Objetivou-se com este trabalho desenvolver sachês incorporados com óleos essenciais avaliando as propriedades físico-químicas e microbiológicas de mangas armazenadas em saco de papel contendo estes sachês em seu interior. Os óleos essenciais de orégano (Origanum vulgaris) e capim-limão (Cymbopogon citratus) foram incorporados em saches, e sua atividade antimicrobiana foi testada nos fungos Colletotrichum gloeosporides, Lasiodiplodia theobromae, Xanthomonas campestris pv. mangiferae indica, Alternaria alternata. Frutos de manga 'Tommy Atkins' foram acondicionados individualmente em sacos de papel contendo em seu interior um sachê antimicrobiano e mantidos a 25 °C ± 2 °C e UR 80% ± 5% por nove dias. Os parâmetros cor da epiderme, firmeza de polpa, sólidos solúveis, acidez titulável e pH sofreram pouca influência dos óleos essenciais de orégano e capim limão, indicando que a presença do óleo essencial não altera as características físico-químicas da polpa de manga. Os sachês ativos incorporados com óleos essenciais de orégano e capim limão apresentaram controle no crescimento dos microrganismos testados, sendo o capim-limão mais eficiente, reduzindo em aproximadamente 2 ciclos Log a contagem de mesófilos aeróbios e fungos filamentosos e leveduras em relação ao tratamento controle empregado.

Objetivou-se avaliar dois métodos de inoculação de Colletotrichum gloeosporioides em maracujá, testar a patogenicidade de diferentes isolados, o efeito fungitóxico e a composição química dos óleos essenciais das espécies medicinais alecrim-pimenta (Lippia sidoides Cham.), capim-santo [Cymbopogon citratus (D. C.) Stapf.], alfavaca-cravo (Ocimum gratissimum L.), no controle da antracnose [Colletotrichum gloeosporioides (Penz.)], associado ao estádio de maturação de frutos de maracujazeiro-amarelo. Avaliaram-se três experimentos, onde se testou a patogenicidade de seis isolados do fungo em delineamento inteiramente casualizado, com seis repetições, outro com o mesmo delineamento em esquema fatorial 2x2 (suspensão de conídios e disco de micélio) e frutos (verdes e maduros), com seis repetições. No tratamento com frutos, utilizou-se o delineamento inteiramente casualizado, em esquema fatorial 5x3+1, sendo cinco concentrações (0; 2; 4; 6 e 8µL mL-1) e três espécies medicinais, mais o tebuconazol, com cinco repetições. Fez-se a caracterização química dos óleos por cromatografia gasosa, com espectrometria de massas. Todos os isolados foram patogênicos. Os frutos maduros apresentaram maior diâmetro das lesões, quando inoculados com suspensão de conídios. O óleo de C. citratus proporcionou o menor diâmetro das lesões nos frutos, até a concentração de 6 µL mL-1. Na concentração de 8 µL mL-1, todos os óleos inibiram o desenvolvimento do fungo. O timol (30,24%), o citral (77,74%) e o eugenol (92,89%) foram componentes majoritários em L. sidoides, C. citratus e O. gratissimum, respectivamente.

Dentre os patógenos veiculados pelas sementes e grãos do milho (Zea mays L.), Fusarium proliferatum causa tombamento e morte de plântulas, podridão de raízes, de colmo, de espiga e grãos. Assim, o tratamento de sementes mostra-se como importante medida para preservar a qualidade. O objetivo do presente trabalho foi avaliar a atividade antifúngica de diferentes concentrações dos extratos de alho (Allium sativum L.) e capim-santo (Cymbopogon citratus Stapf.) visando o controle de F. proliferatum em sementes de milho e observar os efeitos dos tratamentos sobre a germinação de sementes, sanidade e desenvolvimento das plântulas. A partir de grãos de milho, foi feito o isolamento de F. proliferatum, que foi cultivado em meio BDA. Foram avaliados os efeitos dos extratos de alho e capim santo sobre crescimento micelial, nas concentrações 0,5%, 1,0%, 2,5%, 5,0% e 10,0%, medindo-se os diâmetros das colônias do fungo durante oito dias. Esporos de F. proliferatum foram imersos em soluções dos extratos, nas concentrações mencionadas, e avaliados, quanto à germinação de conídios, às 6, 12, 18 e 24 horas de imersão. Sementes de milho foram tratadas em soluções dos extratos e inoculadas com F. proliferatum, sendo avaliada quanto à incidência do fungo, percentagem de germinação e incidência de tombamento e podridão do colmo das plântulas. Os extratos empregados reduziram a taxa de crescimento micelial e a germinação dos esporos, como também a incidência de F. proliferatum em grãos de milho. O extrato de alho, a partir da concentração 2,5%, mostrou maior eficiência em relação aos demais tratamentos. Os produtos vegetais aumentaram a germinação das sementes e também controlaram o tombamento e a podridão do colmo das plântulas de milho.

