to a magnetite precursor. FeO content ranges up to 17 mass % and spontaneous magnetization ranges from 8 to 32 J T-1 kg-1. Zero-field room temperature Mössbauer spectra are rather complex, indicating that the hyperfine field distributions due to Fe3+ and mixed valence Fe3+/2+ overlap. The structural variabilities of the (Mg, Ti)-rich iron oxide spinels is essentially related to the range of chemical composition of its precursor (Mg, Ti)-rich magnetite, and probably to the extent to which it has been oxidized during transformation in soil.

O termo compactação do solo refere-se à compressão do solo não saturado durante a qual existe um aumento da densidade do solo em conseqüência da redução de seu volume, pela expulsão do ar causado pelo manejo inadequado. Com o objetivo de estudar o efeito de diferentes sistemas de manejo na densidade do solo máxima (Ds máx), e na umidade crítica de compactação (Ucrít), realizou-se um experimento em um Latossolo Roxo da Região de Lavras (MG), submetido aos seguintes sistemas de manejo: preparo com arado de discos (AD), preparo com escarificador (CM), semeadura direta (SD), preparo com arado de aivecas (AA) e preparo intensivo com grade (GD). O experimento foi instalado, em novembro de 1994, em blocos casualizados. Foram realizadas três amostragens em 1996: em janeiro, antes da semeadura do feijão; em abril, antes da colheita do feijão, e em novembro, antes da semeadura do milho. As amostras deformadas foram coletadas nas camadas de 0-0,07 e 0,20-0,27 m de profundidade nos sistemas de manejo AD, CM e AA. No sistema de manejo SD, as amostras deformadas foram coletadas nas camadas de 0-0,07 e 0,25-0,32 m de profundidade e, no caso do sistema GD, nas camadas de 0-0,07 e 0,15-0,22 m de profundidade. A partir das curvas de compactação, obteve-se a densidade do solo máxima. Determinaram-se, ainda, o teor de argila e matéria orgânica (MO), o limite de plasticidade (LP) e a água retida a - 0,033 e - 0,01 MPa. Os valores de (Ds máx), (Ucrít) para os diferentes sistemas de manejo, época de coleta e profundidades, não foram estatisticamente diferentes, não sendo, portanto, influenciados pelos teores de argila e M.O. Verificou-se que a Ucrít corresponde a 90% do LP e a 90% da água retida a - 0,01 MPa é aproximadamente igual à água retida a - 0,033 MPa.