A tiririca (Cyperus rotundus) é uma espécie daninha de difícil manejo, causadora da redução do estande e do rendimento em plantios comerciais das mais variadas culturas. Devido à sua agressividade, capacidade de reprodução, alta dispersão e rusticidade, seu controle é difícil e oneroso. Objetivou-se com este trabalho avaliar métodos alternativos de controle da tiririca baseados na alelopatia e na homeopatia. No manejo com alelopatia, testaram-se extratos aquosos de feijão-de-porco (Canavalia ensiformes), mucuna-preta (Stizolobium aterrimum), alecrim-pimenta (Lippia sidoides) e capim-limão (Cymbopogon citratus). Com a homeopatia foi utilizada a escala centesimal hahnemanniana, onde se testaram as dinamizações 3CH, 6CH, 9CH e 12CH. Ambos os experimentos foram conduzidos em caixas gerbox transparentes, contendo areia grossa lavada, grãos de 1 a 3 mm de espessura, e 10 tubérculos sadios de tiririca, distribuídos uniformemente ao longo do recipiente. Após a aplicação dos tratamentos, os recipientes foram dispostos em estufa do tipo BOD a 25 ºC, com fotoperíodo de 12 horas, onde permaneceram por 15 dias até a avaliação final. O extrato que apresentou o melhor manejo da tiririca foi o de alecrim-pimenta, que diminuiu o percentual de emergência e o vigor das plântulas; o extrato dessa espécie ocasionou maior efeito na redução do comprimento das plântulas de tiririca do que 2,5 kg ha-1 i.a. atrazina - herbicida utilizado para comparação. A homeopatia não apresentou diferença entre as dinamizações, não tendo assim efeito satisfatório no controle da tiririca. Os extratos de capim-limão, mucuna-preta e feijão-de-porco não apresentaram efeitos alelopáticos. No entanto, o extrato de alecrim-pimenta é promissor no manejo alternativo de tiririca, sendo necessários novos estudos para elucidação dos princípios químicos envolvidos e da sua real ação no metabolismo da planta.

β-Citronellol is an alcoholic monoterpene found in essential oils such Cymbopogon citratus (a plant with antihypertensive properties). β-Citronellol can act against pathogenic microorganisms that affect airways and, in virtue of the popular use of β-citronellol-enriched essential oils in aromatherapy, we assessed its pharmacologic effects on the contractility of rat trachea. Contractions of isolated tracheal rings were recorded isometrically through a force transducer connected to a data-acquisition device. β-Citronellol relaxed sustained contractions induced by acetylcholine or high extracellular potassium, but half-maximal inhibitory concentrations (IC50) for K+-elicited stimuli were smaller than those for cholinergic contractions. It also inhibited contractions induced by electrical field stimulation or sodium orthovanadate with pharmacologic potency equivalent to that seen against acetylcholine-induced contractions. When contractions were evoked by selective recruitment of Ca2+ from the extracellular medium, β-citronellol preferentially inhibited contractions that involved voltage-operated (but not receptor-operated) pathways. β-Citronellol (but not verapamil) inhibited contractions induced by restoration of external Ca2+ levels after depleting internal Ca2+ stores with the concomitant presence of thapsigargin and recurrent challenge with acetylcholine. Treatment of tracheal rings with L-NAME, indomethacin or tetraethylammonium did not change the relaxing effects of β-citronellol. Inhibition of transient receptor potential vanilloid subtype 1 (TRPV1) or transient receptor potential ankyrin 1 (TRPA1) receptors with selective antagonists caused no change in the effects of β-citronellol. In conclusion, β-citronellol exerted inhibitory effects on rat tracheal rings, with predominant effects on contractions that recruit Ca2+ inflow towards the cytosol by voltage-gated pathways, whereas it appears less active against contractions elicited by receptor-operated Ca2+ channels.

Dermatophytosis is a common zoonosis in urban centers. Dogs and cats have played an important role as its disseminators. Environmental decontamination is essential for the prevention of its propagation to humans and animals. However, sanitizers or disinfectants with antifungal activity, currently available, have high toxicity. The present study evaluated the in vitro effects of an extract of citronella (Cymbopogon nardus) on 31 Microsporum canis isolates from animals and home environments. Susceptibility tests were performed based on document M38-A2 (2008) of the Clinical and Laboratory Standards Institute with modifications for natural products. Although susceptibility variation was observed between the fungus tested, the concentrations that inhibited the growth of 50 and 90% of the microorganisms were low (19.5 and 78 µg/mL, respectively). Thus, this citronella extract showed potent fungistatic and fungicide activities against M. canis isolated from animals and home environments. Therefore, it could be an alternative for dermatophytosis prophylaxis in the home environment.

Strongyloides ratti larval extract was used for the standardization of ELISA to detect genus-specific IgE in human strongyloidiasis. Forty serum samples from monoinfected patients shedding S. stercoralis larvae (Group I), 40 from patients with other intestinal parasites (Group II), and 40 from copronegative healthy subjects (Group III) were analyzed. Genus-specific IgE levels (ELISA Index: EI) were significantly higher in the group I (EI = 1.43) than groups II (EI = 0.70) and III (EI = 0.71), showing positivity rates of 55%, 2.5% and 0%, respectively. Similarly, sera from copropositive patients had significantly higher levels of total IgE (866 IU/mL) as compared to those from group II (302 IU/mL) and III (143 IU/mL). A significant positive correlation was found between levels of Strongyloides specific-IgE and total IgE in sera from patients with strongyloidiasis. In conclusion, S. ratti heterologous extract showed to be a useful tool for detecting genus-specific IgE by ELISA, contributing for a better characterization of the immune response profile in human strongyloidiasis.

Previous experiences have demonstrated the same ABO system and P system antigens in A. lumbricoides extracts and in their hosts. The aim was to show the behavior of an A. lumbricoides extract from an O Group patient against monoclonal antibodies of different specificities. Agglutination Inhibition Tests were carried out facing the extract against monoclonal antibodies (anti A 2.23; anti B 2.54; anti B 2.62; anti AB 2.39 and anti H 2.72) in optimal concentrations. Suspensions of O Group fresh red cells were used as revealing system. The extract only inhibited the agglutination of anti H 2.72 with O erythrocytes. The semiquantitative Agglutination Inhibition Test of the extract was made against two series of anti H 2.72 dilutions by using O Group fresh red cells as revealing system. A difference of five dilutions between the titers of both series has been observed and the presence of H Antigen in the extract has been significantly confirmed. The fact that the extract did not inhibit the agglutination against anti A, anti B and anti AB has corroborated our previous observations about absence of A and B epitopes in A. lumbricoides extracts from O Group patients. The results of the preceding studies and this experience have demonstrated the membrane glycoconjugated importance in A. lumbricoides. They could be involved in molecular mimicry for this parasite.

Cryptococcosis is a worldwide disease caused by the etiological agent Cryptococcus neoformans. It affects mainly immunocompromised humans. It is relatively rare in animals only affecting those that have received prolonged antibiotic therapy. The propolis is a resin that can present several biological properties, including antibacterial, antifungal and antiviral activities. The standard strain C. neoformans ATTC 90112 was used to the antifungal evaluation. The tests were realized with propolis ethanol extract (PEE) G12 in concentrations from 0.1 to 1.6 mg mL-1. The evaluation of MIC and MFC were done according to DUARTE (2002)5. The inhibitory effect of PEE G12 on the fungal growing was seen at the concentration of 0.2 mg mL-1 and 1.6 mg mL-1 was considered a fungicidal one.

The aim of this study was to assess the antioxidant and anti-schistosomal activities of the garlic extract (AGE) and Nigella sativa oil (NSO) on normal and Schistosoma mansoni-infected mice. AGE (125 mg kg-1, i.p.) and NSO (0.2 mg kg-1, i.p.) were administrated separately or in combination for successive 28 days, starting from the 1st day post infection (pi). All mice were sacrificed at weeks 7 pi. Hematological and biochemical parameters including liver and kidney functions were measured to assess the progress of anemia, and the possibility of the tissue damage. Serum total protein level, albumin, globulin and cholesterol were also determined. Malondialdehyde (MDA) and glutathione (GSH) levels were determined in the liver tissues as biomarkers for oxidative and reducing status, respectively. The possible effect of the treatment regimens on Schistosoma worms was evaluated by recording percentage of the recovered worms, tissue egg and oogram pattern. Result showed that, protection with AGE and NSO prevented most of the hematological and biochemical changes and markedly improved the antioxidant capacity of schistosomiasis mice compared to the infected-untreated ones. In addition, remarkable reduction in worms, tissue eggs and alteration in oogram pattern were recorded in all the treated groups. The antioxidant and antischistosomal action of AGE and NSO was greatly diverse according to treatment regimens. These data point to these compounds as promising agents to complement schistosomiasis specific treatment.

In this study, a methanolic extract from Echinaster (Othilia) echinophorus was evaluated for activity against Leishmania amazonensis. The extract showed activity against the promastigote and amastigote forms with IC50 values of 62.9 and 37.5 μg.mL-1 respectively. This extract showed a moderate toxicity on macrophages from BALB/c mice. A dose of 100 mg/kg/day was effective when administered during 15 days by intraperitoneal route to BALB/c mice infected experimentally.

The present work was carried out to evaluate the molluscicidal activity of active ingredient Taraxerol with the acetone extract of Euphorbia tirucalli against Lymnaea acuminata snail. The (fecundity, hatchability and survivability) of snail L. acuminata exposed to this extract was studied. The effects of the tested extracts on life-history traits of harmful snail L. acuminata have also been evaluated, and this study also expounds the inhibitory effects of these extracts singly as well as in binary combination (1:1 ratio). It concluded that these herbal products act as a potential source of molluscicides, and that they would also have the advantage of easy availability, low cost, biodegradability and greater acceptance amongst users than synthetic pesticide.

Candida albicans is an opportunistic human pathogen that is capable of causing superficial and systemic infections in immunocompromised patients. Extracts of Sapindus saponaria have been used as antimicrobial agents against various organisms. In the present study, we used a combination of two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) and matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF MS) to identify the changes in protein abundance of C. albicans after exposure to the minimal inhibitory concentration (MIC) and sub-minimal inhibitory concentration (sub-MIC) of the butanolic extract (BUTE) of S. saponaria and also to fluconazole. A total of six different proteins with greater than 1.5 fold induction or repression relative to the untreated control cells were identified among the three treatments. In general, proteins/enzymes involved with the glycolysis (GPM1, ENO1, FBA1), amino acid metabolism (ILV5, PDC11) and protein synthesis (ASC1) pathways were detected. In conclusion, our findings reveal antifungal-induced changes in protein abundance of C. albicans. By using the previously identified components of the BUTE of S. saponaria(e.g., saponins and sesquiterpene oligoglycosides), it will be possible to compare the behavior of compounds with unknown mechanisms of action, and this knowledge will help to focus the subsequent biochemical work aimed at defining the effects of these compounds.

Plant derived insecticides have considerable potential for mosquito control because these products are safer than conventional insecticides. This study aimed to investigate sublethal activities of Ipomoea carica or railway creeper crude acethonilic extract against life history trait of dengue vectors, Aedes albopictus and Aedes aegypti. The late third instar larvae of Ae. albopictus and Ae. aegypti were exposed to a sublethal dose at LC50 and larvae that survived were further cultured. Overall, Ipomea cairica crude extracts affected the whole life history of both Aedes species. The study demonstrated significantly lower egg production (fecundity) and eggs hatchability (fertility) in Ae. albopictus. The sublethal dose of crude extracts reduced significantly the width of larval head capsule and the wing length of both sexes in both Aedes species. The significance of sublethal effects of I. cairica against Aedes mosquitoes was an additional hallmark to demonstrate further activity of this plant despite its direct toxicity to the larvae. The reduced reproductive capacity as well as morphological and physiological anomalies are some of the effects that make I. cairica a potential candidate to be used as a new plant-based insecticide to control dengue vectors.
